In the present study, we investigate anti-diabetic effect of pectinase-processed ginseng radix (GINST) in high fat diet-fed ICR mice. The ICR mice were divided into three groups: regular diet group, high fat diet control group (HFD), and GINST-treated group. To induce hyperglycemia, mice were fed a high fat diet for 10 weeks, and mice were administered with 300 mg/kg of GINST once a day for 5 weeks. Oral glucose tolerance test revealed that GINST improved glucose tolerance after glucose challenge. Compared to the HFD control group, fasting blood glucose and insulin levels were decreased by 57.8% (p<0.05) and 30.9% (p<0.01) in GINST-treated group, respectively. With decreased plasma glucose and insulin levels, the insulin resistance index of the GINST-treated group was reduced by 68.1% (p<0.01) compared to the HFD control group. Pancreas of GINST-treated mice preserved a morphological integrity of islets and consequently having more insulin contents. In addition, GINST up-regulated the levels of phosphorylated AMP-activated protein kinase (AMPK) and its target molecule, glucose transporter 4 (GLUT4) protein expression in the skeletal muscle. Our results suggest that GINST ameliorates a hyperglycemia through activation of AMPK/GLUT4 signaling pathway, and has a therapeutic potential for type 2 diabetes.

Fatty acid synthase (FAS) is considered as a novel drug target for the development of anticancer and anti-obesity agents. Bioassay-guided fractionation of a n-hexane-soluble extract prepared from the roots of Salvia miltiorrhiza Bunge (Labiatae), using an in vitro enzyme assay, led to the isolation of five abietane diterpenoids: 15,16-dihydrotanshinone I (1), cryptotanshinone (2), tanshinone I (3), tanshinone IIA (4), and dansenspiroketallactone (5). Compounds 1-5 were tested for their in vitro FAS inhibitoryactivity and, except for compound 5 (IC(50) > 100 μM), compounds 1-4 inhibited the enzyme activity with IC(50) values ranging from 12.0 to 30.3 μM. Our findings may be partially related to the anticancer activity of abietane diterpenoids from the plant, suggesting a further study on the anticancer potential of tanshinone derivatives.

In this paper, a series of amphoteric polyacrylamide (PAM) with different molecular weight and charge were synthesized and used as retention and dewatering aids. Better products had been selected and tested under varied conditions. Effects of molecular weight, cationic charge density, degree of hydrolysis<br/> and other factors of PAM on retention and dewatering of papermaking were also considered. Finally, synthesized quaternary and tertiary ammonium amphoteric polyacrylamide were practised in paper mill trial.

This paper dealt with effect of dual component and micropartical retention system on papermaking. First-pass retention under dynamic drainage condition was studied in neutral and alkaline papermaking system. Cationic starches, amphoteric starches and amphoteric polyacrylamide were added prior to high shear force, then anionic microparticle (colloidal silica or bentonite clay) were added. The system is found to be very useful to improve filler retention. For mitigating unfavorable effect of interfering sub-stances [I] , anionic trash catcher (ATC)such as p-DADMAC was tested in this study.

Due to ionic migration and adsorption, an electrical double layer with a thickness scaled by the Debye length is formed at the hydrogel-solution interface. For a thin temperature-pH dual sensitive hydrogel whose chickness is comparable to the Debye length, its mechanical behavior may be affected by the electrical double layer. In this paper, by incorporating electromagnetic field theory into thermodynamic theory, we develop a coupled field theory for a temperature-pH dual sensitive hydrogel, and we take into account the electrical double layer. The proposed model can be used to analyze the in uences of the size effect, ionic concentration, ambient temperature and solution pH value on the swelling of a thin layer of the temperature-pH sensitive hydrogel.

A novel monoclonal antibody (mAb), known as AC10364, was identified from an antibody library generated by immunization of mice with human carcinoma cells. The mAb recognized proteins in lysates from multiple carcinoma cell lines. Cell cytotoxicity assays showed that AC10364 significantly inhibited cell growth and induced apoptosis in multiple carcinoma cell lines, including Bel/fu, KATO-III and A2780. Compared with mAb AC10364 or chemotherapeutic drugs alone, the combination of mAb AC10364 with chemotherapeutic drugs demonstrated enhanced growth inhibitory effects on carcinoma cells. These results suggest that mAb AC10364 is a promising candidate for cancer therapy.
Polymorphisms in the Thymidylate Synthase Gene and Risk of Colorectal Cancer
Gao, Chang-Ming,Ding, Jian-Hua,Li, Su-Ping,Liu, Yan-Ting,Cao, Hai-Xia,Wu, Jian-Zhong,Tajima, Kazuo Asian Pacific Journal of Cancer Prevention 2012 Asian Pacific journal of cancer prevention Vol.13 No.8
To evaluate the relationship between polymorphisms (28 bp repeated sequences in 5'-UTR and 6-bp ins/del in 3'-UTR) in then thymidylate synthetase gene (TS) and risk of colorectal, colon and rectal cancers, we conducted a case-control study with 315 cases of colorectal cancer and 439 population-based controls in Jiangsu province, China. TS genotypes were identified using PCR.RFLP (restriction fragment length polymorphism) methods. Odds ratios (ORs) were estimated with an unconditional logistic regression model. We found that the distributions of 5'-UTR genotypes in TS were significantly different between controls and male colon cases (${\chi}^2$=8.25, P = 0.016). Compared with 3R/3R genotype, individuals with the 2R allele were at an increased risk of colon cancer (age-, BMI-, smoking- and alcohol drinking-adjusted OR=1.98, 95%CI: 1.11-3.53) among men. In ccontrast, the 6-bp ins/del polymorphism at the TS 3'- UTR did not influence risk of the colorectal, colon and rectal cancers. When combined genotypes for both TS 5'-UTR and 3'-UTR polymorphisms were evaluated, individuals with the 5'-UTR 2R allele had a OR of 3.61 (95%CI: 1.38-9.49) for colon cancer among men with the 3'-UTR .6bp/-6bp genotype. These results show that the polymorphism of the 28 bp repeated sequences in TS 5'-UTR could influence susceptibility to colon cancer and that there was a coordinated effect between TS 3'-UTR and 5'-UTR polymorphisms in increasing risk of colon cancer among Chinese men.
