        Mapping Quantitative Trait Loci for Leaf and Heading-related Traits in Chinese Cabbage (Brassica rapa L. ssp. pekinesis)

        The inheritance of leaf and heading-related traits in Chinese cabbage (Brassica rapa ssp. pekinensis) are poorly understood since only a handful of studies of these traits are available till date. In this study, QTL mapping was done for seven leaf and heading related traits that includes gross weight, number of non-wrapper leaves, head weight, head length, head width,head length/head width ratio, and the number of head-forming leaves in 139 F3 families derived from a cross between two diverse Chinese cabbage lines, ‘501’ with small head and ‘601’ with large head. A total of 27 QTL spread over the nine linkage groups were identified for seven leaf and head-related traits, many of which showed major effects clustered in A2, A4,A6, and A8. The results provided a better understanding of the genetic factors controlling leaf and head-related traits in Chinese cabbage.

        Post-Acute Care as a Key Component in a Healthcare System for Older Adults

        Older adults often experience functional decline following acute medical care. This func-tional decline may lead to permanent disability, which will increase the burden on the medical and long-term care systems, families, and society as a whole. Post-acute care aims to promote the functional recovery of older adults, prevent unnecessary hospital readmission, and avoid premature admission to a long-term care facility. Research has shown that post-acute care is a cost-effective service model, with both the hospital-at-home and community hospital post-acute care models being highly effective. This paper describes the post-acute care models of the United States and the United Kingdom and uses the example of Taiwan’s highly effective post-acute care system to explain the bene-fits and importance of post-acute care. In the face of rapid demographic aging and smaller household size, a post-acute care system can lower medical costs and improve the health of older adults after hospitalization.

        Targeting neddylation pathway with MLN4924 (Pevonedistat) induces NOXA-dependent apoptosis in renal cell carcinoma

        Inhibition of protein neddylation pathway has emerged an attractive anticancer strategy in preclinical studies by using Nedd8-activating enzyme (NAE) inhibitor MLN4924 (Pevonedistat). Previous studies have reported the antitumor activity of MLN4924 mediated by its efficacy on apoptosis, autophagy and senescence. However, whether MLN4924 has any effect on renal carcinoma cells (RCC) remains unexplored. Here we reported that MLN4924 specifically inhibited protein neddylation pathway, leading to statistically significantly suppress the proliferation, survival and migration of RCC cells by inducing G<SUB>2</SUB> cell-cycle arrest, followed by apoptosis in a MLN4924 dose-dependent manner. Further mechanistic study revealed that MLN4924-induced apoptosis was mediated by substantial up-regulation of pro-apoptotic NOXA. These findings highlighted the anticancer effects of the neddylation inhibitors (e.g. MLN4924) for the treatment of RCC.

        Effect of different alfalfa cultivars on growth and development of the spotted alfalfa aphid, Therioaphis trifolii(Monell)

        Therioaphis trifolii (Monell) is a well-known pest in alfalfa cultivation in China. Individuals of T. t r i f o l i i were allowed to feed on 10 alfalfa cultivars and their growth, reproduction, and population life table parameters were estimated. Nymphs that fed on Hunter River had the longest development time, whereas nymphs that fed on Defu had the shortest development time. Adults that fed on Debao, Zhongmu No. 2, and Defu had the shortest development time, whereas adults that fed on Golden Empress and Hunter River had the longest development times. Individuals that fed on Zhungeer produced the greatest number of offspring (13.950), and those that fed on Hunter River produced the fewest offspring (6.000). The survival rate of T. t r i f o l i i was the highest on Algonquin and lowest on Hunter River, whereas the survival time was the longest on Golden Empress and shortest on Debao. The population that fed on Algonquin had the highest intrinsic growth rate (0.560) and shortest population doubling time (1.237 days), whereas those that fed on Hunter River had the lowest intrinsic growth rate (0.217) and longest population doubling time (3.194 days). The life table parameters of the experimental T. t r i f o l i i population were used to quantify the resistance strengths of the cultivars. Results indicate that the Hunter River cultivar may effectively resist damage from T. trifolii. This study provides a theoretical basis for breeding aphid-resistant alfalfa cultivars, enabling further research into the prevention and control of aphid infestation.

        Genetic diversity and population structure of the amylolytic yeast Saccharomycopsis fibuligera associated with Baijiu fermentation in China

        The amylolytic yeast Saccharomycopsis fibuligera is a predominant species in starters and the early fermentation stage of Chinese liquor (Baijiu). However, the genetic diversity of the species remains largely unknown. Here we sequenced the genomes of 97 S. fibuligera strains from different Chinese Baijiu companies. The genetic diversity and population structure of the strains were analyzed based on 1,133 orthologous genes and the whole genome single nucleotide polymorphisms (SNPs). Four main lineages were recognized. One lineage contains 60 Chinese strains which are exclusively homozygous with relatively small genome sizes (18.55–18.72 Mb) and low sequence diversity. The strains clustered in the other three lineages are heterozygous with larger genomes (21.85–23.72 Mb) and higher sequence diversity. The genomes of the homozygous strains showed nearly 100% coverage with the genome of the reference strain KPH12 and the sub-genome A of the hybrid strain KJJ81 at the above 98% sequence identity level. The genomes of the heterozygous strains showed nearly 80% coverage with both the sub-genome A and the whole genome of KJJ81, suggesting that the Chinese heterozygous strains are also hybrids with nearly 20% genomes from an unidentified source. Eighty-three genes were found to show significant copy number variation between different lineages. However, remarkable lineage specific variations in glucoamylase and α-amylase activities and growth profiles in different carbon sources and under different environmental conditions were not observed, though strains exhibiting relatively high glucoamylase activity were mainly found from the homozygous lineage.

        The Biocompatibility of Multi-Source Stem Cells and Gelatin-Carboxymethyl Chitosan-Sodium Alginate Hybrid Biomaterials

        BACKGROUND: Nowadays, biological tissue engineering is a growing field of research. Biocompatibility is a key indicator for measuring tissue engineering biomaterials, which is of great significance for the replacement and repair of damaged tissues. METHODS: In this study, using gelatin, carboxymethyl chitosan, and sodium alginate, a tissue engineering material scaffold that can carry cells was successfully prepared. The material was characterized by Fourier transforms infrared spectroscopy. In addition, the prepared scaffolds have physicochemical properties, such as swelling ratio, biodegradability. we observed the biocompatibility of the hydrogel to different adult stem cells (BMSCs and ADSCs) in vivo and in vitro. Adult stem cells were planted on gelatin-carboxymethyl chitosan-sodium alginate (Gel/SA/CMCS) hydrogels for 7 days in vitro, and the survival of stem cells in vitro was observed by live/died staining. Gel/SA/CMCS hydrogels loaded with stem cells were subcutaneously transplanted into nude mice for 14 days of in vivo culture observation. The survival of adult stem cells was observed by staining for stem cell surface markers (CD29, CD90) and Ki67. RESULTS: The scaffolds had a microporous structure with an appropriate pore size (about 80 lm). Live/died staining showed that adult stem cells could stably survive in Gel/SA/CMCS hydrogels for at least 7 days. After 14 days of culture in nude mice, Ki67 staining showed that the stem cells supported by Gel/SA/CMCS hydrogel still had high proliferation activity. CONCLUSION: Gel/SA/CMCSs hydrogel has a stable interpenetrating porous structure, suitable swelling performance and degradation rate, can promote and support the survival of adult stem cells in vivo and in vitro, and has good biocompatibility. Therefore, Gel/SA/CMCS hydrogel is a strong candidate for biological tissue engineering materials.

        Mechanical behavior of stud shear connectors embedded in HFRC

        Hybrid-fiber reinforced concrete (HFRC) may provide much higher tensile and flexural strengths, tensile ductility, and flexural toughness than normal concrete (NC). HFRC slab has outstanding advantages for use as a composite bridge potential deck slab owing to higher tensile strength, ductility and crack resistance. However, there is little information on shear connector associated with HFRC slabs. To investigate the mechanical behavior of the stud shear connectors embedded in HFRC slab, 14 push-out tests (five batches) in HFRC and NC were conducted. It was found that the stud shear connector embedded in HFRC had a better ductility, higher stiffness and a slightly larger shear bearing capacity than those in NC. The experimentally obtained ultimate resistances of the stud shear connectors were also compared against the equations provided by GB50017 2003, ACI 318-112011, AISC 2011, AASHTO LRFD 2010, PCI 2004, and EN 1994-1-1 (2004), and an empirical equation to predict the ultimate shear connector resistance considering the effect of the HFRC slabs was proposed and validated by the experimental data. Curve fitting was performed to find fitting parameters for all tested specimens and idealized load-slip models were obtained for the specimens with HFRC slabs.

        Hybrid medium model for conjugate heat transfer modeling in the core of sodium-cooled fast reactor

        Core-wide temperature distribution in sodium-cooled fast reactor plays a key role in its decay heat removal process, however the prediction for temperature distribution is quite complex due to the conjugate heat transfer between the assembly flow and the inter-wrapper flow. Hybrid medium model has been proposed for conjugate heat transfer modeling in the core. The core is modeled with a Realistic modeled inter-wrapper flow and hybrid medium modeled assembly flow. To validate present model, simulations for a three-assembly model were performed with Realistic modeling, traditional porous medium model and hybrid medium model, respectively. The influences of Uniform/Non-Uniform power distribution among assemblies and the Peclet number within the assembly flow have been considered. Compared to traditional porous medium model, present model shows a better agreement with in Realistic modeling prediction of the temperature distribution and the radial heat transfer between the inter-wrapper flow and the assembly flow.

        Lower ω-6/ω-3 Polyunsaturated Fatty Acid Ratios Decrease Fat Deposition by Inhibiting Fat Synthesis in Gosling

        The objective of the current study was to investigate the effects of dietary ${\omega}-6/{\omega}-3$ polyunsaturated fatty acid (PUFA) ratios on lipid metabolism in goslings. One hundred and sixty 21-day-old Yangzhou geese of similar weight were randomly divided into 4 groups. They were fed different PUFA-supplemented diets (the 4 diets had ${\omega}-6/{\omega}-3$ PUFA ratios of 12:1, 9:1, 6:1, or 3:1). The geese were slaughtered and samples of liver and muscle were collected at day 70. The activities and the gene expression of enzymes involved in lipid metabolism were measured. The results show that the activities of acetyl coenzyme A carboxylase (ACC), malic enzyme (ME), and fatty acid synthase (FAS) were lower (p<0.05), but the activities of hepatic lipase (HL) and lipoprotein lipase (LPL) were higher (p<0.05), in the liver and the muscle from the 3:1 and 6:1 groups compared with those in the 9:1 and 12:1 groups. Expression of the genes for FAS (p<0.01), ME (p<0.01) and ACC (p<0.05) were higher in the muscle of groups fed diets with higher ${\omega}-6/{\omega}-3$ PUFA ratios. Additionally, in situ hybridization tests showed that the expression intensities of the high density lipoprotein (HDL-R) gene in the 12:1 and 9:1 groups were significantly lower (p<0.01) than that of the 3:1 group in the muscle of goslings. In conclusion, diets containing lower ${\omega}-6/{\omega}-3$ PUFA ratios (3:1 or 6:1) could decrease fat deposition by inhibiting fat synthesis in goslings.

        Cardiac-targeted delivery of nuclear receptor RORα via ultrasound targeted microbubble destruction optimizes the benefits of regular dose of melatonin on sepsis-induced cardiomyopathy

        Background Large-dose melatonin treatment in animal experiments was hardly translated into humans, which may explain the dilemma that the protective effects against myocardial injury in animal have been challenged by clinical trials. Ultrasound-targeted microbubble destruction (UTMD) has been considered a promising drug and gene delivery system to the target tissue. We aim to investigate whether cardiac gene delivery of melatonin receptor mediated by UTMD technology optimizes the efficacy of clinically equivalent dose of melatonin in sepsis-induced cardiomyopathy. Methods Melatonin and cardiac melatonin receptors in patients and rat models with lipopolysaccharide (LPS)- or cecal ligation and puncture (CLP)-induced sepsis were assessed. Rats received UTMD-mediated cardiac delivery of RORα/cationic microbubbles (CMBs) at 1, 3 and 5 days before CLP surgery. Echocardiography, histopathology and oxylipin metabolomics were assessed at 16–20 h after inducing fatal sepsis. Results We observed that patients with sepsis have lower serum melatonin than healthy controls, which was observed in the blood and hearts of Sprague–Dawley rat models with LPS- or CLP-induced sepsis. Notably, a mild dose (2.5 mg/kg) of intravenous melatonin did not substantially improve septic cardiomyopathy. We found decreased nuclear receptors RORα, not melatonin receptors MT1/2, under lethal sepsis that may weaken the potential benefits of a mild dose of melatonin treatment. In vivo, repeated UTMD-mediated cardiac delivery of RORα/CMBs exhibited favorable biosafety, efficiency and specificity, significantly strengthening the effects of a safe dose of melatonin on heart dysfunction and myocardial injury in septic rats. The cardiac delivery of RORα by UTMD technology and melatonin treatment improved mitochondrial dysfunction and oxylipin profiles, although there was no significant influence on systemic inflammation. Conclusions These findings provide new insights to explain the suboptimal effect of melatonin use in clinic and potential solutions to overcome the challenges. UTMD technology may be a promisingly interdisciplinary pattern against sepsis-induced cardiomyopathy.
