        Objective : The use of herbal therapy is becoming an increasingly attractive approach for the treatment of various inflammatory disorders. The Alpiniae officinari Rhizoma is popular in Aisa as a traditional herbal medicine. Alpiniae officinari Rhizoma is a species of the ginger family(Zingiberacease). Method : This study was performed to investigate the anti-inflammatory effect of Alpiniae officinari Rhizoma extract by the methods of "carrageenan induced paw edema" and "Lipopolysaccharide-induced inflammatory mediators in mouse macrophage RAW 264.7 cells". Result : We suggest that Alpiniae officinari Rhizoma extract decreased paw volume induced by plantar injection of carrageenan. Also Alpiniae officinari Rhizoma extract inhibited nitric oxide, prostaglandin E₂production and induced nitric oxide synthase, cyclooxygenase-2 protein expression in Mouse macrophage RAW 264.7 cells stimulated with lipopolysaccharide. Conclusion : This study shows that Alpiniae officinari Rhizoma extract seems to have anti-inflammatory effect by inhibition of nitric oxide, prostaglandin E_(2) production and nitric oxide synthase, cyclooxygenase-2 protein expression.

        Primary Hyperoxaluria in Korean Pediatric Patients

        Background: Primary hyperoxaluria (PH), a rare inborn error of glyoxylate meta bolism causing overproduction of oxalate, is classified into three genetic subgroups: type 1-3 (PH1-PH3) caused by AGXT, GRHPR, and HOGA1 gene mutations, respectively. We performed a retrospective case series study of Korean pediatric patients with PH. Methods: In total, 11 unrelated pediatric patients were recruited and their phenotypes and genotypes were analyzed by a retrospective review of their medical records. Results: Mutational analyses revealed biallelic AGXT mutations (PH1) in nine patients and a single heterozygous GRHPR and HOGA1 mutation in one patient each. The c.33dupC was the most common AGXT mutation with an allelic frequency of 44%. The median age of onset was 3 months (range, 2 months-3 years), and eight patients with PH1 presented with end stage renal disease (ESRD). Patients with two truncating mutations showed an earlier age of onset and more frequent retinal involvement than patients with one truncating mutation. Among eight PH1 patients presenting with ESRD, five patients were treated with intensive dialysis followed by liver transplantation (n=5) with/without subsequent kidney transplantation (n=3). Conclusion: Most patients presented with severe infantile forms of PH. Patients with two truncating mutations displayed more severe phenotypes than those of patients with one truncating mutation. Sequential liver and kidney transplantation was adopted for PH1 patients presenting with ESRD. A larger nation-wide multicenter study is needed to confirm the genotype-phenotype correlations and outcomes of organ transplantation.

        Effects of Supercritical Carbon Dioxide Treatment on Meat Quality and Sensory Evaluation in Soy Sauce and Hot-pepper Paste Marinated Pork

        The objective of this study was to investigate the effects of supercritical carbon dioxide (SC-CO2) treatment on meat quality and sensory evaluation of marinated pork. Meat marinated in two traditional Korean marinades, soy sauce and hot-pepper paste, and raw marinated meat were then treated with 7.4, 12.2, or 15.2 MPa CO2 at 31.1oC for 10 min. The SC-CO2 treatments had no effect on the meat pH (p>0.05) or Warner-Bratzler shear force (p>0.05). There was no significant difference (p>0.05) in the total loss (sum of treatment loss and cooking loss) between the control and SC-CO2 treated samples at 15.2 MPa (soy sauce marinated pork: 21.78 vs. 18.97%; hot-pepper marinated pork: 21.61 vs. 18.01%). After the SC-CO2 treatment, lighter surface colors were observed in the treatment samples compared to those of the control samples (p< 0.001). However, tasting panelists were unable to distinguish a difference in color or in overall acceptability of the control and treatment (p>0.05). In the case of soy sauce marinated pork, when SC-CO2 applied at 15.2 MPa and 31.1oC for 10 min, treatment samples showed a tenderer meat than the control samples. Therefore, the SC-CO2 treatment conditions had no adverse effects on the sensory quality characteristics of the marinated meat products.

        Prospective validation of a novel dosing scheme for intravenous busulfan in adult patients undergoing hematopoietic stem cell transplantation

        The objective of this study was to externally validate a new dosing scheme for busulfan. Thirty-seven adult patients who received busulfan as conditioning therapy for hematopoietic stem cell transplantation (HCT) participated in this prospective study. Patients were randomized to receive intravenous busulfan, either as the conventional dosage (3.2 mg/kg daily) or according to the new dosing scheme based on their actual body weight (ABW) (23×ABW<sup>0.5</sup> mg daily) targeting an area under the concentration-time curve (AUC) of 5924 μM∙min. Pharmacokinetic profiles were collected using a limited sampling strategy by randomly selecting 2 time points at 3.5, 5, 6, 7 or 22 hours after starting busulfan administration. Using an established population pharmacokinetic model with NONMEM software, busulfan concentrations at the available blood sampling times were predicted from dosage history and demographic data. The predicted and measured concentrations were compared by a visual predictive check (VPC). Maximum a <i>posteriori</i> Bayesian estimators were estimated to calculate the predicted AUC (AUC<sub>PRED</sub>). The accuracy and precision of the AUC<sub>PRED</sub> values were assessed by calculating the mean prediction error (MPE) and root mean squared prediction error (RMSE), and compared with the target AUC of 5924 μM∙min. VPC showed that most data fell within the 95% prediction interval. MPE and RMSE of AUC<sub>PRED</sub> were -5.8% and 20.6%, respectively, in the conventional dosing group and –2.1% and 14.0%, respectively, in the new dosing scheme group. These findings demonstrated the validity of a new dosing scheme for daily intravenous busulfan used as conditioning therapy for HCT.

        UPLC-DAD-QTOF/MS를 이용한 국내 재배 블루베리(Vaccinium corymbosum)와 복분자(Rubus coreanus)의 플라보노이드 특성 비교

        UPLC-DAD-QTOF/MS를 이용하여 국내에서 재배한 7품종의 하이부시 블루베리와 4지역에서 재배된 복분자 열매에서 총 29종(블루베리 28종, 복분자 4종)의 개별 플라보노이드 성분을 정성 및 정량 분석하였다. 블루베리의 평균 플라보노이드 함량은 143.0 mg/100g DW로 관찰되었고, `Darrow`에서 가장 높았으며, `Nelson`이 가장 낮은 것으로 나타났다. 복분자의 평균 플라보노이드 함량은 95.4 mg/100g DW로 나타났으며, 총 플라보노이드 함량은 정읍 > 순창 > 고창 > 광양순으로 높았으나 정읍, 순창, 고창간에 유의적인 함량차이는 나타나지 않았다. 블루베리는 hyperoside와 isoqercitrin이 각각 총 플라보노이드 함량 중 평균 31.4%, 13.3%을 차지하였으며, 복분자는 rutin과 miquelianin이 각각 총 플라보노이드함량 중 평균 51.4%, 40.2%를 차지하여 두 베리류 열매간 주요 플라보노이드 성분의 특성 차이를 확인할 수 있었다. 블루베리 열매에서 quercetin 3-O-robinobioside, quercetin 3-O-(6``-O-malonyl)glucoside, isorhamnetin 3-O-robinobioside, avicularin, kaempferol 3-O-(6``-O-acetyl)glucoside, quercetin이 처음으로 동정되었다. 본 연구 결과는 국내산 블루베리와복분자를 이용한 기능성 식품 개발에 필요한 기초 자료로 활용될 수 있을 것으로 판단되며, 추후 연구에서는 보다 다양한 베리류에 대한 추가 분석이 필요할 것으로 생각된다. 또한 본 연구 방법은 베리류 뿐만 아니라 다양한 식물에 존재하는 플라보노이드의 정성 및 정량 분석이 가능하여 활용 가치가 높을 것으로 사료된다. BACKGROUND: The objective of this study was to identify and compare the main phenolic compounds (anthocyanins, flavonoids, phenolic acids) in blueberry and black raspberry cultivated in Korea using ultra-performance liquid chromatography -diode array detection -quadrupole time-of-flight mass spectrometry (UPLC-DAD- QTOF/MS). METHODS AND RESULTS: Twenty-nine flavonoids were identified by comparison of ultraviolet and mass spectra with data in a chemical library and published data. Blueberry contained flavonols including kaempferol, quercetin, isorhamnetin, myricetin, and syringetin aglycones. Isorhamnetin 3-O-robinobioside, kaempferol 3-O-(6``-O-acetyl) glucoside, quercetin, quercetin 3-O-arabinofuranoside (avicularin), quercetin 3-O-(6``-O-malonyl) glucoside, and quercetin 3-O-robinobiosidewere detected for the first time in blueberry. The flavonoids in raspberry consisted of quercetin aglycone and its glycosides. The mean total flavonoid content in blueberry [143.0mg/100 g dryweight (DW)]was 1.5-times that in raspberry (95.4mg/100 gDW). The most abundant flavonoid in blueberry was quercetin 3-O-galactoside (hyperoside, up to 76.1 mg/100 g DW) and that in raspberry was quercetin 3-O-glucuronide (miquelianin, up to 55.5 mg/100 g DW). Miquelianin was not detected in blueberry. CONCLUSION: Flavonol glycosides were the main flavonoids in blueberry and black raspberry cultivated in Korea. The composition and contents of flavonoids differed between blueberry and black raspberry, andmay be affected by the cultivar and cultivation conditions.

Main test items are the suitability of the system such as vibration of the rolling stock, noise, current collection, track and signal system. In this article, part of the vibration and noise related test results are introduced such as the vibration of the carbody and bogie, running stability, jerk, interior noise of the passenger car and driver"s cabin.
