The production of therapeutic protein from transgenic domestic animal is the major technology of biotechnology. Insulin-like growth factor-1 (IGF-1) is known to play an important role in the growth of the animal. The objective of this study is construction of knock-in vector that bovine IGF-1 gene is inserted into the exon 7 locus of ${\beta}$-casein gene and expressed using the gene regulatory DNA sequence of bovine ${\beta}$-casein gene. The knock-in vector consists of 5' arm region (1.02 kb), bIGF-1 cDNA, CMV-EGFP, and 3' arm region (1.81 kb). To express bIGF-1 gene as transgene, the F2A sequence was fused to the 5' terminal of bIGF-1 gene and inserted into exon 7 of the ${\beta}$-casein gene. As a result, the knock-in vector is confirmed that the amino acids are synthesized without termination from the ${\beta}$-casein exon 7 region to the bIGF-1 gene by DNA sequence. These knock-in vectors may help to create transgenic dairy cattle expressing bovine bIGF-1 protein in the mammary gland via the expression system of the bovine ${\beta}$-casein gene.
양성자 자기공명분광법을 이용한 우울증 동물모델에서의 항우울제 약물 효능 평가

본 기초연구에서는 강제수영검사(forced swimming test, FST)에 의해 우울증이 유발된 쥐의 해마(hippocampus) 부위에서 뇌 대사물질(brain metabolites)중에 choline 신호의 변화를 관찰하였다. 본 연구의 목적은 우울증이 유발된 쥐에 항우울제인 desipramine-HCl을 투여하여 정상 쥐와 약물을 투여한 쥐 사이의 뇌 대사물질의 차이를 자기공명분광법(magnetic resonance spectroscopy: MRS)을 통하여 알아보는 것이다. 실험대상으로는 6주령의 SD rat을 사용하였으며 우울증을 유발시키고 항우울제의 약물평가를 위한 모델링 방법으로 강제수영검사를 시행하였다. 자기공명분광법을 시행하기 위한 localization 방법으로는 PRESS 펄스시퀀스를 사용하였다(TR: 2,500 ms, TE: 144 ms, averaging: 512, complex data point: 2048, scan time: 25 min). 정상 쥐 그룹과 우울증을 유발시킨 뒤 항우울제를 투여한 쥐 그룹 모두 양쪽 해마부위에서의 NAA/Cr, Cho/Cr 비율(ratio)은 차이가 나지 않았다. 또한 왼쪽 해마에서 정상 쥐와 항우울제를 투여한 쥐 사이에서 NAA/Cr, Cho/Cr 비율도 차이가 나지 않았고, 오른쪽 해마부위에서도 통계적으로 유의한 차이가 없었다. 본 연구를 통하여 anti-immobility효과를 나타내는 항우울제를 투여함으로써 choline 신호가 정상 쥐와 비슷하게 회복되는 것을 관찰하였다. 이는 앞으로의 항우울제 약물 평가에 있어 자기공명분광법을 도입함으로써 기존의 주관적인 판단을 배제할 수 없었던 행동변화분석만으로 항우울제의 효과를 평가하는 방법에 비해 객관적이고 정량적인 분석을 할 수 있다는 점에서 아주 유용한 방법이라 사료된다. In this study, we observed the alteration of choline signal intensity in hippocampus region of the depressive rat model induced by forced swimming test (FST). The purpose of this study was to evaluate the antidepressant efficacy in the depressive animal model using MR spectroscopy. Fourteen experimentally naive male Sprague-Dawley rats weighting $160{\sim}180\;g$ were used as subjects. Drug injection group was exposed to the FST except for control group. The drugs were administered subcutaneously (SC) in a volume equivalent to 2ml/kg. And three injections were administered 23, 5, and 1h before beginning the given test. 1H MR spectra were obtained with use of a point resolved spectroscopy (PRESS) localization sequence performed according to the following parameters: repetition time, 2500 ms; echo time, 144 ms; 512 average; 2048 complex data points; voxel dimensions, $1.5{\times}2.5{\times}2.5\;mm^3$ ; acquisition time, 25min. There were no differences in NAA/Cr and Cho/Cr ratio between the right and the left hippocampus both normal control rats and antidepressant-injected rats. Also, no differences were observed in NAA/Cr and Cho/Cr ratio between the normal control rats and the antidepressant-injected rats both the right and the left hippocampus. In this study, we found the recovery of choline signals in the depressive animal model similar to normal control groups as injecting desipramine-HCl which was antidepressant causing anti-immobility effects. Thus, we demonstrated that MR spectroscopy was able to aid in evaluating the antidepressant effect of desipramine-HCl.
감정자유기법(EFT)의 연구를 중심으로 본 경락기반심리요법의 동향

Objectives : The purpose of this study is to investigate the researches about regarding Emotional Freedom Technique (EFT) and to understand the trends of Meridian-based Psychotherapy. Methods : Every article relevant to EFT was obtained from 'Pubmed' and Korean journal databases. Keywords used for searching included 'EFT' and 'Emotional freedom technique'. Results : 1) 5 reviews, 11 randomized controlled trials, 3 controlled trials, 1 single group comparative study and 4 case studies were searched. 2) Anxiety disorders were most frequently studied with EFT. Other studies included insomnia, depression and pain symptoms. EFT interventions were attempted in many different protocols and assessed with various tools. 3) Review articles indicated that meridian-based psychotherapies, such as EFT, are based on the meridian theory of oriental medicine. They evaluate EFT positively for its effectiveness on psychiatric diseases. Conclusions : EFT is increasingly studied and used in clinical practice of various fields. Objective evaluation tools and standardized intervention protocols are needed for the development of a new guideline for EFT.

In the manufacture of CFRP(Carbon Fiber Reinforced Polymer Composite) composite structures, various independent components join by bolts and pins. Holes for bolts and pins have an effect on the failure strength of such structures, because those act as notches in structures. The failure characteristic of such structures are different from those of plain plate subject to remote load. In this paper, tensile properties of woven CFRP composite plates with laminates of 0°, 30°and 45° were obtained according to ASTM D 3039. By using obtained tensile failure strength and Tan-Cheng failure criterion, tensile failure strength of CFRP laminate with arbitrary fiber angle were evaluated. Also, the degradation of tensile properties by center hole(φ10㎜) with a remote load was evaluated and the failure strengths were applied to Tan's failure criterion, similarly.
폴리옥시에틸렌 메타크릴레이트 반응성 계면활성제를 이용한 수성 접착제 제조

Water-based adhesives are increasingly used in recent years because they are easy to handle and environmentally safe. In order to compare the properties of the water-based adhesive according to the surfactant used, synthetic surfactants were confirmed. Solid content, viscosity, initial adhesive force were compared. The synthesized reactive surfactants were polyoxyethylene(10) oleyl ether methacrylate(POE(10)-MA) and polyoxyethylene(20) oleyl ether methacrylate(POE(20)-MA). In the case of using POE(10)-MA and POE(20)-MA respectively, the solid contents were 68% and 56%. The initial adhesion was measured as No. 32 for adhesives with POE(10)-MA and No. 20 for the use of POE(20)-MA, respectively.

In this study, a simplified FE model for evaluating seismic performance of valve support was suggested and an apparatus for a real structure testing was developed. The seismic performances of three different types of valve supports were evaluated by the real structure testing. By comparing the results between the real structure testing and FEA using the simplified FE model, it was verified that the suggested simplified FE model can be utilized for comparative evaluation of seismic performance of valve supports.
