A new ent-kaurane type diterpenoid glycoside, 17-O-β-D-glucopyranosyl-16α-ent-kauran-19-oic acid (1), together with 17-hydroxy-16α-ent-kauran-19-oic acid (2), 16α,17-dihydroxyl-ent-kauran-19-oic acid (3), and 16α-hydroxy-17-acetoxy-ent- kauran-19-oic acid (4) were isolated from the aerial parts of Inula japonica Thunb. The structure of 1 was determined mainly by use of 1D and 2D NMR spectroscopic techniques including HSQC, 1H-1H COSY, HMBC, and NOESY. In addition, 4 exhibited significant inhibitory activity on NO production in LPS-stimulated RAW264.7 cells with IC50 value of 14.3 μg/mL.

Objective: Tiny cerebral aneurysms are difficult to embolize because the aneurysm’s sac is too small for a single small coil, and coils within the aneurysm may escape from the confinement of a stent. This study was performed to introduce the stent-assisted coil-jailing technique and to investigate its effect on the coil embolization of tiny intracranial aneurysms. Materials and Methods: Sixteen patients with tiny intracranial aneurysms treated with the stent-assisted coil-jailing technique between January 2011 and December 2013 were retrospectively reviewed and followed-up. Results: All aneurysms were successfully treated with the coil-jailing technique, and at the end of embolization, complete occlusion of the aneurysm was achieved in 9 cases (56.3%), incomplete occlusion in 6 (37.5%), and partial occlusion in 1 (6.3%). Intraprocedural complications included acute thrombosis in one case (6.3%) and re-rupture in another (6.3%). Both complications were managed appropriately with no sequela. Follow-up was performed in all patients for 3–24 months (mean, 7.7 months) after embolization. Complete occlusion was sustained in the 9 aneurysms with initial complete occlusion, progressive thrombosis to complete occlusion occurred in the 6 aneurysms with initial near-complete occlusion, and one aneurysm resulted in progressive thrombosis to complete occlusion after initial partial occlusion. No migration of stents or coils occurred at follow-up as compared with their positions immediately after embolization. At follow-up, all patients had recovered with no sequela. Conclusion: The stent-assisted coil-jailing technique can be an efficient approach for tiny intracranial aneurysms, even though no definite conclusion regarding its safety can be drawn from the current data.

In this paper, polycaprolactone (PCL)/collagen (COL) nanofibrous membranes were prepared by electrospinning to study their mechanical properties as well as biocompatibility. The surface morphology was characterized by scanning electron microscopy (SEM) to access the effect of microstructure on the mechanical properties, cytocompatibility, and enzymatic degradation behaviors. In addition, the effect of hydrophilicity of membranes on the antithrombotic properties was also investigated. The results show that tensile strength of PCL/COL10 is higher than that of other samples due to smaller fiber diameters and tight spatial interlacing of PCL/COL10 membranes. In addition, the hydrophobic surface of PCL becomes hydrophilic after the addition of collagen, which affect platelet adhesion and cytocompatibility of nanofibrous membrane samples. The addition of collagen could promote the cell proliferation as well as the enzymatic degradation rate. Overall, PCL/COL10 has good morphology, the best mechanical properties, appropriate degradation rate, and the best antithrombotic effect as well as biocompatibility.
A Study on Major Factors Influencing Dry Cutting Temperature of AISI 304 Stainless Steel

TiN coated and uncoated tools were used to conduct the dry cutting temperatures for AISI 304 stainless steel. Effects of cutting variables on cutting temperatures were investigated using single factor experiment method. The optimal cutting parameters of cutting temperatures were obtained through orthogonal experiment method. The forming process and temperature distribution of chips were analyzed using finite element method (FEM). The results show that cutting temperature generally increases with the increase of cutting speed and feed rate. When depth of cut increases at the range of 0.8-1.5 mm, the cutting temperature increases firstly and decreases subsequently. And the cutting temperature of TiN coated tool is lower than that of uncoated tool. The optimal parameters of cutting temperature are obtained by orthogonal experiment, which is A1B1C3. The highest temperature appears on the second deformation zone by the simulation.

BACKGROUND: Human umbilical cord mesenchymal stem cells (hUCMSCs) have emerged as promising therapy for immune and inflammatory diseases. However, how to maintain the activity and unique properties during cold storage and transportation is one of the key factors affecting the therapeutic efficiency of hUCMSCs. Schisandrin B (SchB) has many functions in cell protection as a natural medicine. In this study, we investigated the protective effects of SchB on the hypothermic preservation of hUCMSCs. METHODS: hUCMSCs were isolated from Wharton’s jelly. Subsequently, hUCMSCs were exposed to cold storage (4 C) and 24-h re-warming. After that, cells viability, surface markers, immunomodulatory effects, reactive oxygen species (ROS), mitochondrial integrity, apoptosis-related and antioxidant proteins expression level were evaluated. RESULTS: SchB significantly alleviated the cells injury and maintained unique properties such as differentiation potential, level of surface markers and immunomodulatory effects of hUCMSCs. The protective effects of SchB on hUCMSCs after hypothermic storage seemed associated with its inhibition of apoptosis and the anti-oxidative stress effect mediated by nuclear factor erythroid 2–related factor 2 signaling. CONCLUSION: These results demonstrate SchB could be used as an agent for hypothermic preservation of hUCMSCs.
Galectin-9 Acts as a Prognostic Factor with Antimetastatic Potential in Hepatocellular Carcinoma

Considerable research has been conducted concerning galectin-9 and carcinomas, but little information is available about any relation with the hepatocellular carcinoma. In this study, we employed a small interfering RNA (siRNA) targeting galectin-9 to down-regulate the expression in HepG2 cells. As a result, after galectin-9 expression was reduced, cell aggregation was suppressed, while other behaviour such as the proliferation, adhesion and invasion to ECM, cell-endothelial adhesion and transendothelial invasion of the cells were markedly enhanced. When tumors of 200 patients with hepatocellular carcinoma were tested for galectin-9 expression by immunohistochemistry, binding levels demonstrated intimate correlations with the histopathologic grade, lymph node metastasis, vascular invasion and intrahepatic metastasis (P<0.05). Moreover, survival analysis indicated that patients with galectin-9 expression had much longer survival time than those with negative lesions, and the Log-rank test indicated that this difference was statistical significant (P<0.0001). The Cox proportional hazards model suggested that negative galectin-9 expression in hepatocellular carcinoma represented a significant risk factor for patient survival. We propose that galectin-9 might be a new prognostic factor with antimetastatic potential in patients with hepatocellular carcinoma.

Hepatitis B virus (HBV) genome P-encoded protein HBV DNA polymerase (Pol) has long been known as a reverse transcriptase during HBV replication. In this study, we investigated the impact of HBV Pol on host cellular processes, mainly apoptosis, and the underlying mechanisms. We showed a marked reduction in apoptotic rates in the HBV Pol-expressed HepG2 cells compared to controls. Moreover, a series of assays, i.e., yeast two-hybrid, GST pull-down, co-immunoprecipitation, and confocal laser scanning microscopy, identified the host factor eEF1A2 to be associated with HBV Pol. Furthermore, knockdown of eEF1A2 gene by siRNA abrogated the HBV Pol-mediated anti-apoptotic effect with apoptosis induced by endoplasmatic reticulum (ER) stress-inducer thapsigargin (TG), thus suggesting that the host factor eEF1A2 is essential for HBV Pol’s anti-apoptosis properties. Our findings have revealed a novel role for HBV Pol in its modulation of apoptosis through integrating with eEF1A2.
RPSA Gene Mutants Associated with Risk of Colorectal Cancer among the Chinese Population

The primary aim of this study was to evaluate the relationship of single nucleotide polymorphisms (SNPs) in ribosomal protein SA (RPSA) gene with colorectal cancer (CRC). A case-control study including 388 controls and 387 patients with CRC was conducted in a Chinese population. Information about socio-demography and living behavior factors was collected by a structured questionnaire. Three SNPs (rs2133579, rs2269349, rs7641291) in RPSA gene were genotyped by Illumina SnapShot method. Multiple logistic regression models were used for assessing the joint effects between tea consumption and SNPs on CRC. The subjects with rs2269349 CC genotype had a decreased risk for CRC (OR=0.60; 95%CI = 0.37-0.99), compared with TT/CT genotype after adjustment for covariates. A similar association of rs2269349 with rectal cancer was observed (OR=0.49; 95%CI=0.24-1.00). Further analyses indicated that this SNP could modify the protective effect of tea drinking on CRC. Among the subjects with rs2269349 TT/CT or rs2133579 AA/GA, there was a marginal significantly lower risk of CRC (OR and 95%CI: 0.63 and 0.39-1.01 for rs2269349; 0.64 and 0.40-1.02 for rs2133579) in tea-drinking subjects in comparison to non-tea-drinking subjects. Mutants in the RPSA gene might be associated with genetic susceptibility to CRC and influence the protective effect of tea consumption in the Chinese population.

Two ergosterins and two triterpenoids were isolated from the dried fruit bodies of Ascomyce Bulgaria inquinans. By means of chemical (hydrolysis) and spectroscopic methods (NMR, EIMS), their structures were established as betuinic acid (1), cerevisterol (2), (24R)ergosta-7, $22Ediene-3\beta,\;5\alpha,\;6\beta-triol-3-O-palmitate$ (3) and ursolic acid (4). Compound 3 is a new compound.
Research on the Cutting Performances and Wear Mechanisms of TiAlCrN Coated Tools during Dry Turning

Physical vapour deposition technique was used to deposit TiAlCrN coatings on the YT15 tungsten carbide inserts. The dry turning tests of 20CrMo steel were carried out to evaluate performances of TiAlCrN coated and uncoated tools on the CA6140A lathe. The effect of the two kinds of tools on cutting forces, cutting temperature, surface roughness and tool wear had been investigated to assess the performance of TiAlCrN coated tools. The results showed that the cutting force and cutting temperature obtained by TiAlCrN coated tools were decreased and the TiACrN coated tools produced a better surface finish in comparison with the uncoated tools. The TiAlCrN coated tools yield working life about 45 min, which was two times of that for uncoated tools. The wear mechanisms of the TiAlCrN coated tools were mainly oxidation and boundary wear, accompanied with diffusion wear.
