        Effects of Traditional Chinese Cooking Methods on Formation of Heterocyclic Aromatic Amines in Lamb Patties

        Different amounts of the potent mutagenic and/or carcinogenic heterocyclic aromatic amines (HAAs) areformed in muscle-based foods under different cookingmethods. HAAs (9 varieties) in lamb patties cooked usingtraditional Chinese cooking methods (roasting, frying, panfrying,and stewing in seasonings) were investigated. Thetotal HAAs contents in roasted, fried, pan-fried, and stewedpatties were 4.39-123.15 ng/g, 3.59-43.24 ng/g, 0.71-10.05ng/g, and 51.07-120.32 ng/g, respectively. Amounts of HAAsincreased as cooking time increased. 2-amino-1-methyl-6-phenylimidazo [4,5-b] pyridine (PhIP) was the dominantHAAs in deep roasted and fried samples, while 1-methyl-9H-pyrido [3,4-b] indole (Harman) and 9H-pyrido [3,4-b]indole (Norharman) were the main HAAs in pan-fried andstewed samples. Types and contents of HAAs formed atdifferent cooking times using different methods are unique. Stewing in seasoning generated a higher HAAs contentthan the high-temperature cooking methods roasting,frying, and pan-frying.

        OSI-027 alleviates rapamycin insensitivity by modulation of mTORC2/AKT/TGF-β1 and mTORC1/4E-BP1 signaling in hyperoxia-induced lung injury infant rats

        Background The mechanism of long time and high-concentration oxygen treatment leading to acute lung injury (ALI) or developmental lung disease in infants is currently unclear. Here we found that compared with the effect of rapamycin, pan-mTOR1/2 inhibitor OSI-027, alleviates hyperoxia-induced lung injury (HILI) by modulation of mTORC2/AKT/TGF-β1 and mTORC1/4E-BP1 signaling in infant rats. Objective Infant rats were treated with continuous inhalation of 90% medical oxygen. Normal saline, rapamycin, or OSI-027 was intraperitoneally injected, and the status of lung injury was tested on days 3, 7, and 14. The activation of mTOR/AKT/TGFβ1 and mTORC1/4E-BP1 signaling was confirmed by immunohistochemistry and Western blot analysis in normal and hyperoxia-treated live precision-cut lung tissues. The inhibitory effect of OSI-027 extended to the active state of other proteins implicated in mTOR1/2 signaling was demonstrated in hyperoxia-induced injured lung tissues. Results Our data demonstrate that hyperoxia-induced serious lung inflammation and fibrosis. OSI-027 significantly attenuated the pathological process of HILI, inhibit the phosphorylation of the primary downstream targets of mTORC1/C2, and reduce the activation of TGF-β1 signaling. Conclusions The results suggest that mTORC2/AKT/TGF-β1 and the rapamycin-insensitive mTORC1/4E-BP1 (Thr37/46) signaling has an important effect during HILI with a potential meaning for the progress of novel anti-hyperoxia-injury strategies.       • Geometric Optimization of a Mathematical Model of Radiofrequency Ablation in Hepatic Carcinoma

        Radio frequency ablation (RFA) is an effective means of achieving local control of liver cancer. It is a particularly suitable mode of therapy for small and favorably located tumors. However, local progression rates are substantially higher for large tumors (>3.0 cm). In the current study, we report on a mathematical model based on geometric optimization to treat large liver tumors. A database of mathematical models relevant to the configuration of liver cancer was also established. The specific placement of electrodes and the frequency of ablation were also optimized. In addition, three types of liver cancer lesion were simulated by computer guidance incorporating mathematical models. This approach can be expected to provide a more effective and rationale mechanism for employing RFA in the therapy of hepatic carcinoma.

        Effect of Thermal Aging on Electrical Properties of Low Density Polyethylene

        The thermal degradation of low density polyethylene (LDPE) will accelerate the production of carbonyl groups (C=O), which can act as the induced dipoles under high voltage. In this paper, we researched the dielectric properties and space charge behavior of LDPE after thermal aging, which can help us to understand the correlation between carbonyl groups (C=O) and electrical properties of LDPE. The spectra results show that LDPE exhibit obvious thermooxidative reactions when the aging time is 35 days and the productions mainly contain carboxylic acid, carboxylic eater and carboxylic anhydride, whose amount increase with the increasing of aging time. The dielectric properties show that the real permittivity of LDPE is inversely proportional to temperature before aging and subsequently become proportional to temperature after thermal aging. Furthermore, both the real and imaginary permittivity increase sharply with the increasing of aging time. The fitting results of imaginary permittivity show that DC conductivity become more sensitive about temperature after thermal aging. On this basis, the active energies of materials calculated from DC conductivity increase first and then decrease with the increasing of aging time. In addition, the space charge results show that the heterocharges accumulated near electrodes in LDPE change to the homocharges after thermal aging and the mean volume charge density increase with the increasing of aging time. It is considered that the overlaps caused by electrical potential area is the main reason for the increase of DC conductivity.

        Genetic diversity and population structure of the amylolytic yeast Saccharomycopsis fibuligera associated with Baijiu fermentation in China

        The amylolytic yeast Saccharomycopsis fibuligera is a predominant species in starters and the early fermentation stage of Chinese liquor (Baijiu). However, the genetic diversity of the species remains largely unknown. Here we sequenced the genomes of 97 S. fibuligera strains from different Chinese Baijiu companies. The genetic diversity and population structure of the strains were analyzed based on 1,133 orthologous genes and the whole genome single nucleotide polymorphisms (SNPs). Four main lineages were recognized. One lineage contains 60 Chinese strains which are exclusively homozygous with relatively small genome sizes (18.55–18.72 Mb) and low sequence diversity. The strains clustered in the other three lineages are heterozygous with larger genomes (21.85–23.72 Mb) and higher sequence diversity. The genomes of the homozygous strains showed nearly 100% coverage with the genome of the reference strain KPH12 and the sub-genome A of the hybrid strain KJJ81 at the above 98% sequence identity level. The genomes of the heterozygous strains showed nearly 80% coverage with both the sub-genome A and the whole genome of KJJ81, suggesting that the Chinese heterozygous strains are also hybrids with nearly 20% genomes from an unidentified source. Eighty-three genes were found to show significant copy number variation between different lineages. However, remarkable lineage specific variations in glucoamylase and α-amylase activities and growth profiles in different carbon sources and under different environmental conditions were not observed, though strains exhibiting relatively high glucoamylase activity were mainly found from the homozygous lineage.

      • Association of TNF-α-308 and -238 Polymorphisms with Risk of Cervical Cancer: A Meta-analysis

        Published data on the associations between tumor necrosis factor-alpha (TNF-${\alpha}$) promoter -308G>A and -238G>A polymorphisms and cervical cancer risk are inconclusive. To derive a more precise estimation of the relationship, a meta-analysis was performed. Data were collected from MEDLINE and PubMed databases. Crude odds ratios (ORs) with 95% confidence intervals (CIs) were calculated in a fixed/random effect model. 13 separate studies including 3294 cases and 3468 controls were involved in the meta-analysis. We found no association between TNF-${\alpha}$-308G>A polymorphism and cervical cancer in overall population. In subgroup analysis, significantly elevated risks were found in Caucasian population (A vs. G: OR = 1.43, 95% CI = 1.00-2.03; AA vs. GG: OR = 2.09, 95% CI = 1.34-3.25; Recessive model: OR = 2.09, 95% CI = 1.35-3.25) and African population (GA vs. GG: OR = 1.53, 95% CI = 1.02-2.30). An association of TNF-${\alpha}$-238G>A polymorphism with cervical cancer was found (A vs. G: OR = 0.61, 95% CI = 0.47-0.78; GA vs. GG: OR = 0.59, 95% CI = 0.45-0.77; Dominant model: OR = 0.59, 95% CI = 0.46-0.77). When stratified by ethnicity, similar association was observed in Caucasian population (A vs. G: OR = 0.62, 95% CI = 0.46-0.84; GA vs. GG: OR = 0.59, 95% CI = 0.43-0.82; Dominant model: OR = 0.60, 95% CI = 0.44-0.83). In summary, this meta-analysis suggests that TNF-${\alpha}$-238A allele significantly decreased the cervical cancer risk, and the TNF-${\alpha}$-308G>A polymorphism is associated with the susceptibility to cervical cancer in Caucasian and African population.

      • Image Quality Assessment with Saliency Map in Nonsubsampled Contourlet Transform Domain

        Many researchers evaluate images by objective image quality assessments instead of subjective ones. Objective image quality assessment sets up mathematical model according to the human visual system, and it evaluates the image quality through the reference image and the distorted image. The Structural Similarity Index (SSIM) is one of the most classical methods in image quality assessment. However, SSIM has several inherent shortcomings. First, SSIM does not take spatial position, spatial frequency, or direction into account. Second, SSIM considers that different regions in an image have equal importance for overall image quality assessment. Third, it is unreasonable to use fixed parameters for various images. To overcome these shortcomings, we propose a new method of image quality assessment based on Nonsubsampled Contourlet Transform (NSCT). Firstly, NSCT is performed to decompose the image into a low-pass map and high-pass ones. Then, low-pass and high-pass maps are respectively assessed with different strategies. In addition, saliency map is added to describe the importance of different regions in an image. Last, we proposed an approach to calculate the adaptive parameters for various images. Experimental comparisons among five public benchmark databases demonstrate that the proposed method is better than other competing methods.

        Influence of Peanut Cultivars and Environmental Conditions on the Diversity and Community Composition of Pod Rot Soil Fungi in China

        Peanut yield and quality are seriously affected by pod rot pathogens worldwide, especially in China in recent years. The goals of this study are to analyze the structure of fungal communities of peanut pod rot in soil in three peanut cultivars and the correlation of pod rot with environmental variables using 454 pyrosequencing. A total of 46,723 internal transcribed spacer high-quality sequences were obtained and grouped into 1,706 operational taxonomic units at the 97% similarity cut-off level. The coverage, rank abundance, and the Chao 1 and Shannon diversity indices of the operational taxonomic units were analyzed. Members of the phylum Ascomycota were dominant, such as Fusarium, Chaetomium, Alternaria, and Sordariomycetes, followed by Basidiomycota. The results of the heatmap and redundancy analysis revealed significant variation in the composition of the fungal community among the three cultivar samples. The environmental conditions in different peanut cultivars may also influence on the structure of the fungal community. The results of this study suggest that the causal agent of peanut pod rot may be more complex, and cultivars and environmental conditions are both important contributors to the community structure of peanut pod rot fungi.
