        Deep sequencing reveals microRNA signature is altered in the rat epididymis following bilateral castration

        Background In the epididymis of bilateral castrated male rat, gene expression profile changed significantly. However, up to date, no study has investigated how these genes were regulated by microRNAs (miRNAs). Objective We investigated the alterations in the miRNA signature of the epididymis from sham-operated and bilaterally castrated rats. Methods By employing deep sequencing technology and qPCR, the global alterations of epididymal miRNA signature between sham-operated (Con-EP library) and bilaterally castrated rats (Cas-EP library) were explored. MiRNA-target interaction networks were annotated by GO and KEGG enrichment. Results We identified 313 and 306 known miRNAs as well as 152 and 114 novel miRNAs in the Con-EP and Cas-EP libraries, respectively. 59 miRNAs were differentially expressed, including 24 up-regulated and 35 down-regulated miRNAs, among which two up-regulated and three down-regulated ones were validated using qPCR. The expression of these miRNAs in the epididymides of rats at different postnatal ages showed regular changes from birth to adult, suggesting they were androgen-regulated. GO analysis showed that many of the miRNA targets were enriched in metabolic processes. KEGG analysis demonstrated that the targets mainly participated in the mitogen-activated protein kinase (MAPK) pathway. Moreover, 3 and 6 functional modules were detected among the up- and down-regulated miRNA target interaction networks, respectively, and these modules were involved in various biological processes. Conclusion This study represents the first systematic investigation of alterations in the miRNA signature of the epididymis from bilaterally castrated rats and will provide useful resources for functional studies of the miRNAs in the male reproductive system.

        Activation of TLR7-mediated autophagy increases epileptic susceptibility via reduced KIF5A-dependent GABAA receptor transport in a murine model

        The pathophysiological mechanisms underlying epileptogenesis are poorly understood but are considered to actively involve an imbalance between excitatory and inhibitory synaptic transmission. Excessive activation of autophagy, a cellular pathway that leads to the removal of proteins, is known to aggravate the disease. Toll-like receptor (TLR) 7 is an innate immune receptor that regulates autophagy in infectious and noninfectious diseases. However, the relationship between TLR7, autophagy, and synaptic transmission during epileptogenesis remains unclear. We found that TLR7 was activated in neurons in the early stage of epileptogenesis. TLR7 knockout significantly suppressed seizure susceptibility and neuronal excitability. Furthermore, activation of TLR7 induced autophagy and decreased the expression of kinesin family member 5 A (KIF5A), which influenced interactions with γ-aminobutyric acid type A receptor (GABAAR)-associated protein and GABAARβ2/3, thus producing abnormal GABAAR-mediated postsynaptic transmission. Our results indicated that TLR7 is an important factor in regulating epileptogenesis, suggesting a possible therapeutic target for epilepsy.

        Strong earthquakes and measurement performance of masonry and adobe structures

        Earthquakes, which are unavoidable natural phenomena in Turkey, have often produced economic and social disaster. The latest destructive earthquakes happened in Van city. Van, Turkey, earthquakes with M = 7.2 occurred on 23 October 2011 at 13:41 (local time), whose epicenter was about 16 km north of Van (Tabanlivillage) and M = 5.6 on 9 November 2011 epicentered near the town of Edremit south of Van in eastern Turkey and caused the loss of life and heavy damages. Both earthquakes killed 644 people and 2608 people were injured. Approximately 4000 buildings collapsed or were seriously damaged. The majority of the damaged structures were seismically insufficient, unreinforced masonry and adobe buildings in rural areas. In this paper, site surveys of the damaged masonry and adobe buildings are presented and the reasons for the caused damages are discussed in detail.

        Optimization of Fermentation Medium for Glycyrrhizin Biotransformation to Monoglucuronyl-glycyrrhetinic Acid by Plackett-Burman and Box-Behnken Design

        Plackett-Burman (PB) and Box-Behnken (BB) experimental designs were used to optimize fermentation variables for the biotransformation of glycyrrhizin (GL) to monoglucuronyl-glycyrrhetinic acid (MGGA). The PB design was first used to screen the important factors among the medium variables. The steepest ascent method was used to approach the optimum range for each of these factors. The BB design was finally used to analyze the response surfaces of the screened factors for further optimization. The optimized conditions for this system were 0.7 g/L MgSO4·7H2O, 1.19 g/L GL, and cultivation for six days. The biotransformation of GL to MGGA could reach up to 35.72%, which is a good result for this kind of transformation.

        Preparation of Nanomaterial Wettable Powder Formulations of Antagonistic Bacteria from Phellodendron chinense and the Biological Control of Brown Leaf Spot Disease

        Brown leaf spot disease caused by Nigrospora guilin- ensis on Phellodendron chinense occurs in a large area in Dayi County, Chengdu City, Sichuan Province, China each year. This outbreak has severely reduced the production of Chinese medicinal plants P. chinense and caused substantial economic losses. The bacterial isolate JKB05 was isolated from the healthy leaves of P. chinense, exhibited antagonistic effects against N. guilinensis and was identified as Bacillus megaterium. The following fermentation medium and conditions improved the inhibitory effect of B. megaterium JKB05 on N. guilinensis: 2% glucose, 0.1% soybean powder, 0.1% KCl, and 0.05% MgSO4; initial concentration 6 × 106 cfu/ml, and a 42-h optimal fermentation time. A composite of 0.1% nano-SiO2 JKB05 improved the thermal stability, acid-base stability and ultraviolet resistance by 16%, 12%, and 38.9%, respectively, and nano-SiO2 was added to the fermentation process. The best formula for the wettable powder was 35% kaolin, 4% polyethylene glycol, 8% Tween, and 2% humic acid. The following quality test results for the wettable powder were obtained: wetting time 87.0 s, suspension rate 80.33%, frequency of microbial contamination 0.08%, pH 7.2, fineness 95.8%, drying loss 1.47%, and storage stability ≥83.5%. A pot experiment revealed that the ability of JKB05 to prevent fungal infections on P. chinense increased considerably and achieved levels of control as high as 94%. The use of nanomaterials significantly improved the ability of biocontrol bacteria to control this disease.

        The Protective Roles of S-adenosylmethionine Decarboxylase (SAMDC) Gene in Melon Resistance to Powdery Mildew Infection

        Powdery mildew caused by Podosphaera xanthii (P. xanthii) is one of important diseases in melon. Wehave previously investigated the differential gene expression in the incompatible P. xanthii-melon interactions andidentified one EST containing homologous sequences to S-adenosylmethionine decarboxylase (SAMDC) cDNA. Giventhis, SAMDC gene of Cucumis melo was cloned and designated as CmSAMDC in this study. It was 1,095 bp longand encoded a 364-amino acid peptide with a molecular mass of 40 kD. By sequence analyzing, the deducedCmSAMDC protein was shown to have two conserved regions of a putative proenzyme cleavage site and a PESTdomain. In addition, the expressions of CmSAMDC in the resistant melon materials increased more sharply than inthe susceptible the melon materials, and the higher polyamines (PAs) and hydrogen peroxide (H2O2) contents inresistant melon materials were found as well, which were accompanied by up-regulation of the stress-responsive defenseenzyme activities. Over-expression of CmSAMDC in Arabidopsis resulted in greatly reduced pathogen infection in theinoculated leaves of transgenic lines, enhanced resistance to powdery mildew, and the enhanced resistance appearedto be associated with pathogen-induced cell death. Taken together, our results suggested that CmSAMDC and perhapsits orthologous genes might be involved in responses of plants to biotrophic pathogens.

        Experimental Investigation of Mechanical Properties, Formability and Force Measurement for AA7075-O Aluminum Alloy Sheets Formed by Incremental Forming

        Incremental sheet forming (ISF) has demonstrated its high potential to shape complex three-dimensional components without using specific tooling, thus enabling product customization, cost reduction and efficiencies. This paper presents an experimental campaign to investigate AA7075-O aluminum alloy sheet forming using single point incremental forming (SPIF) technology and understand the forming process mechanism. Firstly, tensile tests were carried out to characterize the mechanical properties of AA7075-O sheets with three different thicknesses. Then, the effects of tool path types with different incremental steps on the maximum part draw angle and the successful part height were evaluated to clarify the formability. Additionally, a fracture forming limit diagram was developed to give the design limits for strain. Finally, the trends in forming forces were analyzed considering the influence of different draw angles,sheet thicknesses, step-down sizes and sheet orientations. Experimental results showed that formability is affected by the part draw angle and incremental steps of the tool path. However, the likelihood of successful forming also depends on other geometrical design parameters such as the part height. In addition, the influences of process parameters on tool forces provide further insights into the deformation mechanics of AA7075-O sheets. The slope of the force curve after the peak value can be identified as a forming failure prediction indicator regardless of the tool path types when using a truncated cone as a benchmark. Also the influence of sheet orientations on forming forces was investigated in SPIF. The vertical forming force is found to be smaller in a sheet orientation 45to the rolling direction than that of a sheet orientation 0 for three different sheet thicknesses.

      • Anomaly Driving Speed Detection and Correction Algorithm based on Quantiles and KNN

        Driving speed is a key parameter for building the traffic state identification model, its precision directly affects the model reliability and the traffic state identification accuracy. Aiming at the standard normal deviation method’s defects in dealing with the extreme noise data, an anomaly driving speed detection algorithm based on quantiles is proposed, use historical data to establish the exception borders which are used to detect whether an unknown data is abnormal; on the basis of the abnormal data detection, a driving speed prediction algorithm based on improved KNN is proposed, use K-means algorithm to clustering the historical data, and predict the next moment’s speed according to the distance between the data to be predicted and the clusters, the predicted speed can be used to correct the abnormal speed. Experimental results show that the detection rate of the proposed anomaly detection algorithm has improved about 4.25% compared with the standard normal deviation method, and the false detection rate has reduced about 25.51%; the mean relative error of the proposed speed prediction algorithm is 13.69%, it can predict the driving speed well, namely, the anomaly driving speed detection and correction algorithm based on quantiles and KNN is feasible and effective.

      • Urban Road Traffic State Identification Algorithm Based On Particle Filter and Fuzzy Discrimination

        Urban road traffic state identification is a key link to realize the intelligent transportation based on the Internet of Vehicles, and accurately positioning vehicles is the foundation to realize the traffic state identification. Aiming at the problem that GPS has signal blind area in positioning vehicles, a vehicle positioning algorithm based on particle filter was proposed, it could improve the traditional algorithm on degradation and large amount of calculations; Based on vehicle positioning, an urban road traffic state identification algorithm based on fuzzy discrimination was proposed, it could comprehensively consider multiple factors’ influence on traffic state. The experiment results show that the improved particle filter algorithm’s mean squared error has increased about 55.437% compared with GPS method, and the traffic state identification algorithm can accurately identify the traffic state of the study area, it can prove that the urban road traffic state identification algorithm based on particle filter and fuzzy discrimination is feasible and effective.
