During maturation, pancreatic islets achieve their full capacity to secrete insulin in response to glucose, undergo morphological changes in which alpha-cells decrease and beta-cell mass increases, and they acquire the normal alpha- and beta-cell proportion changes that are important for islet functions later in life. In rodents, the first week of postweaning is critical for islet maturation. Multiple studies have documented the detrimental effects of several conditions on pancreatic maturation; however, few studies have addressed the use of pharmacological agents to enhance islet maturation. Biotin might have a potential action on islet maturation. Pharmacological concentrations of biotin have been found to modify islet morphology and function. In a previous study, we found that mice fed a biotin-supplemented diet for 8 weeks after weaning showed an increase in basal and glucose stimulated insulin secretion, enlarged islet size, and modified islet structure. In the present study, we investigated the effect of biotin on maturation features during the first week postweaning. Female BALB/cAnN Hsd mice were fed a control or a biotin-supplemented diet for 1 week after weaning. Compared with the control, biotin-supplemented mice showed an increase in pancreatic islet number and area in addition to an augmented proportion of beta-cells in the islet. These effects were related to an increase in beta-cell proliferation. No differences were found in insulin secretion, blood glucose concentrations, or serum insulin levels. These results indicate that biotin supplementation is capable of affecting beta-cell proliferation and might be a therapeutic agent for establishing strategies for regenerative medicine.

A novel semi-interpenetrating polymer network (s-IPN) based on the entrapment of a thermosensitive polymer, the poly(vinyl methyl ether) (PVME), within a crosslinked 3D structure of poly(vinyl alcohol) (PVA) and poly(vinyl methyl etheralt- maleic acid) (COP) was synthesized by an autoclaving process. The preparation method avoids the use of toxic crosslinkers and allows the simultaneous sterilization of material. The PVA/COP/PVME hydrogel were characterized by Fourier transform infrared spectroscopy, thermal techniques, swelling kinetic measurements, scanning electron microscopy, and rheological analysis. The entrapment of PVME within the hydrated polymer framework significantly modified its transition temperature at pH 7.4 and pH 3 conditions. The swelling kinetics of the s-IPN were dependent on pH (7.4, 3 and 1), and temperature (25 and 37℃). The interpenetrated polymer chains reduced the internal pore sizes of crosslinked network without altering its elastic, solid-like behavior. The loading and in vitro release of 5-fluorouracil, a chemotherapeutic agent, from hydrogel systems were studied at different temperature and pH values. The hydrogels showed a sustained drug release up to 5 h at 37℃, in different pH media. The s-IPN exhibited a promising performance for a range of biomedical applications, in particular, for the controlled drug delivery in response to the pH and temperature conditions.

PCR is a highly accurate technique for confirming thepresence of Mycobacterium avium subsp. paratuberculosis(Map) in broth culture. In this study, a simple, efficient, andlow-cost method of harvesting DNA from Map cultured inliquid medium was developed. The proposed protocol(Universidad Austral de Chile [UACH]) was evaluated bycomparing its performance to that of two traditionaltechniques (a QIAamp DNA Stool Mini Kit andcethyltrimethylammonium bromide [CTAB] method). Theresults were statistically assessed by agreement analysis forwhich differences in the number of cycles to positive (CP)were compared by Student’s t-test for paired samples andregression analysis. Twelve out of 104 fecal pools culturedwere positive. The final PCR results for 11 samples analyzedwith the QIAamp and UACH methods or ones examinedwith the QIAamp and CTAB methods were in agreement. Complete (100%) agreement was observed between datafrom the CTAB and UACH methods. CP values for theUACH and CTAB techniques were not significantlydifferent, while the UACH method yielded significantlylower CP values compared to the QIAamp kit. The proposedextraction method combines reliability and efficiency withsimplicity and lower cost.

Fetal development of the cartilage of the pharyngotympanic tube (PTT) is characterized by its late start. We examined semiserial histological sections of 20 human fetuses at 14-18 weeks of gestation. As controls, we also observed sections of 5 large fetuses at around 30 weeks. At and around 14 weeks, the tubal cartilage first appeared in the posterior side of the pharyngeal opening of the PTT. The levator veli palatini muscle used a mucosal fold containing the initial cartilage for its downward path to the palate. Moreover, the cartilage is a limited hard attachment for the muscle. Therefore, the PTT and its cartilage seemed to play a critical role in early development of levator veli muscle. In contrast, the cartilage developed so that it extended laterally, along a fascia-like structure that connected with the tensor tympani muscle. This muscle appeared to exert mechanical stress on the initial cartilage. The internal carotid artery was exposed to a loose tissue facing the tubal cartilage. In large fetuses, this loose tissue was occupied by an inferior extension of the temporal bone to cover the artery. This later-developing anterior wall of the carotid canal provided the final bony origin of the levator veli palatini muscle. The tubal cartilage seemed to determine the anterior and inferior margins of the canal. Consequently, the tubal cartilage development seemed to be accelerated by a surrounding muscle, and conversely, the cartilage was likely to determine the other muscular and bony structures.

For control of brucellosis in small ruminants, attenuated B. melitensis Rev1 is used but it can be virulent for animals and human. Based on these aspects, it is essential to identify potential immunogens to avoid these problems in prevention of brucellosis. The majority of OMPs in the Omp25/31 family have been studied because these proteins are relevant in maintaining the integrity of the outer membrane but their implication in the virulence of the different species of this genus is not clearly described. Therefore, in this work we studied the role of Omp31 on virulence by determining the residual virulence and detecting lesions in spleen and testis of mice inoculated with the B. melitensis LVM31 mutant strain. In addition, we evaluated the conferred protection in mice immunized with the mutant strain against the challenge with the B. melitensis Bm133 virulent strain. Our results showed that the mutation of omp31 caused a decrease in splenic colonization without generating apparent lesions or histopathological changes apparent in both organs in comparison with the control strains and that the mutant strain conferred similar protection as the B. melitensis Rev1 vaccine strain against the challenge with B. melitensis Bm133 virulent strain. These results allow us to conclude that Omp31 plays an important role on the virulence of B. melitensis in the murine model, and due to the attenuation shown by the strain, it could be considered a vaccine candidate for the prevention of goat brucellosis.

Background/AimsThe evidence suggests that a shorter esophageal length (EL) in gastroesophageal reflux disease (GERD) patients is associated with the presence of hiatal hernia (HH). However, there are no reports of this association in patients with achalasia. The aim is to (1) determine the prevalence of hiatal hernia in achalasia patients, (2) compare achalasia EL with GERD patients and healthy volunteers (HV), (3) measure achalasia manometric esophageal length to height (MELH) ratio, and (4) determine if there are differences in symptoms between patients with and without hiatal hernia. MethodsThis retrospective and cross-sectional study consist of 87 pre-surgical achalasia patients, 22 GERD patients, and 30 HV. High-resolution manometry (HRM), barium swallow, and upper endoscopy were performed to diagnose HH. The EL and MELH ratio were measured by HRM. Symptoms were assessed with Eckardt, Eating Assessment Tool, and GERD–health-related quality of life questionnaires. ResultsThe HH in GERD’s prevalence was 73% vs 3% in achalasia patients (P < 0.001). Achalasia patients had a longer esophagus and a higher MELH ratio than HV and GERD patients (P < 0.001). GERD patients had a lower MELH ratio than HV (P < 0.05). EAT-10 (P < 0.0001) and Eckardt (P < 0.05) scores were higher in achalasia without HH vs HH. ConclusionsThe prevalence of HH in achalasia is significantly lower than in GERD. The longer EL and the higher MELH ratio in achalasia could explain the lower prevalence of HH. Despite the low prevalence of HH in achalasia patients, the surgeon should be encouraged not to rule out HH since the risk of postoperative reflux may increase if this condition is not identified and corrected.
