        hnRNPK-regulated PTOV1-AS1 modulates heme oxygenase-1 expression via miR-1207-5p

        Antisense transcripts were initially identified as transcriptional noise, but have since been reported to play an important role in the quality control of miRNA functions. In this report, we tested the hypothesis that heterogeneous nuclear ribonucleoprotein K (hnRNPK) regulates miRNA function via competitive endogenous RNAs, such as pseudogenes, long non-coding RNAs, and antisense transcripts. Based on analyses of RNA sequencing data, the knockdown of hnRNPK decreased the antisense PTOV1-AS1 transcript which harbors five binding sites for miR-1207-5p. We identified heme oxygenase-1 (HO-1) mRNA as a novel target of miR-1207-5p by western blotting and Ago2 immunoprecipitation. The knockdown of hnRNPK or PTOV1-AS1 suppressed HO-1 expression by increasing the enrichment of HO-1 mRNA in miR-1207-5p-mediated miRISC. Downregulation of HO-1 by a miR-1207-5p mimic or knockdown of hnRNPK and PTOV1- AS1 inhibited the proliferation and clonogenic ability of HeLa cells. Taken together, our results demonstrate that hnRNPKregulated PTOV1-AS1 modulates HO-1 expression via miR- 1207-5p. [BMB Reports 2017; 50(4): 220-225]

      • 행동수정과 유산소성운동에 의한 비만 청소년의 체중 및 혈중 총콜레스테롤량 변화에 관한 연구

        The purpose of this article is to provide a method of losing weight and early managing risky factors of geriatric diseases by having male and female obese juveniles participate in behavioral modification and aerobic exercise, foster their own desirable behavioral habit and confidence in solving obesity and continue their living pattern all their life. In order to do this, 64 subjects over 20% obese degree obtained using the standard weight, belonging within 50% range of weight percentage per height (male : 32, female : 32) were grouped to 4 categories of aerobic exercise therapy, behavioral modification therapy, both therapies combinedly applied and control ones by 8 male and female students allocated to each group respectively, and they were practiced to each program for 12 weeks, and the findings between group were obtained as follows. 1. The male and female students' weight reduced most in the combinedly performed group of behavioral modification therapy and aerobic exercise therapy when 12week was applied to each group, and the descending order of next highest rate was aerobic exercise therapy, behavioral modification therapy and control groups, which indicated a statistically significant difference between the mean of male and female weight (F(1.56)=11.93, p=.001) and between measurements according to the application period of program (f(3.169)=265.77, p=.000). 2. the male and female middle school students' cholesterol amount of blood fat reduced most in the combinedly performed group of aerobic exercise and behavioral modification therapy. 3. It indicated a statistically significant difference between groups (f(3.56)=9.50) and the measurements according to the application period of program as well (f(3.168)=554.94, p=.000). 4. The cholesterol amount per measurement period reduced most6 between 8 and 12weeks.

      • 韓國産 蓼(여뀌)科 植物에 관한 本草學的 硏究 (I)

        For the purpose of developing Korean Herbalogy with the plants of Polygonaceae which grow wild and is planted in Korea, the these and writings on herbalogy, from literature of successive generations, have been thoroughly investigative and the results obtained were as follows: 1. There were totaled to 16 genera and 156 species in Korea and among them medicinal plants are 12 genera, 47 species, some 30% in total, but the number of species may be added because of the similiar plants. 2. According to the oriental names which can be used for medical purpose, the medicinal plants belonging to the Polygonaceae family were classified as Herba 21, Radix 12, Foilum 8, Lignum 5, Flos 4, Fructus 5, Ramulus 2 and Semen 1. Thus it was noticed that Herba was the main kind. 3. According to sum of 156 species in Polygonaceae family, they were classified into Persicaria genera 63, Polygonum genera 23, Rumex genera 22, Bistorta genera 14, Aconogonum genera 6, Fagopyrum genera 5, Bilderdykia genera 4, Amblygonum genera 3, Reynoutria genera 3, Rheum genera 3, Pleuropterus genera 2, Pleuropteropyrum genera 2, Tovara genera 2, Ampelygonum genera 1, Homalocladium genera 1 and Oxyria genera 1. Thus it was noticed that Persicaria genera was the main kind, some 40.3% in total. 4. According to nature and flavour of medicinal plants, they were classified into cold, cool; 23 each, warmth, heat; 11 each, balance 11 and unidentified 5. Thus it was noticed that cold, cool is the main in nature and flavour of medicinal plants. From this result, it was revealed that the plants for medical purpose in Polygonaceae was 30% of the whole, in which HERBA was mostly abundunt and toxic plants was slightly poisonous that it will be used for clinical treatments more easily. It is considered that many experiements and clinical approaches must be continued to use Polygonaceae plants widely.

        The problems in valve-train system are valve bouncing, high seating, velocity jumping, surge, valve acceleration and overshoot in high-speed running. It is essential to estimate them accurately in designing the shape of cam. In this paper, the method for measurement of contact load, which is applied between rocker arm and valve, using strain gauge is provided. In the result of this research, contact load and dynamic characteristics between rocker arm and valve is verified, and the design data for improving the engine performance are provided.

        top quark의 질량을 추정하기 위해 본 연구에서는 광범위하게 중성 K중간자계의 현상을 조사 연구하였다. top quark의 질량이 Frizsch의 질량행렬과 Sctech의 질량행렬로 예언된 상한값을 넘을 수 있다는 실험 결과에 비추어 top quark의 큰 값을 수용할 수 있는 B.L. 질량행렬과 G.J. 질량행렬을 사용하였다. 또한 KL-KS 질량차 Δm과 CP 비보존 매개변수 |ε|을 구속조건으로 사용하였다. G.J. 질량행렬로 얻은 혼합행렬을 실험자료와 비교하여 117Gev∼178Gev의 범위에 top quark의 질량이 존재한다는 것을 얻었다. 중성 Kaon계에서 Δm과 |ε|의 값의 계산에서 C.K.M. 행렬과 이론적 투입 매개변수에 대한 합리적 값을 사용하여 구간 mt의 하한은 185Gev이며, G.J. 질량행렬로 얻은 혼합행렬과 투입 parameter를 써서 구한 mt의 하한은 180Gev였다. B.L. 질량행렬로 얻은 혼합행렬을 사용할 때 mt의 하한의 범위는 220∼260Gev이다. We have investigated the phenomenology of the neutral kaon system in a comprehensive way in order to estimate the top quark mass. In the light of experimental indications that the top quark mass, mt may exceed the upper bound predictioned by the Fritzsch ansatz and Stecch's ansatz for quark mass matrices, we have used B. L. mass matrix and G. J. mass matrix which could accommodate large value of mt. And also we have used the KL-KS mass difference Δm and the CP non-conservation parameters |ε|, as constraints to be satisfied. Comparing the C. K. M. matrix by G. J. ansatz and experimental data, we have obtained mt to be in the range 117Gev - 178Gev. In the neutral kaon system C. K. M. matrix and reasonable values for the theoretical parameters suggest the lower bound of top quark mass might be about 185GeV. And also we have obtained the lower bound of mt to be about 180GeV through the mixing matrix by G. J. ansatz and values of input parameters. When we used the Hiking matrix made by B. L. ansatz we have obtained the lower bound of mt to be in the range 220GeV-260GeV.

      • 다축 제어 시뮬레이터 링크부의 동특성에 관한 연구

        차량개발과정에 있어서 완성차량에 대한 실험방법은 크게 실차 주행실험과 실험실 재현실험의 두가지로 분류할 수 있다. 이 중 실험실 재현실험은 주어진 실험실내의 공간에서 차량의 주행상태를 반복, 재현함으로써 시간의 절감과 함께 공간 및 환경의 제약에서 벗어날 수 있으며, 실험의 지속적 재현성과 실험중인 차량을 수시로 관찰, 분석할 수 있어 취약부분 및 성능의 조기 확인이 가능한 장점이 있다. 이런 이유로 많은 국외 자동차사에서는 다축 제어 시뮬레이터를 이용한 실험실 재현실험을 활성화하여 차량개발기간의 단축은 물론 높은 신뢰성을 가진 고품질 자동차의 개발에 활용하고 있는 실정이나, 아직까지 국내에서는 수입하여 활용측면에서는 아직 초보단계를 벗어나지 못하고 있다. 본 논문에서는 실험실 재현실험의 구동부인 다축 제어 시뮬레이터에 작용하는 4가지 요소중 3가지 요소인 연직·전후·횡 하중등을 복합적으로 재현할 수 있는 링크 구조를 설계하여 동역학 해석 프로그램인 ADAMS를 이용하여 시뮬레이션을 수행하고, 링크부의 안정성 평가 및 최적모델의 사양을 결정하기 위해 FEM 해석을 하였다. Test in the development of vehicle consist of driving test and simulation test. The last one has many advantages. It can reduse time and cost during development, can overcome the spacial and environmental limitation, and can provides repeatabilities of similar experiments and various data. In these reason, the simulation test is widely used in the developed countries for developing new vehicles and analyzing the dynamic characteristics of vehicle developed.

      • Induction of Apoptosis and Inhibition of Invasion in Human Hepatoma Cells by Anthocyanins from Meoru

        <P>Anthocyanins belong to a class of flavonoids exhibiting antioxidant and anti-inflammatory actions as well as a variety of chemotherapeutic effects. However, little is known about the cellular and molecular mechanism of anticancer activity. In this study, we investigated if the anthocyanins (delphinidin-3,5-diglucoside: cyanidin-3,5-diglucoside: petunidin-3,5-diglucoside: delphinidin-3-glucoside: malvdin-3,5-diglucoside: peonidin-3,5-diglucoside: cyanidin-3-glucoside: petunidin-3-glucoside: peonidin-3- glucoside: malvidin-3- glucoside = 27: 63: 8.27: 1:2.21: 6.7: 1.25: 5.72: 1.25) isolated from meoru (Vitis coignetiae Pulliat) exerted antiproliferative and anti-invasive and apoptotic effects on human hepatoma Hep3B cells. It was found that the anthocyanins could inhibit cell growth by 75% at the concentration of 400 microg/mL for 48 h. Flow cytometric analysis showed that the anthocyanins increased the amount of DNA fragments (sub-G1 fraction) in a dose-dependent manner, which is closely related to mitochondrial dysfunction and reduction in antiapoptotic proteins (Bcl-2, xIAP, cIAP-1, and cIAP-2). The anthocyanins also significantly inhibited the migration and invasion of Hep3B cells through a matrigel-coated chamber. Taken together this study indicates that the anthocyanins from meoru have antiproliferative and anti-invasive effects and may induce apoptosis through the activation of the mitochondrial pathway and inhibition of antiapoptotic proteins. This study provides evidence that the anthocyanins isolated from meoru might be useful in the treatment of human hepatitis B-associated hepatoma.</P>
