      • Effect of gel spinning accompanied with cross-linking using boric acid on the structure and properties of high-molecular-weight poly(vinyl alcohol) fiber

        <P>To enhance the thermal stability of poly(vinyl alcohol) (PVA) fiber, the fiber was prepared from the gel spinning of high molecular weight (HMW) PVA by using dimethyl sulfoxide/water (8/2, v/v) as a solvent, accompanied with the cross-link by boric acid (B-PVA). In addition, the structure and properties of the B-PVA fiber were compared with those of the HMW PVA fiber obtained by using the same spinning system without cross-linking (NB-PVA). Through a series of experiments, it turned out that cross-linking actualized by an optimum amount of boric acid (0.3 wt % based on PVA) and zone drawing caused significant changes in the properties of HMW PVA gel fiber. That is, cross-linking increased thermal degradation temperatures at each degradation step and amounts of final residues, resulting in improving thermal properties of the PVA fiber. On the contrary, it was found that in the case of the B-PVA fiber, some broadening of the original PVA unit cell occurred, which was identified by the peak shift to lower angle in X-ray diffractogram.       • Removal of Cochlodinium polykrikoides using a novel material produced from sediment - A field study

        The present study was conducted in three fields at Namhae to examine the removal efficiency of organic free, heavy metal immobilized sediment on Cochlodinium polykrikoidesand on sea water quality. The present study results concluded that removal efficiency was depends on the initial number of red tide cells. There was no drastic change in the sea water quality after sediment spray. For the comparison of effectiveness of betonite, zeolite and lime were mixed with sediment did not show any marked difference in removal. Finally, the present study evaluated 5Og/m² sediment is Sufficient to remove 100% cells density, even though various environmental factors are interfering the mechanism.

        이모작답(二毛作畓)에 있어서 수도이앙기(水稻移秧機)의 실용화(實用化)에 관(關)한 연구(硏究)

        수도재배(水滔栽培)에 기계이앙(機械移秧)을 실시(實施)하기 위(爲)하여 조생통일외(早生統一外) 3개(固) 품종(品種)을 4월(月) 16일(日)부터 10일(日) 간격(間隔)으로 3회(回) 상자육묘(箱子育苗)하고 40일묘(日苗)를 이앙기(移秧機)로 이양(移秧)하였던 바 묘(苗)의 생육(生育)과 기계이앙후본답(機械移秧後本畓)에서의 수도(水滔)의 생육(生育)과 수량(收量)을 조사(調査)한 결과(結果)를 요약(要約)하면 다음과 같다. 1. 40일묘(日苗)의 초장(草長)과 엽수(葉數)는 관행육묘(慣行育苗)에 비(比)해 상자육묘구(箱子育苗區)에서 현저(顯著)히 짧은 경향(傾向)이었으며 상자육묘(箱子育苗)의 경우 물못자리 상자육묘구(箱子育苗區)의 초장(草長) 및 엽수(葉數)가 밭못자리구(區)보다 현저(顯著)히 길었다. 2. 40일묘(日苗)를 단근(斷根)하여 발근력(發根力)을 시험(試驗)한 결과(結果) 상자육묘구(箱子育苗區)의 발근력(發根力)은 관행육묘구(관行育苗區)의 발근력(發根力)에 비(比)해 현저(顯著)히 낮았다. 한편 같은 상자육묘구(箱子育苗區)에서는 물못자리 상자육묘구(箱子育苗區)의 묘(苗)의 발근력(發根力)이 밭못자리에 비(比)해 높은 경향(傾向)이었다. 3. 본시험(本試驗)에 공시(供試)된 유일계(流一系) 품종(品種)은 상자육묘(箱子育苗)의 경우 못자리에서 25~30일(日) 경(頃) 부터 잎의 상부가 황변(黃變) 내지 황적색(黃赤色)으로 변(變)하고 생육(生育)이 거의 정지(停止)되는 현상(現像)을 나타내었다. 4. 상자육묘(箱子育苗)의 기계이앙구(機械移秧區)는 관행육묘이앙구(慣行育苗移秧區)에 비(比)해 활착(活着)이 5~7일(日) 이상(以上) 지연되었으며 출수기(出穗期)도 품종(品種)에 따라 차이(差異)가 있으나 대체로 5~7일(日) 지연되었다. 5. 정조수량(正粗收量)은 유신(維新)의 관행재배구(慣行栽培區)에 비(比)해 모두 약간의 감수(減收)를 보였는데 기계이앙구내(機械移秧區內)에서는 5월(月) 26일(日) 이앙(移秧)의 경우 통일(統一)의 수량(收量)이 가장 높았고 6월(月) 15일(日)의 수량(收量)은 밀양(密陽) 15호(號)가 가장 많았으며 기계이앙재배(機械移秧栽培)를 위(爲)한 품종(品種)의 선택(選擇)은 중요(重要)한 것으로 인정(認定)된다. 6. 수도이앙기(水滔移秧機)로는 $3.3m^2$당(當) 이앙주수(移秧株數)를 73주(株)부터 108주(株)까지 조정(調整)이 가능(可能)하며 파종밀도(播種密度)가 균일(均一)하면 결주율(缺株率) 1% 미만(未滿)의 작업정도(作業精度)로 인력이앙작업(人力移秧作業)에 비(比)하여 2조(條)의 경우 5~6배(倍)의 이앙작업능률(移秧作業能率)을 올릴 수 있으므로 단작지대(單作地帶)에서는 실용화(實用化)가 용역(容易)하리라 믿는다. 7. 이모작지대(二毛作地帶)에서는 이앙시기(移秧時期)가 한정(限定)되어 있으므로 적절(適切)한 품종(品種)의 선택(選擇)과 상자묘(箱子苗)의 육묘기술(育苗技術)이 개발(開發)된다면 수량(收量)은 감소(減少)없이 이앙작업(移秧作業)의 기계화(機械化)가 무난(無難) 할 것으로 전망(展望)된다. In order to investigate the effects of mechanical transplanting on the rice, four rice varieties including Jo-saing Tongil were planted in case nursering bed for three times with 10 days interval beginning from April 16. Fourty days old seedlings were transplanted with rice-transplanter, and growth habits of seedlings in bed and rice plants in main field and yield were examined and summerized as follows: 1. The plant height and number of leaves of the 40 days old seedling in case nursery bed were appeared to be less than those of seedlings grown in ordinary nursery bed. In case nursery, the plant height and number of leaves of seedlings grown in case nursery were apparently higher than those of seedlings grown in upland case nursery. 2. The root of 40 days old seedlings were cut to investigate the rooting ability of seedlings, and the rooting ability of the seedlings grown in the case nursery bed were apparently lower in the rooting ability compared with the seedlings grown in ordinary nursery bed. The rooting ability of the seedlings grown in the lowland case nursery was higher than that of the seedlings grown in the upland case nursery. 3. The tip of leaves of the varieties derived from Indica type were becoming yellow and yellow-red in color 25 to 30 days after planted in the case nursery bed. Those varieties was almost stopped growth. 4. The seedlings transplanted by transplanter had shown more than five to seven days of delay in rooting, compared with the seedlings transplanted by ordinary method. The heading dates was delayed five to seven days when transplanter was used, even though the delay of heading dates was a little different depending on the varieties. 5. The yields of unhulled rice were a little lower than that of Yusin variety which was hand planted. When mechanically transplanted, the yield of Tongil variety was highest in the plot where seedlings were transplanted on May 26. When transplanted on June 15, Milyang#15 was highest in yield, indicating the importance of varietal selection in mechanical transplantation. 6. The rice transplanter could transplant from 73 to 108 seedlings per $3.3m^2$. If the seeds of rice were evenly planted in the bed, the two-row rice transplanter could be used five to six times more effectively with less than one percent of missing plant compared with the hand transplanting. This suggests that rice transplanter could be utilized in the mono cropping area. 7. Since the time of transplantings in the double-cropping area is rather limitted, mechanical transplantation might be possible without reducings yield providing better adapted varieties and improved case-nursery techniques.

        Is Transradial Cerebral Angiography Feasible and Safe? A Single Center's Experience

        Objective : Although a transradial angiography is accepted as the gold standard for cardiovascular procedures, cerebral angiography has been performed via transfemoral approach in most institutions. The purpose of this study is to present our experience concerning the feasibility, efficacy, and safety of a transradial approach to cerebral angiography as an alternative to a transfemoral approach. Methods : Between February 2007 and October 2009, a total of 1,240 cerebral angiographies were performed via a transradial approach in a single center. The right radial approach was used as an initial access route. The procedure continued only after the ulnar artery was proven to provide satisfactory collateral perfusion according to two tests (a modified Allen's test and forearm angiography). Results : The procedural success rate was 94.8% with a mean duration of 28 minutes. All supra-aortic vessels were successfully catheterized with a success rate of 100%. The success rates of selective catheterization to the right vertebral artery, right internal carotid artery, left internal carotid artery, and left vertebral artery were 96.1%, 98.6%, 82.6% and 52.2%, respectively. The procedure was performed more than twice in 73 patients (5.9%), including up to 4 times in 2 patients. The radial artery occlusion was found in 4 patients (5.4%) on follow-up cerebral angiography, but no ischemic symptoms were observed in any of the cases. Conclusion : This study suggests that cerebral angiography using a transradial approach can be performed with minimal risk of morbidity. In particular, this procedure might be useful for follow-up angiographies and place less stress on patients.

      • Comparison of Actin Nucleation in Early Stage Mouse and Bovine Oocytes followed by Vitrification and Slow Freezing

        The cryopreservation of unfertilized oocytes is valuable for both livestock and human. However, freezing procedure to oocyte induce many kinds damages such as cytoskeleton disorganization, and chromosome and DNA abnormalities. Especially, during mammalian oocyte meiotic maturation, a process called oocyte polarization occurs, which leads to a unique asymmetric division. Actin play key roles during oocyte polarization experiences several steps including spindle migration, spindle anchoring and cortical reorganization. However, how chilling stress affect to actin nucleation in oocytes is poorly studied. In this study, we investigated expressions and feature of Arp2/3 (actin activators), WAVE2 and JMY (nucleation promoting factors) in MІІ stage oocytes after different freezing procedure in oocytes. Results indicated that survival, IVF, developmental rates of vitrified-thawed oocytes were significantly higher than those of slow freezed oocytes. Especially, expressions of Arp2/3, WAVE2 and JMY in vitrified-thawed oocytes were similar with normal oocytes. However, expression of actin nucleation related genes in slow freezed group was significantly higher or lower than normal oocytes. Our results may suggested that slow freezing procedure to oocyte induced abnormal actin neucleation results in low survival and developmental rates.

        A novel role for bone-derived cells in ankylosing spondylitis: Focus on IL-23

        <P><B>Abstract</B></P> <P>The main aim of this study are to explore the role of bone-derived cells (BdCs) in ankylosing spondylitis (AS) and determine the underlying molecular mechanisms of IL-23 production. Primary BdCs were isolated from diced bone of facet joints obtained during surgery from seven AS patients and seven disease control (Ct) patients. Osteoblastic activity of BdCs was assessed by measuring their alkaline phosphatase activity and by alizarin red staining. Osteoblast and endoplasmic reticulum (ER) stress-related genes were assessed by quantitative PCR, immunoblotting, immunofluorescence, and immunohistochemistry. In addition, expression of IL-23 in response to BIX (selective BIP inducer X)-induced ER stress was evaluated by qPCR and ELISA. Protein interaction and binding to IL-23 promoter were confirmed by Immunoprecipitation and Chromatin immunoprecipitation, respectively. Transcript levels of genes involved in osteoblast function, as well as of the ER stress marker were higher in the AS group than the Ct group, and elevated RUNX2, BiP and IL-23 expression were observed in the BdCs, serum, and bone biopsies from the AS group. BIX-induced ER stress stimulated osteoblastic activity and IL-23 secretion by upregulating RUNX2 expression. Furthermore, in AS BdCs, RUNX2 interacted with C/EBPβ to bind to IL-23 promoter and RUNX2 knockdown suppressed IL-23 secretion. These finding may provide a molecular mechanism involved in sustained ER stress in AS BdCs stimulates the activation of RUNX2 and C/EBPβ genes, leading to IL-23 production.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Bones and its-derived cells from patients with AS showed an increase in ER stress. </LI> <LI> IL-23 cytokine was significantly higher in AS patients than in healthy controls. </LI> <LI> Inducing ER stress in AS exhibited an increase of bone-related genes. </LI> <LI> Inducing ER stress in AS was accompanied with augmentation of IL-23 cytokine. </LI> <LI> ER stress-induced RUNX2 is involved in IL-23 secretion and bone-related genes. </LI> </UL> </P>

        Background: The serum titer of anti- Helicobacter pylori immunoglobulin G (IgG) antibody starts to increase again when reinfection occurs after a successful eradication. The aim of this study was to evaluate whether the serum pepsinogen (PG) assay is effective for the diagnosis of gastric atrophy after reinfection. Methods: This study included consecutive Korean adults who had a remote H. pylorieradication therapy, but showed positive serum anti- H. pylori IgG antibody test on the day of upper gastrointestinal endoscopy and serum PG test at our center. Advanced chronic atrophic gastritis on endoscopy was defi ned as either closed- or open-type atrophy. Serologic atrophy was defi ned as PG I/II ratio <3.0 and PG I level < 70 ng/ml. Results: Of the 131 subjects who fulfi lled the study inclusion criteria, 50 showed high bacterial loads (serum anti- H. pylori antibody titer =150 AU/ml) and 28 showed low bacterial loads (<50 AU/ml). The high bacterial load group showed highest PG levels and lowest PG I/II ratio (Table 1). Although serologic atrophy was more frequent in thehigh bacterial load group (p=0.034), this group showed the highest serum PG I levels (p=0.001). Conclusions: The severity of gastric atrophy cannot be assessed by serum PG assay after reinfection, as suggested by our fi nding that PG I/II ratio is inversely related to the PG I level. Higher bacterial load induces a larger increase in PG II level than PG I level due to severe infi ammatory reaction upon reinfection. Therefore, a decreased PG I/II ratio is no longer valuable for detecting serologic atrophy in the reinfected subjects after a successful H. pylori eradication.

        Background:There are many report about anti-infi ammatory potentials of melatonin on experimental colitis and ulcerative colitis in human. So we aimed to evaluate the anti-infi ammatory mechanism of melatonin on DSS induced colitis with and without sleep deprivation by gene expression after melatonin treatment. Methods: We used the 5 groups of C57BL/6 mice. Group I: control, Group II: 2% DSS induced colitis for 7days, Group III: 2% DSS induced colitis and melatonin (10mg/kg) treatment, Group IV: 2% DSS induced colitis with sleep deprivation(SD, 20hr/d) and Group V: 2% DSS induced colitis with SD and melatonin treatment. RNA was isolated from the colon of mice to analyze by microarray. We confi rmed signifi cant changes of expression of important genes by RT-PCR. Based on microarray result, we planned to perform immunohistochemical staining and Western blotting. Results: Sleep deprivation worsens body weight reduction of mice and exacerbate the severity of colonic infi ammation. Administration of melatonin reduced the rate of weight loss and severity of mucosa injury compared with saline injection group. 68 genes were signifi cantly changed by 2% DSS, sleep deprivation and melatonin in microarray. In RT-PCR there are signifi cant change of adipoq gene, which are related with adiponectin protein. We also confi rmed adiponectin expression by melatonin injection using immunohistochemical stain and Western blotting. Conclusions: Sleep deprivation acts as an aggravating factor, whereas melatonin acts as an improving factor of infi ammation. Besides sleep control, Melatonin has potent anti-infi ammatory effects on colonic mucosa. It is known that adiponectin also had an anti-infi ammatory effects by inhibiting Nuclear Factor-kappa B, TNF-a and IL-6 and inducing anti-infi ammatory cytokines such as IL-10, and IL-1 receptor antagonists. Adiponectin mRNA was decreased in infi amed colonic mucosa of murine colitis and restored by melatonin. This study showed melatonin directly affect mucosal healing and indirectly modulate adiponectin pathway

        Liver abscess is a pus-filled cyst in the liver. A liver abscess can also develop as a result of surgery or other trauma to the liver. Liver abscess is treated with antibiotics and PCD (Percutaneous Catheter Drainage). PCD have complications such as infection due to primary catheter placement and continued catheterization, bleeding including vessel laceration, pseudoaneurysm, and vascular fi stula formation and rarely spondylitis, intraperitoneal abscess. Here, we report a case of spondylitis and intraperitoneal abscess after PCD in liver abscess patient. A 81 years old female patient visited our hospital complaining of RUQ abdominal pain and diarrhea that started 5 days ago. She had history of angina diagnosed 10 years ago and dementia 1 years ago. A physical examination revealed RUQ tenderness. Serum laboratory tests revealed the following: white blood cells 11,890/mm³, neutrophil 91. 8%, CRP 27. 1 mg/dL, BUN/Cr 50. 6/1. 2 mg/ dL, AST 22 IU/L, ALT 34 IU/L, ALP 255 U/L, GGT 83 IU/L. Abdominal pelvic computed tomography (CT) revealed 7. 5cm multiseptated hepatic abscess in S5/6. We tried PCD for abscess drainage and applied ceftriaxone and metronidazol for antibiotic treatment. But since the abscess was multiseptated, there was no drainage of abscess after percuteneous catheter insertion. And we retried PCD for second time on 10th hospital day but there was no drainage of abscess again. After second percuteneous catheter insertion trial, liver 3 phase CT which was done on 14th hospital day revealed newly noted intraperitoneal abscess and spondylitis which we think that the complication was due to trauma to liver by PCD insertion. Left untreated, however, a liver abscess can burst and spread the infection, leading to sepsis, a life-threatening bacterial blood infection. So we had to transfer patient to the hospital that can operate and manage the complication.
