Purpose: The objective of this study was to describe the clinical phenotypes of children and adolescents with cystic fibrosis (CF); and to assess the role of pancreatic insufficiency and neonatal screening in diagnosis. Methods: A cross-sectional study was conducted, which included 77 patients attending a reference center of CF between 2014 and 2016. Epidemiological data, anthropometric measurements, and the presence of pulmonary, pancreatic, gastrointestinal and hepatobiliary manifestations were evaluated based on clinical data and complementary examinations. Results: Of the 77 patients, 51.9% were male, with a median age of 147 months (7.0-297.0 months), and the majority showed adequate nutritional status. The most common phenotype was pulmonary (92.2%), followed by pancreatic (87.0%), with pancreatic insufficiency in most cases. Gastrointestinal manifestation occurred in 46.8%, with constipation being the more common factor. Hepatobiliary disease occurred in 62.3% of patients. The group with pancreatic insufficiency was diagnosed earlier (5.0 months) when compared to the group with sufficiency (84.0 months) (p=0.01). The age of diagnosis was reduced following implementation of neonatal screening protocols for CF (6.0 months before vs. 3.0 months after, p=0.02). Conclusion: The pulmonary phenotype was the most common, although extrapulmonary manifestations were frequent and clinically relevant, and should mandate early detection and treatment. Neonatal screening for CF led to earlier diagnosis in patients with pancreatic failure, and therefore, should be adopted universally.

Prickly palm cactus husk was used as a solid-state fermentation support-substrate for production of the ligninolytic enzymes laccase, peroxide manganese, and lignin peroxidase by Aspergillus niger. Effects of water activity, temperature, and fermentation time on enzymatic production were evaluated using a central composite rotatable design. Response surface methodology revealed that maximum enzyme production was achieved at 73.38 h of fermentation, a water activity of 0.87 Aw, at 28.74oC for laccase, at 65.33 h, 0.89 Aw, and 28.96oC for lignin peroxidase, and at 70.44 h, 0.91 Aw, and 28.84oC for manganese peroxidase. Optimized enzyme production was 9,023.67 UI/L for laccase, 2,234.75 UI/L for lignin peroxidase, and 8,534.81 UI/L for manganese peroxidase. Thermostability and pH stability were observed for all enzymes. Enzymatic deactivation kinetic experiments indicated that enzymes remained active after freezing of crude extracts.

Paenibacillus larvae and Melissococcus plutonius are the primary bacterial pathogens of honeybees and the causativeagents of American and European foulbrood disease (AFB and EFB) respectively. Such diseases have beengaining importance since there are few therapeutic options beyond the reporting of microorganisms resistantto conventional antibiotics. Due to the inefficiency and/or lowefficacy of some antibiotics, researches with nanotechnologyrepresent, possibly, new therapeutic strategies. Nanostructured drugs have presented some advantagesover the conventional medicines, such as slow, gradual and controlled release, increased bioavailability,and reduced side-effects, among others. In this study, in vitro antimicrobial activity of tea tree oil (TTO) nanoparticlesagainst Paenibacillus species, including P. larvae and M. plutonius strains was evaluated. Minimal inhibitoryconcentration (MIC) in Mueller–Hinton or KSBHI broth by the microdilution method was assessed. TTO registeredMIC values of 0.18–6.25%, while the MIC values obtained for the TTO nanoparticle were of 0.01–0.93%. The possible toxic effect of TTO and TTO nanoparticle has been assessed by the spraying application method inthe concentrations higher than the MICs. Bee mortality was evident only in treatment with TTO and the TTOnanoparticles show no toxic effects after 7 days of observation. Our results showed for the first time that TTOnanoencapsulation presented a high activity against Paenibacillus species and M. plutonius strains showing thatthe use of nanotechnology may represent one alternative way for the treatment or prevention of AFB and EFB.
Atomoxetine and Fluoxetine Activate AMPK-ACC-CPT1 Pathway in Human SH-SY5Y and U-87 MG Cells

Objective Atomoxetine and fluoxetine are psychopharmacologic agents associated with loss of appetite and weight. Adenosine monophosphate-activated protein kinase (AMPK) is the cellular energy sensor that regulate metabolism and energy, being activated by fasting and inhibited by feeding in the hypothalamus. Methods Human brain cell lines (SH-SY5Y and U-87 MG cells) were used to study the outcome of atomoxetine and fluoxetine treatment in the activity of AMPK-acetyl-CoA carboxylase (ACC)- carnitine palmitoyl transferase 1 (CPT1) pathway and upstream regulation by calcium/calmodulin-dependent kinase kinase β (CaMKKβ) using immunoblotting and CPT1 enzymatic activity measures. Results Phosphorylation of AMPK and ACC increased significantly after atomoxetine and fluoxetine treatment in the first 30-60 minutes of treatment in the two cell lines. Activation of AMPK and inhibition of ACC was associated with an increase by 5-fold of mitochondrial CPT1 activity. Although the neuronal isoform CPT1C could be detected by immunoblotting, activity was not changed by the drug treatments. In addition, the increase in phospho-AMPK and phospho-ACC expression induced by atomoxetine was abolished by treatment with STO-609, a CaMKKβ inhibitor, indicating that AMPK-ACC-CPT1 pathway is activated through CaMKKβ phosphorylation. Conclusion These findings indicate that at the cellular level atomoxetine and fluoxetine treatments may activate AMPK-ACC-CPT1 pathways through CaMKKβ in human SH-SY5Y and U-87 MG cells.

To address the diversity of cyclodextrin-producing P. graminis strains isolated from wheat roots and rhizospheres of maize and sorghum sown in Australia, Brazil, and France, restriction fragment length polymorphism analysis of part of genes encoding RNA polymerase (rpoB-RFLP) and DNA gyrase subunit B (gyrB-RFLP) was used to produce genetic fingerprints. A phylogenetic tree based on rpoB gene sequences was also constructed. The isolates originated from Brazil could be separated from those from Australia and France, when data from the rpoB-based phylogenetic tree or gyrB-RFLP were considered. These analyses also allowed the separation of all P. graminis strains studied here into four clusters; one group formed by the strains GJK201 and RSA19T, second group formed by the strains MC22.02 and MC04.21, third group formed by the strains TOD61, TOD 221, TOD302, and TOD111, and forth group formed by all strains isolated from plants sown in Cerrado soil, Brazil. As this last group was formed by strains isolated from sorghum and maize sown in the same soil (Cerrado) in Brazil, our results suggest that the diversity of these P. graminis strains is more affected by the soil type than the plant from where they have been isolated.
Genoprotective Effects of the Green Tea-Derived Polyphenol/Epicatechin Gallate in C6 Astroglial

In vitro and in vivo studies have recently reported significant chemopreventive effects of green tea-derived polyphenols in different diseases. However, it remains unclear how such effects could be triggered. In order to elucidate the effects of epicatechin gallate (ECG) in C6 cells, both by itself and against H2O2-induced genotoxicity, measurements of DNA strand breaks and chromosome loss were performed. DNA damage was measured by comet and micronucleus assays. The present study shows for the first time how ECG, the major green tea-derived polyphenol, is able to exert dose-dependent genoprotective effects in an H2O2-induced toxicity model of C6 astroglial cells. We demonstrate that doses of ECG in a range from 0.1 to 1μM were able to completely prevent H2O2-induced genotoxicity in vitro. In contrast, considerably higher concentrations of ECG (10μM) were able to reverse previous positive effects in a dose- and time-dependent manner. The same results were confirmed by both comet (F3,9=336,148; P<.001) and micronucleus (F3,9=23,228; P<.001) methods. Together, our data show ECG as a dose-dependent genoprotective compound in C6 astroglial cells. This indicates that small doses of polyphenols included in our diet could have beneficial effects on neural cells, contributing to prevention of oxidative stress-associated brain pathologies. In addition, our data highlight the importance of strictly modulating doses and/or consumption of antioxidant-fortified foods or additional supplements containing such beneficial molecules.

Purpose - This paper’s aim is to analyze the technological information in patent databanks as a strategy in prospecting for new technologies. Research design, data, and methodology - We detail the major free electronic database sources for patent information, the patent documents, the patent document structures, INID codes (Internationally Agreed Numbers for the Identification of Data), indexation, references, and classification notions. Additionally, we review and analyze information on the activities of the Center of Dissemination Documentation and Technological Information (CEDIN) from the National Institute of Intellectual Property (INPI) of Brazil for the period 2000 to 2011. Results - The research shows that the technological information contained in the patents could provide a wide range of functionality within companies and universities. Conclusions - In recent years, (CEDIN), a specialist in intellectual property, has been serving internal and external users by providing guidance on the basis of patents and other literature, but the number of users served is still small. In order to familiarize more potential users of such technological information, task forces should be created among INPI, universities, and companies.
