      • 수용성 Polymer-metal Complex의 형성과 중합 개시제로서의 활용성에 관한 연구(Ⅰ)

        The formation of Water-Soluble Poly(Methacrylic acid) (PMA) and poly (acrylamide) (PAAm)-Cu(II) complexes was achieved at various PH. The structures and the optimum conditions for the formation of complexes have been determined through UV, IR and viscosity measurements. In addition, it was found that they can be used as an initiator for vinyl type monomers. Formed polymer-metal complexes are the most stable at around PH=7 and therefore give rise to maximum absorbance in UV spectrum. As PH is increased the conformation of polymer-metal complexes has become more compact. This can be explained in terms of decreasing viscosity with increasing PH. It was also observed that the function as an initiator of the polymer-metal complexes is very active at around PH=7 and the complexes can polymerize vinyl type monomers up to 10% Conversion. These results can explain the fact that the stable complexes were formed at around PH=7 and the formed complexes can be used as good initiators.

        혈소판 농축 혈장이 임플란트 주변 골형성에 미치는 효과에 대한 연구

        Various methods have been used to fill in the defects around the implants. Recently, researchs of developing and purifying various growth factors, and efforts to apply these growth factors in clinical situations have been carried out. Among those tries, platelet-rich plasma(PRP) containing abundant PDGF and TGF-β obtained by the sequestration and concentration of platelets by the gradient density centrifugation became one of the popular and preferred methods becuase of relatively simple procedure and abundant growth factor components. Bone morphogenic protein(BMP), platelet-derived growth factor(PDGF), and transforming growth factor-beta(TGF-β) are growth factors in the PRP that promote the bone formation. To elucidate the effects of PRP on the bone formation in peri-implant bone defects, twenty-seven Steri-Oss implants were inserted in the mandibular premolar areas of 8 adult dogs and artificial bone defects(3×4×4㎜) were prepared at the buccal side of the implants in 1st premolar, at the distal side of the implants in 3rd premolar. The control groups were filled with bovine bone grafts without PRP, and the experimental groups were filled with bovine bone grafts with PRP. Animals were sacrificed at 1, 2, 4, 8, and 12 weeks after the operation and speimens were observed by light and electron microscope. The results were as follows. first, the bone formation was observed at 1st week in the experimental groups, but at 2nd week in the control groups. second, the mature bone was observed at 4th week in the experimental groups. but 8th week in the control groups. the bone growth rate of experimental groups was more rapid than that of control groups. third, at 12th week, bone density ratio was higher in the experimental groups(71.65%) than that of the control groups(48.49%). fourth, Grafted bovine bone were almost completely absorbed at 1st week in experimental groups, but it was still present at 8th week in control groups. From the above results, PRP may accelerate the formation of new bone by promoting absorption of the graft material and increasing the growth rate of bone.

        문학을 통한 슬픈 공간의 창조 -이백(李白)의 추포가(秋浦歌)와 두보(杜甫)의 추흥(秋興)을 중심으로

        This study set out to analyze the spatial characteristics embodied in Song of Qiupu and Autumn Meditation and investigate how space, a background in a literary work, would be filled with what symbolic images and recreated as a unique place containing a poet``s internal sadness. The research efforts led to the following conclusions: First, there was the structure of deung-go-im-su, which implies the symbolism of parting in literary works since Nine Changes by Song Yu during the Warring States Period. In those works, it was often used as spatial composition to express a sad feeling. Second, there was another structure that placed the basic spaces of mountain and water and then included natural features with the symbolism of sorrow. They placed the monkey``s cry, and the sound of bird and hujia in the mountain and a boat, fish, and old man in the river to express a poet``s sad feeling. Third, the poet used his own figure with grey hair and white natural features. Grey hair is accompanied with despair and sad sentiments towards death and offers an image equal to melancholy. White symbolizes innocence and purity and further autumn in the Five Elements. In literature, autumn often represents solitude. In those words, a sad image is conveyed through white in the white heron, monkey, cloud, and frost. Fourth, Du Fu often used the historical images and contrasting expressions of Giju and Jangan in his Autumn Meditation. Such historical images as Begjeseng and Gonmyongji displayed the poet``s internal depression more prominently. Contrast between past and present, flamboyance and war, and center and margin in the two places expresses the poet``s yearning and despair more clearly. Fifth, the poet often moves his eyes from the upper part of space to the lower part in order to describe his loss of hope and internal despair, thus showing his depressed and solitary sensibility.

        "추(秋)"이미지와 비추(悲秋)정서의 연원 고찰 -선진시기의 문학을 중심으로

        This study examined the origin of ``sad autumn(悲秋)`` literature. Among the images of autumn, ``sad autumn`` literature focuses on the gloomy image associated with loneliness and sense of grief which dwell in the human minds. Such images create aesthetic quality which is represented in literature as a sad image. ``Sad autumn`` is one of the ten significant subject matters in Chinese classic literature. Autumn image has changed according to the etymological history of the character, 秋(autumn). From oracle born inscription to gilt lettering, the character 秋 reflected the image of burning a locust with fire hence projecting a factual wornout image. With a writing system called Sojeon, 秋 became a combination of 禾(rice plant) with 火(fire) so that the image of autumn changed into richness in Chin Era. Subsequently autumn was perceived as a romantic season of richness in Han Era based on 『Lexicon of the Origin of Chinese Characters』following ``Seolmun Haeja.`` Finally, the idea of ``sad autumn`` became apparent in 『Erya(爾雅)』annotated by Guo Pu(郭璞) in Wei Chin Era. Such changes of the image of autumn were due to the improved economy, change and expansion of the cultural spectrum. The image of 秋 in 『The Book of Odes(詩經)』 and 『Poetry of Chu(楚辭)』 corresponded to the respective feature of the former being plain realistic and the latter splendid emotional. Thus the image of autumn in 『The Book of Odes』 was realistic merely indicating the season and restrictive implying the power of writing. However a sense of belonging was apparent with autumn scenes and emotion of the author who tried to connect difficulties in life with autumn scenes. In Qu Yuan`s 「Li Sao(離騷)」 and 「Nine Pieces(九歌)」, objects including orchid were integrated with sorrowful instances of 秋 he tried to project. Hence his poems additionally offered moral images such as nobleness, integrity and respect. It was related with his subject matters such as patriotic royalty and responsibility of the ruling class. In Song Yu`s 「Nine Arguments(九辯)」, the agony of an unknown scholar was described as lonely autumn scenes, hence resonating with many other common folks. It became the landmark of autumn literature by equating the season autumn with sadness and loneliness and by transforming into aesthetic quality. Implications of the above gradual changes of the idea of ``sad autumn`` are as follows. Even if the image of ``sad autumn`` was an archetype, it manifested as a literary genre only after the accumulation of culture and an equally sophisticated author.

        Antiadipogenic Activity of Solvent-partitioned Fractions from Limonium tetragonum in 3T3-L1 Preadipocytes

        갯질경이(Limonium tetragonum)는 질경이과에 속하는 여러해살이 풀로 습지에 자생하는 염생식물의 일종이며, 항산화, 항종양 및 간보호 효능이 있는 것으로 알려져 있다. 본 연구에서는 갯질경이 추출물로부터 용매 극성에 따라 분획한 분획물(H₂O, n-BuOH, 85% aq. MeOH 및 n-Hexane)을 이용하여 지방세포내 중성지방 생성 및 지방세포 분화조절 인자 발현에 미치는 영향을 검토하였다. 마우스 유래 지방전구세포3T3-L1을 지방세포로 분화하여 Oil Red O염색법으로 지방 세포 분화정도를 확인한 결과, 갯질경이 분획물에 의해 지방세포의 형성이 농도의존적으로 억제되었다. 또한 지방생성조절에 관여하는 전사인자 PPARγ, C/EBPα 및 SREBP-1c의 발현을 mRNA와 단백질 수준에서 확인한 결과 갯질경이 분획물 처리시 지방세포 분화 인자의 발현이 유의적으로 감소하였다. 지방세포 분화에 관여하는 것으로 알려진 MAPK 신호 전달 경로를 확인한 결과 갯질경이 분획물 처리군에서 p38, ERK 및 JNK의 인산화가 억제되었다. 용매 분획물중에서 H₂O 및 n-Hexane 분획물이 가장 우수한 지방생성 억제활성을 나타내었는데 이는, 분획물 중 페놀 또는 지방 유도체에 의한 것으로 사료된다. 본 연구 결과로부터 갯질경이 분획물의 MAPK 신호전달 경로 억제를 통한 항비만 효과를 확인하였으며, 나아가 건강기능성 식품 소재로서의 개발 가능성이 기대된다. Limonium tetragonum, an edible halophyte that grows on salt marshes in Korea, is thought to possess various health benefits (e.g., antioxidant, antitumor, and hepatoprotective). In the present study, different solvent partitioned subfractions, water (H₂O), buthanol (n-BuOH), 85% aqueous methanol (85% aq. MeOH), and hexane (n-hexane), from crude extract of L. tetragonum were tested for their ability to prevent adipogenesis in differentiating 3T3-L1 preadipocytes. The treatment of differentiating 3T3-L1 preadipocytes with L. tetragonum subfractions (LTFs) resulted in suppressed adipogenesis and reduced expression of adipogenesis-related transcription factors such as peroxisome proliferator-activated receptor gamma (PPARγ), CCAATT/enhancer-binding protein alpha (C/EBPα), and sterol regulatory element- binding protein 1c (SREBP-1c) at both mRNA and protein levels. In addition, the LTF treatment notably decreased the levels of phosphorylated p38, extracellular signal-regulated kinase (ERK), and c-Jun N-terminal kinase (JNK) of the mitogen-activated protein kinase (MAPK) pathway in association with PPARγ-linked adipogenesis. Among all the tested LTFs, H₂O and n-hexane were the most effective in lowering lipid accumulation and regulating the adipocyte differentiation via PPARγ pathway. Taken together, the results indicated that the H₂O and n-hexane LTFs contain bioactive compounds that may exhibit significant antiadipogenesis activity by downregulation of the PPARγ pathway and inactivation of the MAPK signal pathway in 3T3-L1 preadipocytes.

      • A case of adult Gaucher disease improved with the enzyme replacement therapy

        Gaucher disease is an autosomal recessive, lysosomal storage disease caused by the deficiency of glucosylceramidase. Deficiency of this enzyme results in accumulation of glucoceramide in the macrophage and leads to hepatosplenomegaly, pancytopenia, bone damage and sometimes can be fatal. It is usually diagnosed in the first or second decade of life with the appearance of bone pain, splenomegaly and thrombocytopenia. There are several reports of Korean pediatric patient’s case with Gaucher disease instead ofrare adult patient. We report the case of a-40-years old man diagnosed Gaucher disease and successfully improved with the enzyme replacement therapy. A-40-years old man visited hospital with leukopenia and thrombocytopenia. On abdominal CT scan, his spleen was huge and mild hepatomegaly was shown. Initial laboratory test was perfomed including enzyme test, angiotensin-converting enzyme level was high. On bone marrow aspiration examination, there were no hematologic malignancies, just only few gaucher cellswith coarse cytoplasm and eccentric nucleus are found. Also, some collections of large cells with fibrillary cytoplasm were shown on bone marrow biopsy specimen, that suggests Gaucher disease. And he was performed hip MRI because of hip pain, there were marrow edema involving right SI joint. The patient received the enzyme replacement therapy with active enzyme using Cerezyme(imiglucerase), a recombinant form of glucosylceramidase. On enzyme replacement therapy, angiotensin-converting enzyme level was normalized, and spleen size was much decreased. His hip pain was improved with no other abnormal signal intensity on hip MRI. Gaucher disease should be considered in the differential diagnosis of adult patients with unexplained splenomegaly and bicytopenia. The early diagnosis of Gaucher disease would lead to safe and effective treatment with enzyme replacement, which can decrease morbidity.
