        Changes in prostate-specific antigen kinetics during androgen-deprivation therapy as a predictor of response to abiraterone in chemonaïve patients with metastatic castration-resistant prostate cancer

        Purpose: Metastatic castration-resistant prostate cancer (mCRPC) has a poor prognosis. Abiraterone acetate (AA), enzalutamide, and chemotherapy are first-line treatments for patients with mCRPC. This study examined prognostic factors for AA response in the form of prostate-specific antigen (PSA) kinetics throughout androgen-deprivation therapy (ADT) in chemonaïve patients with mCRPC. Materials and Methods: We retrospectively included data from 34 chemonaïve patients with mCRPC who had received AA at some point between January 2017 and December 2018. We separated patients into two study arms according to the decrease in PSA percentages after use of AA for 3 months. We correlated PSA kinetics parameters with response and compared the two study groups with respect to PSA kinetics. Results: The patients’ median age was 77 years. In the total group of patients, 64% had a response to AA, whereas 35% did not. The ratio of the PSA level at nadir to the level during ADT was significantly higher in the AA-sensitive group (19.78 vs. 1.03, p=0.019). Conclusions: Patients who experienced a dramatic change in PSA level during ADT were more likely to be resistant to AA after progression to mCRPC. Chemotherapy rather than AA might be more suitable as a first-line treatment for these patients.

      • A Feature-based Tumor Detection for MRI Breast Imaging

        In this paper, we propose a feature-based scheme composed of the intra-slice, texture and inter-slice analyses to achieve tumor region identification in MRI breast images. Our intra-slice analysis evaluates the intensity and size information of candidate regions. To find a precise region, a region growing algorithm is proposed based on ellipse fitness. In the texture analysis, some texture features are extracted and combined with a neural network to reduce the false alarms. The inter-slice analysis is based on the continuity characteristic to verify the static behavior of tumor regions. The experimental results show that our proposed scheme can correctly identify tumor regions.

        Prolyl endopeptidase remodels macrophage function as a novel transcriptional coregulator and inhibits fibrosis

        Macrophages are immune cells crucial for host defense and homeostasis maintenance, and their dysregulation is involved in multiple pathological conditions, such as liver fibrosis. The transcriptional regulation in macrophage is indispensable for fine-tuning of macrophage functions, but the details have not been fully elucidated. Prolyl endopeptidase (PREP) is a dipeptidyl peptidase with both proteolytic and non-proteolytic functions. In this study, we found that Prep knockout significantly contributed to transcriptomic alterations in quiescent and M1/M2-polarized bone marrow-derived macrophages (BMDMs), as well as aggravated fibrosis in an experimental nonalcoholic steatohepatitis (NASH) model. Mechanistically, PREP predominantly localized to the macrophage nuclei and functioned as a transcriptional coregulator. Using CUT&Tag and co-immunoprecipitation, we found that PREP was mainly distributed in active cis-regulatory genomic regions and physically interacted with the transcription factor PU.1. Among PREP-regulated downstream genes, genes encoding profibrotic cathepsin B and D were overexpressed in BMDMs and fibrotic liver tissue. Our results indicate that PREP in macrophages functions as a transcriptional coregulator that finely tunes macrophage functions, and plays a protective role against liver fibrosis pathogenesis.

        Generation of Selenoprotein with Glutathione Peroxidase Activity by Chemical Modification of the Single-chain Variable Fragment Expressed in a Single-protein Production System and Its Antioxidant Ability

        Glutathione peroxidase (GPX) is one of the important members of the antioxidant enzyme family. It can catalyze the reduction of hydroperoxides with glutathione to protect cells against oxidative damage. Single-chain variable fragment (scFv) can be converted into seleniumcontaining single-chain variable fragment (Se-scFv) by chemical modification of the hydroxyl groups in scFv, thus Se-scFv possesses GPX activity and becomes a prodrug. To improve the expression of scFv and simplify its purification steps, Single-protein production (SPP) system was used to express scFv and chemical modification was used to synthesize Se-scFv. Therefore, we must construct a new scFv-WCD1-lessACA gene, which can express its mRNA not containing any ACA sequences and express its amino acid sequence of target protein (scFv) being same to scFv-WCD1. In this way, the scFv-WCD1-lessACA can be only expressed in SPP system and no other background proteins in the cells could be expressed. The expression results showed that high level of scFv-WCD1-lessACA synthesis was at least sustained for 96 h in the virtual absence of background protein synthesis. Then, selenocysteine (Sec) was incorporated into the scFv-WCD1-lessACA by chemical modification and resulted in Se-scFv-WCD1-lessACA. The enzymatic characteristics of Se-scFv-WCD1-lessACA were determined. GPX activity was 2,563 U/μmol,its binding constant for GSH was 0.687 ×105/mol. Moreover,Se-scFv-WCD1-lessACA was confirmed to have a strong antioxidant ability to protect mitochondria against oxidative damage induced by Vc/Fe2+ (mitochondrial damage model),suggesting that Se-scFv-WCD1-lessACA has potential application for protection of mitochondrial damage induced by reactive oxygen species (ROS).

        De novo assembly and analysis of the Heortia vitessoides transcriptome via high-throughput Illumina sequencing

        Heortia vitessoides Moore, a defoliating pest in Aquilaria sinensis (Loureiro) Sprenger forests, is responsible for significant damage to the trees. There is a lack of nucleotide and protein sequence information for H. vitessoides. Hence, in this study, the transcriptome of H. vitessoides was sequenced using the Illumina Hiseq 2000 platform to evaluate the expression profiles and predict the functional genes. A total of 60,587,900 clean reads were produced and de novo assembled into 66,673 unigenes of average length 957 bp; 28,202 and 27,339 identified unigenes were annotated using the National Center for Biotechnology Information (NCBI) non-redundant (NR) database and Swiss-Prot database, respectively. All assembled unigenes were categorized into 64 biological processes, cellular components, or molecular functions using Gene Ontology (GO). In total, 24,278 unigenes were annotated using a database of eukaryotic orthologous groups (KOG) and assigned to 25 functional categories. Of these, 9172 unigenes had matches in the Kyoto Encyclopedia of Genes and Genomes (KEGG) and were assigned to 344 KEGG pathways. In addition, there were 11,670 simple sequence repeats (SSRs) in 66,673 unigenes and 64 repeated motif types, of which A/T was the most frequent. We also found that 1000 unigenes were related to insecticide resistance. To the best of our knowledge, this is the first comprehensive transcriptome analysis for H. vitessoides, providing valuable genome data sources for the study of the molecular biology of this pest

        Induced expression of three heat shock proteins mediated by thermal stress in Heortia vitessoides (Lepidoptera: Crambidae)

        To gain an insight into the function of heat shock proteins (HSPs) in insects during thermal stress, three HSP cDNAs were identified in the transcriptome of adult Heortia vitessoides, one of the most destructive defoliating pests in Aquilaria sinensis (Loureiro) Sprenger forests. The open reading frames of HvHsp60, HvHsp70, and HvHsp90 were 1,719, 2,070, and 2,151 bp in length, respectively, and encoded proteins with molecular weights of 61.05, 75.02, and 82.23 kDa, respectively. Sequence analysis revealed that all three HSPs were highly conserved in structure. Regarding the stage‐specific expression profiles, HvHsp60, HvHsp70, and HvHsp90 mRNAs were detected in all developmental stages. Regarding the tissue‐specific expression profiles, the expression levels of the three HSP genes were different in various larval and adult tissues. Moreover, the expression patterns of heat‐stressed larvae, pupae, and adults indicated that HvHsp60, HvHsp70, and HvHsp90 were heat‐inducible. In particular, HvHsp60 transcripts increased dramatically in larvae and pupae that were heat‐stressed at 40°C and were upregulated in adults that were heat‐stressed at 35°C and 40°C. The expression of HvHsp70 significantly increased in all of the three different developmental stages at 35°C, 40°C, and 45°C. The expression of HvHsp90 obviously increased at 30°C, 35°C, and 40°C in larvae and could be induced at 35°C in pupae and adults. The results suggest that HSP60, HSP70, and HSP90 play a major role in protecting H. vitessoides against high‐temperature stress.

        Development of Integrated Multi-Station System to Precisely Detect and Mitigate Surface Damage on Fused Silica Optics

        In high power laser facility, irreversible damage on fused silica optics, induced by laser irradiation or processing, seriously affects the service life of optics. Therefore, the work of inhibiting damage growth has been carried out in various countries. In our work, an integrated multi-station system is designed to detect and mitigate surface damage on fused silica. The process of processing fused silica optics include UV laser conditioning, surface damage detection and surface damage mitigation with CO2 laser. UV laser conditioning pre-initiates surface damage on fused silica optics with the laser flux less than Laser-Induced Damage Threshold (LIDT). Images of surface damage acquired from camera are processed by improved global threshold segmentation algorithm to extract damage information. Finally, CO2 laser is applied to process the damage with specific morphology to enhance the laser damage resistance. This integrated multi-station system saves the repeated optics installation time between the workstations with the positioning accuracy of 20 μm. Furthermore, the damage with diameter of 10 μm is mitigated to prolong service life of processed fused silica optics. The efficient and accurate integrated multi-station system is of great significance for off -line detecting and mitigating surface damage of fused silica optics in high power laser facility.

      • Association of Chemotherapy-induced Leucopenia with Treatment Outcomes in Advanced Non-small-cell lung Cancer Cases Receiving the NP Regimen

        Background: Chemotherapy induced leutropenia has been shown to be associated with improved treatment outcomes in selected solid tumors. We studied the association of chemotherapy induced leutropenia with treatment related outcomes in advanced non-small-cell lung cancer. Methods: This is a prospective analysis of patients receiving chemotherapy for advanced NSCLC at the Shandong Cancer Hospital from 2005-07.The chemotherapy included cisplatin $35mg/m^2$, IV on $d_{1,2}$ and vinorelbine $25mg/m^2$ IV on $d_{1,8}$ every 21 days. Patients were stratified into three groups (A) those experiencing grades 0 leucopenia, group (B) grades 1-2 and group (C) grades 3-4. The outcomes studied were response rate (RR), disease control rate (DCR), and time to progression (TTP). Results: 128 patients were studied. The RRs in groups A, B and C were 30.8%, 56.8% and 71.4%, respectively, p=0.010. The DCRs were 61.5%, 83.8% and 92.9%, respectively, p=0.009 and the median TTPs were 150 days (95%CI: 91-209), 189 days (95%CI: 181-197) and 207 days (95%CI: 172-242), p=0.009. The differences in RR and TTP were significant. In patients whose CIL kept on 10 days at least, the TTP was significantly prolonged, p=0.0213, and the same was the case for those experiencing grades 1-2 leucopenia and ECOG 0, p=0.0412. Conclusions: Occurrence of CIL correlated with RR and TTP in patients with advanced NSCLC receiving cisplatin and vinorelbine chemotherapy, especially in patients experiencing grades 1-2 leucopenia and ECOG 0, and the same for those with CIL persisting for 10 days at least. CIL could be a biological measure of drug activity and a marker of efficacy.
