        Comparative analysis of bovine maternal corpus luteum microRNAs with aberrant and normal developed cloned fetus at late gestation

        Background The development efficiency of cloned cattle is extremely low (< 5%), most of them were aborted at late gestation. Based on our previous studies, some recipient cows with a cloned fetus would present as engorged uterine vessels and enlarged umbilical vessels randomly. Abortion involves both maternal and fetal factors. Objective Our aim was to explore this phenomenon by microRNAs expression profile analysis of maternal corpus luteum (CL), which was related to pregnancy maintenance. Methods The present study provided the comparison of maternal CL miRNAs expression of abnormally and normally developed cloned bovine fetus at late gestation (~ 210 days) using RNA-Seq technology. Results We selected two abnormally pregnant cows (abnormal group, AG) and three normally pregnant cows (normal group, NG) and acquired valid reads of 9317,261–12,327,185 (~ 84.53–91.28%) from five libraries. In total, we identified 981 conserved miRNAs and 223 novel miRNAs. 1052 miRNAs were co-expressed, 124 miRNAs were uniquely expressed in AG, and 93 miRNAs were uniquely expressed in the NG. Compared with NG, 11 were significantly overexpressed, and 22 were downregulated (p < 0.05) at AG among 1052 co-expressed miRNAs. The differentially expressed miRNAs-targeted genes were further analyzed by Gene Ontology and KEGG pathway analysis. Notably, the steroid biosynthesis pathway was a significantly enriched term (p < 0.01), which may affect the secretion of progesterone. Conclusion Our research suggested that abnormal miRNAs expression of bovine maternal CL may affect the pregnant status at late gestation.

      • Locating the Change Point of Mean Residual Life of Certain Life Distributions

        A class of life distributions, whose mean residual life keeps stable at its earlier phase and then starts to decrease in time, is proposed to model the life of an element haying survived its burn-in. A strongly consistent estimator and a nonparametric testing procedure are developed to locate the occurrence of the change-point of the mean residual life. Finally, some numerical simulations are employed to be an illustration as well.

        Preload state detection for precision spindle bearings based on multi-level classification

        In this study, a multi-level state classification method based on support vector data description (SVDD) is proposed to detect bearing preload state. Firstly, a three-category classification support vector data description algorithm is proposed to establish the three-state non-aliasing hypersphere model, which can combine the kernel principal component analysis (KPCA) and membership degree. Then, by classifying the first-level training sample model of uniform and non-uniform preload states, the second-level training sample model of non-uniform preload states is established based on SVDD algorithm. Moreover, the balanced multi-label propagation classification criterion is defined that can be used to identify the preload state level based on the training sample model. Finally, a preload state detection system is developed, which can accurately simulate uniform/non-uniform preload states for spindle bearings. The experimental results demonstrate that the proposed algorithm can effectively classify the preload states of spindle bearings with average accuracy higher than 94 %.

        Uplinks Analysis and Optimization of Hybrid Vehicular Networks

        5G vehicular communication is one of key enablers in next generation intelligent transportation system (ITS), that require ultra-reliable and low latency communication (URLLC). To meet this requirement, a new hybrid vehicular network structure which supports both centralized network structure and distributed structure is proposed in this paper. Based on the proposed network structure, a new vehicular network utility model considering the latency and reliability in vehicular networks is developed based on Euclidean norm theory. Building on the Pareto improvement theory in economics, a vehicular network uplink optimization algorithm is proposed to optimize the uplink utility of vehicles on the roads. Simulation results show that the proposed scheme can significantly improve the uplink vehicular network utility in vehicular networks to meet the URLLC requirements.

        Altered Activity and Functional Connectivity of Superior Temporal Gyri in Anxiety Disorders: A Functional Magnetic Resonance Imaging Study

        Objective: The prior functional MRI studies have demonstrated significantly abnormal activity in the bilateral superior temporal gyrus (STG) of anxiety patients. The purpose of the current investigation was to determine whether the abnormal activity in these regions was related to a loss of functional connectivity between these regions. Materials and Methods: Ten healthy controls and 10 anxiety patients underwent noninvasive fMRI while actively listening to emotionally neutral words alternated by silence (Task 1) or threat-related words (Task 2). The participants were instructed to silently make a judgment of each word’s valence (i.e., unpleasant, pleasant, or neutral). A coherence analysis was applied to the functional MRI data to examine the functional connectivity between the left and the right STG, which was selected as the primary region of interest on the basis of our prior results. Results: The data demonstrated that the anxiety patients exhibited significantly increased activation in the bilateral STG than the normal controls. The functional connectivity analysis indicated that the patient group showed significantly decreased degree of connectivity between the bilateral STG during processing Task 2 compared to Task 1 (t = 2.588, p = 0.029). In addition, a significantly decreased connectivity was also observed in the patient group compared to the control group during processing Task 2 (t = 2.810, p = 0.012). Conclusion: Anxiety patients may exhibit increased activity of the STG but decreased functional connectivity between the left and right STG, which may reflect the underlying neural abnormality of anxiety disorder, and this will provide new insights into this disease.

        A positive feedback loop of heparanase/ syndecan1/nerve growth factor regulates cancer pain progression

        Background: The purpose of this research was to assess the role of heparanase (HPSE)/syndecan1 (SDC1)/nerve growth factor (NGF) on cancer pain from melanoma. Methods: The influence of HPSE on the biological function of melanoma cells and cancer pain in a mouse model was evaluated. Immunohistochemical staining was used to analyze HPSE and SDC1. HPSE, NGF, and SDC1 were detected using western blot. Inflammatory factors were detected using ELISA assay. Results: HPSE promoted melanoma cell viability, proliferation, migration, invasion, and tumor growth, as well as cancer pain, while SST0001 treatment reversed the promoting effect of HPSE. HPSE up-regulated NGF, and NGF feedback promoted HPSE. High expression of NGF reversed the inhibitory effect of HPSE down-regulation on melanoma cell phenotype deterioration, including cell viability, proliferation, migration, and invasion. SST0001 down-regulated SDC1 expression. SDC1 reversed the inhibitory effect of SST0001 on cancer pain. Conclusions: The results showed that HPSE promoted melanoma development and cancer pain by interacting with NGF/SDC1. It provides new insights to better understand the role of HPSE in melanoma and also provides a new direction for cancer pain treatment.

        Synthesis of natural glucomannan derivative as a highly-efficient green inhibitor for mild steel in the simulated seawater

        Herein, we report a rational design and synthesis of a highly-efficient and eco-friendly glucomannanderivative (GA-2) by the introduction of the thiazole and Schiff base groups into glucomannan (GA). After confirmation of GA-2 by the spectroscopic methods, it was introduced as corrosion inhibitor withexcellent anti-corrosion action for mild steel (MS) in the simulated seawater. As-obtained results revealthat GA-2 has a significantly enhanced anticorrosion performance, and its inhibitive efficiency reaches98.8% at the 0.5 mmol L1 inhibitor concentration, much higher than that of GA (69.8%) and the intermediatederivative GA-1 (81.2%). The significantly enhanced protection performance could be attributed tothe rich S heteroatoms, aromatic rings, and Schiff base groups in the GA-2, which strongly promotes thechemisorption as well as physisorption between GAD and MS. X-ray photoelectron spectroscopy (XPS)results, show that the appearance of N-Fe and S-Fe bonds confirms the strongly anchoring interactionbetween GA and MS. Furthermore, according to the quantum chemical calculations, the results fromthe lowe energy gap and radical distribution function (RDF) analyses further confirm the strongchemisorption of GA-2 at the surface of steel, which illustrates the excellent inhibition performance atthe molecule and atom level.

        Numerical investigation on seismic performance of reinforced rib-double steel plate concrete combination shear wall

        Double steel plate concrete composite shear wall (SCSW) has been widely utilized in nuclear power plants and high-rise structures, and its shear connectors have a substantial impact on the seismic performance of SCSW. Therefore, in this study, the mechanical properties of SCSW with angle stiffening ribs as shear connections were parametrically examined for the reactor containment structure of nuclear power plants. The axial compression ratio of the SCSW, the spacing of the angle stiffening rib arrangement and the thickness of the angle stiffening rib steel plate were selected as the study parameters. Four finite element models were constructed by using the finite element program named ABAQUS to verify the experimental results of our team, and 13 finite element models were established to investigate the selected three parameters. Thus, the shear capacity, deformation capacity, ductility and energy dissipation capacity of SCSW were determined. The research results show that: compared with studs, using stiffened ribs as shear connectors can significantly enhance the mechanical properties of SCSW; When the axial compression ratio is 0.3–0.4, the seismic performance of SCSW can be maximized; with the lowering of stiffener gap, the shear bearing capacity is greatly enhanced, and when the gap is lowered to a specific distance, the shear bearing capacity has no major affect; in addition, increasing the thickness of stiffeners can significantly increase the shear capacity, ductility and energy dissipation capacity of SCSW. With the rise in the thickness of angle stiffening ribs, the improvement rate of each mechanical property index slows down. Finally, the shear bearing capacity calculation formula of SCSW with angle stiffening ribs as shear connectors is derived. The average error between the theoretical calculation formula and the finite element calculation results is 8% demonstrating that the theoretical formula is reliable. This study can provide reference for the design of SCSW.

        Milling Chatter Mitigation with Projection-Based Robust Adaptive Controller and Active Magnetic Bearing

        Chatter vibration has been a critical phenomenon in milling, and results in poor surface finishes, severe tool wear and shrill noise, which are usually overcome with quite conservative cutting parameters and hence the machining efficiency is significantly affected. In order to suppress the chatter vibration and improve the machining efficiency of milling process, a milling chatter mitigation method is presented with projection-based robust adaptive controller and an active magnetic bearing (AMB) installed in the milling spindle. The AMB is utilized to apply the active force to stabilize the milling process when the originally selected cutting conditions are unstable, with which the chatter-free boundary can be enlarged. Considering the possible parameters’ uncertainties of milling system which result from the nonlinear dynamic behaviors of the spindle system and the possible saturation of actuator caused by the noise, a projection-based robust adaptive controller is designed. Simulations of active chatter mitigation with different degrees of milling system’s uncertainties are performed, and the results show that the boundary of stability lobes diagram (SLD) of milling chatter is significantly enlarged. In addition, the milling experiments are also performed with the AMB installed in a milling spindle, and the results show that the chatter vibration is exactly mitigated with the presented method in this paper.
