Emily Dickinson is one of the poets who attempted to ardently quest for their inner world. She was reared under the influence of New England tradition of moral Calvinism, and quested for her inner world through dissatisfaction with her tradition and unending doubts of human beings and God. She magnified her consciousness toward immortality and eternity in an effort to transcend mortality, while controlling her pain with a very rational and complex vision. This study examines the poems between 1861 and 1865, her most creative period, focusing on the theme of death, immortality, time and eternity. These themes are inseparably related to love, a concentric energy which combines the poles of death and immortality, and time and eternity. Emily Dickinson's existential life starts from death recognized in every moment of life. At this moment, through her magnified consciousness, time is combined with eternity, death, with immortality. And love is the concentric energy through which to drive the cycle of and combine all of the disparate things: time and eternity, death and immortality and the finite and the infinite. The fusion of the contradictions can be made through the poet's extremely magnified consciousness. The consciousness is realized in the dazzling light where time ends. The region of timelessness where time ends implies the consciousness of life which perpetually illuminates the poet's world of poetry. The finite and the infinite, death and immortality, time and eternity are combined together in this radiant consciousness. The radiance of the consciousness which can be attained through love, a concentric energy fusing the contradiction is the ultimate point toward which the poet's soul endlessly aspires.

Development and Application of Cyclodextrin Hydrolyzing Mutant Enzyme Which Hydrolyzes β- and γ-CD Selectively

<P>Cyclodextrins (CDs) are produced from starch by cyclodextrin glucanotransferase (CGTase), which has cyclization activity. Specifically, alpha-CD is an important biomolecule, as it is a molecular carrier and soluble dietary fiber used in the food industry. Upon inspection of the conserved regions of the glycoside hydrolase (GH) 13 family amylases, the amino acids K232 and H233 of CGTase were identified as playing an important role in enzyme reaction specificity. A novel CD hydrolyzing enzyme, cyclodextrin glycosyl transferase.(CGTase)-alpha, was developed using site-directed mutagenesis at these positions. Action pattern analysis using various substrates revealed that CGTase-alpha was able to hydrolyze beta- and gamma-CD, but not alpha-CD. This selective CD hydrolyzing property was employed to purify a alpha-CD from alpha-CD mixture solution. The alpha-CD that remained after treatment with CGTase-alpha and exotype glucoamylase was purified using hydrophobic interaction chromatography with 99% purity.</P>

Effect of Ovarian Extract on Oocyte Maturation and Early Embryonic Development in Pigs

Various factors in the ovary are known to regulate oocyte maturation and hormone secretory functions; however, the effect of ovarian extract (OE) on oocyte maturation and embryonic development in pigs remains unknown. In this study, we first evaluated whether OE supplementation in the in vitro maturation (IVM) medium alters the oocyte maturation capacity by affecting glucose/amino acid metabolites, meiotic maturation, cumulus cell (CC) expansion, and antioxidants. Various OE concentrations (50, 100, 200, 500, and 5000 μg/mL) were included in the IVM medium. Only the oocytes treated with 100 μg/mL OE exhibited an improved meiotic maturation rate when compared with that of the other groups (non-treated group, 78.6 ± 3.0% vs. 100 μg/mL OE-treated group, 81.6 ± 4.3%); however, the difference was not significant. To observe the changes in glucose and amino acid metabolism in the OE-treated oocytes, we measured the amounts of diverse constituents (glucose, lactate, glutamine, and ammonia) in the IVM medium containing OE. Lactate and ammonia levels in the OE-treated group after 44 h of IVM were higher (p < 0.01) than those in the non-treated group. In addition, the expression of the CC expansion factors (Has2 and Tnfaip6) significantly increased (p < 0.05), whereas the mRNA expression levels of antioxidative enzymes (Sod1, Cat, and Gpx1) significantly diminished (p < 0.05) in the OE-treated group. Moreover, mature oocytes treated with 100 μg/mL OE demonstrated increased subsequent embryonic development rates after 144 h of IVM. Thus, the addition of OE in IVM mediums may improve oocyte maturation capacity which could enhance antioxidative enzyme activation, energy metabolism, and expression of the CC expansion factors in porcine oocytes.

류성룡(柳成龍)이 재현한 아버지의 일생 -『종천영모록(終天永慕錄)』 수록 「선고행년기(先考行年紀)」의 글쓰기 방법 연구-

柳成龍은 자신의 아버지 立巖 柳仲郢의 삶을 「先考行年紀」라는 독특한 작품 속에 재현하였다. 「선고행년기」는 류성룡이 형 류운룡과 함께 찬술한 것이다. 류운룡의 주도로 1573년경 「先府君行年紀」의 1차 찬술이 완료되었고, 약 30년 뒤 류성룡은 「선부군행년기」 속 정보의 검증을 통해 아버지의 삶을 총체적으로 재구하였다. 「선고행년기」는 크게 두 가지 측면에서 류운룡의 작품과 차이가 있다. 첫째, 綱目式 연보의 형식에 품계, 날짜, 성명 등의 새로운 정보를 구체적으로 기록하여 사실성을 강화하였다. 해당 연월에 지은 詩文의 제목이나 내용을 인용하여 作品年譜의 역할을 겸하게끔 하였다. 둘째, 특정 행적 전후의 정치적 정황을 상세히 설명하고 대화체를 삽입하거나 같은 시기에 일어난 사건을 한 데 모아 유기적으로 서술함으로써 서사성을 강화하였다. 추상적 평가를 자제하고 묘사의 진실성을 강조하여 명종대의 정치적 혼란상을 날카롭게 고발하였다. 이러한 글쓰기 방법을 통해 아버지에 대한 류성룡의 존모의식과 정치적 암흑기를 견고하게 살아낸 가문의 중요 인물로서 류중영의 면모가 입증된다. Ryu Sung-Ryong(柳成龍) rebuilt his father Ryu Jung-young's(柳仲郢) life in a unique work called Chronology of Father's life(先考行年紀). Chronology of Father's life is a work created by Ryu Sung-Ryong in collaboration with his brother Ryu Woon-Ryong(柳雲龍). About 1573, the first creation of the Chronology was completed, and about thirty years later, the father's life was clarified through constant checking and verification of information. There are two major differences in the Chronology of Father's life from the works of Ryu Woon-Ryong. First, the facts were reinforced by recording specific information such as the name, date. It cited the title and contents of the literary works written in the corresponding month, and also played a role of the annals of the works. Second, the narrative was strengthened by describing the political context before and after the event in detail, inserting dialogue, or organizing events occurring at the same time organically. He refrained from the abstract evaluation and emphasized the truthfulness of the description, accusing the political confusion of King Myeongjong(明宗) sharply. Ryu Sung-Ryong's respect for his father is revealed through such writing methods. In addition, the status of Ryu Jung-young, who survived the political turmoil, has risen.

This paper deals with the complex aspects that are revealed in the process how the poet as a perceiving subject recognizes the object, focusing on her first poetry book, Notes from the Divided Country published in 2003. The poet, whose consciousness is basically rooted in the historical trauma of her divided country, Korea, tries to unify the dissociated consciousness through her own historical imagination. As Suji Kwock Kim said, she gets her poetic materials from the history of her native country, and also gets her poetic inspiration from the circumstances of her divided country. The origin of her consciousness that is derived from the divided country is closely related with the nature of dissociated consciousness of modern man. The characteristics of Suji Kwock Kim’s poetry lie in her historical imagination through which her poetic themes are extended to the universal ones of the human kind.

김종직(金宗直)의 서적 간행 활동 일고 - 지방관 재임기를 중심으로 -

본고는 占畢齋 金宗直의 정치적 이력 가운데 가장 중요한 治績을 남긴 것으로 평가되는 지방관 재임기, 그가 嶺南 지역에서 간행한 8종의 서적들을 목록화하고 그 간행 양상과 특징, 목적을 연구한 것이다. 먼저 김종직이 간행에 관여한 서적들이 문장 학습 및 科擧 준비와 일정부분 관련이 있음을 살피고, 그것이 조선전기 영남 지역의 학술사·서지사적 경향과도 일치함을 분석하였다. 김종직은 經書와 문장학습서를 중심으로 지방의 인재를 고무시키고 향촌의 문화를 유교적으로 쇄신하는 데 일조하고자 하였다. 그는 지방의 인재들이 과거를 통해 중앙 조정으로 진출하여 조선의 문명화를 진행할 것을 주문하였다. 이에 따라 보다 정확한 경전의 독서를 가능하게 하는 각종 注解書를 간행하였다. 그것은 유가의 경전에만 머물지 않고, 당송고문을 학습할 수 있는 문장선집, 예서 등에 이르렀다. 특히 주해서는 조선전기의 경전 학습방법을 유추할 수 있는 자료이기도 하다. 김종직은 자신이 간행·보급하는 서적들이 지방 인재들의 학습교재이자 조선에 유가 문명을 확산시킬 도구로 활용되기를 바랐다. 이 연구를 통해 교육자로서 김종직의 면모를 재발견하고, 조선전기 문학사의 연구사적 공백을 일정 부분 메울 수 있기를 기대한다. This article examined the publication patterns of 8 books published by Kim Jong-jik (金宗直) during his reign at the time of provincial office, which is considered to have left the most important achievement in his political career. First, Kim Jong-jik observed that the books involved in the publication were related to learning sentences and preparin g for examinations(科擧) in part, and analyzed them in line with academic and surgical historic trends in the Yeongnam region of Joseon. Kim Jong-jik wanted to promote local talent and help reform the culture of the home town, focusing on the subject and academic studies. He asked local talents to move on to central government through examinations to promote the civilization of Joseon. Accord ingly, various versions of the book were intended to enable more accurate reading of the Bible. Kim Jong-jik judged that sentences can be refined through accurate reading that can accurately understand the meaning of the scriptures. He also hoped that the books he published and distributed would be used as such teaching materials. Beyond encouragi ng local talents to enter the center, this data can be used to infer the methods of learning from the early Joseon Dynasty. Through this research, I hope that Kim Jong-jik, an educ ator, can be rediscovered and that the research vacuum in Joseon Dynasty can be filled with a certain amount.
