The propose of this thesis was to study the usage of the low-grade Vermiculite from the Chung Chong province and also to increase its Value-added. A series of experiments have been done for separation as well as heat-expansion characteristics of the Chung Chong Vermiculite some conclusions can be drawn as follows; 1. Vermiculite from Chung-Yang and Hong-Shung areas wear better than that from Palabora's mine in South Africa in terms of the usefulness for an organic fertilizer. 2. The ratio of the volume expansion of Vermiculite increased along with the temperature. 3. In general, the ratio of volume expansion of the vermiculite from Chung-Yang, Hong-Shung and Palabora's mine in South Africa increased with temperature. But it decreased temporarily in the vicinity of 850℃. The result seems to be due to temporary decrease in the latent heat as the crystal water in the crystals such as Ca_(10)(Mg,Fe)₂A1₄Si_(9)O₃₄(OH)₄, K(Fe,Mg)₃AlSi₃O_(10)(OH)₂and K (Fe,Mg)₃AlSi₃O_(10)(OH)₂evaporated. 4. Above 1050℃, the crystal structure began to change and resulted in partial destruction to produce some dusts. Finally sintering occurred.

To improve insecticidal activity of B. thuringiensis 2385-1 (Bt 2385-1), a recombinant plasmid, pHT1K-1Ac, was introduced into lepidopteran toxic Bt 2385-1 by electroporation. The presence of the recombinant plasmid in Bt 2385-1 after electroporation was confirmed by PCR. Bt 2385-1 transformant was named as Bt pHT1K-1Ac/2385-1 (1K-1Ac/2385-1). The 1K-1Ac/2385-1 transformant produced bipyramidal-shaped parasporal inclusion as like the wild-type strain, Bt 2385-1, and showed an 130 kDa band of Cry1Ac protein. The insecticidal activity of 1K-lAc/2385-1 against S. exigua was similar to that of Bt 2385-1 but the $LC_{50}$ value of transformant against P. xylostella was 1.8 times lower. Through these bioassay results, it was confirmed that toxicity of Bt 2385-1 transformant showed synergistic effect by introducing Cry1Ac. These results suggested that the multiple expressions of Cry proteins in a promising Bt strain may interact synergistically in insect midgut, resulting in increase of toxicity and expansion of host spectrum.

<P>We report a two-photon fluorescent probe (AHg1) that can be excited by 780 nm femto-second pulses, shows high photostability and negligible toxicity, and can visualize the site of Hg<SUP>2+</SUP> accumulation, but can also estimate trace amounts of mercury ions in fresh fish organs by two-photon microscopy.</P> <P>Graphic Abstract</P><P>We report a two-photon probe (AHg1) that can selectively detect Hg<SUP>2+</SUP> in the live cells and fish organs at 80–150 μm depth without interference from other metal ions and the membrane-bound probes. <IMG SRC='http://pubs.rsc.org/services/images/RSCpubs.ePlatform.Service.FreeContent.ImageService.svc/ImageService/image/GA?id=b922305c'> </P>

The E. coli-B. thuringiensis shuttle vector for expression of cry1Ac, pHT1K-1Ac plasmid was introduced into acrystalliferous B. thuringiensis Cry-B and Spodoptera toxic STB-3 strain. The presence of a recombinant plasmid in transformants after electroporation was confirmed by PCR. The 1K-1Ac/Cry-B (Cry-B transformant) and 1K-1Ac/STB-3 (STB-3 transformant) produced bipyramidal-shaped parasporal inclusion that was 130 kDa in size as like B. thuringiensis subsp. kurstaki HD-73. In P. xylostella bioassay, these transformants showed significantly high toxicity than the wild-type recipients and further, in case of B. thuringiensis STB-3 transformant still had original Spodoptera toxicity. These results suggested that the pHT1K could be successfully applied for generating individual B. thuringiensis strains that produce various combinations of insecticidal proteins to expand their host spectrum and enhance insecticidal activity.

In order to improve the transformation efficiency of the Bacillus thuringiensis (Bt)-Escherichia coli (E. coli) shuttle vector, pHT3101, we intended to minimize replication origin of Bt in pHT3101. For this, two modified shuttle vectors, pHT1K and pHT261, in which 2.9 kb of replication origin of Bt were shortened to 1 kb and 261 bp, respectively as previously reported. Whereas the pHT1K could efficiently transform Bt into the antibiotic resistant, no transformants were obtained with pHT261. Furthermore, pHT1K showed higher transformation efficiency compared to that of parent vector, pHT3101. Therefore, pHT1K might be a very useful Bt-E. coli shuttle vector carrying minimal replication origin of Bt.

The development of Korean Oriental medicine is based upon the accumulation of experience and knowledge gathered over the centuries. The approaches taken are holistic and empirical. There is a need to understand their actions at molecular levels with more rational, objective and scientific studies. Today it appears that Chronic and age-associated diceases may be multifactorial and hence more complex. A different approach may be required. One claimed usage of Korean Oriental medicine is for the treatment and prevention of chronic and age-associated illnesses. Some of the botanical formulas used for this purpose were discovered thousands of years ago and continue to be used today. There are indications that these formulas may indeed be helpful in the treatment or prevention of chronic diseases. This multi-component medicine could not only be very useful meeting the unmet clinical needs but for defining a more synergistic therapy that supports and maintains the bodies natural curative abilities. The potential usefulness of Korean Oriental medicine embodies the belief of maintaining healthy homeostasis of the body through the proper balance of a mixture of chemical at different organs or tissues. This concept is different from western medicine and implies that multiple compounds may act on multiple mechanisms of action to maintain the balance of the complex web of biology. This is very important in view of sciences current direction to integrate fragmented information to develop future medicines. The western and eastern approaches to human health and disease are complementary to each other. The best approach in developing future medicines is to integrate both approaches.
