Identification of a sensitive urinary biomarker, selenium-binding protein 1, for early detection of acute kidney injury

<P>Acute kidney injury (AKI) is associated with increased mortality rate in patients but clinically available biomarkers for disease detection are currently not available. Recently, a new biomarker, selenium-binding protein 1 (SBP1), was identified for detection of nephrotoxicity using proteomic analysis. The aim of this study was to assess the sensitivity of urinary SBP1 levels as an early detection of AKI using animal models such as cisplatin or ischemia/reperfusion (I/R). Sprague-Dawley rats were injected with cisplatin (6 mg/kg, once i.p.) and sacrificed at 1, 3, or 5 days after treatment. Ischemia was achieved by bilaterally occluding both kidneys with a microvascular clamp for 45 min and verified visually by a change in tissue color. After post-reperfusion, urine samples were collected at 9, 24, and 48 hr intervals. Urinary excretion of protein-based biomarkers was measured by Western blot analysis. In cisplatin-treated rats, mild histopathologic alterations were noted at day 1 which became severe at day 3. Blood urea nitrogen (BUN) and serum creatinine (SCr) levels were significantly increased at day 3. Levels of urinary excretion of SBP1, neutrophil gelatinase-associated lipocalin (NGAL), and a tissue inhibitor of metalloproteinase-1 (TIMP-1) were markedly elevated at day 3 and 5 following drug treatment. In the vehicle-treated I/R group, serum levels of BUN and SCr and AST activity were significantly increased compared to sham. Urinary excretion of SBP1 and NGAL rose markedly following I/R. The urinary levels of SBP1, NGAL, TIMP-1, and KIM-1 proteins excreted by AKI patients and normal subjects were compared. Among these proteins, a marked rise in SBP1 was observed in urine of patients with AKI compared to normal subjects. Based upon receiver-operator curves (ROC), SBP1 displayed a higher area under the curve (AUC) scores than levels of SCr, BUN, total protein, and glucose. In particular, SBP1 protein was readily detected in small amounts of urine without purification. Data thus indicate that urinary excretion of SBP1 may be useful as a reliable biomarker for early diagnosis of AKI in patients.</P>

Erythrophagocytosis of Lead-Exposed Erythrocytes by Renal Tubular Cells: Possible Role in Lead-Induced Nephrotoxicity

<P>Background: Nephrotoxicity associated with lead poisoning has been frequently reported in epidemiological studies, but the underlying mechanisms have not been fully described.</P><P>Objectives: We examined the role of erythrocytes, one of the major lead reservoirs, in lead-associated nephrotoxicity.</P><P>Methods and results: Co-incubation of lead-exposed human erythrocytes with HK-2 human renal proximal tubular cells resulted in renal tubular cytotoxicity, suggesting a role of erythrocytes in lead-induced nephrotoxicity. Morphological and flow cytometric analyses revealed that HK-2 cells actively phagocytized lead-exposed erythrocytes, which was associated with phosphatidylserine (PS) externalization on the erythrocyte membrane and generation of PS-bearing microvesicles. Increased oxidative stress and up-regulation of nephrotoxic biomarkers, such as NGAL, were observed in HK-2 cells undergoing erythrophagocytosis. Moreover, TGF-β, a marker of fibrosis, was also significantly up-regulated. We examined the significance of erythrophagocytosis in lead-induced nephrotoxicity in rats exposed to lead via drinking water for 12 weeks. We observed iron deposition and generation of oxidative stress in renal tissues of lead-exposed rats, as well as the histopathological alterations such as tubulointerstitial lesions, fibrosis, and up-regulation of KIM-1, NGAL, and TGF-β.</P><P>Conclusions: Our data strongly suggest that erythrophagocytosis and subsequent iron deposition in renal tubular cells could significantly enhance nephrotoxicity following lead exposure, providing insight on lead-associated kidney damages.</P><P>Citation: Kwon SY, Bae ON, Noh JY, Kim K, Kang S, Shin YJ, Lim KM, Chung JH. 2015. Natural or man-made disasters are serious enough to paralyse the functions of the nation or regional distinct and affect the property and lives of numerous citizens. The first two hours after occurrence of the disaster are critical for determining death or permanent disabilities of the casualties. The current Disaster Medical Service(DMS) System in our country adopts mainly civil defense model which focuses the cause of the accident, compensation of the victims, or reconstruction. So in the initial phase of disaster, the appropriate rescue and emergency treatment is not provided effectively. In order to assess and reorganize the current status of DMS System in Korea, the Department of Emergency Medicine, Ajou University School of Medicine, planned and conducted an emergency response exercise in Suwon city on April 28, 1995. The exercise, which took place near the Kyunggido Culture and Art Center, coordinated the efforts of the Suwon Fire Department and other related agencies. As well as providing training on emergency response, the exercise was valuable for identifying areas of weakness which will be address in the future.

Unity 3D를 이용한 3D어드벤처 퍼즐 게임

This is configured in a 3D adventure puzzle game game , add a story to the fact that can be somewhat boring in the puzzle game , it has given the features that enhance the driving particle size in the game . Map that was produced directly using the 3DS Max uses the Unity 3D engine to produce a dark atmosphere to become the story , was implemented in accordance with the terrain engine and animation in the game . But is not high degree of freedom because the adventure game , since it is possible to know whether or not what to do but rather clearly , It increases even more immersive degree. 3D 어드벤처 퍼즐게임으로 구성된 이 게임은 퍼즐게임에서의 다소 지루할 수 있는 것에 스토리를 추가하여 게임에 몰입도를 높이는 특징을 주고 있다. 3DS Max를 사용하여 직접 제작한 맵은 스토리와 어울리게 어두운 분위기를 연출하고 Unity 3D엔진을 이용해서 지형엔진과 애니메이션을 게임에 맞게 구현하였다. 어드벤처 게임이다 보니 자유도가 높지는 않지만 오히려 뚜렷하게 무엇을 해야 하는지 알 수 있기 때문에 더욱 더 몰입도를 높여준다.

For the photometric determination of silicon in the iron(Ⅲ) chloride, the iron(Ⅲ) was extracted to organic phase with Amberlite LA-2 in the cons. HCl solution. In the residual aqueous solution, the silicon was determined by molybdenum blue method. The method was found to be very useful to determine a low concentration of silicon in the concentrated iron sample.

단속절삭에서 AE신호를 이용한 공구마멸의 감시

Characteristics of AE(Acoustic Emission) signal is related to cutting conditions, tool materials, and tool geometry in metal cutting. Relation between AE signal and tool wear was investigated experimentally. Experiment is carried out by interrupted cutting for SCM420 workpiece with TiN coating tool on HSS material. AE RMS voltage and count per event were increased according to tool wear. The major results are as follows : 1) AE RMS value is nearly constant as cutting speed changes, but is rapidly increase as feed rate increases. 2) AE RMS value and Count per Event increase as tool wear increases. 3) It is more effective to monitor tool wear by Incremental rate of AE RMA value than by Incremental rate of count per event.

感冒辨證에서 희수식 맥진기를 이용한 肺脈觀察의 臨床的 意義

By useing hisusik-makjingi(희수식맥진기) we analysed lung-pulse of common cold patient. The result as fellow ; 1. In case of wind-cold common cold patient, It is short-avove pulse type and when cold transfer to fever it is dry-cold type pulse. 2. In a case of wind-heat, there is a pulse of above-frequent or frequent-stringed. And also we can find out that in a mild fever case there is not a frequent pulse meaning the heat. 3. In case of vutal energy shortage and yabggi deficiency, we can also notice that the pulse is so powerless. 4. The patient who have had an old complaint and a serious illness didn't show the accordance with the pulse and common cold type.

낙상으로 인한 좌심실 파열 후 생존한 환자 1례

Blunt or nonpenetrating chest injuries are common after motor vehicle accidents or falls. Blunt injury to the heart ranges from contusion to rupture. Cardiac rupture, however, is un-common and associated with a very high mortality rate ; left ventricular rupture due to falling is even more rare. We experienced a case of left ventricular rupture of the heart caused by a fall, which the patient survived. A 63-year-old man fell off a ladder and presented with hypotension from left hemothorax and pericardial tamponade. The hemothorax was relieved via an emergency closed thoracostomy but dyspnea and hypotension continued. And so, echocardio-graphy was done and pericardial tamponade was detected. Emergency left thoracotomy was done to repair the rupture site. We believe that patients with cardiac rupture who reach the hospital alive can often be saved by prompt diagnosis and immediate surgical treatment.

Roxythromycin-tylosin-furaltadone 합제의 닭 유래 세균에 대한 시험관내 항균효과

In vitro antimicrobial susceptibility of roxythromycin (Rx), tylosin (Tyl), erythromycin (EM) and furaltadone (Fur) was studied, either by the single type or the combinated type of Rx-Tyl-Fur (RTF) against E. co1i (30 strains), Staph. aureus (20 strains) and Mycoplasma gallisepticum (5 strains) that are the major pathogens in poultry farm. The paper disk diffusion and MIC tests were applied. In screening test by paper disk diffusion method, the single types of antibiotics showed the sensitivity against 55 strains of bacteria in order of Fur, EM, Rx and Tyl. The combinated RTF showed the higher rate of sensitivity (80∼83.3% at 64㎍/disk, 60-75% at 32㎍/disk) and wider range of growth inhibiting zone (19.6±3.7-24.8±4.7mm) than those of the single drugs. In MIC test, the sensitivity against the 55 strains appeared in order of Fur. EM, Rx and Tyl as similar to those of paper disk diffusion method. The MIC ranges of RTF were 0.125∼16㎍/ml for E. coli, 0.25∼16㎍/ml for Staph. aureus and 1-8㎍/ml for Myco. gallisepticum, showing evidently lower than those of the single types. The MIC_(50) and MIC_(90) of RTF were estimated l∼4㎍/ml and 8∼16㎍/ml, respectively, and remarkably lower than those of the single drugs.
