<P>The figleaf gourd (<I>Cucurbita ficifolia</I> Bouché) root system has the ability to take up water and nutrients at low soil temperatures, and in the present paper, we attempt to reveal some of the molecular mechanisms behind this low-temperature tolerance. Exposure of figleaf gourd root system to low temperature induced accumulation of H<SUB>2</SUB>O<SUB>2</SUB> along the plasma membrane but not in the cytoplasm. H<SUP>+</SUP>-ATPase (EC 3.6.1.35) activity of isolated root plasma membranes and root hydraulic conductivity (<I>Lp<SUB>r</SUB></I>) were largely insensitive to externally applied H<SUB>2</SUB>O<SUB>2</SUB>. However, using bromocresol purple, it was shown that the acidification of the medium surrounding the root was strongly inhibited with low temperature- and H<SUB>2</SUB>O<SUB>2</SUB>-treated roots. Addition of catalase (EC 1.11.1.6) to the root medium during low-temperature exposure led to a recovery of H<SUP>+</SUP>-efflux along the root surface and increased <I>Lp<SUB>r</SUB></I>, demonstrating the importance of an H<SUB>2</SUB>O<SUB>2</SUB> detoxification system in the root cells. Additional evidence for an increased <I>Lp<SUB>r</SUB></I> was obtained by the Fenton reaction wherein a warming of the solution increased the activity of the detoxification system. All available evidence suggests that the ability of figleaf gourd root system to maintain a low level of H<SUB>2</SUB>O<SUB>2</SUB> in the cytoplasm and to detoxify reactive oxygen species is related to the maintenance of water transport activity at low temperatures.</P>
Water and H₂O₂ Permeability of Aquaporins Isolated from Cucumber and Figleaf Gourd

Cucumber aquaporin gene, CsPIP1;1 and figleaf gourd aquaporin gene CfPIP2;1 were injected to Xenopus oocytes in order to study water transport activity. While CsPIP1;1 did not transport any water and was not responsive to mercury, oocyte injected with CfPIP2;1 rapidly increased their volume without saturation indicating active water transport. Calculation of water permeability of oocytes clearly distinguished the two genes. In response to exogenous application of H₂O₂, only CfPIP2;1 responded, reducing osmotic water permeability. Transgenic Arabidopsis plants overexpressing aquaporin genes did not respond to applied H₂O₂.

Background: Melanocyte synthesize melanin within melanosomes which are transferred to the surrounding keratinocytes and can be produced in varying sizes, numbers and densities. Skin whitening products have become increasingly popular. EP-2 is a new melanogenesis inhibitor isolated from yeast. Objectives: In this work, EP-2, a melanogenesis inhibitor isolated from yeast was studied to understand its mechanism of melanogenesis inhibition. Methods: EP-2 used in this research was isolated using gel and reverse phase chromatographies from ethyl acetate fraction of ethanolic extract of Saccharomyces cerevisiae. We assessed melanin contents and tyrosinase activity of B16F10 melanoma cell using absorbance and western blot analysis. And expression of PAR-2 was also assessed by western blot analysis. Results: When B16F10 melanoma cell was stimulated with α -MSH, melanin production and tyrosinase activity were increased and cell morphology was dramatically changed to have many dendrites on the cell membrane surface. However, when EP-2 was added, melanin production and tyrosinase activity were inhibited in a dose-dependent manner and cell morphology was not changed in dendrites. Treatment with EP-2 also revealed decreased level of PAR-2 in Mel-Ab melanocytes and HaCat keratinocytes co-culture. Conclusion: EP-2 has an inhibitory effect on melanogenesis and its mechanism is related to the melanin synthesis, tyrosinase activity and transfer of melanosome to keratinocyte.

Background/Aims: The long-term course of Crohn’s disease (CD) has never been evaluated in non-Caucasian population-based cohorts. The aim of the present study was to evaluate the longterm prognosis of Korean CD patients in the well-defined population-based Songpa-Kangdong inflammatory bowel disease cohort. Methods: Outcomes of disease and their predictors were evaluated for 418 patients diagnosed with CD between 1986 and 2015. Results: During a median of 123 months, systemic corticosteroids, thiopurines, and anti-tumor necrosis factor (TNF) agents were administered to 58.6%, 81.3%, and 37.1% of patients, respectively. Over time, the cumulative probability of starting corticosteroids significantly decreased (p=0.001), whereas that of starting thiopurines and anti-TNFs significantly increased (both p<0.001). The cumulative probability of behavioral progression was 54.5% at 20 years, and it significantly decreased during the anti-TNF era. Intestinal resection was required for 113 patients (27.0%). The cumulative probabilities of intestinal resection at 1, 5, 10, 20, and 25 years after CD diagnosis were 12.7%, 16.5%, 23.8%, 45.1%, and 51.2%, respectively. Multivariable Cox regression analysis identified stricturing behavior at diagnosis (adjusted hazard ratio [aHR], 2.70; 95% confidence interval [CI], 1.55 to 4.71), penetrating behavior at diagnosis (aHR, 11.15; 95% CI, 6.91 to 17.97), and diagnosis of CD during the anti-TNF era (aHR, 0.51; 95% CI, 0.35 to 0.76) as independently associated with intestinal resection. The standardized mortality ratio among CD patients was 1.36 (95% CI, 0.59 to 2.68). Conclusions: The long-term prognosis of Korean patients with CD is at least as good as that of Western CD patients, as indicated by the low intestinal resection rate. Moreover, behavioral progression and intestinal resection rates have decreased over the past 3 decades.

Cough is one of the most common symptom of many respiratory diseases. The Korean Academy of Tuberculosis and Respiratory Diseases organized cough guideline committee and cough guideline was developed by this committee. The purpose of this guideline is to help clinicians to diagnose correctly and treat efficiently patients with cough. In this article, we have stated recommendation and summary of Korean cough guideline. We also provided algorithm for acute, subacute, and chronic cough. For chronic cough, upper airway cough syndrome (UACS), cough variant asthma (CVA), and gastroesophageal reflux disease (GERD) should be considered. If UACS is suspicious, first generation anti-histamine and nasal decongestant can be used empirically. In CVA, inhaled corticosteroid is recommended in order to improve cough. In GERD, proton pump inhibitor is recommended in order to improve cough. Chronic bronchitis, bronchiectasis, bronchiolitis, lung cancer, aspiration, angiotensin converting enzyme inhibitor, habit, psychogenic cough, interstitial lung disease, environmental and occupational factor, tuberculosis, obstructive sleep apnea, peritoneal dialysis, and idiopathic cough can be also considered as cause of chronic cough. Level of evidence for treatment is mostly low. Thus, in this guideline, many recommendations are based on expert opinion. Further study regarding treatment for cough is mandatory.
