<P>Ordered morphologies in disulfonated poly(arylene sulfide sulfone nitrile) (SPSN) copolymers were generated via thermal annealing followed by multiblock copolymer synthesis. While SPSN random copolymers (R-SPSN) showed featureless morphologies, the SPSN multiblock copolymers (B-SPSN) exhibited cocontinuous lamellar morphologies with a center-to-center interdomain size of up to 40 nm. In spite of the well-ordered, interconnected hydrophilic domains, the water self-diffusion coefficient (e.g., <I>D</I> = (0.7–2.0) × 10–10 m2 s–1) and proton conductivity (e.g., σ = 0.16–0.20 S cm–1 in deionized water at 30 °C) through B-SPSN were lower than those of the corresponding R-SPSN (e.g., <I>D</I> = (3.5–3.9) × 10–10 m2 s–1 and σ = 0.21 S cm–1) due to the relatively lower water uptake of the B-SPSN after thermal annealing. The reduced water uptake of B-SPSN was beneficial to reduction of peroxide degradation rate. Thermal annealing produced significant gains in morphological ordering and finer control over desired membrane properties for proton conduction applications.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/mamobx/2013/mamobx.2013.46.issue-19/ma400889t/production/images/medium/ma-2013-00889t_0010.gif'></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/ma400889t'>ACS Electronic Supporting Info</A></P>

Next generation sequencing technologies have accelerated the rate at which whole genome sequencing (WGS) data is acquired. The sheer volume of data generated by WGS requires computational annotation to define potential coding regions and chromosomal features. The accuracy of genomic annotation is thus limited by the power of the computational algorithm and the sequence coverage provided by the raw data. Sequencing of the New Zealand White (NZW) rabbit has been performed to a 7× depth of coverage, leaving large gaps in coverage and potential errors within the draft assembly. In the present study, we have resequenced the collagen type I, alpha 1 (Col1A1) gene of Oryctolagus cuniculus (n = 8). We have characterized the full length cDNA, identified splicing errors within the reference sequence, and identified single nucleotide polymorphisms within the gene. These data underscore the need for a higher resolution assembly of the rabbit genome to advance research in this important large animal model.

Background In cases of total knee arthroplasty (TKA) threatened by potential hardware exposure, flap-based reconstruction is indicated to provide durable coverage. Historically, muscle flaps were favored as they provide vascular tissue to an infected wound bed. However, data comparing the performance of muscle versus fasciocutaneous flaps are limited and reflect a lack of consensus regarding the optimal management of these wounds. The aim of this study was to compare the outcomes of muscle versus fasciocutaneous flaps following the salvage of compromised TKA. Methods A systematic search and meta-analysis were performed to identify patients with TKA who underwent either pedicled muscle or fasciocutaneous flap coverage of periprosthetic knee defects. Studies evaluating implant/limb salvage rates, ambulatory function, complications, and donor-site morbidity were included in the comparative analysis. Results A total of 18 articles, corresponding to 172 flaps (119 muscle flaps and 53 fasciocutaneous flaps) were reviewed. Rates of implant salvage (88.8% vs. 90.1%, P=0.05) and limb salvage (89.8% vs. 100%, P=0.14) were comparable in each cohort. While overall complication rates were similar (47.3% vs. 44%, P=0.78), the rates of persistent infection (16.4% vs. 0%, P=0.14) and recurrent infection (9.1% vs. 4%, P=0.94) tended to be higher in the muscle flap cohort. Notably, functional outcomes and ambulation rates were sparingly reported. Conclusions Rates of limb and prosthetic salvage were comparable following muscle or fasciocutaneous flap coverage of compromised TKA. The functional morbidity associated with muscle flap harvest, however, may support the use of fasciocutaneous flaps for coverage of these defects, particularly in young patients and/or high-performance athletes.

Unstitching the Nanoscopic Mystery of Zeolite Crystal Formation

<P>A molecular-scale understanding of crystal growth is critical to the development of important materials such as pharmaceuticals, semiconductors and catalysts. Only recently has this been possible with the advent of atomic force microscopy that permits observation of nanoscopic features on solid surfaces under a liquid or solution environment. This allows <I>in situ</I> measurement of important chemical transformations such as crystal growth and dissolution. Further, the microscope can access not only an accurate height measurement of surface topography, important to deduce structural elements, but also the forces involved during nanoscopic processes. We have discovered that it is possible to use these features to “illuminate” critical nanoscopic chemical events at crystal surfaces and at the same time extract the associated energies and unstitch the details of the stepwise mechanism of growth and dissolution. This approach has been developed using nanoporous crystals of the heterogeneous catalyst zeolite L; however, in principle the approach could be adapted to many crystal growth problems.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/jacsat/2010/jacsat.2010.132.issue-39/ja105593v/production/images/medium/ja-2010-05593v_0015.gif'></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/ja105593v'>ACS Electronic Supporting Info</A></P>
