      • The Impact of Soil Available Phosphorus on the Intensity of the Mycorrhizal Colonization of Root at Different Patches in Northern Steppe of China

        The research was conducted at a degraded steppe in Saibei Adiministrative Region of ZhangJiakou in Hebei province. Four patches dominated by fringed sagebrush (Artemisia frigida), narrowleaf stellera (Stellera chamaejasme), shining speargrass (Achnatherum splendens), white swordflag (Iris lactea), respectively, were sampled. Phosphorus (P) was applied at four levels (0, 5, 10 and 15㎎ ㎏?¹ soil) in each patch. The mycorrhizal infection rate in Stellera chamaejasme species and Achnatherum splendens species was low, differed from other two patches of Artemisia frigid and Iris lactea.

        Epigenetic inactivation of ACAT1 promotes epithelial-mesenchymal transition of clear cell renal cell carcinoma

        Background: Acetyl-CoA acyltransferase 1 (ACAT1) is a key enzyme catalyzing the production of mitochondrial ketone bodies. We have shown that ACAT1 is down-regulated in kidney renal clear cell carcinoma (KIRC) previously. Objective: To investigate the reasons for downregulation of ACAT1 in KIRC and explore the underlying mechanisms involved in metastatic inhibition regulated by ACAT1. Methods: The Gene Expression Omnibus (GEO) database was queried for meta-analysis of ACAT1 mRNA expression in KIRC. The UALCAN website was used to compare the methylation levels of the ACAT1 promoter region in KIRC and normal tissues. RT-qPCR was used to quantitate ACAT1 transcription levels. The GCBI and Tarbase V.8 databases were used to predict miRNAs that may target the mRNA of ACAT1. The correlation between mRNA expression of ACAT1, MMP7 (matrix metallopeptidase 7), CDH1 (E-cadherin), EpCAM (epithelial cell adhesion molecule), and VIM (vimentin) was analyzed. Extracellular MMP7 protein was quantitated using an ELISA assay. Results: The methylation level of the ACAT1 promoter region in KIRC was significantly higher than that in the normal kidney tissues. The ACAT1 mRNA expression in the KIRC cell lines was restored after treatment with 5-aza-dC (p < 0.05). MiR-21-5p is a conserved microRNA targeting ACAT1. It is expressed at a significantly higher level in KIRC than in normal tissues (p < 0.001). MiR-21-5p miRNA expression negatively correlates with ACAT1 mRNA expression. The expression of miR-21-5p is higher at the T3-T4 stages and in the histologic grades G3-G4. Patients with high miR-21-5p expression tended to have lower overall survival, suggesting that miR-21-5p could serve as a potentially valuable diagnostic biomarker for KIRC (AUC = 0.957; p < 0.001). A mimetic of miR-21-5p inhibited the expression of ACAT1 mRNA and protein. In addition, ACAT1 mRNA expression positively correlates with CDH1 and EpCAM but is negatively correlated with VIM. Overexpression of ACAT1 suppresses the secretion of MMP7 in KIRC cells. Conclusion: Expression of ACAT1 in KIRC is controlled at two levels, firstly by the hypermethylation of the ACAT1 promoter region and secondly by overexpression of miR-21-5p. Downregulation of ACAT1 expression correlates with epithelial-mesenchymal transition (EMT).

        Optimal Duty Cycle Method to Suppress the Commutation Torque Ripple of Brushless DC Motor in Braking Mode

        This paper proposes an optimal duty cycle method to suppress the commutation torque ripple under the braking condition of the brushless DC motor (BLDCM). The commutation torque will also fl uctuate in the braking process, but the current loop and change trend are diff erent from that of electric operation. Therefore, it is necessary to study the commutation torque ripple under braking. To directly analyze the interfering factors of commutation torque ripple, the formulas of commutation torque ripple under the standard brake modulation mode are derived in this paper. Based on the mathematical analysis, an optimized duty cycle method is proposed to suppress the commutation torque ripple by dynamically adjusting the duty cycle during commutation period to make the target formula of torque ripple zero. The simulation and experimental results show that this method can eff ectively reduce the commutation torque ripple, and it is helpful for the promotion of the braking performance and the scope of application of the brushless DC motor

        The mutation of Scenedesmus obliquus grown in municipal wastewater by laser combined with ultraviolet

        Mutagenetic breeding is an efficient technique for the enhancement of lipid productivity from microalgae. In this study, oil-rich microalga Scenedesmus obliquus were treated by Laser-UV composite mutagenesis. Among the 35 mutant strains, X5 was primely screened. Afterwards, a twice UV mutagenizing was operated on X5, and the optimal mutant strain X5-H13 was obtained. The growth rate, dry weight, lipid yield and lipid content of X5-H13 were 0.698× 107 cells/mL·d, 0.99 g/L, 0.49 g/L and 48.8% while cultivated in municipal wastewater, respectively, which were increased by 45%, 58%, 109% and 32% than the original strain. The results of the subculture of repeated mutant showed that the biomass and lipid content of strain X5-H13 were up to 0.99 g/L and 48.8%. The growth of each generation was stable. Furthermore, the random amplified polymorphic DNA analysis indicated that the mutant strain X5-H13 was different from the starting strain, with their genetic similarity coefficient value of 0.815.

        Effects of particle size of zero-valent iron (ZVI) on peroxydisulfate-ZVI enhanced sludge dewaterability

        The advanced oxidization process has proven to be an effective conditioning technique for the improvement of sludge dewaterability. Zero-valent iron (ZVI) is often used as the catalyst of the oxidization process. This study applied ZVI with different particle sizes to the ZVI- peroxydisulfate reactions, and investigated their effects on the improvement of sludge dewaterability. It was found that ZVI particles with smaller sizes (100 and 400 meshes) led to slightly higher enhancement of sludge dewaterability (69.1%-72%) than the larger size particles (20-40 meshes) with the reduction rate of CST by 64%. However, after the treatment, the recycle rate of larger size ZVI particles was obviously higher than the small sizes ZVI particles: 98.3% vs. 87.6-89.7%. Different surface areas of the ZVI particles with different sizes might contribute to the phenomenon. For the small ZVI particles with the sizes of 100 and 400 meshes, no obvious differences of oxidization effects and the improvements of sludge dewaterability were found between them, which might be because an oxide layer could have been formed on the surface of fine ZVI particles and led to agglomeration. According to the economical analysis, the small particles (100 and 400 meshes) of ZVI were more economically favorable for the oxidative conditioning process with ZVI-peroxydisulfate than large ZVI particles (20-40 meshes).

        Prevalence and Risk Factors of Cerebral Small Vessel Disease in a Chinese Population-Based Sample

        Background and Purpose Epidemiological data of cerebral small vessel disease (CSVD) in the general population of China are lacking. We report on the prevalence of lacunes, white matter hyperintensity (WMH), and cerebral microbleeds (CMBs) in a community-based sample in China and compare the results with those of other studies. Methods This was a cross-sectional analysis of the population-based Shunyi Study in China. A total of 1,211 stroke-free participants (mean age, 55.6±9.3 years; 37.4% men) with available 3 Tesla (3T) magnetic resonance images were included in this analysis. Demographic information and risk factor data were assessed. The overall and age-specific prevalence of lacunes, WMH, and CMBs was evaluated. Associations between cardiovascular risk factors and the presence of these lesions were analyzed by multiple logistic regression. Results Our study showed a prevalence of 14.5% for lacunes, 72.1% for periventricular hyperintensity (PVH), 65.4% for deep white matter hyperintensity (DWMH), and 10.6% for CMBs. When compared with other community-based samples, individuals in the same age group showed a higher burden of lacunes and a relatively lower prevalence of CMBs. Advanced age was independently associated with the prevalence of these CSVD markers, while the presence of hypertension increased the risk of lacunes, PVH/DWMH, and CMBs in deep or infratentorial locations. Conclusions A higher burden of lacunes but a relatively lower prevalence of CMBs was observed in this Chinese population. This notable result highlights the challenge of CSVD prevention in China. Chinese have a risk factor profile for CSVD similar to those in other populations.

        Gene Expression Profile of Helicobacter pylori in Response to Growth Temperature Variation

        A Helicobacter pylori whole-genome DNA microarray was constructed to study expression profiles of H. pylori in response to a sudden temperature transfer from 37°C to 20°C. The expression level of the genome at each of four time points (15, 30, 60, and 120 min) after temperature downshift was compared with that just before cold treatment. Globally, 10.2% (n=167) of the total predicted H. pylori genes (n=1636) represented on the microarray were significantly differentially expressed (p<0.05) over a 120 min period after shift to low temperature. The expression profiles of the differentially expressed genes were grouped, and their expression patterns were validated by quantitative real-time PCR. Up-regulated genes mainly included genes involved in energy metabolism and substance metabolism, cellular processes, protein fate, ribosomal protein genes, and hypothetical protein genes, which indicate the compensational responses of H. pylori to temperature downshift. Those genes play important roles in adaption to temperature downshift of H. pylori. Down-regulation of DNA metabolism genes and cell envelope genes and cellular processes genes may reflect damaged functions under low temperature, which is unfavorable to bacterial infection and propagation. Overall, this time-course study provides new insights into the primary response of H. pylori to a sudden temperature downshift, which allow the bacteria to survive and adapt to the new host environment.

        Height-thickness ratio on axial behavior of composite wall with truss connector

        Double skin composite walls offer structural and economic merits over conventional reinforced concrete counterparts in terms of higher capacity, greater stiffness, and better ductility. This paper investigated the axial behavior of double skin composite walls with steel truss connectors. Full-scaled tests were conducted on three specimens with different height-to-thickness ratios. Test results were evaluated in terms of failure mode, load-axial displacement response, buckling loading, axial stiffness, ductility, strength index, load-lateral deflection, and strain distribution. The test data were compared with AISC 360 and Eurocode 4 and it was found that both codes provided conservative predictions on the safe side.

      • Prognostic Value of Caveolin-1 Expression in Gastric Cancer: a Meta-analysis

        The relationship between caveolin-1 (Cav-1) and clinicopathological characteristics of gastric cancer is controversial, although Cav-1 plays an important role in tumor metastasis. To evaluate the clinicopathological and prognostic value of expression in patients with gastric cancer, a meta-analysis was performed to investigate the impact on clinicopathological parameters and prognosis in gastric cancer cases. Studies assessing these parameters for Cav-1 in gastric cancer were identified up to June 2014. Finally, a total of six studies met the inclusion criteria. Our combined results showed that Cav-1 expression was significantly associated with the Lauren classification (pooled OR=0.603, 95% CI: 0.381-0.953, P=0.030). Furthermore, we found that Cav-1 expression predicted a better overall survival in gastric cancer patients (pooled OR=0.590, 95% CI: 0.360-0.970, P=0.038, fixed-effect). In conclusion, the overall data of the present meta analysis showed that Cav-1 expression was not correlated with clinicopathological features except for the Lauren classification. Simultaneously, Cav-1 overexpression predicted a better overall survival in gastric cancer. Cav-1 expression in tumors is a candidate positive prognostic biomarker for gastric cancer patients.
