        End Plate–Stiff ener Connection for SHS Column and RHS Beam in Steel-Framed Building Modules

        This paper investigates the mechanical performance of beam-to-column connections for steel-framed building modules with RHS and SHS sections of relatively small member sizes. In the experimental program, three forms of connections are tested under static bending, including a completely welded connection (W), a bolted end plate connection (EP) and the proposed end plate–stiff ener connection (ST), and the failure modes, moment–rotation behaviours and strain responses were recorded during experiments. Results show that ST connection exhibited superior connection strength, stiff ness and ductility compared with the EP connection. In addition, a detailed fi nite element analysis was conducted to model the connection behaviours, considering bolt pretension, contact behaviours and detailed bolt geometry, showing good comparison with experiments. Furthermore, a parametric study was performed on the end plate thickness and stiff ener thickness for the proposed end plate–stiff ener connection. Such results fi nally enable a comparative study to investigate the performance of module during road transportation, highlighting the eff ects of connection stiff ness.

        Numerical Simulation of Heat Transfer Characteristics of Capillary Radiant Heating Floor

        In order to study the temperature transference characteristics of capillary radiant heating ground, a glowing temperature change flooring model is typically established to analyze the influence of different factors on the heat transfer characteristics of the heating system. The results show that the distribution of covering temperature is uneven in the radiant heating system, and the fluctuation amplitude increases with the increase of the pipe spacing. At the same time, the change of the surface temperature is further analyzed for the finishing layer material on the radiant heating storey. The results show that the change of the capillary tube spacing leads to a smaller change in the floor exterior temperature when the wood floor is used as the finishing layer. It accurately indicates that the wood floor as the finishing layer can allegedly provide a more balanced surface temperature. Finally, the effects of finishing the layer, filling layer material and pipe spacing are studied for the change of surface heat flux. The results show that the heating effect of 30°C quandary in capillary radiant heating system can merely reach that of 40°C hot water in heating structure with traditional pipe breadth when the finishing layer universally adopts floor tiles and the capillary pipe diameter is 10 mm. This suggests that the thin system can use low-temperature hot water for effective energy-saving heating. The above research results can thoughtfully provide theoretical guidance for radiant floor heating.

        5-Formylhonokiol exerts anti-angiogenesis activity $via$ inactivating the ERK signaling pathway

        Our previous report has demonstrated that 5-formylhonokiol (FH), a derivative of honokiol (HK), exerts more potent anti-proliferative activities than honokiol in several tumor cell lines. In present study, we first explored the antiangiogenic activities of 5-formylhonokiol on proliferation, migration and tube formation of human umbilical vein endothelial cells (HUVECs) for the first time $in$ $vitro$. Then we investigated the $in$ $vivo$ antiangiogenic effect of 5-formylhonokiol on zebrafish angiogenesis model. In order to clarify the underlying molecular mechanism of 5-formylhonokiol, we investigated the signaling pathway involved in controlling the angiogenesis process by western blotting assay. Wound-healing results showed that 5-formylhonokiol significantly and dose-dependently inhibited migration of cultured human umbilical vein enthothelial cells. The invasiveness of HUVEC cells was also effectively suppressed at a low concentration of 5-formylhonokiol in the transwell assay. Further F-actin imaging revealed that inhibitory effect of 5-formylhonokiol on invasion may partly contribute to the disruption of assembling stress fiber. Tube formation assay, which is associated with endothelial cells migration, further confirmed the anti-angiogenesis effect of 5-formylhonokiol. In $in$ $vivo$ zebrafish angiogenesis model, we found that 5-formylhonokiol dose-dependently inhibited angiogenesis. Furthermore, western blotting showed that 5-formylhonokiol significantly down-regulated extracellular signal-regulated kinase (ERK) expression and inhibited the phosphorylation of ERK but not affecting the total protein kinase B (Akt) expression and related phosphorylation, suggesting that 5-formylhonokiol might exert anti-angiogenesis capacity $via$ down-regulation of the ERK signal pathway. Taken together, these data suggested that 5-formylhonokiol might be a viable drug candidate in antiangiogenesis and anticancer therapies.

      • Changes in Markers of Liver Function in HCV 1b Patients with Compensated Cirrhosis Treated with Ombitasvir/Paritaprevir/ Ritonavir plus Dasabuvir with Ribavirin

        Aims: Patients chronically infected with HCV are at risk of developing extrahepatic manifestations of HCV as well as progressing to compensated or decompensated cirrhosis and HCC. Although current treatments have high rates of SVR, relatively little is known about possible regression of liver fibrosis after achieving an SVR. The ONYX-II trial examined the efficacy and safety of ombitasvir/paritaprevir/ritonavir plus dasabuvir + ribavirin (RBV) in Asian patients with HCV genotype 1b infection and compensated cirrhosis. Here we report changes in key markers of liver fibrosis and function. Methods: Patients with chronic HCV GT1b infection and compensated cirrhosis were enrolled in China, South Korea and Taiwan and received 12 weeks of OBV/PTV/r (25 mg/150 mg/100 mg once daily) and DSV (250 mg twice daily) with weight-based RBV. The primary objective of ONYX-II was to assess efficacy (SVR12) and safety of the regimen. Changes in markers of liver fibrosis and function between baseline (BL) and post-treatment week (PTW) 12 are presented. Results: Overall, 104 patients were enrolled and treated in ONYX-II. All patients (104/104, 100%) achieved SVR12. BL and PTW12 data for FibroTest score, alanine aminotransferase (ALT), aspartate aminotransferase (AST), total bilirubin, albumin, platelet count and alpha fetoprotein (AFP) are shown in Table 1. All selected parameters showed numerical improvements between BL and PTW12. Mean ALT and AST levels returned to within normal range and FibroTest scores demonstrated a numerical improvement, suggesting improvement in liver status. The complete set of data between BL and PTW12 will be presented for these parameters and other liver composite parameters at the conference. Conclusions: Measurement of key liver function markers during the ONYX-II trial showed a numerical improvement within 12 weeks of completion of treatment in HCV GT1b-infected patients with compensated cirrhosis. Further follow-up of these patients will determine the long-term durability of these changes.

        An approach to automatic boundary segmentation of solid models using virtual topology: toward reconstruction of design features

        Boundary segmentation of solid models is the geometric foundation to reconstruct design features. In this paper, based on the shape evolution analysis for the feature-based modeling process, a novel approach to the automatic boundary segmentation of solid models for reconstructing design features is proposed. The presented approach simulates the designer’s decomposing thinking on how to decompose an existing boundary representation model into a set of design features. First, the modeling traces of design features are formally represented as a set of feature vertex adjacent graphs that use low-dimensional vertex entities and their connection relations. Then, a set of Boolean segmentation loops is searched and extracted from the constructed feature vertex adjacent graphs, which segment the boundary of a solid model into a set of regions with different design feature semantics. In the search process, virtual topology operations are employed to simulate the topological changes resulting from Boolean operations in feature modeling processes. In addition, to realize effective search, search strategies and search algorithms are presented. The segmentation experiments and case study show that the presented approach is feasible and effective for the boundary segmentation of medium-level complex part models. The presented approach lays the foundation for the later reconstruction of design features.

        Different Dimensional and Structural Variations in Coordination Compounds of Cadmium, Manganese and Nickel Constructed from the Ligand 2,2'-Bipyidine-3,3',6,6'-tetracarboxylic Acid (H<sub>4</sub>bptc)

        The reactions of hydrated $CdCl_2$, $MnCl_2$, and $NiCl_2$ with 2,2'-bipyidine-3,3',6,6'-tetracarboxylic acid ($H_4bptc$) afforded the mononuclear [$Cd^{II}(H_2bptc)(H_2O)_3]{\cdot}H_2O$ (1), linear $\{[Cd(H_2bptc)(H_2O)]{\cdot}3H_2O\}_n$ (2), 3-D heterobimetallic $[NaCd(Hbptc)(H_2O)]$ (3), layer $[Mn(H_2bptc)(H_2O)]_n$ (4) and a dinuclear compound $[Ni_2(H_2bptc)-(H_2O)_2]{\cdot}6H_2O$ (5). These compounds have been characterized by elemental analysis, IR, and their structures have been determined by X-ray crystallography. The thermal stabilities of 1-3 were measured by thermogravimetric analysis (TGA) and their solid state luminescence properties together with the free ligand $H_4bptc$ were investigated at room temperature.

      • Efficacy/Safety of Ombitasvir/Paritaprevir/Ritonavir and Dasabuvir with Ribavirin in Asian Patients with Genotype 1b HCV-Infected, Compensated Cirrhosis: ONYX-II SVR 24 Results

        Aims: ONYX-II is a phase 3, open-label study of 3-DAA regimen of OBV/PTV/r and DSV with RBV in treatment-naive and experienced patients with genotype 1b HCV infection and compensated cirrhosis in China, South Korea and Taiwan. SVR12 rate was 100% and the favourable safety profile was shown. The present analysis reports efficacy( SVR24) and safety results. Methods: Patients with chronic GT1b HCV infection and compensated cirrhosis received OBV/PTV/r + DSV + RBV for 12 weeks and will be followed for 48 weeks post-treatment. Efficacy was assessed by SVR12 and SVR24. Safety was assessed as the percentage of patients wi th treatment-emergent adverse events (TEAEs) and laboratory evaluation. Results: Total of 104 patients with chronic GT1b HCV infection (62% female, 100% Asian, 58% treatment-experienced) were enrolled from China (n=63), South Korea (n=21) and Taiwan (n=20). All patients received at least one dose of study drugs. The SVR24 rate was 100% (concordant with SVR12), with no patient relapsing between post-treatment week 12 and 24. Most TEAEs were mild in severity. The most common TEAEs (≥10%) were increased blood bilirubin levels (25%), pruritus (15%), anaemia (14%), asthenia (12%), bilirubin conjugated increased (12%), blood bilirubin unconjugated increased (12%), dizziness (11%) and fatigue (11%). Four patients had serious AEs and all were assessed as not being related to the 3-DAA regimen (one was assessed as being possibly related to RBV). One patient discontinued treatment due to TEAEs (elevations in alanine aminotransferase [ALT], aspartate aminotransferase [AST] and blood bilirubin) after 3 weeks of dosing but achieved SVR12 and SVR24. Laboratory abnormalities ≥ grade 3 were infrequent (ALT: 3%; AST 2%; total bilirubin: 7%). No grade 3 haemoglobin decrease was reported. Conclusions: SVR24 and SVR12 rates were concordant (100%) in HCV GT1b-infected Asian patients with compensated cirrhosis. The regimen was generally well tolerated with mostly mild TEAEs reported.
