      • Matrine Reduces Proliferation of Human Lung Cancer Cells by Inducing Apoptosis and Changing miRNA Expression Profiles

        Matrine, a main active component extracted from dry roots of Sophora flavecens, has been reported to exert antitumor effects on A549 human non-small lung cancer cells, but its mechanisms of action remain unclear. To determine effects of matrine on proliferation of A549 cells and assess possible mechanisms, MTT assays were employed to detect cytotoxicity, along with o flow cytometric analysis of DNA content of nuclei of cells following staining with propidium iodide to analyze cell cycle distribution. Western blotting was performed to determined expression levels of Bax, Bcl-2, VEGF and HDAC1, while a microarray was used to assessed changes of miRNA profiles. In the MTT assay, matrine suppressed growth of human lung cancer cell A549 in a dose- and timedependent manner at doses of 0.25-2.5 mg/ml for 24h, 48h or 72h. Matrine induced cell cycle arrest in G0/G1 phase and decreased the G2/M phase, while down-regulating the expression of Bcl2 protein, leading to a reduction in the Bcl-2/Bax ratio. In addition, matrine down regulated the expression level of VEGF and HDAC1 of A549 cells. Microarray analysis demonstrated that matrine altered the expression level of miRNAs compared with untreated control A549 cells. In conclusion, matrine could inhibit proliferation of A549 cells, providing useful information for understanding anticancer mechanisms.

        Electrochemical Investigation of Tryptophan at a Poly(p-aminobenzene sulfonic acid) Film Modified Glassy Carbon Electrode

        A glassy carbon electrode (GCE) modified with poly(p-aminobenzene sulfonic acid) [Poly(p-ABSA)] film is fabricated by voltammetric technique in phosphate buffer solution (pH 8.0) containing $5.0\;{\times}\;10^{-3}\;mol\;L^{-1}$p- ABSA. Electrochemical behaviors of tryptophan at the Poly(p-ABSA) film electrode are investigated with voltammetry. The results indicate that the electrochemical response of tryptophan is improved significantly in the presence of poly(p-ABSA) film. Compared with the bare glassy carbon electrode, the Poly(p-ABSA) film electrode remarkably enhances the irreversible oxidation peak current of tryptophan. Some parameters such as voltammetric sweeping segments for the electrochemical polymerization, pH, accumulation potential and accumulation time are optimized. Under the optimal conditions, the oxidation peak current is proportional to tryptophan concentration in the range of $1.0\;{\times}\;10^{-7}$ to $1.0\;{\times}\;10^{-6}\;mol\;L^{-1}$, and $2.0\;{\times}\;10^{-6}$ to $1.0\;{\times}\;10^{-5}\;mol\;L^{-1}$ with a detection limit of $7.0\;{\times}\;10^{-8}\;mol\;L^{-1}$. The proposed procedure is successfully applied to the determination of tryptophan in a commercial amino acid oral solution.

        Evidence for the Presence of Long-Lived Plasma Cells in Nasal Polyps

        Purpose: Plasma cells and immunoglobulins (Igs) play a pivotal role in the induction and maintenance of chronic inflammation in nasal polyps. During secondary immune responses, plasma cell survival and Ig production are regulated by the local environment. The purpose of the present study was to investigate the presence of long-lived plasma cells (LLPCs) and specific survival niches for LLPCs in human nasal polyps. Methods: Nasal mucosal samples were cultured with an air-liquid interface system and the Ig levels in culture supernatants were analyzed by enzyme-linked immunosorbent assay. The characteristics of LLPCs in nasal polyps were determined by immunohistochemistry and immunofluorescence. The expression of neurotrophins as well as their receptors was detected by quantitative real-time polymerase chain reaction, immunohistochemistry, immunofluorescence, and Western blotting. Results: The numbers of CD138+ total plasma cells and BCL2+ plasma cells were increased in both eosinophilic and non-eosinophilic nasal polyps compared with those in normal tissues. The production of IgG, IgA, and IgE was detected in culture supernatants even after a 32-day culture of nasal polyps. Although the total numbers of plasma cells were decreased in nasal polyps after culture, the numbers of BCL2+ plasma cells remained stable. The expression of nerve growth factor (NGF) as well as tropomyosin receptor kinase (Trk) A, a high-affinity receptor for NGF, was upregulated in both eosinophilic and non-eosinophilic nasal polyps. In addition, BCL2+ plasma cell numbers were positively correlated with NGF and TrkA mRNA expression in nasal mucosal tissues. Polyp plasma cells had the expression of TrkA. Conclusions: Human nasal polyps harbor a population of LLPCs and NGF may be involved in their prolonged survival. LLPCs may be a novel therapeutic target for suppressing the local Ig production in nasal polyps.

        Leaf Wilting Movement Can Protect Water-Stressed Cotton (Gossypium hirsutum L.) Plants Against Photoinhibition of Photosynthesis and Maintain Carbon Assimilation in the Field

        Under severe water stress, leaf wilting is quite general in higher plants. This passive movement can reduce the energy load on a leaf. This paper reports an experimental test of the hypothesis that leaf wilting movement has a protective function that mitigates against photoinhibition of photosynthesis in the field. The experiments exposed cotton (Gossypium hirsutum L.) to two water regimes: waterstressed and well-watered. Leaf wilting movement occurred in water-stressed plants as the water potential decreased to −4.1 MPa, reducing light interception but maintaining comparable quantum yields of photosystem II (PS II; Yield for short) and the proportion of total PS II centers that were open (qP). Predrawn Fv/Fm (potential quantum yield of PS II) as an indicator of overnight recovery of PS II from photoinhibition was higher than or similar to that in wellwatered plants. Compared with water-stressed cotton leaves for which wilting movement was permitted, water-stressed cotton leaves restrained from such movement had significantly increased leaf temperature and instantaneous CO2assimilation rates in the short term, but reduced Yield, qP,and Fv/Fm. In the long term, predrawn Fv/Fm and CO2assimilation capacity were reduced in water-stressed leaves restrained from wilting movement. These results suggest that, under water stress, leaf wilting movement could reduce the incident light on leaves and their heat load, alleviate damage to the photosynthetic apparatus due to photoinhibition,and maintain considerable carbon assimilation capacity in the long term despite a partial loss of instantaneous carbon assimilation in the short term.

        The Characteristics and Survival Outcomes in Patients Aged 70 Years and Older with Nasopharyngeal Carcinoma in the Intensity-Modulated Radiotherapy Era

        Purpose We aim to examine nasopharyngeal carcinoma (NPC) characteristics and survival outcomes in patients aged 70 years and older in the intensity-modulated radiotherapy (IMRT) era. Materials and Methods From 2006 to 2013, 126 non-metastatic NPC patients aged  70 years who were treated with IMRT +/ chemotherapy were included. Adult Comorbidity Evaluation 27 (ACE-27) was used to measure patient comorbidities. The overall survival (OS) and cancer-specific survival (CSS) were calculated with the Kaplan-Meier method, and differences were compared using the log-rank test. The Cox proportional hazards model was used to carry out multivariate analyses. Results For the entire group, only two patients (1.6%) presented stage I disease, and up to 84.1% patients had stage III-IVB disease. All patients had a comorbidity score of 0 in 24 (19.0%), 1 in 45 (35.7%), 2 in 42 (33.3%), and 3 in 15 (11.9%) patients. The main acute grade during radiotherapy was 3-4 adverse events consisting of mucositis (25.4%), bone marrow suppression (16.7%), and dermatitis (8.7%). After treatment, four patients (3.2%) developed temporal lobe injury. Five-year CSS and OS rates were 67.3% (95% confidence interval [CI], 58.6% to 77.4%) and 54.0% (95% CI, 45.6% to 63.9%), respectively. Five-year OS was significantly higher for ACE-27 score 0-1 than ACE-27 score 2-3 (72.9% and 39.9%, respectively; p < 0.001). Multivariate analyses showed ACE-27 score 0-1 was significantly associated with superior OS (hazard ratio [HR], 3.02; 95% CI, 1.64 to 5.55; p < 0.001). In addition, the rate of OS was higher for stage I-III than that of stage IV, with borderline significance (HR, 1.67; 95% CI, 0.99 to 2.82; p=0.053). But no significant advantage was observed in OS when chemotherapy was used (p > 0.05). Conclusion Our findings suggest IMRT +/– chemotherapy has a manageable toxicity and provides an acceptable survival in patients aged  70 years with NPC. ACE-27 score was significantly associated with survival outcomes in this group population.

        The implications of signaling lipids in cancer metastasis

        Metastasis is the most malignant stage of cancer. Lipid metabolic abnormalities are now increasingly recognized as characteristics of cancer cells. The accumulation of certain lipid species, such as signaling lipids, due to the avidity of lipid metabolism may be a causal factor of tumor malignant progression and metastatic behavior. In this review, we first describe signaling lipids implicated in cancer migration, invasion and metastasis. Next, we summarize the regulatory signaling hubs of lipid anabolic and catabolic metabolism. We then address lipid-rich circulating tumor cells (CTCs) and the lipid composition of exosomes budded off from tumor cells. We also present advances in targeting the regulatory hubs of lipid metabolism and signaling lipids in cancer therapy. Given the complexity of metabolic disorders in cancer, the development of significant portfolios of approaches to target signaling lipids by the integration of multiple chemical modulations, as well as molecular imaging modalities, should offer promising strategies for cancer therapy.

        Identification and Characterization of Alternaria iridiaustralis Causing Leaf Spot on Iris ensata in China

        In 2016, a severe leaf spot disease was found on Iris ensata Thumb. in Nanjing, China. The symptom was elliptical, fusiform, or irregularly necrotic lesion surrounded by a yellow halo, from which a small-spored Alternaria species was isolated. The fungus was identified as Alternaria iridiaustralis based on morphological characteristics. The pathogenicity tests revealed that the fungus was the causal pathogen of the disease. Phylogenic analyses using sequences of ITS, gpd, endoPG, and RPB2 genes confirmed the morphological identification. This study is the first report of A. iridiaustralis causing leaf spots on I. ensata in China.

        Hydrothermal synthesis of morphology controllable Cu2O and their catalysis in thermal decomposition of ammonium perchlorate

        Cu2O micro-crystals with different morphologies (including three branching structures) weresynthesized through hydrothermal method by using sodium tartrate as chelating reagent. Branchinggrowth patterns of Cu2O microcrystals have a strong dependence on the amount of sodium tartrate. These different Cu2O microcrystals were used as catalysts to promote the thermal decomposition ofammonium perchlorate (AP). The thermal decomposition of AP in the presence or absence of Cu2Omicro-crystals was investigated non-isothermally through thermogravimetry and differential scanningcalorimetry (DSC). The data obtained from DSC were applied for the calculation and comparison of thekinetic parameters of AP decomposition process through a model-free approach.
