        A Cytosolic Thioredoxin Acts as a Molecular Chaperone for Peroxisome Matrix Proteins as Well as Antioxidant in Peroxisome

        Thioredoxin (TRX) is a disulfide reductase present ubiquitously in all taxa and plays an important role as a regulator of cellular redox state. Recently, a redox-independent, chaperone function has also been reported for some thioredoxins. We previously identified nodulin-35, the subunit of soybean uricase, as an interacting target of a cytosolic soybean thioredoxin, GmTRX. Here we report the further characterization of the interaction, which turns out to be independent of the disulfide reductase function and results in the co-localization of GmTRX and nodulin-35 in peroxisomes, suggesting a possible function of GmTRX in peroxisomes. In addition, the chaperone function of GmTRX was demonstrated in in vitro molecular chaperone activity assays including the thermal denaturation assay and malate dehydrogenase aggregation assay. Our results demonstrate that the target of GmTRX is not only confined to the nodulin-35, but many other peroxisomal proteins, including catalase (AtCAT), transthyretin-like protein 1 (AtTTL1), and acyl-coenzyme A oxidase 4 (AtACX4), also interact with the GmTRX. Together with an increased uricase activity of nodulin-35 and reduced ROS accumulation observed in the presence of GmTRX in our results, especially under heat shock and oxidative stress conditions, it appears that GmTRX represents a novel thioredoxin that is co-localized to the peroxisomes, possibly providing functional integrity to peroxisomal proteins.

      • Analysis of Cancer Incidence in Zhejiang Cancer Registry in China during 2000 to 2009

        Objective: The Zhejiang Provincial Cancer Prevention and Control Office collected cancer registration data during 2000 to 2009 from 6 cancer registries in Zhejiang province of China in order to analyze the cancer incidence. Methods: Descriptive analysis included cancer incidence stratified by sex, age and cancer site group. The proportions and cumulative rates of 10 common cancers in different groups were also calculated. Chinese population census in 1982 and Segi's population were used for calculating age-standardized incidence rates. The log-linear model was used for fitting to calculate the incidence trends. Results: The 6 cancer registries in Zhejiang province in China covered a total of 60,087,888 person-years during 2000 to 2009 (males 30,445,904, females 29,641,984). The total number of new cancer cases were 163,104 (males 92,982, females 70,122). The morphology verified cases accounted for 69.7%, and the new cases verified only by information from death certification accounted for 1.23%. The crude incidence rate in Zhejiang cancer registration areas was $271.5/10^5$ during 2000 to 2009 (male $305.41/10^5$, female $236.58/10^5$), age-standardized incidence rates by Chinese standard population (ASIRC) and by world standard population (ASIRW) were $147.1/10^5$ and $188.2/10^5$, the cumulative incidence rate (aged from 0 to 74) being 21.7%. The crude incidence rate was $209.6/10^5$ in 2000, and it increased to $320.20/10^5$ in 2009 (52.8%), with an annual percent change (APC) of 4.51% (95% confidence interval, 3.25%-5.79%). Age-specific incidence rate of 80-84 age group was achieved at the highest point of the incidence curve. Overall with different age groups, the cancer incidences differed, the incidence of liver cancer being highest in 15-44 age group in males; the incidence of breast cancer was the highest in 15-64 age group in females; the incidences of lung cancer were the highest in both males and females over the age of 65 years. Conclusions: Lung cancer, digestive system malignancies and breast cancer are the most common cancers in Zhejiang province in China requiring an especial focus. The incidences of thyroid cancer, prostate cancer, cervical cancer and lymphoma have increased rapidly. Prevention and control measures should be implemented for these cancers.

      • Associations Between XRCC1 Arg399Gln, Arg194Trp, and Arg280His Polymorphisms and Risk of Differentiated Thyroid Carcinoma: A Meta-analysis

        Background: Associations between Arg399Gln, Arg194Trp and Arg280His polymorphisms of the XRCC1 gene and risk of differentiated thyroid carcinoma (DTC) have been widely studied but the findings are contradictory. Methods: We performed a meta-analysis in the present study using STATA 11.0 software to clarify any associations. Electronic literature databases and reference lists of relevant articles revealed a total of 10, 6 and 6 published studies for the Arg399Gln, Arg194Trp and Arg280His polymorphisms, respectively. Results: No significant associations were observed between Arg399Gln and DTC risk in all genetic models within the overall and subgroup meta-analyses, while the Trp/Trp vs Arg/Arg and recessive model of the Arg194Trp polymorphism was associated with DTC susceptibility, and the dominant model of Arg280His polymorphism contributed to DTC susceptibility in Caucasians. Conclusions: Our meta-analysis suggests that XRCC1 Arg194Trp may be a risk factor for DTC development.

        The role of the genomic mutation signature and tumor mutation burden on relapse risk prediction in head and neck squamous cell carcinoma after concurrent chemoradiotherapy

        Personalized genetic profiling has focused on improving treatment efficacy and predicting risk stratification by identifying mutated genes and selecting targeted agents according to genetic testing. Therefore, we evaluated the role of genetic profiling and tumor mutation burden (TMB) using next-generation sequencing in patients with head and neck squamous cell carcinoma (HNSC). The relapse mutation signature (RMS) and chromatin remodeling mutation signature (CRMS) were explored to predict the risk of relapse in patients with HNSC treated with concurrent chemoradiotherapy (CCRT) with platinum-based chemotherapy. Patients in the high RMS and CRMS groups showed significantly shorter relapse-free survival than those in the low RMS and CRMS groups, respectively (p < 0.001 and p = 0.006). Multivariate Cox regression analysis showed that extranodal extension, CCRT response, and three somatic mutation profiles (TMB, RMS, and CRMS) were independent risk predictors for HNSC relapse. The predictive nomogram showed satisfactory performance in predicting relapse-free survival in patients with HNSC treated with CCRT.

        SITES(Sustainable Sites Intiative) 가이드라인을 활용한 지속가능한 조경설계 원칙에 기반한 도시공원에 관한 연구-서울숲 공원을 중심으로-

        본 연구는 급격한 도시화에 따른 자연환경 문제가 심각하게 대두되는 현 시점에서, 조성되어 있는 도시공원에 대해 지속가능한 조경설계 원칙에 따라 분석을 시도했다. 이를 위해 먼저, 기존에 조성된 도시공원에 지속가능한 조경설계 원칙이 어느 정도 적용되어 있는지를, 문헌연구에 근거하여 조경설계 개념의 요소를 도출해냈다. 또한 SITES(Sustainable Sites Initiative)의 가이드라인 10가지를 정리하였으며, 기존 선행연구를 통해 10가지 중 5가지 요소를 도출한 후 연구 대상지인 서울숲 공원 분석을 진행하였다. 이를 토대로 서울숲 공원에서는 지속가능한 설계원칙이 어떻게 적용되고 있는지에 대한 연구도 수행하였다. 연구를 진행한 결과, 서울숲 공원은 지속가능한 조경설계 원칙이 적절히 적용되어 운영되고 있음을 알 수 있었다. 하지만 인공적인 수목 이식, 토양의 조건 등 현실적인 문제로 향후, 이러한 조건들로 문제가 생길 가능성도 충분히 고려해 방향성을 설정할 필요성이 느껴졌다. 서울숲 공원은 자연환경과 생태계의 균형을 중시한 설계원칙을 가지고 있으며 지역의 문화적인 특성을 고려한 설계로, 주변 지역을 활력있는 공간으로 탈바꿈하며 지속가능한 발전을 촉진시켰다. 서울숲 공원은 SITES 가이드라인의 다섯 가지 지속가능한 조경설계 원칙을 모두 충족하는 훌륭한 사례이다. 자연환경과 생태계를 고려한 설계, 지역의 문화적 특성을 고려한 설계, 지속가능한 자원 활용의 단계적 결정 시스템 이용, 공평한 재생이 가능한 생태 시스템 제공, 시스템 사고 방법 사용 등 다양한 측면에서 지속가능성을 실천하고 있다. 본 연구는 지속가능한 설계원칙이 비교적 잘 적용된 서울숲 공원의 분석을 통해, 지속가능한 조경설계 원칙의 요소를 도출하여 제시하는 한편 도시공원의 지속가능성을 위한 기준을 제공하고자 한다. This study attempted to analyze the existing urban parks according to the principles of sustainable landscape design at a time when the natural environment is facing serious problems due to rapid urbanization. To this end, first, the extent to which sustainable landscape design principles are applied to existing urban parks was studied, and elements of the landscape design concept were derived based on the literature. In addition, the 10 guidelines of the Sustainable Sites Initiative (SITES) were summarized, and five of the 10 elements were derived through previous studies, and then the Seoul Forest Park, the subject of the study, was analyzed. Based on this, we conducted a study on how sustainable design principles are applied in Seoul Forest Park. As a result of the study, we found that Seoul Forest Park is operated appropriately with sustainable landscape design principles. However, due to practical issues such as artificial tree transplantation and soil conditions, it was felt necessary to set a direction that fully considers the possibility of problems arising from these conditions in the future. Seoul Forest Park has a design principle that emphasizes the balance of the natural environment and ecosystem and takes into account the cultural characteristics of the area, transforming the surrounding area into a vibrant space and promoting sustainable development. Seoul Forest Park is an excellent example of how to fulfill all five sustainable landscape design principles of the SITES Guidelines. It practices sustainability in various aspects, including designing for the natural environment and ecosystem, designing for the cultural characteristics of the area, using a phased decision system for sustainable resource utilization, providing an ecological system for equitable regeneration, and using a systems thinking method. Through the analysis of Seoul Forest Park, where sustainable design principles are relatively well applied, this study aims to derive and present elements of sustainable landscape design principles and provide criteria for sustainability of urban parks.
