        Gα<sub>12</sub> gep oncogene deregulation of p53-responsive microRNAs promotes epithelial–mesenchymal transition of hepatocellular carcinoma

        Hepatocellular carcinoma (HCC) has a poor prognosis owing to aggressive phenotype. Gα<SUB>12</SUB> gep oncogene product couples to G-protein-coupled receptors, whose ligand levels are frequently increased in tumor microenvironments. Here, we report Gα<SUB>12</SUB> overexpression in human HCC and the resultant induction of zinc-finger E-box-binding homeobox 1 (ZEB1) as mediated by microRNA deregulation. Gα<SUB>12</SUB> expression was higher in HCC than surrounding non-tumorous tissue. Transfection of Huh7 cell with an activated mutant of Gα<SUB>12</SUB> (Gα<SUB>12</SUB>QL) deregulated microRNA (miRNA or miR)-200b/a/429, -194-2/192 and -194-1/215 clusters in the miRNome. cDNA microarray analyses disclosed the targets affected by Gα<SUB>12</SUB> gene knockout. An integrative network of miRNAs and mRNA changes enabled us to predict ZEB1 as a key molecule governed by Gα<SUB>12</SUB>. Decreases of miR-200a/b, -192 and -215 by Gα<SUB>12</SUB> caused ZEB1 induction. The ability of Gα<SUB>12</SUB> to decrease p53 levels, as a result of activating protein-1 (AP-1)/c-Jun-mediated mouse double minute 2 homolog induction, contributed to transcriptional deregulation of the miRNAs. Gα<SUB>12</SUB>QL induced ZEB1 and other epithelial–mesenchymal transition markers with fibroblastoid phenotype change. Consistently, transfection with miR-200b, -192 or -215 mimic prevented the ability of Gα<SUB>12</SUB>QL to increase tumor cell migration/invasion. In xenograft studies, sustained knockdown of Gα<SUB>12</SUB> decreased the overall growth rate and average volume of tumors derived from SK-Hep1 cell (mesenchymal-typed). In HCC patients, miR-192, -215 and/or -200a were deregulated with microvascular invasion or growth advantage. In the HCC samples with higher Gα<SUB>12</SUB> level, a correlation existed in the comparison of relative changes of Gα<SUB>12</SUB> and ZEB1. In conclusion, Gα<SUB>12</SUB> overexpressed in HCC causes ZEB1 induction by deregulating p53-responsive miRNAs, which may facilitate epithelial–mesenchymal transition and growth of liver tumor. These findings highlight the significance of Gα<SUB>12</SUB> upregulation in liver tumor progression, implicating Gα<SUB>12</SUB> as an attractive therapeutic target.

        Preparation of Bimodal Porous Carbon Supported PtRu Catalysts for Fuel Cells

        <P>In this work, we synthesised the bimodal porous carbon supported electro-catalyst as a novel catalyst for fuel cells. For this purpose, the bimodal porous carbons with different pore sizes were prepared by imprinting method. Three kinds of bimodal porous carbons were fabricated by using the silica spheres with diameters of ca. 50, 100 and 300 nm and SBA-15 particle having 200–250 nm diameter and 700–900 nm length as templates. The BET surface areas of the bimoal porous carbons were determined to be 235–294 m<SUP>2</SUP> g<SUP>–1</SUP>, which are similar to that of Vulcan XC72. To evaluate the electro-catalytic activity, the PtRu nanoparticles of ca. 1.9–2.6 nm were loaded on the bimodal porous carbon supports. The PtRu/C100, synthesised by using SBA-15 and 100 nm silica sphere, had the higher electrochemical surface area (ECSA) and the lower on-set potential and showed comparable performance with the commercial catalyst.</P>

      • Muricauda olearia sp. nov., isolated from crude-oil-contaminated seawater, and emended description of the genus Muricauda

        <P>A Gram-staining-negative, rod-shaped and non-motile bacterium, designated CL-SS4T, was isolated from crude-oil-contaminated seawater from the west coast of Korea and was investigated by means of a polyphasic taxonomic approach. Strain CL-SS4T grew optimally at 25-30 degrees C, at pH 6.8-7.7 and in the presence of 2-3% sea salts. The major fatty acids were iso-C17:0 3-OH, iso-C15:0 and iso-C15:1 G. The major isoprenoid quinone was MK-6. The DNA G+C content was 50.7 mol%. Analysis of the 16S rRNA gene sequence of strain CL-SS4T revealed that it was a member of the genus Muricauda, sharing 95.7-96.9% sequence similarity with the type strains of Muricauda species and being most closely related to Muricauda aquimarina SW-63T. Phylogenetic analyses based on 16S rRNA gene sequences confirmed that strain CL-SS4T formed a deep lineage related to the genus Muricauda. Strain CL-SS4T was distinguishable from recognized Muricauda species on the basis of differential phenotypic characteristics and DNA G+C content. Based on these results, it is proposed that strain CL-SS4T represents a novel species of the genus Muricauda, for which the name Muricauda olearia sp. nov. is proposed. An emended description of the genus Muricauda is also proposed. The type strain is CL-SS4T (=KCCM 90075T=JCM 15563T).</P>

        3D-PTV measurement was conducted on the wake of a sphere. The Reynolds number is 1130 with the diameter(d=30mm). The measurement system consisted of three CCD cameras, an image grabber, an Ar-Ion laser and a host computer. An eigenvalue analysis was introduced for the analysis on the wake. Statistical turbulent properties were quantitatively clarified using the time-consecutive instantaneous three-dimensional vectors obtained by the 3D-PTV system. The spatial distributions of the sphere wake showed strong elations with spiral motions of the vortcies shedded from the surface of the sphere.

        OGLE-2017-BLG-0173Lb: Low-mass-ratio Planet in a “Hollywood” Microlensing Event

        <P>We present microlensing planet OGLE-2017-BLG-0173Lb, with planet-host mass ratio of either q similar or equal to 2.5 x 10(-5) or q similar or equal to 6.5 x 10(-5), the lowest or among the lowest ever detected. The planetary perturbation is strongly detected, Delta chi(2) similar to 10000, because it arises from a bright (therefore, large) source passing over and enveloping the planetary caustic: a so-called 'Hollywood' event. The factor similar to 2.5 offset in q arises because of a previously unrecognized discrete degeneracy between Hollywood events in which the caustic is fully enveloped and those in which only one flank is enveloped, which we dub 'Cannae' and 'von Schlieffen,' respectively. This degeneracy is 'accidental' in that it arises from gaps in the data. Nevertheless, the fact that it appears in a Delta chi(2) = 10000 planetary anomaly is striking. We present a simple formalism to estimate the sensitivity of other Hollywood events to planets and show that they can lead to detections close to, but perhaps not quite reaching, the Earth/Sun mass ratio of 3 x 10(-6). This formalism also enables an analytic understanding of the factor similar to 2.5 offset in q between the Cannae and von Schlieffen solutions. The Bayesian estimates for the host mass, system distance, and planet-host projected separation are M = 0.39(-0.24)(+0.40) M circle dot, D-L = 4.8(-1.8)(+1.5) kpc, and a(perpendicular to) = 3.8 +/- 1.6 au, respectively. The two estimates of the planet mass are m(p) = 3.3(2.1)(+3.8) M circle plus and m(p) = 8(6)(+11) M circle plus. The measured lens-source relative proper motion mu = 6 mas yr(-1) will permit imaging of the lens in about 15 years or at first light on adaptive-optics imagers on next-generation telescopes. These will allow one to measure the host mass but probably will not be able to resolve the planet-host mass-ratio degeneracy.</P>

        Purpose: This study investigated the difference of menopausal knowledge, attitude between nursing students and non-nursing students. The relationships of the level of menopausal knowledge and attitude and the level of closeness to mother were investigated. Methods: A correlational descriptive design was used with a convenience sample. The survey was conducted with 206 female college students, 106 nursing and 100 non-nursing students, between September 1 and September 14, 2014. The instruments were the Knowledge of Menopause scale, the Menopausal Attitude Measurement scale, and the Level of Closeness to Mother scale. SPSS v22.0 is used to statistical analysis of descriptive statistics, independent t-tests, correlation and χ2-tests. Results: The difference in menopause knowledge between nursing students and non-nursing students is statistically significant (t=6.085, p<001). Nursing and non-nursing college students’ menopause knowledge had a positive relationship with attitude (r = 0.241, p<001). In closeness to mother, there is no significant difference was shown between nursing and non-nursing college students, even though the levels of menopause knowledge are different (t=1.336, p=.183). Conclusion: Results showed having menopausal knowledge positively influences students’ attitudes about menopause. Educators should develop programs imparting menopausal knowledge for non-nursing college students to increase positive menopausal attitude.

      • DHCP 기반 임베디드 RFID R/W 시스템 개발

        본 논문에서는 Tag 신호를 IEEE 802.11 통신 프로토콜을 통해서 데이터 및 영상처리가 가능한 PXA255 ARM 칩을 내장한 임베디드 RFID Reader/Writer 시스템과 전송된 Tag 신호를 이용하여 D/B를 검색한 후 이를 IEEE 802.11 통신 프로토콜 통해서 임베디드 시스템에 전송하는 임베다드 RFID R/W 미들웨어를 개발하였다. 개발한 임베디드 형태의 RFID R/W 시스템은 PXA255 ARM칩을 중심으로 13.56Mhz의 RFID Reader/Writer 서버와 데이터 통신을 위한 무선랜 및 TIT-LCD로 구성되어 있다. 임베디드 RFID R/W 시스템은 Tag 신호를 시리얼단자로 통해 입력받으면 이를 무선랜을 이용하여 서버로 데이터를 전송하고 다시 서버로부터 처리된 결과 이미지 데이터를 받아서 TFT-LCD화면에 표시한다. 임베디드 RFID R/W 미들웨어는 RFID R/W 취득한 Tag 신호를 임베디드 시스템에 전송하고, 임베디드 시스템은 클라이언트 소켓 프로그램을 작동시켜 IEEE 802.11 통신 프로토콜을 통해 윈도우 서버에 접속한 후 Tag 신호를 전송한다.

        NiFe / Cr 다층박막의 층수와 자기이방성에 따른 자기저항특성

        Glass\Cr_(40Å)\[Cr_(10Å)\NiFe_(50Å)]_N multilayers (number of layer N = 1, 2, 3, 4, 5, 6, 10) were made by dc magnetron sputtering under magnetic anisotropy of 200 G. Magnetoresistance curve MR(xx), MR(xy) were measured for the parallel and perpendicular current direction to external magnetic field. MR(xx) curves for the number of layer N=1, 5, 10 were almost became about zero percent, however the curves of other numbers appeared the phenomena of positive magnetoresistance that the resistance increased to external magnetic field, and the irregular and reversed curves in the vicinity of H=0 Oe.

        284 KINETICS OF OOCYTE MATURATION AND SUBSEQUENT DEVELOPMENT OF PARTHENOGENETIC PORCINE EMBRYOS AFTER MEIOTIC INHIBITION WITH ROSCOVITINE

        <P> Maturation of mammalian oocytes is a very important process for subsequent embryo development after fertilization. Prolonged maturation time by meiotic inhibitors could be an effective method for improvement in the meiotic and developmental competence of mammalian oocytes. Roscovitine, a cyclin dependent kinase inhibitor, is known to specifically inhibit M-phase promoting factor (MPF) kinase activity and prevent the resumption of meiosis. The aim of this study was to examine the effect of roscovitine on the maturation and subsequent development of porcine oocytes. Ovaries were collected from slaughtered prepubertal gilts and COCs were aspirated from 2- to 5-mm antral follicles. In control, porcine cumulus oocyte complexes (COCs) were cultured in the maturation medium (TCM-199 supplemented with 0.3% BSA, 1 μg/mL FSH, 1 μg/mL LH, and 10 ng/mL EGF) for 44 h. In the experimental group, COCs were cultured in the inhibition medium (TCM-199 supplemented with 0.3% BSA and roscovitine) for 24 h, and then further cultured in the maturation medium for 44 h. Matured oocytes from both groups were activated by electrical pulse (1.2 kV/cm for 30 μs), and then cultured in PZM-3 medium for 6 days. Apoptotic cells in blastocysts were detected by TUNEL assay and total cell number was examined by propidium iodide (PI) counterstaining. Data were analyzed by chi-square and Student's t-test. The first experiment was conducted to determine the effect of roscovitine (0, 12.5, 25, 50, and 100 μM) on meiotic inhibition of GV oocytes. This effect was dose-dependent, and a concentration of 50 μM was sufficient to prevent meiotic resumption in 79.2% (76/96, 5 replicates) of the porcine oocytes after 24 h of culture when compared to 0 (15.4%, 15/97), 12.5 (32.1%, 36/112), 25 (57.4%, 54/94), and 100 μM (77.8%, 77/99). The second experiment was carried out to examine the kinetics of maturation of roscovitine-treated porcine oocytes. The concentration of roscovitine used was 50 μM. A total of 75.8% (50/66, 3 replicates) of roscovitine-treated oocytes reached metaphase II stage compared with 70.8% (46/65) of control. The third experiment was performed to compare embryo development between control and treated group after parthenogenetic activation. No differences (P > 0.05) were found between the control and the treated group in cleavage rate (77.2%, 132/171 vs. 68.0%, 115/169), blastocyst rate (26.9%, 46/171 vs. 17.8%, 30/169), and total (33.7 ± 12.4 vs. 35.1 ± 12.6) and apoptotic (2.2 ± 2.4 vs. 2.2 ± 1.2) cell number per blastocyst (4 replicates). The results suggest that roscovitine can be used to prolong maturation time of porcine oocytes without reducing meiotic maturation but also without significantly decreasing their subsequent developmental competence. Further studies are necessary to improve the developmental competence of porcine oocytes treated with roscovitine.</P>

        A new 3D-PTV algorithm (a Volume PTV) based upon a hybrid fitness function has been constructed. A coherency fitness function is introduced using the information of space and time to sort out the correct particle pairs between the two camera images. The measurement system consists of two-high-definition-cameras(1k × 1k), a Nd-Yag laser and a host computer. The developed algorithm has been employed to investigate the flow features of the cylinder wake. The Reynolds numbers with the cylinder diameter (d=10㎜) are 360, 720, 900 and 1260. Two-dimensional displacements of the particles of each camera’s image and neighbouring constraints were introduced to reduce the calculation loads. More than 10,000 instantaneous 3D vectors have been obtained by the constructed algorithm. The constructed algorithm could recover more than 80~90% of the particle numbers in the image.
