      • Lipoprotein 정량에 미치는 염색에 대한 실험적 검토

        From the results as compared with changes in lipoprotein fractions on the oil red O and sudan black B stainings followings were obtained: 1. In sudan black staining beta-lipoprotin fraction was shown 61.38%, pre-beta 23.13% and alpha 15.50%, while in oil red O staining 58.00%, 20.13% and 21.88%, respectively. Increased figures showed on the beta and pre-beta lipoproteins of former, while increased alpha-lipoprotein of latter showed. 2. Among lipoprotein-cholesterol fractions the most increased fraction was betalipoprotein. And as the other 2 fractions pre-beta and alpha-lipoproteins were 11.24% and 12.64 %, respectively there were, however, showed similar values. 3. The lipid was distributed as beta-lipoprotein was 63.54%, pre-beta 23.27% and alpha 13.09%.

      • 흰쥐에서 Paraquat 독성에 미치는 Selenium의 영향

        In order to investigate effects of selenium on paraquat-induced toxicities. we measured the enzymes involved in the glutathione metabolism, which Includes glutathione s-transferase, r-glutamylcysteine syntheatse and r-glutamyltranspeptidase, and the contents of glutathione in the paraquat-treated and paraquat plus seleniun-treated groups, respectively. 1. The survival rate was great)y diminished in paraquat plus selenium-treated group compared with paraquat-treated group. 2. The content of 81utathione in paraquat plus selenium-treated group was also reduced to 17.5% and 31.7% in liver and kidney, respectively as compared to the corresponding control values. 3 The activity of r-glutamylcysteine synthetase in paraquat-plus selenium group was significantly increased in liver but not affected in the kidney. 4. The activities of glutathione s-transferase in liver and kidney were not affected in both groups. 5. The activity of r-glutamyl transpeptidase in kidney in was increased to 54% in the paraquat plus selenium groups as compared to that of the control group. These results taken together suggest that the coadministration of selenium and paraqua enhances paraquat-induced toxicities, which appear to be associated with the decreased level of glutathione in liver and kidney.

      • Sol-Gel 반응을 통한 Surlyn/유기 실리케이트 나노복합재료 제조 및 물성에 관한 연구

        Transparent organic-inorganic hybrid material was synthesized via the sol-gel method using polyethylene-co-methacrylic acid(PEMA) ionomer and a metal alkoxide, tetraethylorthosilicae(TEOS). The resulting material was characterized using ^(29)Si solid-state nuclear magnetic resonance spectroscopy results prove that silica is formed via the sol-gel reaction. Scanning electron microscopy analysis (SEM) , transmission electron microscopy (TEM) studies indicate that silica particle size is in nanoscopic level (70nm average size), and silica nanoparticles are uniformly dispersed in the polymer matrix after the sol-gel process. Theremogravimetric analysis (TGA) results show that the silica content of the hybrid. The thermal stability of the ionomer increases after hybridization. Material testing machine of the hybrid shows that the stress of ionomer increase after the sol-gel reaction.

      • Hb F 측정법에 관한 연구

        Hb F(태아혈색소)는 태생기의 대부분을 차지하는 혈색소로서 출생 직후 약 80%의 함량을 보이나 이후 점차 감소하여 출생 1년에 2.9%, 2년에 1.8%, 3년 이후에는 정상성인과 같은 수치를 갖는다. Hb F의 측정법은 Hb F가 구조상 alkali에 대한 저항력이 높으므로 Hb A와 쉽게 구별되는 점을 이용하여 측정하였으나 Hb F에 대한 특이성이 없으며 이외 여러가지 단점을 갖고 있다. acid clution법은 Hb A의 산에 의한 용출을 이용한 것으로 남아있는 Hb F를 염색하여 계산하는 것으로 세포간에 함량의 차이를 갖는다든지 그외 여러 단점을 내포하고 있다. 전기영동법을 이용한 방법은 처음에는 여지를 사용하였으나 시간이 오래 소요되며 해독력이 좋지 않았으므로 널리 이용되지 못하였다. 근래에 이르러 한천젤이나 cellulose acetate의 보급으로 여지를 대신하게 되면서 전기영동법이 Hb 측정에 이용되었다. 특히 한천젤 전기영동은 한천이 갖는 특성 때문에 densitometry에도 적합하므로 더욱더 고무적이다.

      • 흰쥐에서 paraquat독성에 미치는 tumor necrosis factor-α의 영향

        Background : The toxicity of paraquat is due to the oxygen-derived radicals formed by the reaction of oxygen with bipyridylium radical cations. Tumor necrosis factor (TNF)-α induces the synthesis of mitochondrial manganese superoxide dismutase (Mn-SOD) and other protective proteins in a variety of tumor cells and these protein contribute to a protect tumor cells from free radical injury. In this study, the effects of TNF-α on paraquat-induced toxicity were investigated in rats. Methods : Experimental animal used was Sprague-Dawley male rat, body weight 150-200g. The paraquat toxicity was determined by survival rate. The SOD, catalse, and glutathione peroxidase(GSHPx) activity were spectrophtomerically determined by Crapo method, Aebi method and Flohe method, respectively. Results : The survival rate of paraquat-intoxicated rats was decreased by subctaneous injection of TNF-α. However, the survival rate of paraquat injected rats were increased by pretreatment of TNF-α, comparing to saline pretreatment. The liver SOD activity was increased significantly by TNF-α treatment, but kidney and lung enzyme was unaltered in TNF-α treated-rats, comparing to normal control rats. The increment of liver SOD activity by TNF-α treatment was dose- and time-dependant SOD activity of kidney and lung of rats were not changed by TNF-α injection. The activities of catalase and glutathione peroxidase and contents of thiobarbituric acid reactive substance(TBARS) in liver were not changed by TNF-α treatment. Conclusion : From these result, TNF-α potentiate paraquat toxicity in rats but TNF-α pretreatment reduce paraquat induced toxicity. Protective effect may be resulted from the increased liver SOD activities by TNF-α.

        The removal of colours from aqueous solution and dispersions has been studied by dispersed-air flotation, in a semi-batch column. Two colours were used for the experiments: Basic Yellow 28 (basic) and Direct Orange 31 (basic). All two were effectively removed by flotation within 8 min. Sodium dodecyl sulfate, sodium oleate and amines were found to be effective as collectors in the removal of colour, which was found to be related to the pH of the solution and the amount of collector added to it with high collector dosages causing the process to become pH-independent.

      • 가토 적혈구에서 철이온에 의해 유도되는 지질과산화에 미치는 paraquat의 영향

        The participation of superoxide in initiating tissue damage from admini stration of a xenobiotics is best illustrated by paraquat intoxication, in which it is known that one electron reduction of para quat leads to the formation of radical species which react with molecular oxygen to give superoxide. Although it has also been argued that superoxide itself is not toxic, the shunting of electrons into the futile reduction of oxygen to hydrogen peroxide can be considered as a toxic process. In the present study, the effect of paraquat on lipid peroxidation in rabbit erythrocyte was investigated, Incubation of a buffered aerobic mixture of erythrocyte in the presence of Fe^(2+) and glucose or NADH resulted in the formation of lipid peroxide as determined as malonydialdehyde. Paraquat inhibited Fe^(2+) induced lipid peroxidation in rabbit erythrocyte. In contrast, NADH and glucose stimulate the Fe^(2+) induced lipid peroxidation of erythrocyte in the presence of paraquat and the levels of erythrocyte NADH and NADPH were decreased by paraquat. These results suggest that the inhibitory effect of paraquat on FeU induced lipid peroxidation in rabbit erythrocyte is mainly due to a decrease in the level of NADH and NADPH.

      • 방사선을 조사한 생쥐 장기에서의 Superoxide Dismutase 분포에 대한 연구

        The cytosolic and mitochondrial supreoxide dismutases (EC 1.15.1.1) were investigated in various organs, thyroid gland, lung, heart, liver, spleen and kidney, of normal and irradiated mice. The mouse organs were fractionated into cytosolic and mitochondrial fractions by differential centrifugation, and the supsroxidp dismutase was measured by the method of McCord and Fridovich. The mouse organs contain two types of superoxide dismutase ; one is localized in the cytosol and contains cupper and zinc, the other in the mitochondria and contains manganese. Superoxide dismutasc activity was found in all organs of mouse investigated, and maximal activity of enzyme was observed in liver. most of superoxide dismutase activity, 79-86%, was present in the cytosolic fraction, and the rest was associated with mitochondria. The activity of cytosolic superoxide dismutase was most prominent in the liver among those tissues assayed. However, that of mitochoadrial enzyme was highest in the heart. Thc specific activity of superoxide dismutase in mitochondria was found about 2-5 folds greater than that of the enzyme in cytosol and cyanide inhibit the activity of cytosolic enzyme but not mitochondrial enzyme, In irradiatcd mice, the enzyme activity and the specific activity of superoxide dismutase in cytosol and mitochondna of various organs were decreased with minimal value on 7th and l4th day after exposured by whole body-irradiation.
