        생체내 흰쥐 해마 CA1 세포에서 가바성 억제에 대한 전기생리학 특성

        가바성 억제 신경세포는 해마의 정상적인 기능을 조절하는데 중요한 역할을 하며 해마 병변을 유발하는 중요한 요소이다. 본 연구는 in vivo실험법을 사용하여 해마 CAI영역에서의 전기 생리학적 반응을 측정함으로써 가바성 신경세포의 기능을 분석하고 이를 슬라이스 실험법과 비교하고자 하였다. Fimbria-fonix 전기자극시 전형적인 Population spike가 나타났고 10μm bicuculline 존재하에서는 전기자극에 의해 burst 형태의 population spike가 나타났다. Population spike의 크기는 자극 강도에 비례하였으며 그 숫자도bicuculline 전극사용시와 같이 동일한 양상을 보였다. CAI영역의 흥분성 수준을 측정하기 위해 paired-pulse자극을 하였는데 짧은 자극 간격에서 억제성 반응을 보였고 burst형태의 afterdischarge를 나타내었다. CAI영역에서 in vivo 실험법을 사용한 가바성 신경세포반응의 결과는 추체세포의 흥분성 조절을 효과적으로 분석할 수 있으며 in vitro 실험법에 비해 기능적 평가가 더욱 이상적임을 알 수 있다. Inhibitory cells are critically involved in shaping normal hippocampal function and are thought to be important elements in the development of hippocampal pathologies. The present study was carried out in hippocampal CA1 area in vivo to compare with hippocampal slice studies. Intracellular and extracellular recordings with or without bicuculline electrodes were obtained in the intact brain of anesthetized rats, and cells were intracellularly labelled with neurobiotin. Electrical stimulation of fimbria-fornix resulted in an initial short-latency population spike. In the presence of 10μM bicuculline, orthodromic stimulation resulted in bursts of population spike. The amplitude of population spikes in the CA1 region increased with stimulus intensity. as did the number of population spikes when the field recording electrode contained 10μM bicucilline. We measured the level of excitability in the CA1 area, using a paired-pulse stimulus paradigm to evoke population spokes. Population spikes showed strong paired-pulse stimulus. These result suggest that hippocampal CAI inhibitory interneurons can affect the excitabillity of pyramidal neurons that can not be appreciated in conventional in vitro preparation.

        적외선열화상장치를 이용한 Buchanan plugger 표면의 온도상승 분석

        This study was performed to evaluate the temperature rise on various position of the Buchanan plugger, the peak temperature of plugger's type and the temperature change by its touching time of heat control spring. The heat carrier system 'System B' (Model 1005. Analytic Technologies, USA) and the Buchanan's pluggers of F, FM, M and ML sizes are used for this study. The temperature was set to 200℃ which Dr. Buchanan's "continuous wave of condensation" technique recommended on digital display and the power level on it was set to 10. In order to apply heat on the Buchanan's pluggers, the heat control spring was touched for 1, 2, 3, 4 and 5 seconds respectively. The temperature rise on the surface of the pluggers were measured at 0.5 mm intervals from tip to 20 mm length of shank using the infraed thermography (Radiation Thermometer-IR temper, NEC San-ei Instruments, Ltd, Japan) and TH31-702 Data capture software program (NEC San-ei Instruments, Ltd, Japan). Data were analyzed using a one way ANOVA followed by Duncan's multiple range test and linear regression test. The results as follows. 1. The position at which temperature peaked was approximately at 0.5 mm to 1.5 mm far from the tip of Buchanan's pluggers (p<0.001). The temperature was constantly decreased toward the shank from the tip of it(p<0.001). 2. When the pluggers were heated over 5 seconds, the peak temperature by time of measurement revealed from 253.3±10.5℃ to 192.1±3.3℃ in a touch for 1 sec, from 218.6±5.0℃ to 179.5±4.2℃ in a touch for 2 sec, from 197.5±3.0℃ to 167.6±3.7℃ in a touch for 3 sec, from 183.7±2.5℃ to 159.8±3.6℃ in a touch for 4 sec and from 164.9±2.0℃ to 158.4±1.8℃ in a touch for 5 sec. A touch for 1 sec showed the highest peak temperature, followed by, in descending order, 2 sec, 3 sec, 4 sec. A touch for 5 sec showed the lowest peak temperature (p<0.001). 3. A each type of pluggers showed different peak temperatures. The peak temperature was the highest in F type and followed by, in descending order, M type, ML type. FM type revealed the lowest peak temerature (p<0.001). The results of this study indicated that pluggers are designed to concentrate heat at around its tip, its actual temperature dose not correlate well with the temperature which Buchanan's "continuous wave of condensation" technique recommended, and finally a quick touch of heat control spring for 1 sec reveals the highest temperature rise.

        This study was conducted to investigate the reference value of mercury (Hg) in liver and kidney of Korean population. The mercury concentration in 244 'sudden and unexpected death' autopsies (male: 180, female: 64) aged from 0 to87 years was analyzed. The concentration of mercury was measured by atomic absorption spec(Perkin -Elmer Model 5100) with mercury/hydride generating system (FIAS 400). The contents of mercury in liver and kidney fitted well the log-normal distribution rather than normal distribution. Geometric mean concentration of mercury in liver and kidney was 0.115 ㎍/g wet weight and 0.149 μg/g wet weight, respectively. Geometric mean concentration of mercury in female was higher than in male (p<0.01). The mercury content in liver and kidney increased with age up to the forties and slightly decreased there-after, The regression model of mercury deposit in liver and kidney by age was predicted as the following equation : Log KHg=-1.0576+0.0045·Age -0.0001·Age²+0.0873·Sex, Log KHg=-1.0576+0.0152·Age-0.0002·Age²+0.1935·Sex. The liver burden of mercury was estimated to be 158.3~161.3μg in male and 163.0-166.9 ㎍ in female. The kidney burden of mercury was estimated to be 42.0~42.9 ㎍ in male and 55.5 ∼57.1 μg in female.

      • DCT를 이용한 영상의 윤곽선 추출을 위한 양자화 테이블 비교분석

        본 논문에서는 DCT(discrete consine transform)를 이용하여 영상 데이터를 공간 영역에서 주파수 영역으로 변환하여 주파수 영역별로 분해한 뒤, 윤곽선 추출과 압축을 동시에 하기 위한 여러 양자화 테이블을 적용하고, IDCT(Inverse DCT)를 이용하여 복원한 결과를 비교 분석함으로써, 보다 정확한 윤곽선을 가진 영상을 추출할 수 있는 양자화 테이블을 구했다. 이 양자화 테이블을 DCT 처리된 영상에 적용했을 때 윤곽선과 압축 효과를 가진 영상을 얻을 수 있었다. In this paper, we transform image data from spatial domain to frequency domain using DCT, decompose it into each frequency band. We apply various quantization table for edge detection and compression simultaneously. We researched a quantization table for extracting image with exact edge, as compare and analyze restored images using IDCT. When we applied this quantization table to the DCT transformed image, acquired the image with edge and compression effect.

      • 경직성 뇌성마비아동의 도형-배경 색상역전과 시지각 및 시각-운동 수행능력

        The present study was designed to investigate the effect of color reversal of figure-ground on visual perceptional and visuo-motor performance of cerebral palsied. The 20 spastic cerebral palsied and the 20 non-cerebral palsied children participated in the study as subjects. Perceptual subtest of perceptuo-Motor Development Diagnostic Test(PMDT) and Beery-Buktenica Development Test of Visual-Motor Intergration(VMI) were administered to assess visual perceptual and visuo-motor performance, respectively. The main conclusions from the study were as follows. First, the cerebral palsied subjects performed more poorly in both visual perceptional and in visuo-motor tasks than non-cerebral palsied counterparts. However, it was revealed that the relatively poor performance of the former in visual perceptual tasks were primarily due to Synthesis of Figure factor. Second, for spastic cerebral palsied subjects, the color reversal of testing materials had the facilitating effect on visual perception but not on visuo-motor. However, any improvement as a function of color reversal was evidenced neither in visual perceptioanl nor in visuo-motor performance of the non-cerebral palsied. Finally, presentation order did not produce significant improvement of visual perceptional and visuo-motor performance in either group. It suggest thath improvement by color reversal did not result from presentation order.

        흰쥐 안면신경핵 세포의 전기생리학적 및 형태학적 특성

        흰쥐의 안면신경핵을 구성하는 신경세포들의 시냅스 연결 양태 및 세포막 특성을 규명하기 위해 in vivo 필드전위 및 세포내 전위 측정법을 이용하여 전기생리적 반응을 관찰하였다. 말초 안면신경 분지를 역행성으로 전기자극시 자극세기에 비례하여 전위의 크기가 증가되었고 필드 전위의 양태는 두 가지 반응으로 나타났는데 전기자극 직후 1ms 부근에서 정점을 나타내는 양태와 이와 더불어 7~8ms 부근에서 후기 정점을 동반하는 양태가 있었다. 안면신경핵은 염색시 내측, 배외측, 중간측 및 외측등 4부분의 소핵으로 구분되었다. Neurobiotin으로 채워진 단일 신경세포를 형태학적으로 재구축하였는데 세포체는 추체형태를 나타내었고 주 수상돌기는 모든 방향으로 뻗어져 있었고 각 수상돌기의 영역은 해당 소핵 내에 한정되어 있었다. 일련의 과분극 전류(-1.2~+1.2nA)를 세포내에 가하였을 때 동반되는 세포내 전위변화를 입력저항 값으로 계산하였을 때 그 기울기가 직선형으로 나타났다. 탈분극 전류를 세포내 주입시 지속적인 활동성 전위가 나타났으며 전류의 크기에 비례하여 각 전위의 개수가 증가하였고 spike-빈도 적응 현상이 나타났다. 그러나 시간 의존성 내향성 정류현상은 관찰되지 않았고 anodal break excitation이 나타났다. 이상의 실험결과로 보아 안면신경핵을 구성하고 있는 세포들 사이의 시냅스는 다양한 형태로 존재할 가능성이 있다고 사료되며 이들 시냅스간의 변화를 통하여 안면 신경마비, 반쪽 안면 경련, hypoglossal-facial anastomosis등에서 나타날 수 있는 임상적 신경성 증상 기전을 설명할 수 있을 것으로 여겨진다. 주요어 : 안면신경핵, 시냅스, 신경세포막 특성, 안면신경마비, 반쪽 안면 경련 This study used in vivo intracellular and extracellular fIeld potential recording to evaluate the intrinsic membrane properties and connection pattern within facial nucleus. 1.There were four subdivisions of medial, intermediate, lateral, and dorsolateral in facial nucleus. 2.Principal cells in the facial nucleus was recorded from and filled with neurobiotin in anesthetized rats. The extent of their dendrites and the characteristics of cell body were examined. 3.Principal cells had a large amplitude action potential and afterhyperpolarization was followed a single action potential. 4.The response from facial motonucleus to electrical stimulation of the facial nerve was mainly a monophasic wave, with a latency of 1 msec, which was assumed to reflect antidromic activation of facial motoneurons. In some of rats the response in addition showed late components at a latency of about 7-8 msec, but its amplitude was small. 5.Mest of cells exhibited accommodation of spike discharge upon depolarization of membrane by 0.8 nA for 400 ms. Our results support the hypothesis that there normally are weak connections between different parts of the facial motonucleus to explain pathophysiology of hemifacial spasm and facial nerve paralysis.

        To detect shapes in noisy data, the fuzzy Hough transform is introduced. This technique finds shapes by approximately fitting the data points, which avoids the spurious shapes detected when using the conventional Hough transform. An efficient implementation of this method is found in detecting lines and circles.

      • TiN 코팅이 전기도금한 교정용 스테인레스스틸 선재의 세포독성에 미치는 영향

        This study was performed to investigate whether TiN coating could reduce the cytotoxicity of electroplated orthodontic stainless steel wire which had been known to be cytotoxic. Nickel plating was done on stainless steel wire using a electroplating technique. Some of the electroplated wires were coated by TiN film and 4 groups of specimen were prepared (Non-coated electroplated wire, TiN coated electroplated wire, Titanium which was known to have no cytotoxicity, Copper which was known to have high cytotoxicity). Each wire was incubated for 72 hours on the medium. The release of the metal ion was measured by ICP-AES. The incubated medium was diluted 75 %, 50%, 25 % and added on each microplate with Balb/c 3T3 mouse fibroblast for MTT analysis and compared with medium only. Through the measuring of the change in absorbency value of each wire group and the difference of absorbency value according to change of dilution, following results were obtained. 1. The results of ICP-AES analysis showed that lots of nickel ions were isolated from non-coated wires but not isolated from TiN coated wires. 2. The inverted microscope showed that there was a decrease in cell count in non-coated wires but no significant change in TiN coated wires. 3. The results of MTT analysis showed that there was no difference in the absorbency value between TiN coated wires and medium and between titanium and medium but the absorbency value of non-coated wires and copper were lower than that of medium only at all dilution and showed statistically significant differences(p<0.001). 4. Assessment as per ISO 10993, part 5, non-coated wires and copper investigated corresponded to "moderately cytotoxic" and "highly cytotoxic" ; whereas titanium and TiN coated wires were alloted to "non-cytotoxic". The results of the present study indicate that the cytotoxicity of the electroplated orthodontic wire can be changed from "moderately cytotoxic" to "nontoxic" by TiN coating and electroplated wire can be used clinically without the concern of cytotoxicity.
