        D-RADA16-RGD-Reinforced Nano-Hydroxyapatite/Polyamide 66 Ternary Biomaterial for Bone Formation

        BACKGROUND: Nano-hydroxyapatite/polyamide 66 (nHA/PA66) is a composite used widely in the repair of bone defects. However, this material is insufficient bioactivity. In contrast, D-RADA16-RGD self-assembling peptide (DRADA16- RGD sequence containing all D-amino acids is Ac-RADARADARADARADARGDS-CONH2) shows admirable bioactivity for both cell culture and bone regeneration. Here, we describe the fabrication of a favorable biomaterial material (nHA/PA66/D-RADA16-RGD). METHODS: Proteinase K and circular dichroism spectroscopy were employed to test the stability and secondary structural properties of peptide D-RADA16-RGD respectively. Scanning electron microscopy (SEM) and transmission electron microscopy (TEM) were used to characterize the surface of these materials. Confocal laser scanning (CLS), cell counting kit-8 tests (CCK-8), alizarin red S staining, cell immunofluorescence analysis and Western blotting were involved in vitro. Also biosafety and bioactivity of them have been evaluated in vivo. RESULTS: Proteinase K and circular dichroism spectroscopy demonstrated that D-RADA16-RGD in nHA/PA66 was able to form stable-sheet secondary structure. SEM and TEM showed that the D-RADA16-RGD material was 7–33 nm in width and 130–600 nm in length, and the interwoven pore size ranged from 40 to 200 nm. CLS suggests that cells in nHA/PA66/ D-RADA16-RGD group were linked to adjacent cells with more actin filaments. CCK-8 analysis showed that nHA/PA66/ D-RADA16-RGD revealed good biocompatibility. The results of Alizarin-red S staining and Western blotting as well as vivo osteogenesis suggest nHA/PA66/D-RADA16-RGD exhibits better bioactivity. CONCLUSION: This study demonstrates that our nHA/PA66/D-RADA16-RGD composite exhibits reasonable mechanical properties, biocompatibility and bioactivity with promotion of bone formation.

        중국의 한국어『정독』과목에 대한 요구 분석 -졸업 진로에 대한 요구를 중심으로

        Korean Language Education has been classified into Minor Language Education at universities in China. Differs from Major Language Education such as English, Korean Language Education however, has unique difficulties including shorter educational time duration, as well as larger educational planned duties. Besides, students have to face lingual influences from Mandarin, English, and even local languages and languages from ethnic groups. Even though, the social reality requires Korean Language students achieving similar advanced level compare with those whom major in English after undergraduate school. One of the most significant questions for discussion in Korean Language Education studies, therefore, should emphasize the way of co-existence of improving teaching efficiency and ensuring quality teaching content. As the subject occupies the largest time duration in Korean Language Education, Intensive Reading therefore, has gained significant impact on improving teaching quality as well as efficiency. In order to improve the efficiency and practicability of Korean Language Education, this article would accurately demonstrate the contents in need for intensive readings, by conducting series of survey researches from students of Korean Language Education from terms of 2007, 2008, and 2009, respectively.

        중국 내 학원에서의 한국어 학습자의 학습 실태 분석

        1992년 중·한 수교 이후, 한국 드라마의 유입과 한류 열풍으로 중국 각지에서 한국어 학원이 우후죽순처럼 생겨났다. 한국어 학원 교육은 대학교의 한국어정규교육에 비해 그 수가 많고 분포지역이 넓으며 학습자가 다양하고 학습동기도 다양해, 전국의 한국어학원 학습자를 통합적으로 분석하기는 어렵다. 이 때문에. 본고에서는 중국 한 도시의 한국어학원 학습자를 대상으로 하여 무작위 설문조사를 했다. 설문내용은 크게 세 부분으로 나눌 수 있다. 첫 번째 부분은 학습자의 개인정보이다. 이 부분을 통해 학원에서 한국어를 학습하는 학습자의 성별, 나이, 직업 및 수업 이외의 한국어 학습시간 등 주요 특징을 살펴보고자 한다. 두 번째 부분은 학습동기 유형이다. Gardner & Lam bert는 사회언어학적인 시각에서 학습동기를 통합적인 동기(integrative movation)와 도구적인 동기(instrumental motivation)로 나누었다. 본고에서는 학습동기의 이런 분류를 근거로 하여 학원에서 한국어를 학습하는 학습자들의 학습동기를 살펴보고자 한다. 세 번째 부분은 학습 방식이다. 이 부분에서는 학원에서 한국어를 학습하는 학습자들이 선호하는 수업 외 시간의 학습방식을 생활적 방식과 학교적 방식 두 가지로 나누어 살펴보고자 한다. 이 연구의 결과가 중국 내의 한국어 학원에서 공부하는 한국어학습자들의 학습상황을 밝혀, 중국 내 한국어학원의 설립과 발전에 도움이 되기를 바란다. In 1992 China and South Korea established diplomatic relationship which paved the path for the cultural communication between them. Since then "Korean Wave" culture rushed to China along with the acceptance and favor of Korean dramas among Chinese, meanwhile Korean language institutes have mushroomed. However compared to formal Korean language education units Korean language institutes have the following characteristics: large numbers, wide spread, diversity of study motivation and personal situations. Given these reasons; it is imperative to complete statistical analysis on study motivations of language learners in China. Hence this research focuses on a study on Korean language enthusiast learners in a city in China via sampling survey in terms of three aspects. First, individual personal information including gender, age, occupation and study time to find out the basic features of enthusiast learners in Korean language institutes. Second, learning motivation patterns: referring to Gardner & Lam bert`s theory of study motivation in social linguistics contexts. Korean language enthusiast learners` motivation falls into the following two types: integrative motivation and instrumental motivation. Third, study patterns: this parts involves research on learners` study patterns during their spare time which will be divided into survival pattern and academic pattern. This study aims to improve comprehension of the study situation of Korean language enthusiast learners` in China and therefore attributing to the development of Korean language institutes.

        Effects of α-Galactosidase Supplementation on Performance and Energy Metabolism for Broilers Fed Corn-non-dehulled Soybean Meal Diets

        To study the effects of ${\alpha}$-galactosidase (${\alpha}$-Gal) supplementation on performance and energy metabolism, 216 Arbor Acres male broilers were placed in 36 cages of 6 birds each and allotted to 4 diets for 42 d, with 0-21 d as starter period and 22-42 d as grower period. The 4 diets were based on corn non-dehulled soybean meal in a $2{\times}2$ factorial arrangement, with 2 levels of ${\alpha}$-Gal (0 vs. 60 U/kg feed) and 2 levels of ME (normal metabolizable energy (NME) and low metabolizable energy (LME)). Bird performance was obtained at 21 and 42 d of age with samples of feces collected for nutrient digestibility from 19-21 d and 40-42 d. At 21 and 42 d, 1 bird from 6 cages of each treatment was killed to determine liver weight, intestinal pH and chyme viscosity. With the addition of ${\alpha}$-Gal the 42 d body weight (BW) and 0-42 d average daily gain (ADG) were significantly improved (p<0.05). Average daily feed intake (ADFI) of birds fed the LME diet was significantly increased compared to those fed the NME diet during starter (p<0.01) and grower (p<0.05) periods and overall (p<0.01). There was an interaction of ${\alpha}-Gal{\times}ME$ on 0-21 d ADFI (p<0.01). Supplementation of ${\alpha}$-Gal significantly improved (p<0.01) feed efficiency during the grower period and overall. Feed efficiency of birds fed the LME diet was significantly decreased (p<0.05) compared to those fed the NME diet during the starter period and overall. With the addition of ${\alpha}$-Gal apparent metabolizable energy (AME) was improved (p<0.01) by 2.1% and 1.8% during starter and grower periods, respectively. There was a main effect (p<0.05) of ${\alpha}$-Gal on the digestion of neutral detergent fiber (NDF) during the starter period and crude protein (CP), NDF and acid detergent fiber (ADF) during the grower period. With the addition of ${\alpha}$-Gal, the relative weight of liver was reduced (p<0.01) during the two phases. The duodenal and jejunal pH were significantly decreased (p<0.01) with the supplementation of ${\alpha}$at the two phases. ${\alpha}$-Gal addition reduced (p<0.01) chyme viscosity of the ileum during the starter and grower periods. Overall, ${\alpha}$-Gal showed a major effect on nutrient efficiency, improved ADG and feed efficiency, whereas LME decreased feed efficiency. The incorporation of ${\alpha}$-Gal into a LME diet could at least partially offset ME deficiency of non-dehulled soybean meal.

        Expression and evaluation of porcine circovirus type 2 capsid protein mediated by recombinant adeno-associated virus 8

        Background: Porcine circovirus type 2 (PCV2) is an important infectious pathogen implicated in porcine circovirus-associated diseases (PCVAD), which has caused significant economic losses in the pig industry worldwide. Objectives: A suitable viral vector-mediated gene transfer platform for the expression of the capsid protein (Cap) is an attractive strategy. Methods: In the present study, a recombinant adeno-associated virus 8 (rAAV8) vector was constructed to encode Cap (Cap-rAAV) in vitro and in vivo after gene transfer. Results: The obtained results showed that Cap could be expressed in HEK293T cells and BABL/c mice. The results of lymphocytes proliferative, as well as immunoglobulin G (IgG) 2a and interferon-γ showed strong cellular immune responses induced by Cap-rAAV. The enzyme-linked immunosorbent assay titers obtained and the IgG1 and interleukin-4 levels showed that humoral immune responses were also induced by Cap-rAAV. Altogether, these results demonstrated that the rAAV8 vaccine Cap-rAAV can induce strong cellular and humoral immune responses, indicating a potential rAAV8 vaccine against PCV2. Conclusions: The injection of rAAV8 encoding PCV2 Cap genes into muscle tissue can ensure long-term, continuous, and systemic expression.

        Privet golden leaves adapt unexpectedly well to light changes

        Golden-leaf privet ( Ligustrum × vicaryi ) is widely used as a horticultural shrub because of its upper golden leaves, butits lower leaves are green. However, the putative mechanisms of its upper golden leaves and the leaf color changes inresponse to light shifts have not been well studied so far. Here, chlorophylls (Chl), carotenoids, and Chl precursors from bothgolden and green leaves grown in full sunlight (approximately 1200 μmol photons m −2 s −1 at noon) or low-light conditions(180 μmol m −2 s −1 ) were determined spectrophotometrically. In addition, their gas exchange parameters and Chl fl uorescencewere measured in situ. Metabolic fl ux analysis of chlorophyll intermediates indicated that the conversion of prochlorophyllideto chlorophyllide was signifi cantly blocked in golden leaves when compared with green leaves. Green leaves showed higherphotosynthetic capacity in low light than golden leaves, but golden leaves presented unexpectedly stronger photosyntheticcapacity and lower reactive oxygen species accumulation under the high-light condition. Furthermore, golden leaves showeda higher level of nonphotochemical quenching (NPQ) after the light-to-dark shift and presented a stronger adaptive abilityto a broad range of light environments. Higher NPQ values and less oxidative damage in golden leaves may be correlatedwith their higher carotenoid levels. The results imply that lower chlorophyll levels and higher carotenoid levels in canopyleaves may help privet plants acclimate better to illumination changes. This study demonstrates the key role of irradiance ingenerating the two types of Ligustrum × vicaryi leaves and sheds a light on cultivation of other ornamental foliage plants.

      • The small GTPase CDC42 regulates actin dynamics during porcine oocyte maturation

        <P> The mammalian oocyte undergoes an asymmetric division during meiotic maturation, producing a small polar body and a haploid gamete. This process involves the dynamics of actin filaments, and the guanosine triphosphatase (GTPase) protein superfamily is a major regulator of actin assembly. In the present study, the small GTPase CDC42 was shown to participate in the meiotic maturation of porcine oocytes. Immunofluorescent staining showed that CDC42 was mainly localized at the periphery of the oocytes, and accumulated with microtubules. Deactivation of CDC42 protein activity with the effective inhibitor ML141 caused a decrease in actin distribution in the cortex, which resulted in a failure of polar body extrusion. Moreover, western blot analysis revealed that besides the Cdc42-N-WASP pathway previously reported in mouse oocytes, the expression of ROCK and p-cofilin, two molecules involved in actin dynamics, was also decreased after CDC42 inhibition during porcine oocyte maturation. Thus, our study demonstrates that CDC42 is an indispensable protein during porcine oocyte meiosis, and CDC42 may interact with N-WASP, ROCK, and cofilin in the assembly of actin filaments during porcine oocyte maturation.</P>
