Molecular Characterization of Rathi and Tharparkar Indigenous Cattle (Bos indicus) Breeds by RAPD-PCR

        Sharma, Amit Kumar,Bhushan, Bharat,Kumar, Sanjeev,Kumar, Pushpendra,Sharma, Arjava,Kumar, Satish Asian Australasian Association of Animal Productio 2004 Animal Bioscience Vol.17 No.9

Random amplification of polymorphic DNA-Polymerase Chain Reaction (RAPD-PCR) analysis was carried out using DNA samples of 30 animals of Rathi cattle and 42 animals of Tharparkar cattle. Genomic DNA was isolated as per standard protocol and evaluated for its quality, purity and concentration. Twenty three random primers were screened out of which 15 primers yielded satisfactory amplifications and were used for further analysis. Average numbers of polymorphic fragments per primer were 7.07${\pm}$0.86 in Rathi and 6.80${\pm}$0.61 in Tharparkar cattle. The percentage of polymorphic bands in these two cattle breeds were 86 and 87%, respectively. Within breed genetic similarities for pooled over primers in the animals of Rathi and Tharparkar breeds were .577${\pm}$0.30 and 0.531${\pm}$0.02, respectively on the basis of band frequency (BF) and 0.645${\pm}$0.04 and 0.534${\pm}$0.04, respectively on the basis of band sharing (BS). Averages of between breed genetic similarities for pooled over primers were 0.97 and 0.92 according to BF and BS, respectively, which reflect higher degree of genetic similarity between Rathi and Tharparkar cattle breeds. Index of genetic distance based on BF and BS for pooled over primers was 0.030${\pm}$0.011 and 0.088${\pm}$0.031, respectively. Percentage of polymorphic bands and within-breed genetic similarities on the basis of band frequency (BF) and band sharing (BS) for pooled over primers revealed higher genetic similarity in Rathi than Tharparkar cattle population. High estimates of between breed genetic similarities for pooled over primers indicated that either Rathi is having decent from Tharparkar or both the cattle breeds are having common descent. Low value of Index of genetic distances between these two cattle breeds may be due to the fact that Rathi and Tharparkar cattle breeds are the native of Thar Desert in Northwest India. The results of between breed genetic distances also confirm the existence of high degree of genetic similarity between these two breeds of cattle.

A review on the mechanical properties of polymer composites reinforced by carbon nanotubes and graphene

        Amit Kumar,Kamal Sharma,Amit Rai Dixit 한국탄소학회 2021 Carbon Letters Vol.31 No.2

Compared to carbon nanotubes (CNTs), graphene possesses high strength due to wrinkled surface texture caused by a high density of surface defects which benefits more contact with the polymer material than a rolled-up CNT. In the present review, we have discussed and compared the various properties of CNTs (1-D) and graphene (2-D) obtained in experimental results. The effects of covalent and non-covalent functionalization of CNTs and graphene on the properties of its composites have also been reviewed and compared. A comparative analysis has been carried out between CNTs and graphene-reinforced polymer composites. Furthermore, the synergetic effects of CNTs and graphene hybrid nanofiller on the mechanical properties of polymer composites have also been briefly discussed. Finally, this review concludes with the potential application and future challenges are discussed with regards to filler and their polymer composites.

Black Carbon Concentration during Spring Season at High Altitude Urban Center in Eastern Himalayan Region of India

        Khushboo Sharma,Rakesh Kumar Ranjan,Sargam Lohar,Jayant Sharma,Rajeev Rajak,Aparna Gupta,Amit Prakash,Alok Kumar Pandey 한국대기환경학회 2022 Asian Journal of Atmospheric Environment (AJAE) Vol.16 No.1

This study analyzed the BC associated with PM1 and the contribution of biomass burning to the BC using a portable seven-channel Dual spot Aethalometer in and around Gangtok, the capital city of Sikkim, India, during April 2021. Additionally, CO2 and meteorological parameters (Temperature, Pressure, and Relative Humidity) was measured. The minimum concentration of BC was found in rural areas where the contribution of biomass burning to the BC is highest. The observed spatial variability of BC over Gangtok Municipal Corporation (GMC) area is minimal. Five days back-trajectory analysis was done using the Hybrid Single Particle Lagrangian Integrated Trajectory (HYSPLIT) model to understand the regional influences of air masses at Gangtok. The air mass of the studied region is under influence of trans-regional transport from Indo-Gangetic Plains affecting the BC concentration over the studied region. The black carbon presence in the ambient air near the glacier heights in the Eastern Himalayan region may significantly cause localized warming, thereby enhancing glacier melts. The results have significant bearing for the policy-makers to take corrective steps in addressing the issue of rising BC concentration in high altitude regions. A further detailed study is needed to examine the effect of BC on radiative forcing and its large-scale effect on the East Asian summer monsoon using regional climate models.

Effect of Magnesium Addition on Mechanical Properties of Al -Fly Ash Green Composite Produced under Green Ultrasonic Vibration Process

Pankaj Kr. Sharma,Shashi Prakash Dwivedi,Ajay Kumar,Amit Kumar Sharma 한국정밀공학회 2019 International Journal of Precision Engineering and Vol.6 No.3

In the present investigation an effort has been made to produce Al-Fly ash green composite using liquid state stir casting technique followed by green ultrasonic vibration such that it can improve wettability of fly-ash with aluminium by adding magnesium in the presence of argon gas. Addition of magnesium increases wettability of fly-ash particle in the molten aluminium and improves mechanical properties of Al-Fly ash green composite to a great extent. Incorporation of magnesium enhances the homogeneous allocation of fly ash particles in the pure aluminium alloy. More uniform distribution of Fly ash provides better mechanical properties. In this study, sample of Al/2 wt.% Mg/8 wt.% Fly ash provides most excellent outcome amongst all the selected composition. Microstructures show that the distribution of reinforcements is more uniform in this composition. TEM image shows proper wettability between aluminium and reinforcements. Density, cost estimation, hardness, toughness, ductility and tensile strength were also calculated to observe the result of magnesium and fly ash addition in aluminium matrix.

Estimation of carbon stocks of Balganga Reserved Forest, Uttarakhand, India

Amit Kumar,M.P. Sharma 한국산림과학회 2015 Forest Science And Technology Vol.11 No.4

Forest carbon (C) stock estimation was carried out for tree species of Balganga Reserved Forest (BRF) in district Tehri Garhwal, Uttarakhand, India. The highest total biomass density (TBD) and total carbon density (TCD) were estimated at site III in the altitudinal range of 18002600 m as 108.26 and 53.45 Mgha ¡1 followed by site II in the range of 16001800 m as 83.92 and 41.96 Mgha ¡1, and the least at site I in the altitudinal range of 10001400 m as 57.22 and 28.61 Mgha ¡1 with an average of 83.13 and 41.56 Mgha ¡1, respectively. The study found average TBD and TCD of the BRF are 23% more when compared to forests located in the same ecoregion due to difference in diameter at breast height and height of tree species. It also found that the C stock increases with increasing altitude due to fine root area, fine root length growth, and decreasing temperature. The BRF was found to have 16.65 million tons of forest organic C stocks in the year 2014. Accordingly, the C transported to the reservoir from the catchment undergoes aerobic/anaerobic degradation to release greenhouse gases to atmosphere. The study also reveals that oak, devdar, and fir have greater C sequestration capacity compared to other species found in BRF.

        Kumar Mongre, Raj,Sharma, Neelesh,Singh Sodhi, Simrinder,Ghosh, Mrinmoy,Kumar Singh, Amit,Kim, Nameun,Park, Yang Ho,Shin, Young Gyu,Kim, Sung Jin,Jiao Jiao, Zhang,Huynh, Do Luong,Jeong, Dong Kee EDITIONS SCIENTIFIQUES ELSEVIER 2017 BIOMEDICINE AND PHARMACOTHERAPY Vol.87 No.-

<P><B>Abstract</B></P> <P>Hepatocellular carcinoma (HCC) is a major threat to human health worldwide and development of novel antineoplastic drug is demanding task. BRM270 is a proprietary combination of traditional medicinal herbs, has been shown to be effective against a wide range of stem-like cancer initiating cells (SLCICs). However, the underlying mechanism and antitumor efficacy of BRM270 in human hepatocellular carcinoma (HCC) cells have not been well elucidated till date. Here we studied the tumoricidal effect of BRM270 on human-CD133+ expressing stem-like HepG-2 and SNU-398 cells. Gene expression profiling by qPCR and specific cellular protein expressions was measured using immunocytochemistry/western blot analysis. <I>In vivo</I> efficacy of BRM270 has been elucidated in the SLCICs induced xenograft model. In addition, 2DG-(2-Deoxy-<SMALL>D</SMALL>-Glucose) optical-probe guided tumor monitoring was performed to delineate the size and extent of metastasized tumor. Significant (<I>P</I> <I><</I> 0.05) induction of Annexin-V positive cell population and dose-dependent upregulation of caspase-3 confirmed apoptotic cell death by pre/late apoptosis. In addition, bright field and fluorescence microscopy of treated cells revealed apoptotic morphology and DNA fragmentation in Hoechst33342 staining. Levels of c-Myc, Bcl-2 and c-Jun as invasive potential apoptotic marker were detected using qPCR/Western blot. Moreover, BRM270 significantly (<I>P</I> <I><</I> 0.05) increased survival rate that observed by Kaplan-Meier log rank test. In conclusion, these results indicate that BRM270 can effectively inhibit proliferation and induce apoptosis in hepatoma cells by down-regulating CyclinD1/Bcl2 mediated c-Jun apoptotic pathway.</P>

<P><B>Abstract</B></P> <P>The nanoformulation of pesticides has the potential to increase food productivity, while resolving the drawbacks of conventional agrochemicals, which have negative environmental impacts. In this study, herbicide (metsulfuron methyl)-loaded pectin (polysaccharide) nanoparticles were synthesized and evaluated for herbicidal activity and cytotoxicity. The optimum formulation of nanoparticles was obtained using the Central Composite Design. The basic properties (mean particle size, stability, morphology, and interaction between polymer and herbicide) were characterized using a particle size analyzer (PSA), zeta potential, transmission electron microscopy (TEM), and Fourier Transform infrared spectroscopy (FTIR), respectively. The nanoparticles were found to be in size range of 50–90nm with zeta potential value of −35.9mV. The herbicide loading and herbicide encapsulation efficiency of the nanoparticles were determined to be 6.30% and 63±2%, respectively. The cytotoxicity of the herbicide-loaded nanoparticles was evaluated using healthy cell lines (Vero cell lines) and compared with that of commercial herbicide. In addition, an in-field evaluation of our nanoformulation's effects on the <I>Chenopodium album</I> plant was performed using a pectin nanocarrier.
