        산소 플라즈마 처리 후 ZnO 박막에 대한 PL 연구

        The optical properties of ZnO thin film, being treated by O-plasma, have been studied using Photoluminescence(PL) spectroscopy with the change of temperature from 10 K to 290 K. Two characteristic peaks were identified at 10 K : 3.357 eV($D^{\circ}X$) and 3.324 eV(TES). The peak of $D^{\circ}X$ is believed to be due to neutral donor bound excitons where the donor is in the ground state. However, the TES(Two Electron Satellite) peak indicates the excited state of the donor(excitation energy was ~30 meV). The donor binding energy was estimated to be 44 meV, which indicates the possible presence of the neutral donor bound excitons at RT. The thermal effect including thermal broadening was identified from temperature evolution of the spectrum. Both the peak intensity and the peak energy have decreased as the temperature increases. As the temperature approaches to RT, the two peak merges into one broad peak, which is considered a combination of multiple peaks having different physical origins.

        관개기 시험구 논에서의 오염물질의 농도특성

        The pollutant concentrations at experimental paddy plots with three (excessive, standard, reduced) different fertilization rates were investigated during 2001-2002 irrigation seasons. Mean concentrations of pollutants in ponded water were not significantly different among three experimental plots, but the T-N concentrations in percolated water significantly depended on fertilization rates. The T-N, T-P and $COD_{Cr}$, concentrations in ponded water during early irrigation season (late May to mid-June) were much higher than those during later irrigation season likely due to fertilization and low uptake by young rice crops. The T-N concentrations decreased but the concentrations of T-P and $COD_{Cr}$, increased three days after tillering fertilization. The removal rates of T-N by paddy plots were $0.13-0.16g/m^2{\cdot}d$ for an excessive fertilization plot, $0.08-0.25g/m^2{\cdot}d$ for a standard fertilization plot, and $0.03-0.34g/m^2{\cdot}d$ for a reduced fertilization plot three days after tillering fertilization. On the other hand, T-P and $COD_{Cr}$, were released three days after tillering fertilization.

        플라즈마 처리가 ZnO 박막의 물리적 특성에 미치는 영향

        The characteristic changes in ZnO thin film according to H- and O- plasma treatments have been studied by Photoluminescence (PL) spectroscopy at room temperature. The red shift of UV peak by 20-30 meV in PL spectra after plasma treatments is identified, which indicates that there are changes in the binding energy of bound exciton and/or the movement of energy levels of lattice defects and impurities. The width of UV peak is decreased after plasma treatments, which is believed to be closely related to the crystal quality of ZnO film. The increase of UV peak intensity after H-plasma treatment is also observed, and this could mean that the radiative recombination is strengthened because the hydrogen atoms in the plasma diffuse into the film where they passivate and neutralize the defects and the impurities.

        인간 배아줄기세포로부터 분화된 세포에서 MACS 방법을 이용하여 분리한 세포의 특성에 대한 연구

        목 적: 인간 배아줄기세포는 재생 의학이나 조직공학에 있어서 큰 잠재적인 능력을 가지고 있는 것으로 알려져 있다. 이들은 다양한 growth factors 처리나 유전자 발현을 변화시켜 특정 세포로 유도 분화 및 분리가 가능하지만 그 효율성은 아직까지 낮은 상태이다. 본 연구에서는 인간 배아줄기세포로부터 비특이적으로 분화된 세포들을 특정 세포 표면 항체를 이용한 magnetic cell sorting (MACS) 방법으로 분리, 배양하여 그들의 특성을 살펴보았다. 연구방법: 미분화 배아줄기세포주(Miz-hESC4)를 물리적인 방법으로 계대 배양하였으며, 부유 배양법으로 배아체 형성을 유도하였다. 배아체의 자발적인 분화를 위해 DMEM에 10% FBS를 첨가하여 2주 동안 배양하였다. 이렇게 분화된 세포들을 CD34, human epithelial antigen (HEA), human fibroblast (HFB)에 대한 항체를 이용한 MACS system으로 각각의 항체에 대한 양성 또는 음성 세포를 분리하였다. 이러한 MACS 분리 세포를 4주 동안 배양하면서 형태적인 변화를 관찰하고 특이 유전자의 발현 양상을 분석하였다. 결 과: 분리 배양한 CD34 양성 세포들은 배양 초기에는 둥근 형태를 나타내다가 배양 후기에는 작은 다각형의 형태로 관찰되었으며, HEA 양성 세포들은 큰 다각형의 형태를 나타내었고, HFB 양성 세포들은 전형적인 방추체 형태로 관찰되었다. 특이 유전자에 대한 RT-PCR 결과에서, CD34 양성 세포들과 HFB양성 세포들에서는 내배엽과 중배엽 관련 유전자의 발현하는 것을 확인할 수 있었고, HEA 양성 세포들에서는 외배엽 관련 유전자인 NESTIN과 NF68KD의 발현을 관찰할 수 있었다. 배양기간이 경과함에 따라 CD34 양성 세포의 특이 유전자 발현 양상이 변화되었다. 결 론: 이상의 결과는 비특이적으로 분화된 인간 배아줄기세포로부터 특이 세포를 MACS 방법을 이용하여 성공적으로 분리할 수 있음을 보여주었다. 따라서, MACS 방법과 특이 세포에 대한 항체는 인간 배아줄기세포의 유도 분화와 특이 세포의 분리에 매우 유용할 것으로 생각된다. Objective: Human embryonic stem (ES) cells have a great potential in regenerative medicine and tissue engineering. The human ES cells could be differentiated into specific cell types by treatments of growth factors and alterations of gene expressions. However, the efficacy of guided differentiation and isolation of specific cells are still low. In this study, we characterized isolated cells from differentiated human ES cells by magnetic activated cell sorting (MACS) system using specific antibodies to cell surface markers. Methods: The undifferentiated hES cells (Miz-hESC4) were sub-cultured by mechanical isolation of colonies and embryoid bodies were spontaneously differentiated with DMEM containing 10% FBS for 2 weeks. The differentiated cells were isolated to positive and negative cells with MACS system using CD34, human epithelial antigen (HEA) and human fibroblast (HFB) antibodies, respectively. Observation of morphological changes and analysis of marker genes expression were performed during further culture of MACS isolated cells for 4 weeks. Results: Morphology of the CD34 positive cells was firstly round, and then it was changed to small polygonal shape after further culture. The HEA positive cells showed large polygonal, and the HFB positive spindle shape. In RT-PCR analysis of marker genes, the CD34 and HFB positive cells expressed endodermal and mesodermal genes, and HEA positive cells expressed ectodermal genes such as NESTIN and NF68KD. The marker genes expression pattern of CD34 positive cells changed during the extension of culture time. Conclusion: Our results showed the possibility of successful isolation of specific cells by MACS system from undirected differentiated human ES cells. Thus, MACS system and marker antibodies for specific cell types might be useful for guided differentiation and isolation of specific cells from human ES cells.

        [논문철회]위암 수술 후 외래환자의 영양상태 평가: 5가지 영양검색도구의 비교연구

        Objectives: This study aimed to examine the characteristics of patients according to their nutritional status as assessed by five nutritional screening tools: Patient-Generated Subjective Global Assessment (PG-SGA), NUTRISCORE, Nutritional Risk Index (NRI), Prognostic Nutritional Index (PNI), and Controlling Nutritional Status (CONUT) and to compare the agreement, sensitivity, and specificity of these tools. Methods: A total of 952 gastric cancer patients who underwent gastrectomy and chemotherapy from January 2009 to December 2012 at the Samsung Medical Center were included. We categorized patients into malnourished and normal according to the five nutritional screening tools 1 month after surgery and compared their characteristics. We also calculated the Spearman partial correlation, Cohen's Kappa coefficient, the area under the curve (AUC), sensitivity, and specificity of each pair of screening tools. Results: We observed 86.24% malnutrition based on the PG-SGA and 85.82% based on the NUTRISCORE among gastric cancer patients in our study. When we applied NRI or CONUT, however, the malnutrition levels were less than 30%. Patients with malnutrition as assessed by the PG-SGA, NUTRISCORE, or NRI had lower intakes of energy and protein compared to normal patients. When NRI, PNI, or CONUT were used to identify malnutrition, lower levels of albumin, hemoglobin, total lymphocyte count, total cholesterol, and longer postoperative hospital stays were observed among patients with malnutrition compared to those without malnutrition. We found relatively high agreement between PG-SGA and NUTRISCORE; sensitivity was 90.86% and AUC was 0.78. When we compared NRI and PNI, sensitivity was 99.64% and AUC was 0.97. AUC ranged from 0.50 to 0.67 for comparisons between CONUT and each of the other nutritional screening tools. Conclusions: Our study suggests that PG-SGA and NRI have a relatively high agreement with the NUTRISCORE and PNI, respectively. Further cohort studies are needed to examine whether the nutritional status assessed by PG-SGA, NUTRISCORE, NRI, PNI, and CONUT predicts the gastric cancer prognosis.

        The effect of fertiltzer application on ponded and percolation water in rice paddy were investigated at the experimental plots during irrigation period. We laid out three experimental plots such as 150% Excessive, 100% Standard and 70% Reduced. The concentration of T-N and T-P in ponded water were increased rapidly after application. The concentration of T-N and COD in percolated water increased with fertilizer application rates, while T-P concentration were almost constant.

        Characteristics of pollutant load during irrigation and non-irrigation periods was investigated for streamwater from a rural watershed. Water was sampled and discharge was measured at 5-days intervals at outlet of study area. The mean concentrations of TN and TP in an irrigation period are higher than in an non-irrigation period, while mean COD concentration in an irrigation period is lower than in a non-irrigation period. For increasing discharge during an irrigation period, TN concentration increase, TP concentration is nearly unchanged, and COD concentrations decreases.
