        Statistical Optimization of Medium Composition for Growth of Leuconostoc citreum

        Leuconostoc citreum is one of the representative strains of Leuconostoc spp. thatshow fast growth rates in fermented vegetables. Sequential experimental designs including the Plackett-Burman design, fractional factorial design, steepest ascent analysis, central composite design and response surface methodology were introduced to optimize and improve the medium for L. citreum. Fifteen medium ingredients were examined and glucose (20 g/L), yeast extract (12.5 g/L), sodium acetate trihydrate (6.12 g/L), potassium phosphate (42.55 g/L) and dibasic ammonium citrate (4.12 g/L) were chosen as the best components to give a critical and positive effect for cell-growth. The biomass was increased to 2.79 g/L (169%), compared to the 1.65 g/L in MRS medium.

        장수명 공동주택을 대상으로 한 전통디자인요소의 현대화방안에 관한 연구 - 실내공간구성을 위한 가변요소를 중심으로 -

        This research is aimed at the development of Long Life Housing, which can realize sustainable construction and environment. It also promotes practical use of Long Life Housing with evaluation of the field application through Mockup House. This research is one of the important tools to realize Long Life Housing and deducts various traditional design's element technology to cope with demands of change of life cycle and style of residents. this study set three elements of plans for modernization in the traditional design factors that opening, wall and bottom as space composition elements of interior facts. first, Jang-ji wall and storage wall system separated from elements of wall that applied modernize elements of the wall. second, element of opening was restructure swing-lift-up opening system into modernize. third, maru and toet-maru system separated from elements of bottom that was applied modernize elements of the bottom. each elements segregation with four types as basic type, separate type, movable type and mixed type. This research make a proposal of plans for modernization in the traditional design factors, aimed at Long Life Housing through these four types blend. In addition, it points to promote realization of the sustainable 'Korea-style Long Life Housing' through modernization.

      • Pharmacological and Biochemical Characterization of Cells Isolated from Fetal Rat Calvaria

        Transforming growth factor ${\beta}(TGF-{\beta})$는 많은 세포의 증식, 분화 및 여러가지 세포기능에 대해 다양한 조절기능을 갖고 있는 multifunctional polypeptide로 알려져 있다. $TGF-{\beta}$는 골기질에 상당량 존재하며 골조직 대사에 대해 광범위한 영향을 나타낸다. 여러가지 연구결과에 의해 기질과 연관된 $TGF-{\beta}$는 골세포 자체의 산물로 여겨지고 있으나 골조직세포군중 어느종류의 세포가 $TGF-{\beta}$를 형성하는지에 대해서는 논란이 되고 있다. 본 연구는 $TGF-{\beta}$를 형성하는 골조직세포를 규명하고 $TGF-{\beta}$가 서로 다른 세포들에 미치는 영향을 관찰하고자 하였다. 본 실험에 필요한 특정골조직세포군을 얻기 위하여 백서태자두개관을 연속효소처리하여 수집한 골조직세포군의 생화학적 특성규명을 시행하였다. 효소 처리후 초기에 유리되는 세포는 섬유아세포의 특성을 보이며 후기에 유리되는 세포는 acid 및 alkaline phosphatase 활성과 부갑상선호르몬, calcitonin, prostaglandin $E_{2}$에 대한 c-AMP의 반응 및 교원 단백질합성등을 통해 볼때 조골세포유사세포로 보여진다. 골조직과 두개관세포 추출물의 Polyacrylamlde gel과 immunoblot analysis에 의해 골조직내에 $TGF-{\beta}$의 존재와 골세포에 의한 $TGF-{\beta}$의 생성을 확인하였다. 두개관 세포 추출물의 분석에서 모든 세포군에서 $TGF-{\beta}$가 합성되는 것을 관찰하였다. 외부에서 $TGF-{\beta}$를 가한 경우 무혈청 배지에서 골조직 증식에 대해 두가지 양상의 반응을 보였다. 조골세포 유사세포에서는 촉진하는 반응을 보였으나 섬유아세포군에서는 억제반응을 나타냈다. 반면에 교원 및 비교원 단백질 합성에 있어서는 모든 두개관세포군이 촉진반응을 보였다. 단백질 합성증가는 교원단백에 특이성이라기 보다는 일반적인 증가로 보여진다. 또한 단백질합성증가에 대한 $TGF-{\beta}$의 영향은 세포증식과의 관련성이 없는 것으로 사료된다. 결국 골세포에 의한 $TGF-{\beta}$의 생성과 여러 세포군에 대한 서로 다른 작용으로 보아 $TGF-{\beta}$는 autocrine과 paracrine 양상으로 세포기능을 조절함으로써 골조직 대사를 조절하는 중요한 기능을 발휘하는 것으로 사료된다. Transforming growth factor ${\beta}(TGF-{\beta})$ is a multifunctional polypeptide with diverse effects on the proliferation, differentiation and other functions in many cell types. $TGF-{\beta}$ is highly abundant in bone matrix and induces divergent responses in many aspects of bone cell metabolism . Several lines of investigation indicate that matrix-associated $TGF-{\beta}$ is the products of bone cells themselves. However, exact bone cell type reponsible for the production of $TGF-{\beta}$ is still in controversy, The present study was undertaken to determine the cellular origin of matrix-associated $TGF-{\beta}$ and to assess how different bone cells respond to $TGF-{\beta}$. As a prerequisite for this, 5 bone cell populations of distinct phenotype were isolated from fetal calvaria with sequential enzyme digestion protocol and biochemical characterization. Calvarial cell populations released in early stage showed fibroblastic features whereas populations relesed later was enriched with osteoblast-like cell as judged by their acid and alkaline phosphatase activities, cAMP responsiveness to parathyroid hormone, calcitonin and prostaglandin $E_2$ and collagen synthesis rate. By polyacylamide gel and immunoblot analysis of bone and calvarial cell extracts, presence of $TGF-{\beta}$ in bone tissues and production of $TGF-{\beta}$ by bone cells were confirmed again. Subsequent analysis of calvarial cell extracts prepared as individual population revealed that all calvarial cell populations synthesize $TGF-{\beta}$. Exogenously added $TGF-{\beta}$ induced biphasic response upon bone cell proliferation under serum-free condition. In osteoblastic cell populations, it was stimulatory whereas inhibitory in fibroblastic cell populations. In contrast, collagen and noncollagen protein synthesis of all calvarial cell populations were stimulated by $TGF-{\beta}$. Enhancement of protein synthesis was found to be more general rather than specific for collagen synthesis. In addition, effects of $TGF-{\beta}$ on protein synthesis were independent to its effects on cell proliferation. In summary, production of $TGF-{\beta}$ by bone cells and differential actions on various cell populations observed in this study suggest that $TGF-{\beta}$ may play an important role in the regulation of bone metabolism by modulating the specific cellular functions in autocrine and paracrine fashion.

        열공망막박리에서 일차유리체절제술의 결과

        Purpose: To report the anatomic and visual results of vitrectomy without scleral buckling in rhegmatogenous retinal detachment. Methods: Primary vitrectomy without scleral buckling for the treatment rhegmatogenous retinal detachment was done in 41 eyes (41 patients) and followed up for a mean period of 21.5 months(range 6~65 months). Indications of primary vitrectomy were uncertain preoperative breaks, the presence of large breaks, the presence of breaks near equator, multiple breaks, proliferative vitreoretinopathy not related to breaks and presence of inferior vitreous hemorrhage. Results: The anatomic success rate after a single operation was 90.2%. Visual acuity was improved or stable in 37 eyes (90.2%). Progression of lens opacity (35.7%) and formation of epiretinal membrane (12.2%) constituted the major complications after primary vitrectomy. Conclusions: Primary vitrectomy without scleral buckling can be a safe, effective method to repair primary rhegmatogenous retinal detachments in selective cases.

        해저자원개발을 위한 Riser의 동력학적 연구

        해저지하자원개발시 사용되는 riser의 파랑하중에 대한 구조해석방법에 대하여 연구하였다. 본 논문에서 비교 연구한 방법들은 규칙파를 이용한 정력학적해법, 규칙파 및 불규칙파를 이용한 시간영역에서의 동력학적해법과 두가지의 다른 선형화기법을 이용한 주파수영역에서의 동적해법이다. 구조해석은 길이가 다른 두 개의 Riser를 예로 하여 수행되었으며, 이때 유한요소법에 의한 구조모형이 사용되었다. 연구결과 종래의 규칙파에 의한 정력학적해법은 가장 정확한 불규칙파에 의한 동력학적해법에 비하면, 부재의 최대응력은 대소 크게 예측하지만, Riser 운영상 중요한 인자인 하단각도를 상당히 작게 산정하는 경향을 보여 사용에 주의를 요함을 보였다. A comparative study between several methods for the marine drilling riser analysis is carried out. One static analysis method and four dynamic methods are studied. The dynamic analysis methods used are two time domain methods using regular and random waves, and two frequency domain methods using the conventional and an improved linearization techniques. Two different sizes of risers are investigated. The analysis model of the structure is based on the beam-column element with lateral wave/current loads in a vertical plane. The forces on the riser are calculated using a modified farm of the Morison's equation. The finite element method is used to solve the equation for several wave/current conditions.

        장수명 공동주택 설계 프로세스 구축을 위한 보금자리주택 계획사례 연구 - 내곡지구 현상설계를 중심으로 -

        Recently, apartments have had several fundamental problems in planning uniformity caused by fixed space programs and difficulties of maintenance. So decision-makings become more important in design process which were essential to performance realization of long-life housing. Therefore, this study aims to analyze the plans of the Bogeumjari Nae-gok based on the applied designs, and suggests improvements for advanced design process of the long-life housing in the domestic market. This research procedure are as follows: (1) Analyze design guide, district land development plan and local code which have influence on design limitation. (2) Compare partial plans of joined designs and analyze each design characters (3) Implement case studies of design process due to design timetable and design elements of Nae-gok. In result, this study suggests significant fundamental materials for advanced decision-making in design process of long-life housing: details credibility and noise control, etc. This research could be useful in making integrated design process applied for long-life housing.

Through this architectural research deduce elements of architectural planing that can be research utilization with in Check-list composition.
