        훈증소독제, Fumagari OPP<sup>®</sup>의 Staphylococcus aureus에 대한 살균효과

        This study was performed to evaluate the bactericidal efficacy of Fumagari OPP$^{(R)}$, fumigation disinfectant, containing 20% ortho-phenylphenol against Staphylococcus aureus (S. aureus). In this research, efficacy test of fumigant against S. aureus was carried out according to French standard NF T 72-281. S. aureus working culture suspension number (N value), all of the colony numbers on the carriers exposed with the fumigant (n1, n2, and n3), the number of bacterial test suspentions by pour plate method (N1), the number of bacterial test suspentions by filter membrane method (N2) and the mean number of bacteria recovered on the control-carriers (T value) were obtained from the preliminary test. In addition, the reduction number of S. aureus exposed with the fumigant (d value) was calculated using T value, the mean number of bacteria in recovery solution (n'1) and the mean number of bacteria on carriers plated in agar (n'2). N value was $4.0{\times}10^8$ CFU/mL, and n1, n2, and n3 were higher than 0.5N1, 0.5N2 and 0.5N1, respectively. Additionally, T value was $3.4{\times}10^6$ CFU/mL. In the bactericidal effect of the fumigant, the d value was 6.43 logCFU/mL. According to the French standard for the fumigant, the d value for the effective bactericidal fumigant should be over than 5 logCFU/mL. The results indicated that Fumagari OPP$^{(R)}$ had an effective bactericidal activity against S. aureus, then the fumigant can be applied to disinfect food materials and kitchen appliances contaminated with pathogenic bacteria. 본 연구는 20% ortho-phenylphenol을 함유한 훈증소독제, Fumagari OPP$^{(R)}$의 Staphylococcus aureus (S. aureus)에 대한 살균효과를 평가하기 위해, French standard NF T 72-281에 따라 수행하였다. 배양 현탁액 중 S. aureus의 균수 (N 값), 훈증소독제에 노출된 각 담체의 균수 (n1, n2, n3), 평판배지법에 의한 시험균주 현탁액 중 균수 (N1), 여과법에 의한 시험균주 현탁액 중 균수 (N2), 그리고 대조담체의 회복 균수의 평균값 (T 값)을 예비실험을 통해 구하였다. 또한, 훈증소독제에 노출된 S. aureus의 감소 균수 (d 값)는, T 값, 회복액 중 균수의 평균값 (n'1) 그리고 배지의 담체에서 증식한 균수의 평균값 (n'2) 등을 이용하여 산출하였다. N 값은 $4.0{\times}10^8$ CFU/mL이었으며, n1, n2, n3은 각각 0.5N1, 0.5N2, 0.5N1 보다 높게 나타났다. 그리고 T값은 $3.4{\times}10^6$ CFU/mL이었다. 훈증소독제의 살균효과에 있어서, d 값은 6.43 logCFU/mL이었다. 훈증소독제에 대한 프랑스 기준에 따르면, 효과적인 살균력을 갖는 훈증소독제의 d 값이 5 logCFU/mL 이상이어야 하는 것으로 규정하고 있다. 본 연구의 결과로부터, Fumagari OPP$^{(R)}$는 S. aureus에 대해 높은 살균효과를 갖는 것으로 나타나, 병원성 세균으로 오염된 식품재료와 주방기기의 소독에 적용할 수 있을 것으로 사료된다.

        Anticoccidial Efficacy of Coccimuel-S composed with Diclazuril on Experimental and Field Coccidiosis in Broiler Chickens

        The efficacy of water soluble formulation of diclazuril (Coccimuel-S 0.5%) was tested against Eimeria spp. infection broiler chickens. The experiment was performed both experimentally infection and in the field test. Coccimuel-S composed with diclazuril induced a marked inhibitory effect on the different stages of Eimeria life cycle in experimentally infected broiler chickens treated with the drug. The tested dosage levels of Coccimuel-S (0.5 ml/L,equivalent to diclazuril 2.5 ppm) in drinking water showed the significant effect compared with the control group in controlling coccidial infection and reducing the total oocyst numbers, lesion and fecal scores (p < 0.001). In addition,testing of Coccimuel-S (0.25 and 0.5 ml/L) in naturally infected poultry farms (1,200 broiler chickens), showed the significant anticoccidial effect compared to control (p < 0.001). In conclusion, addition of Coccimuel-S at the dose of 0.25 and 0.5 ml/L in the drinking water, induced efficacious effect for the treatment of coccidiosis in naturally coccidia infected broiler chickens.

        Antimicrobial Efficacy of the Disinfectant Solution Nanoxil® Against Fish Pathogenic Bacteria

        Fish pathogenic bacteria are a considerable danger of farmed fish and a source of economic loss in the fish farming industry. In this study, Nanoxil® was compared to hydrogen peroxide and a silver colloid in terms of disinfection efficacy against E. tarda, V. anguillarum and S. iniae. A bactericidal efficacy test conducted by a broth dilution method was used to determine the lowest effective dilution of the disinfectant following exposure to test bacteria for 30 min at 4oC. Nanoxil® and test bacteria were diluted with distilled water (DW), hard water (HW) or an organic matter suspension (OM) according to the treatment condition. Under the OM condition, the bactericidal activity of Nanoxil® against E. tarda exhibited a lowered efficacy compared to that under the DW and HW conditions. Nanoxil® at 500 fold under all of the conditions demonstrated a high bactericidal efficacy against S. iniae. As Nanoxil® possess bactericidal efficacy against fish pathogenic bacteria such as E. tarda, V. anguillarum and S. iniae,this disinfectant solution can be used to limit the spread of fish bacterial diseases.

        Ortho-phenylphenol을 주성분을 하는 훈증소독제의 Clostridium perfringens 아포에 대한 살아포 효과

        This study was performed to evaluate the sporicidal efficacy of a fumigation disinfectant containing 20% ortho-phenylphenol against Clostridium perfringens (C. perfringens) spores. In this research, efficacy test of fumigant against C. perfringens spores was carried out according to French standard NF T 72-281. C. perfringens spores working culture suspension number (N value), all the spore numbers on the carriers exposed with the fumigant (n1, n2, and n3), the number of bacterial spore suspensions by pour plate method (N1), the number of bacterial spore suspensions by filter membrane method (N2) and the mean number of bacterial spore recovered on the control-carriers (T value) were obtained from the preliminary test. In addition, the reduction number of C. perfringens spores exposed with the fumigant (d value) was calculated using T value, the mean number of bacterial spore in recovery solution (n'1) and the mean number of bacterial spore on carriers plated in agar (n'2). N value was $4.3{\times}10^7spores/mL$, and n1, n2, and n3 were higher than 0.5N1, 0.5N2 and 0.5N1, respectively. Additionally, T value was $4.9{\times}10^5spores/carrier$. In the sporicidal effect of the fumigant, the d value was 4.52log reduction. According to the French standard for the fumigant, the d value for the effective sporicidal fumigant should be over than 3log reduction. The results indicated that Fumagari $OPP^{(R)}$ had an efficient sporicidal activity against spores of C. perfringens, then the fumigant can be applied to disinfect food materials and kitchen appliances contaminated with bacterial spores. 본 연구는 20% ortho-phenylphenol을 함유한 훈증소독제, Fumagari $OPP^{(R)}$의 Clostridium perfringens (C. perfringens) 아포에 대한 살아포 효과를 평가하기 위해, French standard NF T 72-281에 따라 수행하였다. 배양 현탁액 중 C. perfringens의 아포수(N 값), 훈증소독제에 노출된 각 담체의 아포수(n1, n2, n3), 평판배지법에 의한 시험 아포 현탁액 중 아포수(N1), 여과법에 의한 시험 아포 현탁액 중 아포수(N2), 그리고 대조 담체의 회복 아포의 평균값(T 값)을 예비실험을 통해 구하였다. 또한, 훈증소독제에 노출된 C. perfringens의 감소 아포수(d 값)는, T 값, 회복액 중 아포수의 평균값(n'1) 그리고 배지의 담체에서 증식한 아포수의 평균값(n'2) 등을 이용하여 산출하였다. N 값은 $4.4{\times}10^7spores/mL$이었으며, n1, n2, n3은 각각 0.5N1, 0.5N2, 0.5N1 보다 높게 나타났다. 그리고 T 값은 $4.9{\times}10^5spores/carrier$이었다. 훈증소독제의 살아포 효과에 있어서, d 값은 4.52 이었다. 훈증소독제에 대한 프랑스 기준에 따르면, 효과적인 살균력을 갖는 훈증소독제의 d 값이 3 이상이어야 하는 것으로 규정하고 있다. 본 연구의 결과로부터, Fumagari $OPP^{(R)}$는 C. perfringens 아포에 대해 높은 살아포 효과를 갖는 것으로 나타나, 병원성 세균의 아포로 오염된 식품재료와 주방기기의 소독에 적용할 수 있을 것으로 사료된다.

        Virucidal efficacy of a disinfectant solution composed of n-alkyl-dimethyl-benzyl-ammonium chloride against porcine epidemic diarrhea virus

        Porcine epidemic diarrhea virus (PEDV) is the causative agent of porcine epidemic diarrhea (PED) and causes a considerable economic loss in swine industry. In this study, the virucidal efficacy of the dis-infectant composed to n-alkyl-dimethyl-benzyl-ammonium chloride (n-ADBAC) was investigated against PEDV. A virucidal efficacy was determined with the viability of PEDV contacted with the disinfectant in Vero cells. The disinfectant and PEDV were reacted on the hard water (HW) or organic matter sus-pension (OM) condition. On HW condition, PEDV was inactivated with 50 fold dilutions of the disinfectant. When the antiviral effect on OM condition was evaluated, the antiviral activity of the dis-infectant showed on 10 fold dilutions against PEDV. As the disinfectant possesses the virucidal efficacy against PEDV, the disinfectant solution can be used to limit the spread of animal viral diseases.

        Comparative analysis of blood concentrations for orally administered micronized and non-micronized florfenicol in Sprague-Dawley rats

        Florfenicol is a fluorinated analog of thiamphenicol and has been used for the treatment of animal respiratory infectious diseases with a mechanism of action that inhibits bacterial protein synthesis. The aim of this study was conducted to investigate the effect of micronized and non-micronized-florfenicol prepared with different comminution techniques on the change in the blood concentration of florfenicol in rats. Forty adult male Sprague Dawley rats (6 weeks of age, body weight 126.8±9.52 g) were randomly allocated to two treatments groups: MF treated with micronized florfenicol powder (particle size,over 90% was 10 μm) and NMF treated with non-micronized florfenicol powder (particle size, over 70% was 100 μm) were given 97.5 mg/kg body weight once daily for four days. The results showed that MF at day 2, 3 (p<0.05) and 4 (p<0.001)had a significantly increased serum concentration of florfenicol compared to NMF. The results show that the micronization of florfenicol may provide higher oral bioavailability of the drug compared to its normal form.

        Application of a Lateral Flow Immunoassay to Determine Ampicillin Residues in Muscle Tissue of Olive Flounder (Paralichthys olivaceus)

        Antibiotic Detection Kit (Combination I), a lateral flow immunoassay (LFIA) developed for the detection of antibiotic residues in milk, was utilized for the analysis of antibiotic residues in the muscle tissue of olive flounder. After 60-min treatment by dipping in water dosed with ampicillin (200-g/ton water), the residue depletion of ampicillin was investigated in 25 cultured olive flounder (Paralichthys olivaceus). Muscles of fish were sampled on the 1st, 2nd, 3rd, 4th and 5th day after drug treatment. The concentration of ampicillin in the muscle was determined by LFIA. The absorbance ratio of the sample to the control blank (Bs/Bo) was employed as an index to determine the muscle residues in olive flounder. To investigate the recovery rate, standard solutions were added to muscle samples to give final concentrations in the muscle of 4 and 8 ng/ml. The recovery rates of all spiked samples were > 96% of the spiked value. Ampicillin was detected in the muscle of fish treated with the drug until the 2nd day of the withdrawal period. The present study showed that the LFIA can be easily adopted to predict ampicillin residues in tissue of farmed fishes.

        Bactericidal Efficacies of an Aquatic Disinfectant Tablet Composed to Calcium Hypochlorite Against Vibrio anguillarum and Streptococcus iniae

        Vibrio spp. and Streptococcus spp. have caused a considerable disease of farmed fish and economic loss in fish farming and seafood industry. In this study, the efficacy of an aquatic disinfectant tablet composed to calcium hypochlorite was evaluated against V. anguillarum and S. iniae. A bactericidal efficacy test by broth dilution method was used to determine the lowest effective dilution of the disinfectant following exposure to test bacteria for 30 min at 4oC. An aquatic disinfectant tablet and test bacteria were diluted with distilled water (DW), hard water (HW)or organic matter suspension (OM) according to treatment condition. V. anguillarum on the DW, HW and OM condition was completely inactivated with 16,000 15,000 and 13,000 fold dilutions of the disinfectant, respectively. On the DW, HW and OM condition, S. iniae was absolutely inactivated with 17,000 16,000 and 14,000 fold dilutions of the disinfectant, respectively. As an aquatic disinfectant tablet possesses bactericidal efficacy against fish pathogenic bacteria such as V. anguillarum and S. iniae this disinfectant solution can be used to control the spread of fish infective bacterial diseases.

        Antiviral Efficacy of an Aquatic Disinfectant Tablet Composed to Calcium Hypochlorite Against Red Sea Bream Iridovirus

        In this study, the veridical efficacy of an aquatic disinfectant tablet composed to calcium hypochlorite against red sea bream iridovirus (RBIV). A veridical efficacy was determined with the viability of RBIV contacted with the disinfectant in viral stock cultured in fat head minnow cell line. An aquatic disinfectant tablet and RBIV were reacted on the distilled water (DW), hard water (HW) or organic matter suspension (OM) condition. On DW and HW condition, RBIV was inactivated with 25,000 fold dilutions of an aquatic disinfectant tablet. With the investigation of the antiviral effect of the disinfectant on OM condition, RBIV was inactivated on 22,000 fold dilutions of an aquatic disinfectant tablet. As an aquatic disinfectant tablet possesses veridical efficacy against RBIV, the disinfectant solution can be used to limit the spread of cultured marine fish viral disease.

        Bactericidal Efficacy of a Disinfectant Composed of Povidone-iodine Against Clostridium Perfringens and Mycobacterium Fortuitum

        Clostridium perfringens (C. perfringens) and Mycobacterium fortuitum (M. fortuitum) are associated with considerable diseases in animals and human. In this study, the disinfection efficacy of a commercial disinfectant composed to povidone-iodine (PVI) was evaluated against C. perfringens and M. fortuitum. A bactericidal efficacy test by broth dilution method was used to determine the lowest effective dilution of the disinfectant following exposure to C. perfringens and M. fortuitum for 30 min at 4°C. The disinfectant and test bacteria were diluted with hard water (HW) or organic matter suspension (OM) according to treatment condition. On HW condition, the bactericidal activity of the disinfectant against C. perfringens and M. fortuitum was 50 and 80 fold dilutions, respectively. On OM condition, the bactericidal activity of the disinfectant against both C. perfringens and M. fortuitum was 15 fold dilutions. As the disinfectant composed to PVI possesses bactericidal efficacy against C. perfringens and M. fortuitum, the disinfectant solution can be used to control the spread of bacterial diseases.
