Microarray Analysis of Oxygen-Glucose-Deprivation Induced Gene Expression in Cultured Astrocytes

Since astrocytes were shown to play a central role in maintaining neuronal viability both under normal conditions and during stress such as ischemia, studies of the astrocytic response to stress are essential to understand many types of brain pathology. The microarray system permitted screening of large numbers of genes in biological or pathological processes. Therefore, the gene expression patterns in the in vitro model of astrocytes following exposure to oxygen-glucose deprivation (OGD) were evaluated by using the microarray analysis. Primary astrocytic cultures were prepared from postnatal Swiss Webster mice. The cells were exposed to OGD for 4 hrs at 37oC prior to cell harvesting. From the cultured cells, we isolated mRNA, synthesized cDNA, converted to biotinylated cRNA and then reacted with GeneChips. The data were normalized and analyzed using dChip and GenMAPP tools. After 4 hrs exposure to OGD, 4 genes were increased more than 2 folds and 51 genes were decreased more than 2 folds compared with the control condition. The data suggest that the OGD has general suppressive effect on the gene expression with the exception of some genes which are related with ischemic cell death directly or indirectly. These genes are mainly involved in apoptotic and protein translation pathways and gap junction component. These results suggest that microarray analysis of gene expression may be useful for screening novel molecular mediators of astrocyte response to ischemic injury and making profound understanding of the cellular mechanisms as a whole. Such a screening technique should provide insights into the molecular basis of brain disorders and help to identify potential targets for therapy.

Forme fruste choledochal cyst (FFCC) is one of the diverse types of choledochal cyst with little or no dilatation of the extrahepatic bile duct (EHBD). It is considered that FFCC has to do with the pancreatobiliary malunion (PBM). In children, 3 to 6 millimeters of EHBD is assumed to be normal. Even though there is no clear-cut definition, FFCC is likely to be associated with bile duct dilatation less than 10 millimeters. Almost all cases have PBMs and symptoms of the pancreatitis or cholangitis. We experienced a case of FFCC in a 4-year-old boy. His EHBD measures 10 mm diameter. He had symptoms of pancreatitis and elevated hepatic transaminases. The pancreatobiliary common channel was 28 millimeters. He underwent EHBD resection and Roux-en-Y hepaticojejunostomy and was discharged with no specific complications.

In this study, the species composition of Decapods community was inquired and investigated at five stations in Muan tidal flat, from July 22, 2006 to October 1, 2006. The Decapods uncovered through the study, were identified and classified into two suborders, fifteen families, forty-three species. Among them, Macruras were identified and classified into three families, eleven species, Anomuras into four families, five species, Bachyuras into eight families, twenty-seven species, respectively. The diversity of Decapods community is composed of Bachyuras(62.79%), Macruras(25.58%) and Anomuras(11.63%). Inspecting the species composition over each station, thirty species of decapods were uncovered at station 1(Macruras: eight species, Anomuras: three species, Bachyuras: nineteen species), Twenty species(Macruras: nine species, Anomuras: one species, Bachyuras: ten species) at station 2, twenty-five species(Macruras: four species, Anomuras: three species, Bachyuras: eighteen species) at station 3, twenty-one species(Macruras: five species, Anomuras: two species, Bachyuras: fourteen species) at station 4, and twenty-five species(Macruras: five species, Anomuras: three species, Bachyuras: seventeen species) at station 5. Regarding species diversity, station 1 recorded the highest mark among the five stations, while station 2 marked the lowest. According to the classified community, Macruras were recorded high diversity at station 2, Anomuras at station 1, 3, 5, Bachyuras at station 1, respectively. Among the newly uncovered seven species, two species of Macruras - (Alpheus brevicristatus, Alpheus bisincisus), - five species of Bachyuras (Macrophthalmus japonicus, Macrophthalmus dilatatus, Charybdis japonica, Hemigrapsus penicillatus Helice tridens sheni) were appeared at every station. But fourteen species (Macruras: three species, Anomuras: two species, Bachyuras: nine species) were appeared only at one station. Compared with the results of preceding studies (twenty-three families, sixty-two species in total), the new species uncovered this time were seven families, twelve species (Anomuras: one family, one spesies, Bachyuras: six families, eleven species - Pagurus samuelis, philyra yangmataoensis, Orithyia sinica, Macromedaeus distinguendus, Heteropanope makiana, Tritodynamia horvathi, Uca lactea, Scopimera globosa, Scopimera longidactyla. Paracleistoma cristatum, Sesarma pictum, Sesarma pilacatum). Especially, Orithyia sinica, Philyara yangmataoensis, Scopimera longidactyla, Tritodynamia rathbuni, Tritodynamia horvathi, Camptandrium sexdentatum, etc, have high worth of preservation, because they were not inhabited in other areas. So far, in this investigation over Muan tidal flat and Hampyeong Bay, newly uncovered Decapods were twenty-three families, seventy-four species(Macruras: seven families, twenty-four species, Anomuras: six families, eleven species, Bachyuras: ten families, thirty-nine species) in all.

N/A Adenoma of the gallbladder is very rare pathologic condition and small in size, usually dis- covered incidentally during the laparotomy or diagnostic procedure for other intraabdominal lesions. Fifteen cases of the gallbladder adenoma were found from 1982 to 1993 at Depart- ment of Surgery, Masan Koryo General Hospital. The incidence of gallbladder adenoma to cholecystectomy during the same period was 0.76%(15:1985). The male to female ratio was 1;3.7 and the peak incidence was 6th decade. Pertaining to size, seven cases were less than 10mm in diameter, three were 10 to 14mm and remained five were larger than 15mm. For in- vestigating gallbladder adenoma, ultrasonography was a highly sensitive method (sensitivity'. 53.3%). There was a case of adenoma with focal adenomatous carcinoma, but no definite evi- dence of sequence suspecting malignant change from the underlying adenoma. In conclusion, the ultrasonography was a standard imaging technique in the preoperative diagnosis and as- sessing the surgical indications of the gallbladder adenoma,and there had been no definite ma- lignant evidence in our cases.
