The serum free medium was developed and used for the suspension culture of mammalian cells. Although there were the problems of the longer lag time and the smaller maximum cell concentration achievable, the higher specific productivity as well as other advantages of the serum free medium can make it a more realistic alternative. The existence of a staggering period in glucose concentration vs. time profile in the batch culture can be a practical indicating signal for performing fed batch culture. The concentration dependence of the effects of the additives in the serum free medium as well as its economic feasibility was also tested.

The recombinant human growth hormone (rHGH) was expressed in Saccharomyces cerevisiae. The aggregated rHGH molecules were solubilized by rasing pH of the cell lysates. When pH of the solution was lowered to around 6.5, substantial amount of the contaminating yeast proteins was effectively removed. The rHGH was further purified by successive chromatographic steps including DEAE cellulose, Sephacryl S-200, DE-52, and Phenyl-sepharose. The specific activity of the purified rHGH was 2.7IU per mg, when assessed by radioreceptor assay. This value was higher than that of pituitary-derived international standard HGH, indicating that rHGH was correctly folded.
Porcine Somatotropin(pST)이 비육돈의 성장생산성과 육질에 미치는 영향

The effects of porcine somatotropin(pST) on the growth performance and meat quality were investigated in the finishing pigs. Fourteen crossbred barrows weighing 60.4㎏ were allotted to either the control or pST group. The control group received no injection and pST group was injected daily for 41 days with 100㎍ of locally produced recombinant pST/㎏ BW. Administration of pST increased average daily gain by 15%(P$lt;0.05) and improved feed efficiency by 24%(P$lt;0.01). compared to control group. Feed consumption was reduced in pST group by 10%(P$lt;0.05), Four pigs from each treatment were slaughtered to measure carcass traits and meat quality. pST increased loin eye area (P$lt;0.11) and decreased backfat thickness by 19%(P$lt;0.05). There were no differences in texture, color, tenderness, juiciness and flavor of pork between control and pST group.

The current study was undertaken to evaluate the effects of a locally-produced recombinant porcine somatotropin (pST) on growth perfermance and pork quality of finishing pigs. A total of 32 barrows weighing average 61kg were employed for the 6-week growth trial : 8 barrows were allocated into the control group (CONT): 8 barrows into the slow release-low dose pST group (SR-L); 8 barrows into the solve release - high dose pST group (SR-H); 8 barrows into the daily pST group(DAILY). CONT pigs received no pST. SR-L and SR-H pigs were treated with pST twice during 6 week trial(Each dose contained the pST equavalent to 50 ㎍ and 100 ㎍ per day, respectively). DAILY pigs received 100 ㎍ pST/㎏ B W daily. All pigs were fed a commercial feed containing 0.87% lysine ad lib. There was no significant difference in average daily gain among treatments. Feed/gain was reduced by 20% in DAILY (3.09 vs 3.85; p$lt;0.05) and 11% in SR-H (3.43 vs 3.84; p$lt;0.05) compared to the CONT. Backfat of the DAILY group was 20% thinner than that of the CONT and loin eye area was wider by 17% (p$lt;0.05) in DAILY group, compared to the CONT. The extent of the improvement in growth perfermance of SR groups was less (p$lt;0.05) than that of DAILY group. There were no significant differences in the color, juiciness, flavor and texture of pork among treatments, with the tenderness of DAILY group being less desirable compared to that of CONT group. Moisture content of DAILY group was higher (p$lt;0.05) than that of CONT group, while lipid content was reduced in DAILY group. SR group showed the middle values between CONT and DAILY group in most criteria on growth performance and pork quality investigated. These results indicated that the recombinant pST produced by LG Chem., Ltd., Korea stimulated growth performance in growing pigs and did not show any major negative effects on pork quility, suggesting that slow release pST could be used as a growth stimulant for growing pigs.
