작약(Paeonia lactiflora) 뿌리 추출물의 대식세포에서 p62/SQSTM1 증가를 통한 자가포식 유도

Autophagy contributes to enhancing the immune system (innate and adaptive immune system) against foreign pathogens. Autophagy of macrophages is used as a major indicator for developing vaccine adjuvants to increase the adaptive immune response. In this study, PLR activated autophagy and increased p62/SQSTM1. The knockdown of p62/SQSTM1 attenuated PLR-mediated autophagy. Inhibition of TLR4 blocked PLR-mediated increase in p62/SQSTM1 level and autophagy induction. In addition, inhibition of PI3K blocked HSL-mediated increase of p62/SQSTM1. PLR increased Nrf2 level and the inhibition of TLR4 and PI3K reduced PLR-mediated increase of Nrf2. Taken together, it is believed that PLR may induce autophagy through upregulating p62/SQSTM1 via TLR4/PI3K/Nrf2 signaling pathway.

Although inflammation acts as host defense mechanism against infection or injury and is primarily a self limiting process, inadequate resolution of inflammatory responses leads to various chronic disorders. This work aimed to elucidate the anti-inflammatory effects of 2-methoxy-4-vinylphenol (2M4VP) isolated from pine needles in LPS-stimulated RAW264.7 cells. Some key pro-inflammatory mediators including nitric oxide (NO), prostaglandins (PGE_2), inducible NO synthase (iNOS), and cyclooxygenase-2 (COX-2) were studied by sandwich ELISA and western blot. In addition, suppression of NF-κB and MAPK activation, and histone acetylation was studied by western blot analysis and immunostaining. 2M4VP dosedependently inhibited NO and PGE_2 production and also blocked LPS-induced iNOS and COX-2 expression. In addition, 2M4VP potently inhibited the translocation of NF-κB p65 into the nucleus by IκB degradation following IκB-α phosphorylation and the phosphorylation of MAPKs such as p38, ERK1/2, and JNK. Also, 2M4VP inhibited hyper-acetylation of histone H3 (Lys9/Lys14) induced by LPS. Taken together, our results suggest that 2M4VP, a naturally occurring phenolic compound, exert potent anti-inflammatory effects by inhibiting LPSinduced NO, PGE_2, iNOS, and COX-2 in RAW264.7 cells. These effects are mediated by suppression of NF-κB and MAPK activation and histone acetylation.

In this study, we investigated in vitro immunostimulatory activity of fruit extracts from Pyrus ussuriensis var. hakunensis (Nakai) T.B. Lee (PUF) using mouse macrophage RAW264.7 cells. PUF increased the production of immunostimulatory factors such as NO, iNOS, IL-1β, IL-6 and TNF-α, and phagocytic activity in RAW264.7 cells. The inhibition of TLR2 and TLR4 blocked PUF-mediated production of immunostimulatory factors in RAW264.7 cells. In addition, the inhibition of MAPKs signaling pathway reduced PUF-mediated production of immunostimulatory factors. From these results, PUF may have immunostimulatory activity via TLR2/4-mediated activation of MAPKs signaling pathway. Therefore, PUF expected to be used as a potential immune-enhancing agent.
도깨비부채 잎 추출물의 NF-κB 활성화를 통한 대장암 세포 HCT116에 대한 세포생육 억제활성

In this study, we evaluated the anti-cancer activity and potential molecular mechanism of 70% ethanol extracts of leaves from Rodgersia podophylla against human colorectal cancer cells, HCT116. RPL dose-dependently decreased the cell viability through RPL-induced apoptosis in HCT116 cells. RPL induced inactivation the nuclear factor κB(NF-κB) through blocking IκB-α degradtion and P65 nuclear accumulation. The inhibition of GSK3β by LiCl attenuated RP-L-mediated NF-κB signaling inactivation. In addition, RP-L induced GSK3β activation. Based on these findings, RPL may be a potential candidate for the development of chemopreventive or therapeutic agents for human colorectal cancer

Recently, as a natural substance has been emphasized interest in research to enhance the immune function. Greenlettuce (Lactuca sativa L.) is a popular vegetable used fresh and it contains various phytochemicals and antioxidantcompounds, and has been reported to have various physiological activities such as antibacterial, antioxidant, antitumor andanti-mutagenic. However, only a few studies have investigated on the mechanism of action of immune-enhancing activityof lettuce. Therefore, in this study, the immunomodulatory activities and potential mechanism of action of Green lettuceextracts (GLE) were evaluated in the murine macrophage cell line RAW264.7. GLE significantly increased NO levels byRAW264.7 cells, as well as expressions of immunomodulators such as iNOS, COX-2, IL-1β, IL-6, IL-12, TNF-α andMCP-1. Although GLE activated ERK1/2, p38, JNK and NF-κB, GLE-mediated expressions of immunomodulators wasdependent on p38, JNK and NF-κB. In addition, TLR4 inhibition blocked GLE-mediated expressions of immunomodulators and activation of p38, JNK and NF-κB. Taken together, these results demonstrated that TLR4-MAPK/NF-κBsignalling pathways participated in GLE-induced macrophage activation and GLE could be developed as a potentialimmunomodulating functional food.
건조비지분말의 물리적 특성에 대한 단백질과 지질 함량의 영향

The effects of chemical compositions (protein, lipid, and dietary fiber) on the physical properties of dried biji powders were investigated. The raw biji was freeze-dried (control) and hot-air dried (untreated). The untreated biji was further defatted and deproteinated. The prepared biji powders were analyzed for the proximate composition, total dietary fiber (TDF), water absorption index (WAI), water solubility index (WSI), swelling power, solubility (including the quantification of soluble carbohydrate and protein fractions), and final viscosity (using a rapid visco analyzer). Control and untreated biji powders exhibited the similar chemical compositions. The defatted biji possessed higher TDF, although its protein content did not significantly differ for control and untreated ones. The deproteinated biji consisted mainly of TDF. WAI and swelling power increased in the order: deproteinated > defatted > control > untreated biji powders. WSI and solubility increased in the order: control > untreated > defatted > deproteinated biji powders. The similar patterns were observed for soluble carbohydrate and protein fractions. The deproteinated biji revealed the highest viscosity over applied temperatures, while the untreated one was lowest. Overall results suggested that the physical properties of the dried biji powder were reduced by protein and fat, but enhanced by dietary fiber.
Anti-oxidant Effect of Agastache rugosa on Oxidative Damage Induced by H2O2 in NIH 3T3 Cell

The plant Agastache rugosa Kuntze has various physiological and pharmacological activities. Especially, it has been regarded as a valuable source for the treatment of anti-inflammatory and oxidative stress-induced disorders. However, little has been known about the functional role of it on oxidative damage in mammalian cells by ROS. In this study, we investigated the DPPH radical, hydroxyl radical, hydrogen peroxide and intracellular ROS scavenging capacity, and Fe2+ chelating activity of the extracts from Agastache rugosa. In addition, we evaluated whether the extract can be capable of reducing H2O2-induced DNA and cell damage in NIH 3T3 cells. These extracts showed a dose-dependent free radical scavenging capacity and a protective effect on DNA damage and the lipid peroxidation causing the cell damage by H2O2. Therefore, these results suggest that Agastache rugosa is useful as a herbal medicine for the chemoprevention against oxidative carcinogenesis.
팥(Phaseolus angularis) 열수 추출물의 산화적 DNA와 세포 손상 억제 효과

In this study, we evaluated the protective effects of the hot water extract from red bean (Phaseolus angularis) against oxidative DNA and cell damage induced by hydroxyl radical. The antioxidant activities were evaluated by hydroxyl radical and hydrogen peroxide scavenging assay, and Fe2+-chelating assay. Although the extract with hot water didn’t scavenge the hydroxyl radical, it removed and chelated hydrogen peroxide and ferrous iron necessary for the induction of hydroxyl radical by 71% and 64% at 200 μg/ml, respectively. Its protective effect on oxidative DNA damage was carried using ψX-174 RF I plasmid DNA comparing the conversion level of supercoiled form of the plasmid DNA into open-circular form and linear form and the expression level of phospho-H2AX in NIH 3T3 cells. In ψX-174 RF I plasmid DNA cleavage assay, it inhibited oxidative DNA damage by 96% at 200 μg/ml. Also, it decreased the expression of phospho-H2AX by 50.1% at 200 μg/ml. Its protective effect against oxidative cell damage was measured by MTT assay and the expression level of p21 protein in NIH 3T3 cells. In MTT assay for the protective effect against the oxidative cell damage, it inhibited the oxidative cell death and the abnormal cell growth induced by hydroxyl radical. Also, it inhibited p21 protein expression by 98% at 200 μg/ml. In conclusion, the results of the present studies indicate that hot water extract from red bean exhibits antioxidant properties and inhibit oxidative DNA damage and the cell death caused by hydroxyl radical. 본 연구에서는 열수 팥 추출물이 hydroxyl 라디칼에 의해 유도되는 산화적 스트레스에 미치는 영향을 알아보기 위하여 항산화활성과 DNA 및 세포의 산화적 손상 억제 효과를 조사하였다. 팥 열수 추출물의 DPPH 라디칼과 hydroxyl 라디칼의 제거능은 다소 낮았으나, Fe2+-chelating과 과산화수소 제거효과는 높게 나타나 활성산소의 생성을 억제하는 데 효과적인 것으로 확인되었다. 또한 팥 열수 추출물의 in vitro DNA cleavage, DNA migration 및 H2AX의 인산화비 억제활성은 높은 활성을 보여주고 있어 라디칼에 의한 DNA 손상 억제에 효과적으로 작용하였다. 또한 지질과산화와 p21의 발현율을 통해 세포의 산화적 손상에 미치는 영향을 살펴보면 지질과산화 억제능과 p21의 발현율에 매우 효과적으로 작용하고 있어 라디칼에 의한 산화적 스트레스로부터 세포를 보호할 것으로 생각된다.

In this study, water extracts from Rosa acicularis leaves (RAL) increased the production of immunostimulatory mediators and phagocytic activity in RAW264.7 cells. RAL increased p62/SQSTM1 expression. Inhibition of TLR4, JNK, and PI3K/AKT blocked RAL-mediated increase of p62/SQSTM1. RAL activated JNK and PI3K/AKT signaling. RAL-mediated activations of JNK and PI3K/AKT signaling were reversed by TLR4 inhibition. Taken together, it is believed that RAL-mediated autophagy may be dependent on activating via TLR4-dependent activation of JNK and PI3K/AKT signaling in macrophages.
