Background: Rifampicin (RFP) is one of the principal first-line drugs used in combination chemotherapies against Mycobacterium tuberculosis , and its use has greatly shortened the duration of chemotherapy for the successful treatment of drug-susceptible tuberculosis. Compensatory mutations have been identified in rpoC that restore the fitness of RFPresistant M. tuberculosis strains with mutations in rpoB . To investigate rpoC mutation patterns, we analyzed 93 clinical M. tuberculosis isolates from patients in South Korea. Methods: Drug-resistant mycobacterial isolates were cultured to determine their susceptibility to anti-tubercular agents. Mutations in rpoC were identified by sequencing and compared with the relevant wild-type DNA sequence. Results: In total, 93 M. tuberculosis clinical isolates were successfully cultured and tested for drug susceptibilities. They included 75 drug-resistant tuberculosis species, of which 66 were RFP-resistant strains. rpoC mutations were found in 24 of the 66 RFP-resistant isolates (36.4%). Fifteen different types of mutations, including single mutations (22/24, 91.7%) and multiple mutations (2/24, 8.3%), were identified, and 12 of these mutations are reported for the first time in this study. The most frequent mutation involved a substitution at codon 452 (nt 1356) resulting in amino acid change F452L. Conclusion: Fifteen different types of mutations were identified and were predominantly single-nucleotide substitutions (91.7%). Mutations were found only in dual isoniazid- and RFP-resistant isolates of M. tuberculosis . No mutations were identified in any of the drug-susceptible strains.

A recent study reveals that missense mutations of EWSR1 are associated with neurodegenerative disorders such as amyotrophic lateral sclerosis, but the function of wild-type (WT) EWSR1 in the central nervous system (CNS) is not known yet. Herein, we investigated the neuroanatomical and motor function changes in Ewsr1 knock out (KO) mice. First, we quantified neuronal nucleus size in the motor cortex, dorsal striatum and hippocampus of three different groups: WT, heterozygous Ewsr1 KO (+/-), and homozygous Ewsr1 KO (-/-) mice. The neuronal nucleus size was significantly smaller in the motor cortex and striatum of homozygous Ewsr1 KO (-/-) mice than that of WT. In addition, in the hippocampus, the neuronal nucleus size was significantly smaller in both heterozygous Ewsr1 KO (+/-) and homozygous Ewsr1 KO (-/-) mice. We then assessed motor function of Ewsr1 KO (-/-) and WT mice by a tail suspension test. Both forelimb and hindlimb movements were significantly increased in Ewsr1 KO (-/-) mice. Lastly, we performed immunohistochemistry to examine the expression of TH, DARPP-32, and phosphorylated (p)-DARPP-32 (Thr75) in the striatum and substantia nigra, which are associated with dopaminergic signaling. The immunoreactivity of TH and DARPP-32 was decreased in Ewsr1 KO (-/-) mice. Together, our results suggest that EWSR1 plays a significant role in neuronal morphology, dopaminergic signaling pathways, and motor function in the CNS of mice.

Background and Objectives:but also because of other additional injuries around the nose. The purpose of this study is to explore the incidence, type, location and direction of nasal septal fracture in nasal bone fracture patients who were diagnosed by nasal bone CT. Subjects and Method:We analysed the medical records and films of 135 nasal bone fracture patients who were diagnosed by computerized tomography from January 2005 to September 2005. The nasal bone fracture was classified by six types on nasal bone CT:unilateral, bilateral, open book, impacted, greenstick, comminuted. The external deviation of nasal bone (20 degre below or up) and corelation between septal fracture and nasal bone fracture were analysed by chi-square test (p<0.05). Results:The incidence of each type of nasal bone fracture is unilateral (41% ), bilateral (18% ), open book (8% ) (6% ), gren stick (17% ), and comminuted (10% ). The incidence of combined septal fracture was high in comminuted and impacted rather than in unilateral and bilateral (p<0.05). And there was no corelation between the directions of the trauma force and nasal septal devi-ation. Conclusion:The incidence of concurrent nasal septal fracture when nasal bone fracture ocur is relatively high, so we must (Korean J Otorhinolaryngol-Head Neck Surg 2007 ;50 :903-6)

Combinatorial biosynthesis can be defined as modifying biosynthetic pathways generally by combining the natural or engineered biosynthetic enzyme activities from disparate sources and exploiting substrate promiscuity of biosynthetic machinery to produce “unnatural” natural products with altered structures with aims to expand the structural diversity of natural products and improve their pharmaceutical properties. In the broad sense, combinatorial biosynthesis would encompass precursor-directed biosynthesis, which incorporates chemically synthesized precursor in vivo into a natural products by the endogenous natural biosynthetic catalysts using their inherent substrate promiscuity, and mutasynthesis, where the gene producing the natural substrates is disrupted thus more efficiently incorporating the modified precursors. Some recent examples for combinatorial biosynthesis of approaches to create the structural diversity of natural products including polyketides and aminoglycosides will be presented.
