        허혈성 뇌손상 백서에서 공자대성침중방(孔子大聖枕中方)이 학습과 기억에 미치는 영향

        Objectives: The purpose of this study is to evaluate the effect of Gonjadaesungchimjoongbang on spatial learning abilities and memories in ischemic brain injury. Methods: Rats were separated into three groups; (1) Normal, (2) Saline medication after ischemic brain injuries (control), (3) Gonjadaesungchimjoongbang medication after ischemic brain injuries (experiment). Ischemic brain injuries was induced by MCA occlusion and reperfusion. Morris water maze test was conducted for spatial learning and memory tests. Then, the change of BDNF in the hippocampus($7^{th}$, $14^{th}$ day) was examined by immunohistoche- mistry. Results: In Morris water maze test, spatial learning abilities and memory functioning were considerably increased in the experiment group as oppose to control group on $7^{th}$ and $14^{th}$ day(p<0.01). Moreover, immunohistochemistric response of BDNF in the hippocampus indicated that the more increased immune reaction was found in the experiment group as oppose to the control group on $7^{th}$ and $14^{th}$ day. Conclusions: Gonjadaesungchimjoongbang can improve the learning abilities and memories in ischemic brain injury.

        Mycoplasma 폐렴 환아 치험 1례

        Objectives: The purpose of this study is to find the effect of Keumsuyukgunjeongamibang and aromatherapy on a child diagnosed with mycoplasma pneumonia. Methods: The child had been treated with herbal medicine and aromatherapy for 8 days. In order to examine the improvement, chest X-ray and blood test were performed. Results: Various symptoms, such as, coughing, rhinorrhea, dyspnea, was disappeared. The series of 4 X-ray images and blood test proved that the patient has improved. Conclusions: The result of this study shows that administration of Keumsuyukgunjeongamibang and aromatherapy are effective in treatment of mycoplasma pneumonia.

        복부 초음파 검사로 진단받은 급성 충수염 환아 증례 보고

        Objectives: This study was to evaluate the effects of herbal medication and acupuncture therapy for children with acute appendicitis diagnosed by abdominal ultrasonography. Methods: Kamieuiyeeintang, a Korean Herb medicine, and acupuncture were given for four days. The abdominal ultrasonography and VAS scale were used to check the improvements. Results: Right lower abdominal pain, nausea and anorexia were totally gone after the treatments, and the abdominal ultrasonography showed normal. Conclusions: The result of this study showed that Kamieuiyeeintang medication and acupuncture treatment have cured acute appendicitis.

        소아 본태성 진전 치험 1례

        Objectives This clinical study was aimed to examine the effects of herbal medicine and acupuncture on early onset - essential tremor and report on the therapeutic progress. Methods Herbal medication (Kamiyangsintang) and acupuncture were used for treatment. Also, VAS scale were used to check the improvement. Results The hands tremor was relieved during 4 months of treatment. However, the symptom was aggravated after one and a half years. The patient has shown improvement after retreatment. Conclusions This study showed that child patient diagnosed with essential tremor who received Kamiyangsintang and acupuncture treatments was improved. However, it also can be aggravated depending on the patients and their body condition. More cases and long-term observation in relation to early onset of tremor are required.

        산사(山?)가 DNCB로 유발된 생쥐의 알레르기성 접촉피부염에 미치는 영향

        Allergic contact dermatitis (ACD) is a type IV delayed hypersensitivity reaction that results from exposures and subsequent sensitization to an environmental chemical. Crataegus Pinnatifida (CP) is commonly used to improve spleen function, remove retention of food, and promote blood circulation. This study is designed to investigate the effects of CP on ACD induced by 2,4-dinitrochlorobenzene (DNCB) in mice. In this experiment, the effects of CP on changes in body weights, ear and dorsum skin thicknesses, ear weights, clinical aspects on the dorsum skin, histopathological changes, spleen weights, cytokines were investigated. In addition, the effects on the proliferation rates of splenocytes were also investigated in vivo and vitro study. In results, CP spread (CPS) group and CP spread and administered (CPS+Adm) group showed decrease in spleen weights. In CPS+Adm group, dorsum skin thicknesses were decreased significantly compared to control group. CP treatment diminished erythema, desquamation and keratosis which were induced by repeated painting of DNCB. In histopathological observation, spongiosis and edema were diminished in CPS and CPS+Adm group. CP led to decrease in the proliferation rates of splenocytes in vivo and vitro. In conclusion, these data suggest that CP can decrease symptoms of ACD, so CP is useful to treat patient with ACD.

        거금도 내 소하천의 담수어류군집 특성에 관한 연구

        The freshwater fish fauna for the whole water system of Geogeumdo Island was surveyed at a total 10 sites from March 2018 to February 2019. During the surveyed period, 13 species belonging to 5 families and 4 orders were collected. Cyprinidae (68.84%) and Gobiidae (18.21%) were the dominant family, and the dominant species was Zacco temminckii (53.71%), and the subdominant species was Rhynchocypris oxycephalus (10.78%). Kichulchoia brevifasciata (1.27%) that endangered species were found in this study, but the habitat was confirmed in one site in Sinpyeongcheon compared to previous literature. Three endemic species of Korea were found in this island such as Iksookimia hugowolfeldi (5.07%), Liobagrus somjinensis (2.99%) and K. brevifasciata (1.27%). According to analysis of fish community structure based on average diversity, evenness index, dominance and richness indices in Geogeumdo Island were 1.666, 0.649, 0.645 and 1.713, respectively. 2018년 3월부터 2019년 2월까지 10개 지점의 거금도 전수계 담수 어류상을 조사하였으며, 조사 기간 동안 4목 5과 13종이 채집되었다. 조사 결과 잉어과 Cyprinidae (68.84%)와 망둑어과 Gobiidae (18.21%)가 우세하였으며, 우점종은 갈겨니 Zacco temminckii (53.71%), 아우점종은 버들치 Rhynchocypris oxycephalus (10.78%)로 나타났다. 본 조사에서 멸종위기 야생생물 I급인 좀수수치 Kichulchoia brevifasciata (1.27%)가 출현하였으나 이전의 문헌과 비교하여 서식지가 줄어 신평천 일부 지점에서만 서식이 확인되었다. 또한 거금도의 한국 고유종으로는 남방종개 Iksookimia hugowolfeldi (5.07%), 섬진자가사리 Liobagrus somjinensis (2.99%), 좀수수치 K. brevifasciata (1.27%) 3종이 확인되었다. 군집지수를 분석한 결과 다양도 지수는 1.666, 균등도 지수 0.649, 우점도 지수 0.645, 종 풍부도 지수 1.713으로 나타났다.

        율천의 어류상과 어류군집구조의 분석

        본 연구에서는 2020년 4월부터 2020년 10월까지 섬진강의 오수천으로 유입되는 율천의 주요 지점 9곳을 3회에 걸쳐 조사하여 어류상 및 군집구조를 분석하였다. 그 결과 율천 일대에서 5과 19종의 어류가 확인되었다. 한국 고유종은 7종 (Rhodeus uyekii, Squalidus gracilis majimae, Microphysogobio yaluensis, Odontobutis interrupta, Cobitis tetralineata, Zacco koreanus, Squalidus chankaensis tsuchigae)이 조사되었다. 어류의 과별 종 구성을 살펴 보면, 잉어과가 15종으로 가장 많았고, 나머지 어종은 망둑어과, 동사리과, 미꾸리과, 버들붕어과에서 각각 1종씩 나타났다. 우점종은 Z. platypus였으며, 아우점종은 Z. temminckii로 확인되었다. 군집지수는 우점도 0.499, 다양도 2.279, 균등도 0.774 및 종 풍부도 2.594로 나타났다. 지점 3에서 14종으로 전체 지점 중 가장 많은 종 수를 보였으며 본 지점은 우점도 (0.480)가 낮고 다양도 (2.173)와 종 풍부도 (2.701)는 가장 높았다. 군집분석 결과, 하천의 구조에 따라 군집이 일치하는 경향을 보였으나 비교적 적은 수의 개체수를 확인한 St. 9은 상류구간과 유사한 결과로 확인되었다. St. 5는 많은 종 수와 개체수에 의해 별도의 cluster를 형성하고 있었다. In this study, nine major sites of Yulcheon stream, a principal tributary of the Seomjingang River, were surveyed from April 2020 to October 2020, and their fish fauna and community structure were analyzed. A total of five families and 19 species of fish were identified. Among the 19 species, seven species were found to be endemic to Korea: Rhodeus uyekii, Squalidus gracilis majimae, Microphysogobio yaluensis, Odontobutis interrupta, Cobitis tetralineata, Zacco koreanus, and Squalidus chankaensis tsuchigae. Among five families, the family Cyprinidae had the largest number of species (15), whereas the rest of the four families: Gobiidae, Odontobutidae, Cobitidae, and Osphronemidae, had one species each. The two dominant species in the family Cyprinidae were identified as Z. platypus and Z. temminckii. The community index of nine major sites of Yulcheon stream was 0.499 dominance, 2.279 diversity, 0.774 evenness, and 2.594 species richness. The number 3 site had 14 species, the largest number of species among all sites, and the dominance rate (0.480) was low, whereas diversity (2.173), and species richness (2.701) were the highest. As a result of cluster analysis, clusters matched according to the structure of the river types, but St. 9, which confirmed a relatively small number of populations, showed a structure similar to that of the upstream section. In the case of St. 5, a separate cluster was formed by a large number of species and populations.

        Purification of the Cry1Ac protein of Bacillus thuringiensis and assessment against the Plutella xylostella and soil microbial community

        The insecticidal toxin gene of Bacillus thuringiensis (Bt) is the most commonly used to develop insect-resistant living modified organisms (LMOs). Insecticidal proteins produced in transgenic plants are released into the soil from the roots. In this study, possible effects of crystal 1Ac (Cry1Ac) protein on the soil microbial community in Korea were studied. To purify the insoluble Cry1Ac protein expressing Escherichia coli cells, we performed repeated sonication and PBS washing of the insoluble part and Cry1Ac protein was isolated in soluble form from the insoluble form using 100 mM Na2CO3 buffer (pH 9.6) without affinity bead. Also, sizeexclusion chromatography (SEC) was performed to increase the purity of the isolated Cry1Ac protein. The final protein product was identified as Cry1Ac protein through MALDI-TOF. Insecticidal activity of Cry1Ac protein was demonstrated through the death of Plutella xylostella treated with Cry1Ac protein. Purely isolated Cry1Ac protein showed the same insecticidal activity as Cry1Ac expressed in LM crops. To investigate the change of soil microbial distribution using maize field soils treated with Cry1Ac protein, we isolated high quality metagenomic DNAs from buffer- and Cry1Ac protein-treated soil groups, and analyzed the distribution of soil microorganisms through next-generation sequencing (NGS) analysis. NGS results showed a similar microbial distribution in both buffer- and Cry1Ac protein-treated samples. These results suggest a useful risk assessment method for domestic targeted insect and soil microorganisms using the Cry1Ac protein.

        Bacillus thuringiensis 유래 Vip3Aa 단백질 순수분리 및 꿀벌(Apis mellifera)에 대한 위해성평가

        Most insect-resistant LMOs have been produced by applying Cry and Vip3Aa proteins. Vip3Aa protein is activated during the vegetative stage of Bacillus thuringensis (Bt) and the inhibitory activity of the Vip3Aa protein against pathogenic attacks from lepidopteran insect species is well known. However, a risk assessment of the Vip3Aa protein compared to the Cry protein has not been conducted in South Korea. This study demonstrates a possible risk assessment method for Vip3Aa protein against honeybees (Apis mellifera). For the risk assessment of the protein, we purified the recombinant Vip3Aa protein in Escherichia coli. The survival rate and symptoms of general intoxication of 4 months honeybees were measured after Vip3Aa exposure. These results indicated that there was no significant difference in the survival rate and the symptom between Vip3Aa and the control buffer. In this study, we established standard methods of Vip3Aa protein purification and oral adult toxicity test using A. mellifera as an LMO risk assessment technique for preserving the natural ecosystem of South Korea. 본 연구는 LMO 유전산물의 위해성평가를 위해 바실러스로부터 증폭된 Vip3Aa 유전자를 이용하여 대장균에서 단백질 순수분리 하였으며, MALDI-TOP 분석법을 통해 기존의 알려진 살충성 Vip3Aa 단백질과 동등한 단백질임을 증명하였다. 순수 분리한 Vip3Aa 단백질을 이용하여 꿀벌 과독성 급성섭식독성평가를 수행하였다. 그 결과 무처리군, Hepes buffer, Vip3Aa 단백질 처리군 모두 치사 및 일반중독증상을 보이는 개체는 발견되지 않았다. 이 결과를 통해 Vip3Aa 단백질은 꿀벌에 위해성을 나타내지 않는다는 결론을 얻을 수 있었다. 본 연구 결과는 향후 국내 LMO 유전자 산물 위해성평가에 유용하게 활용될 것이라 사료된다.

        자연산과 양식산 동자개 Pseudobagrus fulvidraco 정소의 생식소발달 비교 연구

        Male gonadal development of the yellow catfish, Pseudobagrus fulvidraco, one of the most popular fish species in Korean aquaculture performance, was investigated by histological observation of monthly collected specimens to make comparisons between wild and cultured individuals. Their reproductive cycle was classified into the successive developmental stages as follows: a growing stage (April), a spawning stage (May), a degeneration stage (June to July), and a resting stage (August to October) in the wild and outdoor-cage individuals; a growing stage (April to June), a spawning stage (July to August), a degeneration stage (September), and a resting stage (October) in the indoor-cage ones. Values of gonadosomatic index (GSI) of wild and outdoor cages peaked in May, followed by a sudden decline in August~September and June~August, respectively. In contrast, GSI values of the indoor-cage individuals peaked in September and were followed by a sudden drop. Remarkable seasonal variation in condition factor (CF) was undetectable, peaking in June in the wild-cage individuals and November in the wild ones. Overall, our results suggest that it is suitable to use the male of the outdoor-cage individuals for artificial fertilization and that it is efficient to perform artificial fertilization in May, such as reproductive cycle of wild. 양식업에서 인기 있는 어종 중 하나인 동자개를 자연산과 양식산을 구분하여 생식소 발달을 관찰하였다. 자연산과 양식산 동자개는 2018년 4월에서 10월에 채집하여 수컷의 생식소지수, 비만도지수 및 생식소 발달을 연구하였다. 광학현미경으로 관찰된 월별 생식소 발달과 GSI를 기반으로 생식소 발달단계는 자연산과 실외 사육장은 4월 성숙기, 5월 산란기, 6월에서 7월 퇴화기, 8월에서 10월 휴지기로 관찰되었다. 실내 사육장은 4월에서 6월까지 성숙기, 7월에서 8월 산란기, 9월 퇴화기, 10월 휴지기로 구분되었다. GSI 값은 자연산과 실외 사육장은 5월에 가장 높은 값을 보였고, 실내 사육장은 9월에 가장 높은 값을 나타내 차이를 보였다. CF는 자연산과 양식산 사이에 유의한 차이가 없었으나 자연산은 6월에 가장 높은 값을 보였으며, 실외 사육장은 11월에 가장 높은 값을 보였다. 본 실험 결과, 실외 사육장 동자개 수컷을 인공 수정에 사용하는 것이 적합하며, 자연산 생식주기와 같은 5월에 인공 수정을 하는 것이 효율적일 것으로 판단된다.
