Calcium chloride 피브로인 용해물의 Gel Filtration Chromatography에 의한 순수분리 및 효소 가수분해 효과

The pure-separation of calcium chloride-treated fibroin hydrolysates could be carried out using gel filtration chromatography. Also, the effect of its enzymatic hydrolysis was investigated in order to find out the enhancement of their functionality. The average molecular weight(Mw), solubility and free amino acid compositions of three hydrolysates samples (calcium chloride, calcium chloride-flavo-urzyme and calcium chloride-thermoase) were measured to compare their characteristics. The molecular weight of calcium chloride hydrolysate was about Mw 46,800 and it can be reduced to Mw12,500 and 1,070 upon the enzymatic hydrolysis by flavourzyme and thermoase, respectively. A solubility of calcium chloride-treated samples shows about 60% while calcium chloride/enzyme-treated samples are perfectly soluble (100% solubility). The total amino acid composition of calcium chloride enzymatic hydrolysates are much higher than that of calcium chloride hydrolysate.

The bond direction between fiber axis and N-H bond direction of $^15N$-Gly labeled silk fibroin fiber and poly-p-phenyleneterephthalamide(PPTA) oriented were obtained from the dipolar-bipolar interaction information, respectively. Theoretical method for the determination of molecular bond directions relative to the axis of fiber was derived on the basis of solid state nuclear magnetic resonance dipolar-dipolar interaction tensor. The oriented samples were placed parallel to the applied magnetic field, and the $^15N-^1H$ hetero-nuclear dipolar-dipolar splitting values were obtained. Substituting this values into the derived equation, the N-H bond angles ($$\theta$_NH$=83$^{\circ}$ and 97$^{\circ}$) are calculated. Also, the N-Hbond direcyions to fiber axis of PPTA are 44$^{\circ}$ and 67- These obtained values compared with the values from the angular dependation spectra simulation of oriented PPTA sample.

The collagen growth effects in rats using B. mori fibroin sponge sheet, pure fibroin solution and chitosan sponge sheet were investigated. Histopathological inspection of the wound 12 days later showed the increase of a vascular ingrowth and the absence of inflammatory cells. Wound healing effects were accelerated in the order of Fibroin sponge sheet ≥Fibroin dressing solution > Chitosan sponge sheet > Control. The mean percentage of collagen production in rats after 12 days was 46.12±4.58% and 42.11±5.67% in fibroin sponge sheet and dressing solution-treated rats, respectively. These values were higher than the relative content in the chitosan sponge sheet and control groups, which was 23.87±3.24 and 2.42±0.32%, respectively.

The behavior of tyrosine(Tyr.) residue of Bombyx mori silk fiber from yellow fluorescent cocoon has been examined for the dependence of pH in aqueous silk solution under the presence of orange II salt. Through the peak separation of angular dependence of spectral pattern of 15N-Tyr. and (1-13C)-Tyr. between the fiber axis and the molecular bond direction, N-H bond in fiber as well as the orientation distribution around the fiber axis were analyzed. Also, and sericin component was obtained from these angular dependence of oriented spectral pattern. The pH dependence of the 13C NMR chemical shift of B. mori silk fibroin was examined in aqueous solution in the presence of orange II hydrophobic interaction such as stacking between the Tyr.-ring and the orange ll. All peaks of orange II are broad at pH ≥ 7.0. However, these become sharper at pH ≥8.0 and remain sharp at higher pH. In these higher pH range, a chemical shift change occurs due to the deprotonation of the Tyr. side group of fibroin. At higher pH, such a hydrophobic cluster is destroyed because of the electrostatic interaction according to the deprotonation of the Tyr.-OH group.

The preparation of skin care gauze could be make to mixing separated cellulose from waste milk park and degummed silk fibroin fiber. Also, its wound covering and anti-barterial activity were investigated in order to find cut the enhancement of their functionality. By the 30% silk fibroin fiber including skin care gauze. the anti-bacterial activity values of Sraphylococcus stran are much 4 times higher than of0~10% silk fibroin fiber including skin care gauze. The average yield of cellulose from waste milk pack was obtalned 50-6O%, and their morphologie,, physical properties, modulus and biodegradation ratios are studies, respectively.
효소처리에 의한 Bombyx mori fibroin 가수분해물의 분자량 및 구조 변화

본 연구는 누에 실 샘으로부터 얻은 Bombyx mori 피브로인의 효소 가수분해 시 그 분자량 및 구조적 특성을 파악하고자 하였다. Gel filtration chromatography(GFC)에 의해 B. mori 피브로인을 순수 분리한 결과 회수율이 약 70% 이상으로 기존의 방법(막과 튜브 분리법)에 비해 약 40% 이상 높았다. 이는 GFC법이 고 순수ㆍ고 수율의 B. mori 피브로인을 얻을 수 있는 방법임이 확인되었다. Gel permeation chromatography에 의한 분자량 측정 결과 누에 체내 피브로인, 염화칼슘 용해 물 및 효소 가수분해물이 각각 평균분자량 108,000 Da 내외, 65,000 Da 내외 및 1,000 Da 내외임을 알 수 있었다. 또한 효소 가수분해에 의한 구조적 변화를 ¹³C-NMR로 알아본 결과, B. mori 피브로인의 메인 골격에 해당되는 glycine, alanine 및 serine 등의 골격 피크가 관찰되어졌으며, 특히 효소가수분해 시 glycine를 의미하는 40-44 ppm 부근의 피크가 높게 나타난 것으로 보아 -G-A-G-A-반복 구조가 분해되었고, 이로 인해 분자량이 현저히 저하되었을 것으로 생각되어진다. This study examined the enzymatic digestion, average molecular weight distribution and structural changes of Bombyx mori silk gland fibroin using gel permeation chromatography and nuclear magnetic resonance methods. The pureseparation of calcium chloride-treated fibroin hydrolysates was carried out by gel filtration chromatography. Also, the effects of fibroin’s enzymatic hydrolysis were investigated using an edible enzyme. The average molecular weight of three hydrolysate samples (silk gland fibroin (SF), SF-calcium chloride (SFC), and SFC-enzyme) were measured to compare their characteristics. The molecular weights of SF, SFC, and SFC-enzyme were approximately 108,000, 65,000, and 1,000 Da, respectively. Finally, we determined the structural characteristic changes of the different enzymatically digested samples by ¹³C nuclear magnetic resonance methods. For the enzymatically digested fibroin, the glycine ¹³C<SUP>α</SUP> resonance indicated that the amino acid was dramatically changed and/or separated out; however, this was not shown for the normal Bombyx mori silk gland fibroin.
