배뇨조절과 관련된 감각세포에서 Purinergic Receptor와 Vanilloid Receptor의 분포와 상관관계

방광의 배뇨작용 조절에 관계하는 감각신경에 발현하는 신경전달물질 수용체로서 중요한 역할을 할 것으로 추측되는 것 중에는 ATP를 ligand로 하는 P2X3 수용체와 capsaicin을 ligand로 하는 vanilloid 수용체(VR1)가 있다. 이 두 가지 수용체를 이용한 과민성방광 등 질환의 새로운 치료방법의 가능성을 알아보기 위해 두 수용체의 분포 및 두 가지 수용체 사이의 상관관계를 알아보았다. 생후 8주 된 Sprague-Dawley 흰쥐의 방광벽에 retrograde nerve tracer를 주입하고 그 중 방광자극군의 방광에는 0.5% acetic acid를 주입하여, 1주일 뒤에 정상대조군과 방광자극군 흰쥐의 L6-S1 척수신경절 표본에 P2X3 수용체와 VR1에 대한 면역조직화 학염색과 Western blot을 시행하였다. 정상대조군과 달리, 실험군의 척수신경절에서 방광의 감각을 담당하는 신경세포에는 VR1 발현의 증가와 P2X3 수용체의 down-regulation이 나타났다. 이것으로 보아 자극을 받은 방광에서 과민성을 일으키는 데에 P2X3 수용체와 VR1이 중요한 작용을 할 것이라는 단서를 얻게 되었고, 이를 이용해 방광의 상피밑층 신경섬유와 상피세포에서 P2X3 수용체와 VR1의 작용을 조절하면 과민성 방광의 증상을 경감시킬 수 있음이 예상된다. 이 연구결과를 통해 P2X3 수용체와 VR1의 발현 조절은 배뇨근 과반응에 대한 적은 부작용과 높은 치료효과를 얻을 수 있는 새로운 치료법 개발에 응용 할 수 있을 것으로 보인다. Two of the synaptic receptors involved in the regulation of micturition, P2X3 receptor, which is operated by ATP, and vanilloid receptor 1 (VR1), which is operated by capsaicin, are regarded as newcomers. To investigate the possibility that these receptors act as therapeutic targets for treatment of an overactive bladder, we investigated their distribution and reciprocal relationship. Eight-week-old Sprague Dawley rats were injected with retrograde nerve tracer within the bladder wall, and 15 rats were injected with 0.5% acetic acid inside the bladder. After a week, the animals were killed, and their dorsal root ganglia (DRG) at the levels of L6 and S1 were harvested. Immunohistochemistry or Western blot analysis of P2X3 and VR1 were performed on the DRG. The DRG neurons with afferent fibers from the bladder had increased expression of VR1 and downregulated P2X3 receptors. The P2X3 receptor and VR1 seemed to account for the important parts of the hypersensitivity of the inflammatory bladder. We conclude that the simultaneous modulations of both P2X3 receptor and VR1 may have a synergic effect on the treatment of overactive bladder and may produce greater response rates.

The purpose of this study is to use waste resources and improve the physical properties of concrete by crushing LCD waste glass for which an appropriate recycling method has not been developed and mixing it into concrete. LCD waste glass was pulverized into two sizes with an average particle diameter of 5㎛ and 10㎛ using a ball mill, and cement paste and concrete were produced by substituting 10% and 20%. Cement paste was used for XRD analysis, and concrete was used for compressive strength test, micropore structure test, and repid chloride penetration test. As a result of analyzing the hydrate composition by XRD, the reduction of calcium hydroxide due to LCD substitution was roughly confirmed, and in the result of compressive strength, concrete including LCD powder showed higher strength than OPC, and was up to 24% higher at 5㎛-20%. In addition, the replacement of the LCD powder reduced the size of the pores inside the concrete, and reduced the penetration rate of chloride. The reason for this result is believed to be due to the pozzolanic reaction caused by SiO2 contained in a large amount of waste LCD glass. Calcium hydroxide produced by the hydration reaction reacts with SiO2 to form C-S-H gel, and it is thought that the gel fills the voids in the concrete to increase the compressive strength and prevent the penetration of harmful ions.
