        Anti-asthma and antitussive effects of a fermented extract of a mixture of Ramulus mori, Anthriscus sylvestris, and Salvia plebeian

        Respiratory immunity is getting more importantrecently due to outbreak of respiratory diseases andincreasing the concentration of fine dust. The aim of thisstudy was to investigate respiratory protection effect of afermented extract of medicinal plants (FEMP) containingRamulus mori, Salvia plebeia, and Anthriscus sylvestris. The expression levels of IL-8 and IL-17 in LPS/poly-Larginine(PLA) and FEMP-cotreated A549 cells were lowerthan those in LPS/PLA only-treated cells. The levels ofIgE, IL-17, and IL-4 in the bronchoalveolar lavage fluid(BALF) and serum of FEMP-treated mice with ovalbumin/LPS-induced asthma were lower than the control levels. The lung inflammation score and the number of inflammatorycells in the BALF decreased by FEMP treatment. Inthe citric acid-induced coughing guinea pig, the FEMPtreatment decreased the number of coughs. Therefore,FEMP shows anti-asthmatic and antitussive activitieswithout hepatotoxicity and can be used as a compoundaiming to improve respiratory health.

        The protective effect of Areca Semen and Toosendan Fructus mixture in a chronic model of reflux esophagitis

        Objective:The aim of present study was to clarify the effect of Areca Semen and Toosendan Fructus Mixture (AT-mix) on chronic reflux esophagitis (CRE) in rats. Methods:The antioxidant activity of AT-mix was measured through DPPH and ABTS radical scavenging activities in vitro. CRE was induced in SD rats (5 weeks, male) by ligating the border forestomach and granular portion with 2-0 silk and the duodenum near the pyloric portion was covered with 2-mm wide piece of 18-Fr Nélaton catheter. And then rats were treated AT-mix 200 mg/kg one daily for 14 days. The anti-oxidant and inflammatory protein levels were evaluated using western blotting. Results:Gross lesion of esophageal mucosa after AT-mix treatment showed a superior enhancement compared with that of CRE control rats. AT-mix treatment strongly reduced both DPPH and ABTS radical scavenging activities (DPPH, IC50 8.15±0.14 g/mL; ABTS, IC50 24.69±0.03 g/mL, repspectively). Levels of the NADPH oxidase subunit including NOX4 and p22phox increased in CRE control rats. Otherwise, AT-mix treatment significantly reduced. The activation of Nuclear factor-erythroid 2-related factor 2 (Nrf2) led to significantly the up-regulation of HO-1. The inhibition of IB phosphorylation led to NF-B inactivation. Subsequently, NF-B inactivation significantly induced the decrease of COX-2, iNOS, TNF-, and IL-6 protein expressions. Conclusion:Taken together, these results suggest that AT-mix treatment can attenuate the esophageal mucosal ulcer though inhibiting NF-B pathway and enhancing Nrf2/HO-1 pathway. Thus, the additional mechanism study about AT-mix would need for the development as a safe herbal therapy for CRE.

        닭고기와 잘 어울리는 췌장라이페이즈 억제능을 가진 한약재의 비교평가

        Objectives:In this study, the inhibitory activities of Korean Medicine against pancreatic lipase in vitro and biochemical analyses in vivo were measured to determine its possibility as a well-matched sauce material with chicken. Methods:The inhibitory activity on pancreatic lipase enzyme of 11 samples were evaluated in vitro and then 5 samples were selected. The activity of pancreatic lipase was investigated using orlistat as a positive control. Animals were divided into eight groups (n =7). The experimental groups except for normal group were fed 60% high-fat diet for 7 days. 5 samples were orally administered at a dose of 200 ㎎/㎏ body weight and orlistat were orally administrated at a dose of 60 ㎎/㎏ body weight for 7 days. Biochemical anaylses of 5 samples were executed based on lipid parameters analysis. Results:Korean Medicines with an IC50 of below 1 ㎎/㎏ were Scutellariae Radix, Gardeniae Fructus, Theae Folium Coptidis Radix, and Mori Cortex Radicis. Body weight change of Mori Cortex Radicis reduced significantly, however fecal triglyceride couldn`t regulate effectively. The most excellent inhibitory effect of pancreatic lipase showed in Scutellariae Radix treatment and also regulated significantly serum triglyceride and total cholesterol. Moreover, the supplementation of Coptidis Radix excreted meaningfully triglyceride to fece. Conclusions:In conclusion, Coptidis Radix may exert anti-obesity effect by directly inhibiting pancreatic lipase, which would prevent the absorption of lipid from the small intestine. Besides, Mori Cortex Radicis may led to the decrease of the body weight via the different pathway.

        MIA로 유발된 골관절염 동물모델에서 산수유 추출물의 골관절염 개선 효과

        Objectives: Osteoarthritis (OA) is a chronic and degenerative joint disease characterized by progressive degeneration of articular cartilage. Inflammation is a recognized and important factor of OA progression. The present study was designed to investigate the protective effect of Corni Fructus water extract (CFW) on a monosodium iodoacetate (MIA)-induced rat model of OA. Methods: Osteoarthritis was induced by injection of MIA (50 µL; 80 mg/mL) into the knee joint cavity of rats. After an adaptation period for seven days, the rats were divided into 4 groups (n=8/group): normal, control, indomethacin-treated (5 mg/kg), and CFW-treated (200 mg/kg) groups. The rats were treated orally for 14 days. Pain was evaluated by determining hind paw weight distribution. For biochemical analyses, we measured the changes in reactive oxygen species (ROS) and peroxynitrite (ONOO-) in the knee joint. The presence of anti-oxidant proteins and inflammatory proteins was determined by western blotting. Results: The administration of CFW significantly improved the hind paw weight distribution. The ROS and ONOO- levels of knee joint were significantly decreased in the CFW group. CFW inhibited the production of inflammatory mediators, such as COX-2, and inflammatory cytokines, including IL-6 and IL-1β, via the NF-κB signaling pathway. The expression of anti-oxidant enzymes, such as catalase and GPx-1/2 also increased significantly. Conclusions: The findings indicate that CFW has a therapeutic and protective effect on OA by suppression of inflammation. Therefore, CFW could represent a potential and effective candidate for OA treatment.

        지지면 변화에 따른 교각운동이 복횡근 두께와 균형능력에 미치는 영향에 관한 연구

        Purpose: The purpose of this study was to investigate the effect of transverse abdominis ratio and balance ability during bridge exercises on different support surface. Methods: The subjects were 20 students at G University. Ultrasound equipment was used to measure the transverse abdominisratio and the computerized balance measurement equipment was used to measure the balance ability before and after bridge exercises on different support surface. Results: Transverse abdominis ratio change between the groups was a significant difference only in the unstable support surface training group (p<.05). Normal standing eye close balance ability change between the groups was significantly different only in the unstable support surface training group (p<.05). The changes in the transverse abdominis ratio after 7 days of exercise and 14 days after exercise were significantly different between the groups (p<.05). Normal standing eye close balance ability showed a significant difference at 7 days after exercise and 14 days after exercise (p<.05). Conclusions: It is more effective to balance ability and ratio of the transverse abdominis when bridge exercises on an unstable support surface than to perform bridge exercises on a stable support surface. Therefore, we propose an effective program for patients with instability of the lumbar in clinical practice. bridge exercises on different support surface.
