Verotoxin을 산생하지 않은 Escherichia coli O157과 verotoxin을 산생하는 E coli(VTEC)를 건강한 소의 분변에서 분리하여 생화학적 및 유전적인 특성에 대해서 비교하였다. E coli O157 : nonH7(운동성은 있으나 H혈청형이 7이 아님)의 sorbitol 분해능과 ${\beta}-glucurondase$ 활성은 E coli O157 : H7이 나타내는 것과는 차이가 있었다. 그리고 uidA 유전자는 verotoxin 산생능과 상관없이 sorbitol과 ${\beta}-glucuronidase$ 음성인 E coli O157 : H7에서 특이적으로 검출되었다. 한편 6개 목장에서 수거한 소 분변 45예에서 VTEC를 분리한 결과, 7주(15.6%)가 분리되었으며 이들은 모두 sorbitol을 분해하였으며 ${\beta}-glucurondase$ 활성이 있었으나 장벽 부착인자를 지배하는 eaeA 유전자가 없었다. 비록 소가 VTEC의 보균원으로 추정되나, 정상 소에서 분리한 VTEC는 eaeA 유전자가 결여된 균주가 많으므로 공중위생학상 eaeA 유전자를 보유한 E coli 보다 위해성이 낮으며, 이러한 결과는 왜 사람에서 유행하는 VTEC 혈청형과 소에서 유행하는 것과 차이가 있는지를 일부 설명해준다. Verotoxin non-producing E coli O157 strains have been isolated from cattle feces and compared in particular regard to biochemical properties and genotypes with verotoxin-producing E coli (VTEC). E coli O157 : nonH7 strains had different phenotypes in sorbitol fermentation and ${\beta}-glucuronidase$ activity from E coli O157 : H7. Regardless of verotoxin production ability of E coli O157 : H7, uidA gene was uniquely detected from sorbitol and ${\beta}-glucuronidase$ negative E coli O157 : H7. Forty five fecal samples from 6 dairy farms were obtained and VTEC was detected as 15.6% (7 strains) of the samples. Most VTEC isolates were positive for sorbitol fermentation and ${\beta}-glucuronidase$ activity but negative for eaeA gene. This study suggested that cattle could be a reservior for VTEC. However, absence of eaeA gene in VTEC isolates from most of healthy cattle suggested that they might be less virulent than eaeA-positive E coli against human health.

The scale of renewable power generation facilities such as wind power and solar power generation is being enlarged. Due to the increased scale of renewable power generation facilities, the system stability has been significantly affected by the increased distributed power sources, and various countries such as Germany and the United States are enacting standards to prevent deterioration of system stability. In this paper, we analyze the LVRT (Low Voltage Ride Through) conditions and reactive current injection standards established by Germany and Korea Electric Power Corporation (KEPCO). And, the LVRT control algorithm is applied to the designed inverter, and the operation of the grid-connected inverter was confirmed through simulation based on the LVRT boundary. In addition, the injection characteristic algorithm1 of active and reactive current in LVRT situation was modeled, and the modeled algorithm was verified by checking the simulation performance.

With the increase in renewable power generation facilities, strict regulations on grid connection standards are being presented. The standards of Low Voltage Ride Through(LVRT) and High Voltage Ride Through(HVRT), which are representative grid connection standards to improve grid stability by maintaining grid connection in low and high voltage situations when a grid failure occurs, are gradually being subdivided. In this paper, the LVRT and HVRT control algorithms suggested in the transmission and distribution system connection regulations of KEPCO(Korea Electric Power Corporation) are applied to the grid-connected 3-level TNPC(T-Type Neutral Point Clamped) inverter. And in the VRT situation, the active and reactive current injection strategies were applied to the grid connected inverter algorithm. In addition, various simulation cases were analyzed, such as LVRT & HVRT conditions of transmission and distribution system. In the distribution system LVRT simulation, the grid voltage operated at 150ms and 1500ms, respectively, within 0.5p.u. and 0.75p.u. In the transmission system simulation, the LVRT operation and the HVRT operation were also simulated. By analyzing various simulation cases, the characteristics of the VRT cases was analyzed in detail and VRT control performance of the designed TNPC inverter was verified.

A lateral flow immunoassay kit based on antigen-antibody interactions was developed to detect residues of beta-lactams, quinolones, tetracyclines, and sulfonamides in farmed fish. Group-specific antibodies showing cross-reactivity with other antibiotics in the same group were produced in rabbits. The rabbits were immunized eight times to obtain the maximum titers. Antibodies were extracted from the antisera collected from the immunized rabbits and produced group-specific reactions with antibiotics from the four groups. A kit was prepared that optimize conditions for the antigen-antibody reaction, using colloidal gold conjugated antibodies, and was designed to detect the four groups of antibiotics simultaneously. The kit enabled the detection of antibiotics in the four groups at below maximum residue limits (MRLs), which were 200 μg/kg for tetracyclines, 100 μg/kg for sulfonamides, 50 μg/kg for beta-lactams, and 100 μg/kg for quinolones. The cross-reactivity of the antibodies ranged from 10-80% for the sulfonamides, 20- 100% for tetracyclines, 38-100% for quinolones, and 20-100% for the beta-lactams, confirming that the antibodies were group specific. The test kit was used 30 times to examine spiked antibiotics at the limits of detection (LODs) and all produced positive results, indicating high sensitivity. The LODs for the assay ranged from 4-20 ng/mL for beta-lactams, 25-50 ng/mL for sulfonamides, 20-100 ng/mL for tetracyclines, and 30-80 ng/mL for quinolones, and there were no false negative reactions at above these LODs. In addition, all of the LODs of the developed kit were correlated with high-performance liquid chromatography (HPLC) data. Our lateral flow immunoassay kit can simultaneously detect antibiotic residues from a large number of fish samples rapidly, strengthening the safety of domestic farmed and imported fish.
Cephalosporin C 발효에서 세포고정의 최적화를 위한 통계학적 실험계획법

A 24 factorial design was performed to immobilize Cephalosporium acremonium in optimized alginate beads. Factors include alginate concentration, hardening solution, polyethyleneimine(PEI) concentration, and bead size. The statistical analyses of factorial effects resulted in finding two important parameters. bead size and PEI concentration, which influenced the diffusion of cephalosporin C(CPC) . According to Pareto chart. the main effect of bead size was 6.5times higher than that of PEI concentration. When C. acremonium cells were immobilized in optimized alginate beads, the immobilized cells produced CPC more than 300 hours continuously without any loss of cell activities.
