      • DADI/MIKE Spectrometry를 이용한 Peptides의 아미노산 배열결정

        Metastable transition studies로 peptides의 아미노산 배열을 추적하였다. 본 연구에서는 metastable transitions를 확인하기 위해 DADI/MIKE spectrometry를 적용하였다. pentapeptide인 enkephalin 유도체, oligopeptide들의 mixture 및 isomeric peptides mixture에 stepwise DADI/MIKE spectrometry를 적용한 결과 배열결정에 주요한 metastable ionpeaks를 얻을 수 있었다. 이러한 방법으로 얻어진 DADI/MIKE spectra로 용이하게 아미노산 배열을 결정할 수 있었다. Sequence analysis of peptides was investigated by metastable transition studies. In the present work, metastable transitions of peptides were detected by direct analysis of daughter ions (DADI) or mass analyzed ion kinetic energy (MIKE) spectrometry. Stepwise DADI/MIKE spectrometry of enkephalin derivative, oligopeptides mixture, and isomeric peptides mixture were examined. Since prominent metastable ion peaks were observed, it was possible to elucidate peptide sequences from the DADI/MIKE spectra.

        Essential oil components were isolated from Korean aromatic tobaccos by using a simultaneous distillation and extraction apparatus. The essential oils were analyzed by GC/M S and date system. Forty-two volatile components were identified on the basis of their mass spectra. Determination of contents of essential oil components from Hyangcho and Sohyang was achieved by pseudo-multiple ion selection (Ml S) technique. Varietal differences were detected from the quantitative comparison of the MIS data.

      • A Correlation between Oxygen Radical-Induced Mutations and DNA Adducts Formation in Salmonella typhimurium

        산소라디칼에 의한 8-OH-dG의 형성으로 반영된 DNA의 산화손상이 돌연변이, 발암 및 노화의 주요원인으로 추정되고 있다. 본 연구에서는 산소라디칼 발생물질로 처리된 살모넬라균의 DNA에서의 8-OH-dG의 형성을 최근 개발된 HPLC-EC 방법으로 우선 동정하였다. 산소라디칼을 발생시키는 물질들의 처리에 의해 살모넬라 TA100, TA102에서 8-OH-dG가 형성됨을 확인되었다. 동일한 처리에 의한 돌연변이 활성을 Ames test로 측정한 결과 reversion 부위에 GC 염기쌍을 가진 TA100에서 돌연변이 정도가 증가함을 확인함으로서 8-OH-dG가 돌연변이 유발 부위일 가능성이 제기되었다. 한편, reversion 자리에 AT 염기상을 가진 TA102에서도 돌연변이가 유발됨을 확인함으로서 8-OH-dG외의 염기 손상도 산소라디칼에 의한 돌연변이에 관여함을 추정되었다. 또한 oxyR gene이 결여된 변이주의 결과로부터도 산소라디칼에 의한 DNA 손상과 돌연변이 유발간의 상관관계가 추정되었다. Oxidative damage to DNA, refected in the formation of 8-hydroxy-2'-deoxyguanosine (8-OH-dG), may be important in mutagenesis, carcinogenesis and aging process. We have applied a recently developed high-performance liquid chromatography with electrochemical detection (HPC-EC) method for 8-OH-dG adducts to assess their formation in DNA from Salmonella typhimurium exposed to chemical oxidants. 8-OH-dG adducts in DNA were formed in Salmonella tester strains, TA100 and TA102, under the treatment with prooxidants such as hydrogen peroxide and t-butyl hydroperoxide, which can produce oxygen free radicals. Using the liquid pre-incubation assay, oxidant-induced mutations were also found in TA100 and TA102. A correlation between the prooxidant-induced 8-OH-dG formation and mutations in TA100, which contains GC base pairs at the site of reversion,suggests that the 8-OH-dG adduct may be a potentially mutagenic lesion. That mutations were also seen in TA102 which contains AT base pairs at the site of reversion, suggesting that adducts of bases other than deoxyguanosine may also be important in the mutagenic ability of prooxidants. The result with TA4117 $(oxyR^+)$ and TA4119 $(oxy{\Delta})$, oxyR gene-deficient mutant, also suggests that there may be a correlation between active oxygen-induced mutations and DNA adducts formation.

      • 향끽미종 잎담배 성분조성에 관한 연구 III. 정유성분의 특성조사

        새로이 고안된 distillation/solvent extraction방법을 이용하여 오리엔트산 향끽미종과 한국산 향끽미종 잎담배의 정유성분을 분리하였다. 정유성분들을 고성능 가스 크로마토그라지로 분석한 결과 각 잎담배로 부터 특징적인 GC 프로파일을 얻을수 있었다. 얻어진 GC분석자료를 정량적으로 비교함으로써 품종간의 상이점이 확인되었다. Essential oil components were isolated both from Oriental and- Korean using a modified distillation/solvent extraction method. The essential oils were then analyzed by high-resolution glass capillary gas chromatography, to give characteristic GC profile from each tobacco. Varietal differences were detected from the quantitative comparison of the GC data.

        Less-volatile organic components in leaves from aromatic tobaccos of different varieties, both Oriental and Korean types were isolated and concentrated using a simple apparatus with fewer manipulations. Each less-volatile concentrate was then subjected to spectrophotometric recording in the visible range, to thin-layer chromatographic group separation, and high-performance liquid chromatographic profile analysis. The methods allow detection of significant quantitative differences in visible absorption spectra, TLC patterns, and high resolution HPLC profiles among varieties. 오리엔트산 및 한국산 향끽미종 잎담배의 휘발성이 적은 성분들을 간단한 장치와 간단한 조작을 통해 분리 농축하였다. 각 휘발성이 적은 농축물들은 가시 영역에서 흡광도를 측정하고 박층 크로마토그라피 (TLC)와 고성능 액체 크로마토그라피 (HPLC)에 의해서 상세한 조성분석을 하였다. 각 품종간의 정량적인 차이가 가시광선 흡수 스펙트라, TLC 패턴, HPLC프로파일 상에서 확인되었다.

        Xanthine oxidase 활성 및 형전환에 미치는 구리이온의 영향

        Copper intoxication and disturbance of copper metabolism induced various oxygen- derived free radicals related damages. The effect of copper ion on xanthine oxidase activity and type conversion of the enzyme which is concerned to generation of reactive oxygen species, was investigated. It was observed that xanthine oxidase activity was increased by addition of copper ion in the reaction mixture in proportional to the concentration of the metal ion until 60 mcM while the enzyme activity was inhibited in higher concentration of copper treatment. On the other hand, xanthine dehydrogenase activity was inhibited by copper ion addition with concentration dependently. Preincubation of enzyme source with 30 mcM of copper ion, which concentration marked increased the xanthine oxidase activity, unchanged the enzyme activity and type conversion compare to control in vitro system. It was also observed that copper induced xanthine oxidase activity and the enzyme type conversion was protected by dithiothreitol and penicillamine. These results indicate that the increment of the type conversion of xanthine oxidase necessarilly need the presence of copper ion in enzyme assay system.
