The current study was undertaken to determine the effects of age and feeding level of the concentrates on lipogenesis of adipose tissue and adipocyte size in the Hanwoo bulls. A total of 48 bulls was allocated into 3 feeding groups, 85% 100%, 115% of the requirement for Hanwoo cattle. Four animals per feeding level were killed at 15, 18, 24, 30 months of age for the measurement of lipogenesis and adipocyte size. Lipogenesis was measured with subcutaneous, intermuscular adipose tissue explants in culture by the amount of acetate or glucose converted into the total lipid and adipocyte diameters were measured with subcutaneous, intermuscular, and perirenal fat tissues at each slaughtering age. Initially, we measured the effect of insulin on lipogenesis of Hanwoo adipose tissue to establish an optimal condition for measuring lipogenesis. Insulin had no effect on lipogenesis. As the bulls get older, lipogenesis from acetate was reduced and similar trend was found for glucose. Lipogenesis from acetate of subcutaneous adipose tissue was higher than that of intermuscular adipose tissue. However, there was no significant difference in lipogenesis from glucose between the two adipose tissues. Adipocyte diameter was continuously increased from 15 to 30 months of age in the Hanwoo bulls. Adipocytes from perirenal adipose tissue were bigger than those from subcutaneous or intermuscular adipose tissues. Feeding level of concentrates did not affect either lipogenesis or adipocyte size.

The current study was undertaken to determine the effects of sex and energy restriction on lipogenesis and lipogenic enzyme activities in the pigs. Lipogenesis was measured as glucose conversion to total lipid and activities of glucose-6-phosphate dehydrogenase(GPDH), 6-phosphogluconate dehydrogenase (6-PGDH) and NADP-malate dehydrogenase(ME) were determirfed by measuring NADPH production. The lipoprotein lipase (LPL) activity and blood lipid pattern were also measured. There were no significant differences between gills and boars in lipogenesis, 6-PGDH, ME and LPL activities. Energy restriction(25%) for four weeks did not affect lipogenesis and the activities of GPDH, 6-PGDH and ME. However, the LPL was lower in the energy restricted gills (p $lt; 0.001). On the other hand, fasting for 24 hr reduced lipogenesis(p $lt; 0.01) and ME(p $lt; 0.01) and tended to reduce GPDH and 6-PGDH. The LPL was also reduced in the fasted pigs(p $lt; 0.001). Serum total cholesterol, HDL-C, LDL-C and HDL-C/T-C ratio were not significantly affected by sex, energy restriction and fasting. Boars showed lower serum triglyceride(TG) concentration than gills and fasted pigs showed lower serum TG concentration than fed pigs(p $lt; 0.05). These results indicated that LPL activity represented the lipogenic status in the pig better than other lipogenic enzymes.

The current study was undertaken to determine the effects of increasing body weight, sex, and energy restriction on the LPL mRNA abundance and LPL activity of adipose tissue in the finishing pigs. Serum lipoprotein pattern. triglyceride concentrations and their correlation with LPL activity were also investigated. Serum total cholesterol(C). HDL-C. LDL-C and HDL-C/T-C ratio were not significantly affected by body weight. sex or energy restriction. The triglyceride concentration increased with increasing body weight, being greater in gills than in boars(p$lt;0.05). The serum triglyceride concentration of full-fed pigs tended to be higher than that of energy-restricted pigs. Heavier pigs showed higher LPL activity than lighter pigs(p$lt;0.05), gills showing higher LPL activity than boars. and full-fed pigs showing higher LPL activity than energy-restricted pigs. Gills showed higher LPL mRNA abundance than boars, full-fed pigs showing higher LPL mRNA abundance than energy-restricted pigs. However. there was no significant difference between heavier and lighter pigs in the LPL mRNA abundance. The LPI_ activity was correlated with the triglyceride concentration, but not with the HDL-C, in individual experiments except the energy-restriction experiment. These results suggest that LPL is an important indicator for lipogenesity in pigs.
