        ZNF552, a novel human KRAB/C2H2 zinc finger protein, inhibits AP-1- and SRE-mediated transcriptional activity

        In this study, we report the identification and characterization of a novel C2H2 zinc finger protein, ZNF552, from a human embryonic heart cDNA library. ZNF552 is composed of three exons and two introns and maps to chromosome 19q13.43. The cDNA of ZNF552 is 2.3 kb, encoding 407 amino acids with an amino-terminal KRAB domain and seven carboxyl-terminal C2H2 zinc finger motifs in the nucleus and cytoplasm. Northern blotting analysis indicated that a 2.3 kb transcript specific for ZNF552 was expressed in liver, lung, spleen, testis and kidney, especially with a higher level in the lung and testis in human adult tissues. Reporter gene assays showed that ZNF552 was a transcriptional repressor, and overexpression of ZNF552 in the COS-7 cells inhibited the transcriptional activities of AP-1 and SRE, which could be relieved through RNAi analysis. Deletion studies showed that the KRAB domain of ZNF552 may be involved in this inhibition. [BMB reports 2010; 43(3): 193-198]

        Amelioration of radiation-induced liver damage by p-coumaric acid in mice

        Radiation-induced liver damage (RILD) is a spiny problem in radiotherapy or other circumstances that exposure to radiation. The need for radioprotective agent is increasing to protect liver tissue. This study aimed to explore the hepatoprotective effect of p-coumaric acid (CA) against RILD. C57BL/6 male mice were exposed to 4 Gy irradiation and administrated with CA for 4 days starting on the same day of irradiation. Mice were sacrificed to obtain blood and liver tissues on day 3.5 or 14 post irradiation, respectively. The blood and liver tissues were collected. As compared with the only irradiated group, CA supplementation improved liver morphology, decreased serum alanine aminotransferase and aspartate aminotransferase, inhibited BCL2-associated X (BAX) protein expression, and improved the mice hematopoietic function. CA at the dose of 100 mg/kg body weight showed better effect compared to the other doses. Thus, CA might possess potential to protect against RILD.

      • Prediction of tensile strength degradation of corroded steel based on in-situ pitting evolution

        Steel is becoming increasingly popular due to its high strength, excellent ductility, great assembly performance, and recyclability. In reality, steel structures serving for a long time in atmospheric, industrial, and marine environments inevitably suffer from corrosion, which significantly decreases the durability and the service life with the exposure time. For the mechanical properties of corroded steel, experimental studies are mainly conducted. The existing numerical analyses only evaluate the mechanical properties based on corroded morphology at the isolated time-in-point, ignoring that this morphology varies continuously with corrosion time. To solve this problem, the relationships between pit depth expectation, standard deviation, and corrosion time are initially constructed based on a large amount of wet-dry cyclic accelerated test data. Successively, based on that, an in-situ pitting evolution method for evaluating the residual tensile strength of corroded steel is proposed. To verify the method, 20 repeated simulations of mass loss rates and mechanical properties are adopted against the test results. Then, numerical analyses are conducted on 135 models of corrosion pits with different aspect ratios and uneven corrosion degree on two corroded surfaces. Results show that the power function with exponents of 1.483 and 1.091 can well describe the increase in pit depth expectation and standard deviation with corrosion time, respectively. The effect of the commonly used pit aspect ratios of 0.10–0.25 on yield strength and ultimate strength is negligible. Besides, pit number ratio α equating to 0.6 is the critical value for the strength degradation. When α is less than 0.6, the pit number increases with α, accelerating the degradation of strength. Otherwise, the strength degradation is weakened. In addition, a power function model is adopted to characterize the degradation of yield strength and ultimate strength with corrosion time, which is revised by initial steel plate thickness.

      • Risk Assessment of Breast Cancer in Guangdong, China: A Community-based Survey

        Objectives: Compared with Western countries, the incidence rates for breast cancer in China are still low. However, breast cancer appears to be hitting Chinese women at a much younger age, with a peak between 40 and 50 years. Furthermore, breast tumors of Asian women have molecular and genetic characteristics that are different from those of Caucasian women. Methods: A community-based study was designed to evaluate the relationship between lifestyles and breast cancer risk in Chinese women residing in Guangzhou. 16,314 subjects completed the questionnaire. Potential confounding factors included sociodemographic characteristics. Results: 33 individuals reported a history of breast cancer, yielding a prevalence rate of 202.3/100000. Associations between subjects' demographic and breast cancer risk factors were assessed. Breast cancer is associated with family history of breast cancer, X-rays received, benign breast disease and hyperlipoidemia or hypercholesteremia with elevated odds ratios. Conclusions: Family history of breast cancer, X-ray received benign breast disease and hyperlipoidemia or hypercholesteremia were significantly associated with risk of breast cancer and may havepotential for breast cancer risk assessment.

        Identification and Validation of Four Novel Promoters for Gene Engineering with Broad Suitability across Species

        The transcriptional capacities of target genes are strongly influenced by promoters, whereas few studies have focused on the development of robust, high-performance and cross-species promoters for wide application in different bacteria. In this work, four novel promoters (P<sub>k.r</sub>tufB, P<sub>k.r</sub>1, P<sub>k.r</sub>2, and P<sub>k.r</sub>3) were predicted from Ketogulonicigenium robustum and their inconsistency in the -10 and -35 region nucleotide sequences indicated they were different promoters. Their activities were evaluated by using green fluorescent protein (gfp) as a reporter in different species of bacteria, including K. vulgare SPU B805, Pseudomonas putida KT2440, Paracoccus denitrificans PD1222, Bacillus licheniformis and Raoultella ornithinolytica, due to their importance in metabolic engineering. Our results showed that the four promoters had different activities, with P<sub>k.r</sub>1 showing the strongest activity in almost all of the experimental bacteria. By comparison with the commonly used promoters of E. coli (tufB, lac, lacUV5), K. vulgare (Psdh, Psndh) and P. putida KT2440 (JE111411), the four promoters showed significant differences due to only 12.62% nucleotide similarities, and relatively higher ability in regulating target gene expression. Further validation experiments confirmed their ability in initiating the target minCD cassette because of the shape changes under the promoter regulation. The overexpression of sorbose dehydrogenase and cytochrome c551 by P<sub>k.r</sub>1 and P<sub>k.r</sub>2 resulted in a 22.75% enhancement of 2-KGA yield, indicating their potential for practical application in metabolic engineering. This study demonstrates an example of applying bioinformatics to find new biological components for gene operation and provides four novel promoters with broad suitability, which enriches the usable range of promoters to realize accurate regulation in different genetic backgrounds.

      • Effects of Partner Characteristic, Partnership Quality, and Partnership Closeness on Cooperative Performance: A Study of Supply Chains in High-tech Industry

        Owing to the rapid development of information technology, change of supply chain structures, trend of globalization, and intense competition in the business environment, almost all enterprises have been confronted with unprecedented challenges in recent years. As a coping strategy, many of them have gradually viewed suppliers as “cooperative partners”. They drop the conventional strategy of cooperating with numerous suppliers and build close partnerships with only a small number of selected suppliers. This paper aims to explore partnerships between manufacturers and suppliers in Taiwan’s hightech industry. Through a review of literature, four constructs, including partner characteristic, partnership quality, partnership closeness, and cooperative performance are extracted to be the basis of the research framework, hypotheses, and questionnaire. The questionnaire is administered to staff of the purchasing and quality control departments in some high-tech companies in Taiwan. The proposed hypotheses are later empirically validated using confirmatory factor analysis (CFA) and structural equation modeling (SEM). This paper expects to provide substantial suggestions to enterprises in the high-tech industry and help them develop mutually beneficial partnerships.

      • Differential Protein Expression Profile Between CD20 Positive and Negative Cells of the NCI-H929 Cell Line

        At present, multiple myeloma (MM) remains an incurable disease and cologenic cells may be responsible for disease relapse. It has been proposed that CD20+/CD138- NCI-H929 cells could be hallmarks of MM clonogenic cells. Here, the immunology phenotype of NCI-H929 cells is described. Only a small population of CD20+/CD138- cells (<1%) was found in the NCI-H929 cell line, but CD20+/CD138- cells were not detected. We found that CD20+/CD138+ cells were able to exhibit cologenic capacity by colony formation assay and continuous passage culture. Proteins were analyzed by 1D-SDS-PAGE and TMT based quantitative differential liquid chromatography tandem mass spectrometry (LC-MS/MS). 1,082 non-redundant proteins were identified, 658 of which were differentially expressed with at least a 1.5-fold difference. 205 proteins in CD20+ cells were expressed at higher levels and 453 proteins were at lower levels compared with CD20- cells. Most proteins had catalytic and binding activity and mainly participated in metabolic processes, cell communication and molecular transport. These results proved that there are different biological features and protein expression profile between CD20+ and CD20- cells in the NCI-H929 cell line.

      • Development of Novel Pineapple Peel Juice Through Lactic Acid Fermentation for Enhancing Antioxidant Capacity and Stability

        Pineapples are tropical fruit that are rich in vitamins, enzymes and fibers with many health benefits not only in pulps but also peels. However, the peels of pineapples are normally removed and dropped. The pulps and peels of four pineapple cultivars, Milk, Honey Fragrance, Smooth Cayenne and Golden Diamond pineapples, were juiced, filtered and analyzed, and the Golden Diamond pineapple was found to have highest antioxidant capacity. The pulp and peel juices of Golden Diamond pineapple were inoculated with Enterococcus faecium LYC91 and various concentration calcium carbonate to ferment at 37°C for 24 hours follow by bacterial and antioxidant capacity analysis. The results indicated that the fermented pineapple peel juice with calcium carbonate had highest bacterial number, total polyphenol content, DPPH radical scavenging ability, Fe2 + chelating ability and reducing power activity than pulp juice and unfermented pineapple juice. The viable bacterial number of fermented pineapple pulp and peel juices with calcium carbonate remain 8.0 log CFU/mL after stored at 4°C for 1 month. The freeze-dried powder of the fermented pineapple pulp and peel juices with calcium carbonate retained the viable cell number of 9.0 log CFU/g after stored at 4°C for three months. These results showed that the fermented pineapple pulp and peel juices with calcium carbonate were novel pineapple lactic acid fermented drinks with growth potential.
