To analyze the effects of clearance joint number and dynamic interaction on responses of the flap actuation system, a dynamic analysis model is proposed based on a modified contact force model. The modified contact force model can take contact properties with small clearance, heavy load, large contact area and variable contact stiffness coefficient into consideration for the flap actuation system. Numerical results show that the actuation system presents violent fluctuation due to the dynamic interaction between multiple clearance joints, and the clearance joint closest to the input part suffers more serious contact effects. Furthermore, the combination motion modes between the multiple clearance joints are helpful to judge and analyze the motion state and dynamic behaviors of actuation systems. These simulation findings can provide a theoretical foundation for the optimization design, control strategy and engineering experiments of the actuation systems with multiple clearances.

The diagnosis of malignant mesothelioma (MM) remains a clinical challenge and the fluorescence in situ hybridization (FISH) assay has been reported to be one promising tool. The present meta-analysis aimed to establish the overall diagnostic accuracy of FISH for diagnosing MM. After a systematic review of English language studies, the sensitivity, specificity and other measures of accuracy of FISH in the diagnosis of MM were pooled using random-effects models. Summary receiver operating characteristic curves were applied to summarize overall test performance. Nine studies met our inclusion criteria, the pooled sensitivity and specificity for FISH for diagnosing MM being 0.72 (95% CI 0.67-0.76) and 1.00 (95% CI 0.98-1.00), respectively. The positive likelihood ratio was 34.5 (95% CI 14.5-82.10), the negative likelihood ratio was 0.24 (95% CI 0.16-0.36), and the diagnostic odds ratio was 204.9 (95% CI 76.8-546.6), the area under the curve being 0.99. Our data suggest that the FISH assay is likely to be a useful diagnostic tool for confirming MM. However, considering the limited studies and patients included, further large scale studies are needed to confirm these findings.

A closed-loop coupling model at the system-level is developed to analyze the effects of joint clearance on the dynamic responses of electromechanical aileron actuation system. The proposed model considers the coupling effects between the electromechanical actuator (EMA) control performance and dynamic characteristics of linkage mechanism with joint clearance. Besides, the experiments are conducted in a test rig, which verifies the effectiveness of the proposed closed-loop coupling model. The nonlinear contact force model and modified Coulomb friction model are adopted in the joint clearance of the linkage mechanism, and the influences of clearance size on the dynamic behaviors of electromechanical aileron actuation system are studied. The numerical and experimental results indicate that the novel closed-loop coupling model, considering the EMA control performance and dynamics of linkage mechanism with joint clearance at the same time, is an effective model to predict the dynamic characteristics of electromechanical aileron actuation with joint clearance, which provides a practical method to analyze the dynamic performance of electromechanical coupling multibody systems with joint clearance.

LBH is a transcription factor as a candidate gene for CHD associated with partial trisomy 2p syndrome. To identify potential LBH-interacting partners, a yeast two-hybrid screen using LBH as a bait was performed with a human heart cDNA library. One of the clones identified encodes αB-crystallin. Co-immunoprecipitation and GST pull-down assays showed that LBH interacts with αB-crystallin, which is further confirmed by mammalian two-hybrid assays. Co-localization analysis showed that in COS-7 cells, αB-crystallin that is cytoplasmic alone, accumulates partialy in the nucleus when co-transfected with LBH. Transient transfection assays indicated that overexpression of LBH or αB-crystallin reduced the transcriptional activities of p53 and p21, respectively, Overexpression of both αB-crystallin and LBH together resulted in a stronger repression of the transcriptional activities of p21 and p53. These results showed that the interaction of LBH and αB-crystallin may inhibit synergistically the transcriptional regulation of p53 and p21. [BMB reports 2010; 43(6): 432-437]

Zinc finger-containing transcription factors are the largest single family of transcriptional regulators in mammals, which play an essential role in cell differentiation, cell proliferation, apoptosis, and neoplastic transformation. Here we have cloned a novel KRAB-related zinc finger gene, ZNF424, encoding a protein of 555aa. ZNF424 gene consisted of 4 exons and 3 introns, and mapped to chromosome 19p13.3. ZNF424 gene was ubiquitously expressed in human embryo tissues by Northern blot analysis. ZNF424 is conserved across species in evolution. Using a GFP-labeled ZNF424 protein, we demonstrate that ZNF424 localizes mostly in the nucleus. Transcriptional activity assays shows ZNF424 suppresses transcriptional activity of L8G5-luciferase. Overexpression of ZNF424 in HEK- 293 cells inhibited the transcriptional activity of NFAT and p21, which may be silenced by siRNA. The results suggest that ZNF424 protein may act as a transcriptional repressor that suppresses NFAT and p21 pathway to mediate cellular functions. [BMB reports 2010; 43(3): 212-218]

Zinc finger (ZNF) proteins play a critical role in cell growth, proliferation, apoptosis, and intracellular signal transduction. In this paper, we cloned and characterized a novel human KRAB-related zinc finger gene, ZNF425, which encodes a protein of 752 amino acids. ZNF425 is strongly expressed in the three month old human embryos and then is almost undetectable in six month old embryos and in adult tissues. An EGFP-ZNF425 fusion protein can be found in both the nucleus and the cytoplasm. ZNF425 appears to act as a transcription repressor. Over-expression of ZNF425 inhibits the transcriptional activities of SRE, AP-1, and SRF. Deletion analysis indicates that the C2H2 domain is the main region responsible for the repression. Our results suggest that the ZNF425 gene is a new transcriptional inhibitor that functions in the MAPK signaling pathway. [BMB reports 2011; 44(1): 58-63]
Effect of Texture and Microstructure on Tensile Behaviors in the Polycrystalline Pure Niobium

Pure polycrystalline niobium had different texture and microstructure due to the RCD-60, RCD-84 and CD-84 fabricationprocesses and recrystallization at 1000 °C for 2 h. The tensile behaviors and texture evolutions of the niobium were investigatedto reveal the correlation of initial texture/microstructure and tensile properties. The crucial differences of microstructureand orientations after recrystallization would influence tensile properties of the niobium. The differences of tensile behaviorsduring tension testing correspond to the different microstructure and texture evolution in the niobium. The enhanced ⟨110⟩ //RD texture during the tensile deformation indicates that orientations of sub-structures are arranging along the tensile axis inthe RCD-60 niobium. The RCD-84 and CD-84 niobium present the other phenomenon, that sub-structures near the fractureoccur a significant rotation and the ⟨110⟩ // RD texture has been slightly changed after tensile failure. The recrystallizedniobium fabricated by the RCD-84 obtains excellent tensile properties with ultimate tensile strength of 364 MPa and elongationof 55%. The strain hardening rate of the CD-84 niobium is the highest during the initial stage of tensile deformation,and becomes lower than that of the RCD-84 niobium after the 16% strain. Furthermore, the analysis of Taylor factors isapplied to reveal the difference of hardening rates during tensile deformation due to different fabrication and recrystallizationprocesses of the polycrystalline niobium.
Geft is dispensable for the development of the second heart field

Geft is a guanine nucleotide exchange factor, which can specifically activate Rho family of small GTPase by catalyzing the exchange of bound GDP for GTP. Geft is highly expressed in the excitable tissue as heart and skeletal muscle and plays important roles in many cellular processes, such as cell proliferation, migration, and cell fate decision. However, the in vivo role of Geft remains unknown. Here, we generated a Geft conditional knockout mouse by flanking exons 5-17 of Geft with loxP sites. Cre-mediated deletion of the Geft gene in heart using Mef2c-Cre transgenic mice resulted in a dramatic decrease of Geft expression. Geft knockout mice develop normally and exhibit no discernable phenotype, suggesting Geft is dispensable for the development of the second heart field in mouse. The Geft conditional knockout mouse will be a valuable genetic tool for uncovering the in vivo roles of Geft during development and in adult homeostasis. (BMB reports 2012; 45(3): 153-158)

In this study, we report the identification and characterization of a novel C2H2 zinc finger protein, ZNF552, from a human embryonic heart cDNA library. ZNF552 is composed of three exons and two introns and maps to chromosome 19q13.43. The cDNA of ZNF552 is 2.3 kb, encoding 407 amino acids with an amino-terminal KRAB domain and seven carboxyl-terminal C2H2 zinc finger motifs in the nucleus and cytoplasm. Northern blotting analysis indicated that a 2.3 kb transcript specific for ZNF552 was expressed in liver, lung, spleen, testis and kidney, especially with a higher level in the lung and testis in human adult tissues. Reporter gene assays showed that ZNF552 was a transcriptional repressor, and overexpression of ZNF552 in the COS-7 cells inhibited the transcriptional activities of AP-1 and SRE, which could be relieved through RNAi analysis. Deletion studies showed that the KRAB domain of ZNF552 may be involved in this inhibition. [BMB reports 2010; 43(3): 193-198]
