The transcriptional capacities of target genes are strongly influenced by promoters, whereas few studies have focused on the development of robust, high-performance and cross-species promoters for wide application in different bacteria. In this work, four novel promoters (Pk.rtufB, Pk.r1, Pk.r2, and Pk.r3) were predicted from Ketogulonicigenium robustum and their inconsistency in the -10 and -35 region nucleotide sequences indicated they were different promoters. Their activities were evaluated by using green fluorescent protein (gfp) as a reporter in different species of bacteria, including K. vulgare SPU B805, Pseudomonas putida KT2440, Paracoccus denitrificans PD1222, Bacillus licheniformis and Raoultella ornithinolytica, due to their importance in metabolic engineering. Our results showed that the four promoters had different activities, with Pk.r1 showing the strongest activity in almost all of the experimental bacteria. By comparison with the commonly used promoters of E. coli (tufB, lac, lacUV5), K. vulgare (Psdh, Psndh) and P. putida KT2440 (JE111411), the four promoters showed significant differences due to only 12.62% nucleotide similarities, and relatively higher ability in regulating target gene expression. Further validation experiments confirmed their ability in initiating the target minCD cassette because of the shape changes under the promoter regulation. The overexpression of sorbose dehydrogenase and cytochrome c551 by Pk.r1 and Pk.r2 resulted in a 22.75% enhancement of 2-KGA yield, indicating their potential for practical application in metabolic engineering. This study demonstrates an example of applying bioinformatics to find new biological components for gene operation and provides four novel promoters with broad suitability, which enriches the usable range of promoters to realize accurate regulation in different genetic backgrounds.

Objectives: TCR-gamma-delta+T cells (${\gamma}{\delta}$ T cells) are non-conventional T lymphocytes that can recognize and eradicate tumor cells. Our previous studies showed that infiltration and function of ${\gamma}{\delta}$ T cells were substantially attenuated in hepatocellular carcinoma (HCC). However, their prognostic value was not clarified. Methods: The association between ${\gamma}{\delta}$ T cells and the clinical outcomes was determined by immunohistochemistry (IHC) in a HCC patient cohort (n = 342). Results:Immunohistochemistry showed decreased infiltration of ${\gamma}{\delta}$ T cells in tumoral tissues compared with paired peritumoral tissues. The counts of ${\gamma}{\delta}$ T cells in peritumoral tissues were negatively correlated with tumor size (P = 0.005). Survival analysis showed that the levels of peritumoral ${\gamma}{\delta}$T cells were related to both time to recurrence (TTR) and overall survival (OS) (P = 0.010 and P = 0.036, respectively) in univariate analysis, and related to TTR in multivariate analysis (P = 0.014, H.R. [95% CI] = 0.682 [0.502-0.927]). Furthermore, the level of peritumoral ${\gamma}{\delta}$ T cells showed independent prognostic value for TTR in Barcelona Clinic Liver Cancer (BCLC) stage A patients (P = 0.038, H.R. [95% CI] = 0.727 [0.537-0.984]). However, tumoral ${\gamma}{\delta}$ T cells did not show independent prognostic value for either TTR or OS in HCC patients. Conclusions: Low counts of ${\gamma}{\delta}$ T cells in peritumoral liver tissue are related to a higher incidence of recurrence in HCC and can predict postoperative recurrence, especially in those with early-stage HCC.

Objective: The aim of this study is to compare the cross-cultural differences in product preference among users from different countries, taking Mi band 3 as a case study. Background: With the development of global market, more and more products and services are sold across the globe. Users from different cultures have different behaviors, cognitive styles, and value systems. Therefore, product should be designed to meet the needs and preferences of users from different cultural groups. Method: Compared to traditional research method such as survey questionnaire or interview that requires variety of foreigners as participants, opinion mining from online reviews saves much more cost and time. We collected review data from the following three websites: Naver of South Korea, Jingdong of China, and Amazon of the United States. Firstly, product aspects were extracted from reviews according to word frequency. This indicates how much users are paying attention to different aspects of the product. Then, fine-grained sentiment analysis was conducted to find out customer satisfaction with different product aspects. Lastly, cross-cultural differences among three countries from the result were observed and compared. Results: The product aspects that rank within top 10 in all three countries were battery life, price, step count, and sleep monitoring. What the users from three countries complain the most about was the battery life that is shorter than its advertised duration. Many users also complain about how the screen becomes invisible under the sunlight. Korean think of Korean on the display, application, and manual as the most important feature of the smart band product. Chinese are more concerned about the variety of functions. American tend to use smart band mostly as activity tracker as they care the most about battery, heart rate, and step count and prefer to have the tracker mode for bicycle and swimming on smart band product. Conclusion: Although there exist similar issues among South Korea, China, and the United States, cross-cultural differences are shown in different product aspects such as language, aesthetics, gift, activity tracker function, outdoor visibility, and screen. Application: This research can suggest implications for making strategies in product internationalization and product localization for the global marketing of smart band.

Two new phenylpropanoids, retusiusines A (1) and B (2), and a pair of new phenylpropyl enantiomers, (±)-retusiusine C (3a and 3b), together with eight known compounds, dihydroconiferyl dihydro-p-coumarate (4), methyl 3-(4-hydroxyphenyl) propionate (5), 3-(4-hydroxyphenyl)-propanoic acid (6), dihydroferulic acid (7), methyl 3-(4-methoxyphenyl) propionate (8), 3-(3,4-dimethoxyphenyl)-2-propenal (9), trans-p-coumaric acid (10) and dihydroconiferyl alcohol (11), were isolated from the tubers of Bulbophyllum retusiusculum. The absolute configurations of the new compounds were determined by calculating their electronic circular dichroism (ECD), spectra and specific optical rotations and comparing the calculated values with the experimental data. Compound 2 exhibited potent antifungal activity against Candida albicans (16 μg/mL). Compound 3 showed moderate antibacterial activity against Bacillus subtilis (64 μg/mL).

The study compares the outcome of semantic network analysis based on consumer reviews in two different units of text: paragraph and sentence. Taking Apple Watch Series 4 as a case study, review data was collected from Amazon. The overall process consists of text preprocessing, centrality measures, and cluster analysis. In centrality measures, consumer reviews in paragraph unit can yield more affective adjectives that reveal emotions or feelings of consumers towards the product. On the other hand, more nouns representing product features can be generated from reviews in sentence unit. Comparing the results of cluster analysis, the performance of review data in sentence unit is better. Topics can be created in each cluster based on reviews in sentence unit, and the average number of related words in clusters based on reviews in sentences are bigger. In conclusion, the sentence unit is suggested for semantic analysis when the information about significant product features and topics is required, and the paragraph unit is suggested when the emotions or feelings of users toward the product are required.
Improving Scanner Data Collection in P4-based SDN
Yun-Zhan Cai,Chih-Hao Lai,Yu-Ting Wang,Meng-Hsun Tsai 한국통신학회 2020 한국통신학회 APNOMS Vol.2020 No.09
Port scanning is a well-known behavior when a botnet searches target devices. To detect port scanning accurately, data with high discriminatory power are indispensable. Most related works, however, focus on data analysis methods but neglect storage limitations of switches, which makes their methods impractical. Therefore, we propose a new data collection method for collecting network information of port scanning in P4-based SDN named 0-replacement. Through simulations, we compare the 0-replacement method with two classic data collection methods. Results show that the 0-replacement method improves the true positive ratio by at least 25 percentage points but only consumes 0.36% memory space.
The Effect of Different Inflows on the Unsteady Hydrodynamic Characteristics of a Mixed Flow Pump
Yun, Long,Dezhong, Wang,Junlian, Yin,Youlin, Cai,Chao, Feng Korean Society for Fluid machinery 2017 International journal of fluid machinery and syste Vol.10 No.2
The problem of non-uniform inflow exists in many practical engineering applications, such as the elbow suction pipe of waterjet pump and, the channel head of steam generator which is directly connect with reactor coolant pump. Generally, pumps are identical designs and are selected based on performance under uniform inflow with the straight pipe, but actually non-uniform suction flow is induced by upstream equipment. In this paper, CFD approach was employed to analyze unsteady hydrodynamic characteristics of reactor coolant pumps with different inflows. The Reynolds-averaged Naiver-Stokes equations with the $k-{\varepsilon}$ turbulence model were solved by the computational fluid dynamics software CFX to conduct the steady and unsteady numerical simulation. The numerical results of the straight pipe and channel head were validated with experimental data for the heads at different flow coefficients. In the nominal flow rate, the head of the pump with the channel head decreases by 1.19% when compared to the straight pipe. The complicated structure of channel head induces the inlet flow non-uniform. The non-uniformity of the inflow induces the difference of vorticity distribution at the outlet of the pump. The variation law of blade to blade velocity at different flow rate and the difference of blade to blade velocity with different inflow are researched. The effects of non-uniform inflow on radial forces are absolutely different from the uniform inflow. For the radial forces at the frequency $f_R$, the corresponding amplitude of channel head are higher than the straight pipe at $1.0{\Phi}_d$ and $1.2{\Phi}_d$ flow rates, and the corresponding amplitude of channel head are lower than the straight pipe at $0.8{\Phi}_d$ flow rates.
