      • Genome-wide association studies approach and post-GWAS study in rice

        AGenome-wide association studies (GWAS) have proven a useful technique for identifying genetic loci responsible for natural variation in rice. With the fast developed next-generation sequencing technology, it is possible for people to carry out GWAS by phenotyping different traits. However, how to make full use of huge data, abandon unnecessary data, and solve the problem of data application effectively seems still an obstacle for many researchers. Taking the case of whole-genome resequencing of Korean authentic rice core set, here we present a general technological path of GWAS including: 1) a schematic view of sequencing-based GWAS in rice; 2) a user-friendly and interactive web application for GWAS in rice by the aid of experience from Arabidopsis; 3) Haplotype and association analysis of candidate genes in a certain mechanism pathway, giving 10 starch synthesis genes as example; and 4) functional validation by Trans- and Mata-Omics analysis.

        Prefrontal cortex miR-29b-3p plays a key role in the antidepressant-like effect of ketamine in rats

        Ketamine has a rapid, obvious, and persistent antidepressant effect, but its underlying molecular mechanisms remain unknown. Recently, microRNAs (miRNAs) have emerged as important modulators of ketamine’s antidepressant effect. We investigated the alteration in miR-29b-3p in the brain of rats subjected to ketamine administration and chronic unpredictable mild stress (CUMS), and a sucrose preference test and forced swimming test were used to evaluate the rats’ depressive-like state. We used recombination adeno-associated virus (rAAV) or lentivirus-expressing miR-29b-3p to observe the change in metabotropic glutamate receptor 4 (GRM4). Cell culture and electrophysiological recordings were used to evaluate the function of miR-29b-3p. Ketamine dramatically increased miR-29b-3p expression in the prefrontal cortex of the normal rats. The dual luciferase reporter test confirmed that GRM4 was the target of miR-29b- 3p. The miR-29b-3p levels were downregulated, while the GRM4 levels were upregulated in the prefrontal cortex of the depressive-like rats. The ketamine treatment increased miR-29b-3p expression and decreased GRM4 expression in the prefrontal cortex of the depressive-like rats and primary neurons. By overexpressing and silencing miR-29b-3p, we further validated that miR-29b-3p could negatively regulate GRM4. The silencing of miR-29b-3p suppressed the Ca2+ influx in the prefrontal cortex neurons. The miR-29b-3p overexpression contributed to cell survival, cytodendrite growth, increases in extracellular glutamate concentration, and cell apoptosis inhibition. The overexpression of miR- 29b-3p by rAAV resulted in a noticeable relief of the depressive behaviors of the CUMS rats and a lower expression of GRM4. The miR-29b-3p/GRM4 pathway acts as a critical mediator of ketamine’s antidepressant effect in depressive-like rats and could be considered a potential therapeutic target for treating major depression disorder.

        Synergistic Effect of Bone Marrow-Derived Mesenchymal Stem Cells and Platelet-Rich Plasma in Streptozotocin-Induced Diabetic Rats

        Background: Diabetic wounds are a major clinical challenge, because minor skin wounds can lead to chronic, unhealed ulcers and ultimately result in infection, gangrene, or even amputation. Studies on bone marrow derived mesenchymal stem cells (BMSCs) and a series of growth factors have revealed their many benefits for wound healing and regeneration. Platelet-rich plasma (PRP) may improve the environment for BMSC development and differentiation. However, whether combined use of BMSCs and PRP may be more effective for accelerating diabetic ulcer healing remains unclear. Objective: We investigated the efficacy of BMSCs and PRP for the repair of refractory wound healing in a diabetic rat model. Methods: Forty-eight rats with diabetes mellitus induced by streptozotocin were divided into four groups: treatment with BMSCs plus PRP, BMSCs alone, PRP alone, phosphate buffered saline. The rate of wound closure was quantified. A histopathological study was conducted regarding wound depth and the skin edge at 7, 14, and 28 days after surgery. Results: Wound healing rates were significantly higher in the BMSC plus PRP group than in the other groups. The immunohistochemistry results showed that the expression of platelet/endothelial cell adhesion molecule 1, proliferating cell nuclear antigen, and transforming growth factor-β1 increased significantly in the BMSC plus PRP group compared to the other treatment groups. On day 7, CD68 expression increased significantly in the wounds of the BMSC plus PRP group, but decreased markedly at day 14 compared to the controls. Conclusion: The combination of BMSCs and PRP aids diabetic wound repair and regeneration. (Ann Dermatol 26(1) 1∼10, 2014)

        Dynamic Cloud Resource Reservation Model Based on Trust

        Aiming at the problem of service reliability in resource reservation in cloud computing environments, a model of dynamic cloud resource reservation based on trust is proposed. A domain-specific cloud management architecture is designed in which resources are divided into different management domains according to the types of service for easier management. A dynamic resource reservation mechanism (DRRM) is used to test users' reservation requests and reserve resources for users. According to user preference, several resources are chosen to be candidate resources by fuzzy cluster analysis. The fuzzy evaluation method and a two-way trust evaluation mechanism are adopted to improve the availability and credibility of the model. An analysis and simulation experiments show that this model can increase the flexibility of resource reservation and improve user satisfaction.

        Efficient RNA interference for three neuronally-expressed genes in Nilaparvata lugens (Stål) (Hemiptera: Delphacidae)

        In the brown planthopper Nilaparvata lugens, introduction of double-stranded RNA (dsRNA) successfully knocks down genes transcribed in several types of tissues. However, whether RNA interference (RNAi) can effectively reduce the transcripts of neuronally-expressed genes remains undetermined. In the present paper, we selected three neuronally-expressed genes respectively encoding prothoracicotropic hormone (NlPTTH), pigment-dispersing factor (NlPDF) and short neuropeptide F (NlsNPF). The transcripts of the three genes were detectable throughout all tested developmental stages, and were mainly restricted to the nervous system of N. lugens. We introduced dsRNA originated from each of the three neuronally-expressed genes at a dose of 300 ng into the nymphal body cavity by microinjection. The target mRNA was successfully knocked down. Moreover, silencing of NlPTTH downregulated the transcripts of two ecdysteroidogenesis genes (NlPHM and NlDIB), and surpressed the expression of an ecdysone-response genes (NlFTZ-F1). Furthermore, RNAi of NlPTTH caused typical 20- hydroxyecdysone deficient phenotypes: nymphal development was delayed and nymphal lethality occurred. In addition, injection of a series of dsNlsDPF solutions at the doses of 300, 60, 12, 2.4 and 0.5 ng into the fourthinstar nymphs decreased its target transcript in a dose-dependent manner: the mRNA levels reduced by 94%, 86%, 57%, 33% and 29%, respectively. Therefore, delivery of dsRNA by microinjection can effectively repress the transcripts of the neuronally-expressed genes in N. lugens.

        Chia Seed Supplementation Reduces Senescence Markers in Epididymal Adipose Tissue of High-Fat Diet-Fed SAMP8 Mice

        Adipose tissue is a key organ with substantial senescent cell accumulation under both obesity and aging conditions. Chia seed is an ancient seed and is the richest plant source of α-linolenic acid. We aimed to determine how cellular senescence markers will be altered in adipose tissue of senescence-accelerated mouse-prone 8 (SAMP8) mice fed with high-fat diets (HFDs); and how chia seed can affect the above markers. SAMP8 mice and their control senescence-accelerated mouse-resistant 1 (SAMR1) were divided into four groups, that is, SAMR1 low-fat diet group (R1LF), SAMP8LF group (P8LF), SAMP8 high-fat group (P8HF), and SAMP8HF group supplemented with 10% chia seed (P8HC). At the end of the intervention, body composition was measured through T1-weighted magnetic resonance imaging, and epididymal (EPI) and subcutaneous (SC) adipose tissues were dissected for further analysis. Compared with the R1LF group, the P8HF and P8HC groups had significantly increased body fat mass. In EPI fat, p16, CD68 and PAI-1 mRNA expression from P8HF group were significantly increased; chia seed partially reduced p16 and CD68 mRNA expression. The P8LF group has increased p16 and CD68, and the P8HF group has increased p16, p21, and CD68; and P8HC group has increased p16 mRNA expression. The protein expression of p-AMPK in EPI and SC fat from the P8HF group was reduced. In conclusion, reductions in AMPK activity might be partially responsible for elevation in HFD-induced senescence markers in both EPI and SC fat, and chia seed supplementation is able to reduce senescence-associated markers at least in EPI adipose tissue.

        Application of KF/MgO as a heterogeneous catalyst in the production of biodiesel from rapeseed oil

        Biodiesel was synthesized from rapeseed oil by transesterification over Magnesium oxide loaded with KF. The catalytic activity strongly depends on the loading amount of KF and calcined temperature. We found that the reaction reached a 79.37% yield when the loading amount was at 35 wt% and calcined at 500 oC. The simply dried 30% KF/MgO at 80 oC was found to give equally good results from the catalyst calcined at 500 oC, avoiding the usual activation at high temperature. The catalysts were characterized by the Hammett indicator method, BET, TG/DTG, XRD, NMR, EDS, and FT-IR. According to the instrumental analysis, the activity in the transesterification probably belonged to coordinately unsaturated F− and liberation of hydroxide during preparation. The effects of methanol/oil ratio and catalyst amounts on the conversion were also studied in this paper.

      • Enhanced Antitumor Effect of Curcumin Liposomes with Local Hyperthermia in the LL/2 Model

        Curcumin previously was proven to inhibit angiogenesis and display potent antitumor activity in vivo and in vitro. In the present study, we investigated whether a combination curcumin with hyperthermia would have a synergistic antitumor effect in the LL/2 model. The results indicated that combination therapy significantly inhibited cell proliferation of MS-1 and LL/2 in vitro. LL/2 experiment model also demonstrated that the combination therapy inhibited tumor growth and prolonged the life span in vivo. Furthermore, combination therapy reduced angiogenesis and increased tumor apoptosis. Our findings suggest that the combination therapy exerted synergistic antitumor effects, providing a new perspective fpr clinical tumor therapy.
