      • Molecular Cloning and Bioinformatic Analysis of SPATA4 Gene

        Full-length cDNA sequences of four novel SPATA4 genes in chimpanzee, cow, chicken and ascidian were identified by bioinformatic analysis using mouse or human SPATA4 cDNA fragment as electronic probe. All these genes have 6 exons and have similar protein molecular weight and do not localize in sex chromosome. The mouse SPATA4 sequence is identified as significantly changed in cryptorchidism, which shares no significant homology with any known protein in swissprot databases except for the homologous genes in various vertebrates. Our searching results showed that all SPATA4 proteins have a putative conserved domain DUF1042. The percentages of putative SPATA4 protein sequence identity ranging from 30% to 99%. The high similarity was also found in 1 kb promoter regions of human, mouse and rat SPATA4 gene. The similarities of the sequences upstream of SPATA4 promoter also have a high proportion. The results of searching SymAtlas (http://symatlas.gnf.org/SymAtlas/) showed that human SPATA4 has a high expression in testis, especially in testis interstitial, leydig cell, seminiferous tubule and germ cell. Mouse SPATA4 was observed exclusively in adult mouse testis and almost no signal was detected in other tissues. The pI values of the protein are negative, ranging from 9.44 to 10.15. The subcellular location of the protein is usually in the nucleus. And the signal peptide possibilities for SPATA4 are always zero. Using the SNPs data in NCBI, we found 33 SNPs in human SPATA4 gene genomic DNA region, with the distribution of 29 SNPs in the introns. CpG island searching gives the data about CpG island, which shows that the regions of the CpG island have a high similarity with each other, though the length of the CpG island is different from each other.This research is a fundamental work in the fields of the bioinformational analysis, and also put forward a new way for the bioinformatic analysis of other genes.

        A Duplex PCR Assay for Rapid Detection of Phytophthora nicotianae and Thielaviopsis basicola

        A duplex PCR method was developed for simultaneous detection and identification of tobacco root rot pathogens Phytophthora nicotianae and Thielaviopsis basicola. The specific primers for P. nicotianae were developed based on its internal transcribed spacer (ITS) regions of ribosomal gene, ras gene and hgd gene, while the specific primers for T. basicola were designed based on its ITS regions and ${\beta}$-tubulin gene. The specificity of the primers was determined using isolates of P. nicotianae, T. basicola and control samples. The results showed that the target pathogens could be detected from diseased tobacco plants by a combination of the specific primers. The sensitivity limitation was $100fg/{\mu}l$ of pure genomic DNA of the pathogens. This new assay can be applied to screen out target pathogens rapidly and reliably in one PCR and will be an important tool for the identification and precise early prediction of these two destructive diseases of tobacco.

        Flexible Cu2O/Cu film self-powered photodetector for high visible light selection and fast response

        Spectral selection is one of the key performance parameters for visible light photodetectors (VLPD). However, as the absorbing layer of the VLPD, the semiconductor has strong ultraviolet light absorption, leading to poor visible light selectivity of the VLPD. This work fabricated a flexible photovoltaic VLPD based on Cu2O/Cu Schottky junction by annealing Cu foil. Under zero bias, the Cu2O/Cu VLPD showed the responsivity of 14 mA/W, the response time of 1 ms, and the detectivity of 8.8 × 1011 Jones to 620 nm light. Moreover, the detection band of the device is in the range of 500–640 nm, indicating excellent visible light selectivity. Optical and electrical characterization shows that the high visible selectivity of the device is mainly attributed to the fact that the penetration depth of visible light in Cu2O film matches the position of the Cu2O/Cu Schottky junction. This, in turn, induces the effective separation of the photogenerated carriers excited by the visible light. In addition, the bend detection performance of the device is demonstrated. This work provides a reference for improving the spectral selectivity of visible photodetectors.

        Preparation and properties of bio-based intumescent flame retardant containing chitosan functionalized ammonium polyphosphate for polyurethane

        In this study, chitosan (CS) from fish waste was used to prepare bio-based environmentally friendly flameretardants. After the hydroxyl group (OH) of CS and the ammonium group (NH4 + ) of ammoniumpolyphosphate (APP) underwent reactions, they were filtered and dried to obtain CS-APP. The aminogroup of CS-APP then reacted with the epoxy group of 4,40-methylenebis(N,N-diglycidylaniline) (NDY)to form CS-APP-NDY. Isophorone diisocyanate, polyol, and 3-aminopropyltriethoxysilane reacted to formsilicon polyurethane. CS-APP-NDY was then mixed with Si-PU to prepare polymer composites. To determinethe structure, thermal properties, flame retardancy, and mechanical properties of the composites,Fourier-transform infrared spectroscopy, thermogravimetric analysis, limiting oxygen index (LOI), conecalorimetry, UL-94, thermal analysis-FTIR (TA-FTIR) spectroscopy, universal machine testing, scanningelectron microscopy, X-ray photoelectron spectroscopy, and Raman spectroscopy were performed. TheTGA results revealed that, after the addition of CS-APP-NDY, char yield increased from 0.5 wt% to 25.8wt%, and the thermal stability of pristine PU also improved. In addition, the LOI and UL-94 results indicatedthat, after the addition of CS-APP-NDY, the LOI increased from 18.2% to 26.3%, and the UL-94 levelimproved from ‘‘Fail” to V-1. Overall, these results indicated that the addition of CS-APP-NDY to pristinePU increased its flame-retarding performance.

        Game Theoretic Approach for Joint Resource Allocation in Spectrum Sharing Femtocell Networks

        In this paper, we study the joint price and power allocationin spectrum sharing macro-femtocell networks. The proposedgame theoretic framework is based on bi-level Stackelberg gamewhere macro base station (MBS) works as a leader and underlaidfemto base stations (FBSs) work as followers. MBS has fixed datarate and imposes interference price on FBSs for maintaining itsdata rate and earns revenue while FBSs jointly adjust their powerfor maximizing their data rates and utility functions. Since theinterference from FBSs to macro user equipment is kept under agiven threshold and FBSs compete against each other for powerallocation, there is a need to determine a power allocation strategywhich converges to Stackelberg equilibrium. We consider two casesfor MBS power allocation, i.e., fixed and dynamic power. MBS canadjust its power in case of dynamic power allocation according toits minimum data rate requirement and number of FBSs willing toshare the spectrum. For both cases we consider uniform and nonuniformpricingwhereMBS charges same price to all FBSs for uniformpricing and different price to each FBS for non-uniform pricingaccording to its induced interference. We obtain unique closedform solution for each case if the co-interference at FBSs is assumedfixed. And an iterative algorithm which converges rapidly isalso proposed to take into account the effect of co-tier interferenceon interference price and power allocation strategy. The results areexplained with numerical simulation examples which validate theeffectiveness of our proposed solutions.

        Cloning and Expression Analysis of Gonadogenesis-associated Gene SPATA4 from Rainbow Trout (Oncorhynchus mykiss)

        Gonadogenesis is a complicated process which involves multi-gene interactions. A rainbow trout (Oncorhynchus mykiss) gene spermatogenesis associated 4 (SPATA4) was cloned and characterized from adult rainbow trout testis. The cDNA sequence of rainbow trout SPATA4 contains an open reading frame of 1, 081 nucleatides encoding a putative protein of 259 amino acids. The putative protein from rainbow trout shares a 76.8% homology with zebrafish SPATA4. No trans-membrane regions or signal peptide were detected using bioinformatics methods. Subcellular localization analysis revealed that rainbow trout SPATA4 was a nuclear protein with highest possibility (39.1%). Multi-tissue reverse transcriptase PCR (RT-PCR) was performed to examine the distribution of rainbow trout SPATA4 in eleven organs of adult rainbow trout. The result demonstrated that this gene express specifically in testis and slight amount of expression was detected in ovary. Further analysis of SPATA4 characterization and function in rainbow trout may provide insight into the understanding of gonadogenesis process.

        Influence of synergistic effect of phosphate on corrosion resistance of self-assembled superhydrophobic composite film on 6061 aluminum alloy surfaces

        Aluminum (Al) alloys are used in a wide range of applications due to their advantages. However, theexisting corrosion protection methods have insufficient protection ability and cannot achieve longtermprotection. In this paper, reactive protective film on the surface of aluminum alloys using 1-hydroxyethyl-1,1-diphosphonic acid (HEDP) molecules. Then, Lauric acid/Graphene (LA/rGO) film is usedto prepared on the surface of HEDP film. SEM, XRD, EDS, Raman, XPS and WCA were conducted to analyzephysical and chemical features of coatings. The total resistance of the film reaches 1.0 1020 Xcm2, in asalt spray test using 5wt% NaCl as the corrosion medium, corrosion spots appeared on day 28 with a corrosioninhibition efficiency of 99.99%. Theoretical studies with quantum chemical calculations haveshown that HEDP/LA/rGO composite films can be adsorbed on Al alloy surfaces through P-O-Al and PO-C chemical bonding. The influence of phosphate synergistic effect on the corrosion resistance of theself-assembled superhydrophobic composite film on the surface of 6061 aluminum alloy is discussed. This study presents a scientific and innovative design concept to develop coatings with excellent corrosionprotection for aluminum alloy, and then extend their application to certain areas.
