        질산화 작용이 있는 Aeromonas hydrophila의 동정 및 특성

        폐수처리 중 생물학적 처리에 활용할 수 있는 질소분해 능력을 가진 미생물을 분리하여 동정하고자 경기도내 하천 6지점에서 채취한 시료로부터 50개 균주를 선별하였다. 형태학적, 생화학적 및 배양학적 실험결과 Bergey's mannual of systematic bacteriology의 색인을 통하여 Aeromonas hydrophila로 동정하였다. Aeromonas hydrophila(AH-1). (AH-3), (AH-4), (AH-6) 균이 질산화 능력이 우수하였다. 4개 균주 모두 amoxillin, ampicillin, cephalothin과 ticarcillin에 내성을 나타내었다. 본 실험에서 분리한 .Aeromonas hydrophila의 질산화의 최적조건은 균 농도 1.0×l0exp(6) cells/ml, 배양온도 37℃로 나타났다. For the purpose of the isolation of microorganisms which have the capability of nitrification, we isolated the microorganisms in 6 samples collected from the stream of Kyonggi area. 60 strains were isolated. The selected strain were identified as a Aeromonas hydrophila based on the data obtained from the morphological, biochemical and cultural characteristics defined experiments. Among them Aeromonas hydrophila (AH-1), (AH-3), (AH-4), (AH-6) showed the highest nitrification capability. All isolates were resistant to amoxillin, ampicillin, cephalothin and ticarcillin. Optimum culture conditions of isolates were 37℃ and 1×10exp(8) cells/ml for 4 hours in the nitrate medium.

        Preparation of new natural silk non-woven fabrics by using adhesion characteristics of sericin and their characterization

        <P><B>Abstract</B></P> <P>Electro-spun regenerated silk webs have been extensively studied for biomedical applications because of the simplicity of their fabrication methods However, the productivity of the electro-spinning process is low for web fabrication and the mechanical properties of the electro-spun silk web are not satisfactory, which restricts its commercialization. In this study, a new silk non-woven fabric was successfully fabricated by wetting and hot press treatments using the excellent binding characteristic of sericin. The effects of the press temperature and residual sericin content on the preparation, structure, and properties of the silk non-woven fabric were examined. A press temperature of 200°C was optimum for obtaining non-woven fabrics with best mechanical properties, without yellowing. The silk non-woven fabric could not be fabricated without sericin, and a minimum of 8% sericin was required to fabricate it. As the sericin content was increased, the strength and Young’s modulus of the silk non-woven fabric increased, while the tensile elongation remained constant. Regardless of the press temperature and sericin content, all the silk non-woven fabrics showed good cell viability, comparable to that of the tissue culture plate (TCP) used as a control until 4days, which however decreased compared to that of TCP after 7days.</P>

      • Effect of Empowerment on Perceived Person-Environment Fit and Work Engagement: Focusing on Online Travel Platform Employees

        Purpose - COVID-19 has dramatically changed the business landscape, and the travel industry is no exception. The industry is undergoing a paradigm shift to the platform business, and traditional travel agencies are moving their businesses to online platforms. However, in this irreversible global paradigm shift, it is not easy for travel companies to hire and retain employees, especially companies in the IT sector. Studies typically study consumers, such as selection attributes and satisfaction with travel platforms. However, to the best of our knowledge, there have been almost no studies on employees. This study explored how the empowerment of online travel platform employees influences their perceived person-environment (P-E) fit and work engagement. Design/Methodology/Approach - We analyzed data from 352 employees of 26 domestic online travel platforms who were surveyed in January and February 2021. After coding the collected data, we analyzed it using frequency, exploratory factor, reliability, correlation, and multiple regression analyses using IBM SPSS Statistics, ver. 20.0. Findings - First, enhancing the meaningfulness of work and organizational autonomy influences personjob (P-J) fit and person-organization (P-O) fit. Second, enhancing the meaningfulness of work significantly influences vigor, dedication, and absorption in terms of work engagement, while organizational autonomy significantly influences work engagement vigor. Third, P-J fit and P-O fit influence the vigor of work engagement, and P-O fit influences dedication and absorption. Research Implications - This study demonstrated that the empowerment of travel platform employees can improve perceived P-E fit and work engagement. Thus, if organizations guarantee some degree of empowerment, employees will be energized and engaged more in their workplace. Finally, this helps expand the scope of online travel platform research from the perspective of personnel organization.

      • The Improvement or Regression of Fibrosis after Treatment with Sofosbuvir Plus Ribavirin for HCV Genotype 2 Infection

        Aims: Sofosbuvir (SOF) and ribavirin (RBV) for 12 or 16 weeks is recommended for treatment of patients with HCV genotype (GT) 2 infected patients in KASL guideline based on clinical trials. Whether sustained virological response of HCV infection has any beneficial effect on improvement of liver fibrosis associated with chronic HCV infection remains unclear. We investigated to assess the effects on liver fibrosis of at least 2 years through follow-up liver biopsy and fibroelastography after treatment with sofosbuvir plus ribavirin for HCV G2 infection. Methods: Three tertiary center, prospective observational cohort study evaluates clinical practice data (Korea university Anam, Guro, and Ansan) between January 2015 and December 2019. Clinical data were centrally collected from medical records. The efficacy outcome was sustained virological response 12 weeks after therapy (SVR12). The degree of liver fibrosis was evaluated by APRI score, FIB-4, fibroelestography and paired liver tissue biopsy. Results: 146 patients were visited and 131 patients were treated SOF plus RBV during 12 weeks (n=122) or 16 weeks (n=9). Overall, EVR, ETR, and SVR12 by ITT analysis were 90.0%, 96.2% and 87%. In addition, EVR, ETR, and SVR12 by PP analysis 97.5%, 99.2% and 96.3%, respectively. In subgroup analysis, SVR12 in patients with treatment-naïve and treatment-experience were 97.2% or 94.7%, respectively. SVR12 in patients with and without cirrhosis were 94.4% and 97.4%, respectively. Finally 106 patients showed sutained virological response and follow up 2 year after treatment. The mean APRI score was 0.57 before treatment, but improved to 0.27 at 2 year after treatment (P=0.004). The mean FIB-4 score was 2.15 before treatment, but improved to 1.53 at 2 year after treatment (P=0.004). The mean fibroelastography value was 10.81 kPa before treatment, but improved to 4.75 kPa at 2 year after treatment (P=0.008). Follow-up liver tissue biopsies were done in 8 patients and fibrosis was evaluated by Metavir score (F0-4). Almost all patient except one showed improvement of cirrhosis (≥1 unit decrease in Metavir score, P=0.031). Conclusions: SOF and RBV was safe and effective for treatment of patients with HCV GT2 infection. In patients with sustained virological response 2 year after SOF and RBV, the improvement or regression of liver fibrosis could be observed in biochemical and histological aspect.

        Altered ganglioside patterns accompany the Anti-inflammatory activity of luteolin in Lipopolysaccharide-stimulated Raw 264.7

        Flavonoids have a range of biological activities, including anti-allergic, anti-inflammatory, anti-microbial, and anti-cancer activities, as demonstrated by in vitro studies. In this study, we investigated whether luteolin can be applied to suppression of lipopolysaccharide (LPS)-stimulated inflammatory responses in murine macrophages. Luteolin was found to reduce nitric oxide (NO) production in LPS-stimulated Raw 264.7 cells. In addition, expression of inducible nitric oxide synthetase (iNOS), cyclooxygenase-2 (COX-2), and pro-inflammatory cytokine tumor necrotic factor-α (TNF-α) at the mRNA and protein levels were decreased. These inhibitory effects were found to be caused by the blockade of nuclear factor kappa-light- chain-enhancer of activated B cells (NF-κB) activation and phosphorylation of mitogen-activated protein (MAP) kinase family, extracellular signal-regulated kinases 1/2 (ERK1/2), c-Jun N-terminal kinase (JNK), and p38 MAP kinase. In addition, pre-treatment with luteolin resulted in reduced ganglioside expression levels and inhibited expression of GT1b in Raw 264.7 cells. On the basis of these observations, we suggest that luteolin has potential as an anti-inflammatory drug candidate, and ganglioside GT1b may play a role in the inflammatory process.

        Natural History of Chronic Urticaria in Korea

        Background: Data on the natural history and prognostic variables of chronic urticaria (CU) are rare and information about spontaneous remission of CU is limited. Objective: This study evaluated the natural history of CU and identified predictors for remission. Methods: Total 329 Korean patients with CU, who had follow-ups more than 6 months after diagnosis during a 7-year period in the department of dermatology in three university hospitals were enrolled. Clinical data and laboratory findings obtained by medical records and telephone interviews were analyzed, retrospectively. Results: The proportion recovered in 1, 3, and 5 years after the onset of CU was 10.8%, 18.8%, and 32.9%, respectively. The mean duration of CU was 6.3 years. There were no significant differences in median recovery time depending on sex, age group, severity of CU, and type of CU. The presence of angioedema was significantly related to CU severity. There were no differences in prognosis with respect to the presence of dermographism or angioedema. Patients with atopic dermatitis (AD) had a significantly worse prognosis than patients without a history of AD; but not in patients with the history of allergic rhinitis or asthma. Patients with abnormal laboratory findings did not differ significantly in prognosis. Conclusion: CU remission rate significantly differ according to the presence of AD. This study provides information about the natural course of CU of Korean patients. (Ann Dermatol 32(1) 38∼ 46, 2020)

        가족치아를 이용한 골이식술

        The use of autogenous tooth bone graft material has been commercialized since 2008. Autogenous tooth bone grafts always require that the tooth of the patient be extracted, and thus, the use of graft material are limited in many cases. For solution of limitation in quantity and concurrent use of autogenous tooth bone graft material, the grafting of familial teeth has been suggested. It has the following advantages: the teeth of family members are used as bone graft materials, the genetic composition is identical, and potential genetic and infectious risks can be minimized. Because the teeth of family members are used, a good tissue affinity is obtained, and thus, superior bone generation rates compared to those observed for allogenic or xenogenic bones can be anticipated. We used familial tooth bone-graft materials for alveolar ridge augmentation, socket preservation, and maxillary sinus graft in some cases. In most cases, the impacted third molars of their children were prepared as bone graft material and were used for surgery. In one case, the impacted third molar from the patient`s brother was used as bone graft material. We obtained satisfactory result and these cases are reported herein.

        Agaricus bisporus Attenuates Dextran Sulfate Sodium-Induced Colitis

        Agaricus bisporus (white button mushroom, WBM) is widely consumed in most countries and is reported to have anti-inflammatory and antioxidant activities. However, little is known regarding its effects in dextran sulfate sodium (DSS)-induced colitis, which are related to dysfunction of intestinal immunity. The aim of the present study was to investigate the effects of WBMs in an animal model of DSS-induced colitis. Male, 4-week-old ICR mice (n = 10 per group) were fed a normal diet with or without 10% WBM for 4 weeks, and colitis was induced by 3% DSS in drinking water for 7 days. WBMs prevented DSS-induced shortening of colon length (P = .033) and diminished diarrhea (P = .049) and gross bleeding (P = .001), resulting in a decreased disease activity index. Results of histological analysis showed that WBMs suppressed mucosal damage. In addition, WBMs attenuated the DSS-induced increase in myeloperoxidase activity (P = .012) and upregulation of proinflammatory cytokine tumor necrosis factor-a (P = .020) in the colon segment. Taken together, these findings suggest a possible role for the WBM as an immunomodulator that can prevent and/or treat ulcerative colitis.

        계피로부터 Malassezia 종에 대한 항균물질 분리 및 동정

        To overcome side effects of dandruff medications, many researches on natural anti-dandruff compounds are actively being conducted. Here, we tried to isolate and identify antifungal compounds against four Malassezia species (M. restricta, M. globosa, M. furfur, M. sympodialis) from Cinnamomoum cassia Blume. Cinnamomum cassia Blume was extracted with 80% (v/v) ethanol for 7 days. The extracts were purified by solvent fractionation, silica gel column chromatography and HPLC. The purified compound was identified for the active peak with 1H-NMR and <SUP>13</SUP>C-NMR. Among 5 solvent fractions, only hexane fraction (CHF) showed antifungal activity. The data showed that CHF has higher activity than that of Zinc pyrithione (ZPT) in M. furfur, and lower than that of ZPT in M. restricta, M. globosa and M. sympodialis. Thus, CHF has been further purified by silica gel column chromatography and Prep-HPLC. The analysis of 1H-NMR and <SUP>13</SUP>C-NMR revealed that structure of isolated compound is coumarin. Coumarin showed the antifungal activity of 11.5 mm, 10.1 mm, 14.2 mm and 19.8 mm for M. furfur, M. restricta, M. globosa and M. sympodialis, respectively. MIC50 values of coumarin were 79.09 μg/mL, 133.91 μg/mL, 63.41 μg/mL and 19.72 μg/mL for M. furfur, M. restricta, M. globosa and M. sympodialis, respectively. The anti-inflammatory activity of coumarin at concentrations of 0.013∼0.855 nM showed that coumarin inhibited nitric oxide production by 18∼22% in RAW 264.7 cell. These data suggest that Cinnamomum cassia Blume extracts and its isolated coumarin are good candidates for dandruff medications.
