It is known that alcoholics have significantly lower mitochondrial aldehyde dehydrogenase (ALDH)s' activity than do normal subjects or nonalcoholics with liver disease. However, there are only few reports that explain the reasons behind this reduction of ALDHs' activities. In this study, ALDH activity is inhibited by acetaldehyde, a substrate for ALDH. However, the addition of glutathione (GSH) protected ALDH activities against the inhibitory effects of acetaldehyde in vitro. Furthermore, when GSH depletion is induced using diethyl maleate (DEM) in rats by 24% in cytosol and 43% in mitochondria, ALDH activities were also depressed by 31% and 63%, respectively compared to non-treated rats without significant reductions in other hepatic enzymes. These results suggest that ALDHs' activities are closely related to the concentration of acetaldehyde and/or cellular GSH contents. Therefore in alcoholic liver disease, increased productions of acetaldehyde and decreased contents of mitochondrial GSH may involved in the depression of ALDHs' activities.

<P>Maintaining an appropriate balance between subsets of CD4<SUP>+</SUP> helper T cells and T regulatory cells (Tregs) is a critical process in immune homeostasis and a protective mechanism against autoimmunity and inflammation. To identify the role of vitamin A-related compounds, we investigated the regulation of interleukin (IL)-17-producing helper T cells (Th17 cells) and Tregs treated with all-trans-retinal (retinal). CD4<SUP>+</SUP>T cells or total cells from the spleens of C57BL/6 mice were stimulated under Treg-polarizing (anti-CD3/CD28 and TGF-β) or Th17-polarizing (anti-CD3/CD28, TGF-β, and IL-6) conditions in the presence or absence of retinal. To analyze their suppressive abilities, retinal-induced Tregs or TGF-β-induced Tregs were co-cultured with responder T cells. Collagen-induced arthritis (CIA) was established in interferon (IFN)-γ knockout mice. On day 13, retinal-induced Tregs were adoptively transferred to mice with established CIA after second immunizations. Compared with TGF-β-induced Treg cells, retinal-induced Tregs showed increased Foxp3 expression and mediated stronger suppressive activity. Under Th17-polarizing conditions, retinal inhibited the production of IL-17 and increased the expression of Foxp3.Retinal-induced Tregs showed therapeutic effects in IFN-γ knockout CIA mice. Thus, we demonstrated that retinal reciprocally regulates Foxp3<SUP>+</SUP> Tregs and Th17 cells. These findings suggest that retinal, a vitamin A metabolite, can regulate the balance between pro- and anti-inflammatory immunity. A better understanding of the manipulation of Foxp3 and Tregs may enable the application of this tremendous therapeutic potential in various autoimmune diseases.</P>

Phellinus linteus has traditionally been used as a medicinal mushroom in Oriental countries. Bioactive phytochemicals from the fruiting body of P. linteus were investigated using the on-line HPLC-DPPH system. The system was used to separate 10 antioxidants: protocatechuic acid (1) protocatechualdehyde, (2) caffeic acid (3), ellagic acid (4), hispidin (5), davalliallactone (6), interfungins A (7), hypholomine B (8), inoscavin A (9), and methyldavallialactone (10). Of these, compounds 6 and 7 showed the strongest inhibitory effect against the DPPH radical and superoxide anion radical-scavenging capacity. On comparison with quercetin ($IC_{50}$, 44.0 ${\mu}M$), the $IC_{50}$ value of compounds 6 and 7 for DPPH radical-scavenging capacity were 19.6 ${\mu}M$ and 18.5 ${\mu}M$. Moreover, the antioxidative capacity of compound 6 at 40mM was 2.95 nmol Trolox equivalents per mg dry wt. These results indicated that hispidin dimers such as hispidin with hispidin or hispolon had a stronger effect on radical-scavenging capacity than hispidin itself.

The purpose of this study is to investigate the moderating effect of mother’s discipline style on the relationship between young children’s difficult temperament and young children’s peer interaction. To verify this, 307 mothers with 36-72 months of young children were asked to measure their difficult temperament, peer interaction, and mother’s discipline. The results of this study are summarized as follows. First, young children’s difficult temperament showed a significant negative correlation with young children’s peer interaction, the mother’s positive discipline showed a significant positive correlation with young children’s peer interaction, and the mother’s negative discipline showed a significant negative correlation with young children’s peer interaction. Second, young children’s difficult temperament was found to affect young children’s peer interaction. Third, mother’s positive and negative discipline had an effect on young children’s peer interaction. Fourth, the moderating effect of mother’s positive discipline style was proved in the relationship between young children’s difficult temperament and young children’s peer interaction. Fifth, the moderating effect of mother’s negative discipline style was proved in the relationship between young children’s difficult temperament and young children’s peer interaction. Based on these, the implications and limitations of this study and the direction of follow-up research were discussed.
