        Osmotic Pump모델 당뇨병 실험동물에서 침자극이 혈당조절 및 베타세포 보호작용에 미치는 영향

        Objective : To evaluate the effect of acupuncture on streptozotocin(STZ)-treated rats by subcutaneous implantation of osmotic pump. Methods : STZ was administered to rats at a low dose with osmotic pump to induce beta cell death and diabetes (STZ osmotic pump model). The experimental animals were divided into 4 groups: 1. The control group which was not treated in the STZ osmotic pump model 2. The sham group which was acupunctured at an arbitrary point in the STZ osmotic pump model 3. The sample A group which was acupunctured at the Chung-wan(CV12) in the STZ osmotic pump model 4. The sample B group which was acupunctured at the Chok-samni(ST36) in the STZ osmotic pump model. The effect of acupuncture in the STZ osmotic pump model was observed by measuring the serum glucose level and immunostaining of pancreatic tissue of the rats. Results : STZ injection by subcutaneous implantation of osmotic pump caused hyperglycemia by destroying the pancreatic beta cell selectively. Acupuncture at the Chung-wan acupuncture point(CV12) and Jhok- samni acupuncture point (ST36) in the STZ osmotic pump model separately resulted in a decrease of the serum glucose level. In addition, the cyto-protective effect of the pancreatic beta-cell was detected in the STZ osmotic pump model by acupuncture. And there were few differences between the effects of acu- puncture at the CV12 and ST36. Conclusion : Acupuncture at the CV12 and ST36 had beneficial effects on Type Ⅱ diabetes mellitus, and action mechanism of the effect was thought to be concerned with secretion of endogenous beta- endorphin.

        Osmotic pump를 이용한 암컷 뱀장어(Anguilla japonica)의 성성숙 유도

        뱀장어(Anguilla japonica)의 인공종묘생산은 연어뇌하수체추출물(SPE, salmon pituitary extract)을 암컷 뱀장어에게 지속적인 투여를 하고 인위적으로 성성숙을 유도로 얻은 난과 정자를 인공수정을 하고 부화 시켜 생산한다. 하지만 SPE를 반복적으로 복강에 주사하는 방법은 암컷 뱀장어에게 많은 스트레스를 주며, 결국 스트레스로 인하여 완전한 성성숙을 하지 못하고 폐사에 이르게 되거나 배란한 난의 질이 좋지 않아 부화율과 자어의 생존율이 떨어지는 문제점을 갖고 있다. 본 연구에서는 뱀장어의 성성숙 유도에 유효하다고 알려진 호르몬이 주입된 osmotic pump를 복강에 삽입 후 암컷 뱀장어의 성성숙 유도를 하였다. 본 연구결과 SPE를 투여한 실험어의 GSI가 hCG, GnRHa, MT를 각각 또는 함께 투여한 실험구 보다 유의하게 증가하였으며 조직학적 분석결과에서도 난소의 난모세포가 핵이동기 단계로 발달되었음을 관찰할 수 있었다. 본 결과를 통해 osmotic pump를 이용한 암컷 뱀장어 인위적 성성숙 유도 방법으로 이용 가능할 것으로 생각된다. For the artificial induction of the sexual maturation of Anguilla japonica, salmon pituitary extract (SPE) is continuously injected into females, and the eggs obtained from artificial sexual maturation are artificially fertilized with sperms and hatched. However, repeated injection of SPE in the abdominal cavity causes tremendous stress in females, which may prevent their complete sexual maturation and reduce the immune system function, ultimately resulting in death. In addition, the poor quality of the ovulated eggs can reduce the hatching and survival rate of larvae. In the present study, sexual maturation of females was induced by inserting an osmotic (OS) pump containing hormone analogs known to effectively induce sexual maturation into the abdominal cavity of female eels, and the effect of the OS pump on the induced sexual maturation was investigated. Our study results showed that the gonadosomatic index (GSI) was significantly higher in the fish subjected to SPE injection than those subjected to human chorionic gonadotropin (hCG), gonadotropin-releasing hormone analog (GnRHa), and methyl testosterone (MT) injections, either separately or in combination. In addition, a histological analysis showed that the oocytes in the SPE OS pump groups were more mature (entered the nuclear shift stage) than those in the other groups. These results suggest that an osmotic pump containing hormone analogs can be used to induce sexual maturation in female A. japonica artificially.

      • Ionic liquid flow along the carbon nanotube with electric field

        Liquid pumping can occur along the outer surface of an electrode under a DC electric field. For biological applications, a better understanding of the ionic solution pumping mechanism is required. Here, we fabricated CNT wire electrodes (CWEs) and tungsten wire electrodes (TWEs) of various diameters to assess an ionic solution pumping. A DC electric field created by a bias of several volts pumped the ionic solution in the direction of the negatively biased electrode. The resulting electro-osmotic flow was attributed to the movement of an electric double layer near the electrode, and the flow rates along the CWEs were on the order of picoliters per minute. According to electric field analysis, the z-directional electric field around the meniscus of the small electrode was more concentrated than that of the larger electrode. Thus, the pumping effect increased as the electrode diameter decreased. Interestingly in CWEs, the initiating voltage for liquid pumping did not change with increasing diameter, up to 20 ㎛. We classified into three pumping zones, according to the initiating voltage and faradaic reaction. Liquid pumping using the CWEs could provide a new method for biological studies with adoptable flow rates and a larger ‘Recommended pumping zone’.

        A study on the thermal aging characteristics of an electro-osmotic pump using a liquid crystal polymer as a packaging material

        The generation of bubbles due to the poor barrier property of packaging materials with regard to gas or water permeability can be detrimental to the performance and stability of electro-osmotic (EO) pumps, as their lifetime decreases with the increase in the bubble volume. Water vapor transmission rate tests were conducted on three different polymers, butyl rubber, polycarbonate, and liquid crystal polymer (LCP), to develop a suitable packaging material for EO pumps. LCP showed the most pronounced barrier performance for water vapor permeability before and after an accelerated thermal aging test. Field emission scanning electron microscopy, energy-dispersive X-ray spectroscopy, thermogravimetric analysis, and X-ray diffraction measurement were performed, and it was found that LCP had no significant microstructural, compositional, and crystallographic changes, and thermal degradation after the accelerated thermal aging test. An EO pump was assembled using LCP cases, and it was then tested under various aging temperatures (50 °C, 60 °C, 70 °C, and 80 °C). Moreover, the pump’s thermal aging characteristics were analyzed using the Arrhenius model. Based on the obtained results, the reaction rate constant (k) rapidly increased with the increase in the aging temperature, and the calculated value of the activation energy (Ea) was 69.2 kJ/mol.

        Osmotically regulated asymmetric capsular system for sustained delivery of indomethacin

        The aim of the current study was to develop a sustained release asymmetric membrane capsular system for oral delivery of indomethacin. The capsule membrane was prepared using fabricated glass mold pins by phase inversion technique. Cellulose acetate was used as the semi permeable membrane. The capsule contains pore-forming water-soluble additives, which after coming in contact with aqueous medium, dissolves, resulting in an in situ formation of a micro porous structure. The effect of different formulation variables, like, ratio of drug to osmogen, solubilizing agent and level of pore former, stirring rate on the in vitro release was studied. Scanning electron microscopy of the membrane confirmed its porous, dense asymmetric nature. It was found that drug release rate increased with the increase in amount of osmogen and solubilizer, and independent of stirring rate. Indomethacin release was,directly proportional to the level of pore former, in the membrane. In the present investigation, efforts have been made to increase the release of poorly water soluble drug by means of porosity osmotic pump.

        Design and Optimization of Solid Dispersed Osmotic Pump Tablets of Aceclofenac, A Better Approach to Treat Arthritis

        The aim of this work was to prepare porous osmotic pump tablets for controlled delivery of Aceclofenac. Aceclofenac solid dispersion was prepared to improve the solubility by using the drug-carrier (Mannitol) ratio of 1:1. The osmotic pump tablets were prepared using the solid dispersed product of Aceclofenac. The formulation contains potassium chloride as osmotic agent, cellulose acetate as semipermeable membrane, poly ethylene glycol (PEG 4000) as pore former and sodium lauryl sulphate (SLS) as solubility enhancer. The formulations were designed by the general factors such as osmotic agent and pore former. All formulations were evaluated for various physical parameters and, the in vitro release studies were conducted as per USP. The drug release kinetic studies such as zero order, first order, and Higuchi and Korsmeyer peppas were determined and compared. All the formulations gave more controlled release compared to the marketed tablet studied. Numerical optimization techniques were applied to found out the best formulation by considering the parameter of in vitro drug release kinetics and dissolution profile standards. It was concluded that the porous osmotic pump tablets (F7) composed of Aceclofenac solid dispersion/Potassium chloride/Lactose/Sodium lauryl sulphate/Magnesium Stearate (400/40/95/10/5, mg/tab) and coating composition with Cellulose acetate/PEG 4000 (60/40 %w/w) is the most satisfactory formulation. The porous osmotic pump tablets provide prolonged, controlled, and gastrointestinal environment-independent drug release.

        Drug infusion pump is classified as electronic and mechanical. Electronic drug infusion pump precision control is possible by using a motor, but it is not easy to carry and it is expensive. The mechanical drug infusion pump can be portable and operations to use the easy. However, mechanical infusion pump flow rate is not constant. Therefore, there was a lot of attempts to solve the problems of the Mechanical infusion pump, there is a combination of elastomeric and osmotic pressure of the drug infusion. but this of elastomeric and osmotic infusion pumps is important to elasticity and concentration of difference because of injecting by interaction between elastic and osmotic pressure. Therefore, in this paper indicates interaction between the elasticity and concentration differences. Performance of the infusion pump is optimized by simulation programs.

      • Obtaining Healthy Oocytes from Superovulation of Spontaneous Diabetes Mellitus (sDM) Rats using Osmotic Pump

        Superovulation is a technique to acquire rats to produce many eggs than normal rats. Superovulated eggs were used to make cloned animals through somatic cell nuclear transfer technology (SCNT). Healthy and valuable oocyte retrieval is essential for successful somatic cell nuclear transfer. Superovulation is also essential to maximize to the yield of IVF-derived rat eggs. Osmotic pumps (Alzet®) are miniature in order to provide research with a convenient, and reliable alternative to chronic injections. Acquiring superovulated oocytes through osmotic pump in minimizing irritation to the uterus and ovaries are competitive. We investigated the effects of pregnant mare serum gonadotropin (PMSG) and human Chorionic Gonadotropin (hCG) using osmotic pump in sDM rat. Adult female rats at 11 wks of age were used for superovulation. The response to PMSG and hCG were examined by osmotic pump of 150 IU/kg PMSG + 75 IU/kg hCG or 150 IU/kg PMSG + 150 IU/kg hCG or 300 IU/kg PMSG + 150 IU/kg hCG or 300 IU/kg PMSG + 300 IU/kg hCG. HCG was administered 48 hrs later after administration of PMSG. Oocytes were collected from the oviducts 16–18 hrs after hCG administration. Superovulation was significantly higher in rats administrated 150 IU/kg PMSG + 75 IU/kg hCG. This study demonstrated that healthy oocytes were produced in DM rat by PMSG and hCG that flowed through the osmotic pump ameliorate on uterus and ovaries.

        Evaluation on Clinical Application of Osmotic Pump with Dorzolamide in Normal Dogs

        This study was designed to compare the effects of continuous release of an anti-glaucoma drug applied by an osmotic pump and by conventional eye drop instillation in normal beagle dogs, by measuring drug concentration in the blood and monitoring intra-ocular pressure (IOP). In group 1, an osmotic pump filled with Trusopt® was implanted subcutaneously over the right eye of each dog and the IOP was measured. In group 2, the right eye of each dog was administered with 2% dorzolamide (Trusopt®, Merck, USA) three times per day. Blood was sampled once per week in all groups. The IOP of the end of this study was 16.7 ± 0.58 mmHg in group 2 and 17.7 ± 2.52 mmHg in group 2. Dorzolamide concentration in blood increased continuously over 4 weeks up to 585.8 ± 79.42 μg/ L in group 1, and 747.3 ± 27.89 μg/L in group 2. We achieved satisfactory results in the osmotic pump group, which had a similar effect on IOP, and low fluctuations in IOP. Therefore, the results of this study should allow osmotic pumps to be consider as an alternative method to eye drops for the effective, safe, and convenient treatment of glaucoma.

        Electrodeposition of Mn-Ni Oxide/PEDOT and Mn-Ni-Ru Oxide/PEDOT Films on Carbon Paper for Electro-osmotic Pump Electrode

        $MnO_2$, a metal oxide used as an electrode material in electrochemical capacitors (EDLCs), has been applied in binary oxide and conducting polymer hybrid electrodes to increase their stability and capacitance. We developed a method for electrodepositing Mn-Ni oxide/PANI, Mn-Ni oxide/PEDOT, and Mn-Ni-Ru oxide/PEDOT films on carbon paper in a single step using a mixed bath. Mn-Ni oxide/PEDOT and Mn-Ni-Ru oxide/PEDOT electrodes used in an electro-osmotic pump (EOP) have shown better efficiency compared to Mn-Ni oxide and Mn-Ni oxide/PANI electrodes through testing in water as a pumping solution. EOP using a Mn-Ni-Ru oxide/PEDOT electrode was also tested in a 0.5 mM $Li_2SO_4$ solution as a pumping solution to confirm the effect of the $Li^+$ insertion/de-insertion reaction of Ruthenium oxide on the EOP. Experimental results show that the flow rate increases with the increase in current in a 0.5 mM $Li_2SO_4$ solution compared to that obtained when water was used as a pumping solution.
